      • Rotavirus 전기영동형별을 위한 RNA 추출과정 및 조건의 비교연구

        Rotaviruses (Rv) are the major cause of severe dehydrating dearrhea among young children in Korea, where they are frequently responsible for 68% of the cases requiring hospitalization. Laboratory diagnosis of rotaviral infection in Korea depends mainly on enzyme-linked immunosorbent assay (ELISA) that employs expensive commercial diagnostic kits purchased from abroad and that shous the presence or absence of group A Rv only. PACE have however provided epidemiologic evidence to demonstrate the sxistence of different strains (both group A and atypical Rv) during diarrgeal outbreak, the appearance of new strains and desappearance of old ones from a community, shifts in the prenalence of strains, and the persistance of a single strain in newborns. For such reasons, we have felt that it is essential to raise the sensitivity and shorten the processing time for PAGE. We therfore have devised a rapid method for Rv RNA extration by directly extracting the Rv RNA from the specimens (that is, skipping freon extraction and running as ultracectrifuge) and using an urea-containing dissociation mixture to dissolve dried RNA for PAGE. We have also shorten the PAGE diagnostic results by sequential staining of the same gel with ethidium bromide and silver stain. With the use of the rapid method, it was feasible to increase the efficacy of the PAGE with reduced cost and working time. Phenol extration of stools, however, was essential in maintaining the sensitivity of the method.

      • 경정배양에 의한 카네이션(Dianthus caryophyllus L.)의 기내증식과 순화

        본 실험은 카네이션의 경정배양을 통한 기내증식체계를 확립하고 기내증식과정 중 발생하는 투명묘의 발생을 억제하기 위하여 실시하였으며, 발근과 순화과정을 통합함으로써 그에 따르는 경비와 노동력을 절감할 수 있는 방안을 마련하는 한편, 기내에서 얻은 묘를 직접 재배에 이용할 수 있는 방법을 개발하기 위하여 실시하였다. 경정배양에서 생존율은 Benzyladenine (BA) 또는 Kinetin (KI) 0.1㎎/ℓ 첨가한 모든 배지에서 80%이상이었으나 그 이상의 농도에서는 농도가 증가할수록 생존율이 감소하였으며, Naphthaleneacetic acid (NAA) 또는 Indoleacetic acid (LAA) 0.1㎎/ℓ 혼합첨가시에는 2.0㎎/ℓ까지는 농도에 관계없이 모든 처리구에서 생존율이 75%이상이었다. 경정배양에서 얻은 신초의 증식배양에서 LAA나 NAA의 첨가에 관계없이 KI보다 BA를 첨가하면 신초발육이 더 좋았다. MS배지의 무기염류를 4배로 첨가한 4MS배지에서는 신초가 모두 고사하였고, 1 MS배지에서 신초의 발육이 가장 좋았다. Sucrose 무첨가 배지에서 신초의 발육이 가장 좋았다. Sucrose 무첨가 배지에서 신초의 발육은 거의 이루어지지 않았고, sucrose 20㎎/ℓ첨가배지에서 신초의 발육이 가장 좋았다. 한천보다 겔라이트 첨가배지에서 신초의 발육이 좋았다. 배지 pH는 5.0에서 신초의 발육이 가장 양호하였다. 투명화묘의 발생은 경정배양과 계대배양 전과정에서 발생하였고, 한천 또는 겔라이트의 농도를 높여주었을때에 억제되었다. 발근 및 순화단계에서 신초의 발육과 근형성은 sand, vermiculite배지에서 보다 perlite배지에서 더 좋았다. This study was carried out to establish a system for clonal multiplication and preventing vitrification in shoot tip culture of carnation. Combining the two steps of rooting and acclimatization, it may possibly reduce the cost and save the labor for clonal propagation. In addition, the plantlets raised by this one step rooting and acclimatization could be directly transferred to soil. The survival rate was over 80% in the media containing 0.1㎎/ℓ benzyl-adenin (BA) or kinetin (KI), but decreased as the concentration increased. When auxins were added in combination with cytokinins, the survival rate was over 75% regardless of the cytokinin concentration up to 2.0㎎/ℓ. When the in vitro shoots were subcultured, shoot development was better in the media containing BA than those containing KI regardless of addition of indoleacetic acid (LAA) of α-naphthalene acetic acid (NAA). Shoot development was the best in the standard MS medium(1xMS), while all the shoots on the medium containing 4 times the inorganic salts of MS medium(4xMS) died. Shoot development in MS medium free of sugar was not noticed. The MS medium containing 20g/ℓsucrose resulted in the best shoot growth. Shoot development was better on the media containing gelrite than those containing agar. The MS medium at pH 5.0 favored the shoot development. Vitrification of shoots occurred during the initial shoot culture as well as during the subculture of the in vitro shoots, but it was suppressed when the concentration of gelrite or agar in the media was increased. As the substrate for rooting and acclimatization of the in vitro shoots, perlite was better than sand or vermiculite.

      • 복소함수 T_c(z)=z^n+c의 쥴리아 집합

        복소 함수의 쥴리아 집합이란 무한대로 발사하는 점들의 집합의 경계를 말한다. 이것은 쥴리아 집합의 어떤 점은 무한대로 발산하지 않는 궤도를 갖고 있으나, 이 궤도의 근방에서 무한대로 발산하는 궤도를 갖는 점이 존재함을 의미한다. 본 논문의 목적은 T_c(z)=z^n+c 형태의 복소 함수의 쥴리아 집합에 대한 연구에 있다. 이 때 z와 c는 복소수이고, n≥2이다. 특히, 매개 상수 c 에 대한 여러 가지 형태의 쥴리아 집합에 대하여 살펴보고, 또한 쥴리아 집합에 대한 주기성과 대칭성을 조사하였다.

        Varicella-zoster virus, an unusual pathogen of community acquired pneumonia in adult, may causes respiratory distress and death. Since the first description by Waring et al. in 1942, the disease has been recognized by roentgenography, pathologic study, and virus studies as a special syndrome of the varicella virus. Immunocompromized conditions such as leukemia, malignant lymphoma and corticosteroid therapy predispose patients to varicella pneumonia. It occurs rarely in immunocompetent patients. The clinical manifestations of varicella zoster pneumonia exhibit a wide spectrum of disease from asymptomatic to death. Acyclovir is the recommended treatment and corticosteroid can be used in severe varicella pneumonia. We report a case of varicella pneumonia developed in a immunocompetent patient.

        A successful treatment of infants and children with congenital heart defects depends upon several factors: diagnosis must be accurate, the operative technique, including techniques of perfusion and myocardial preservation, must be suited to the needs of patients and intensive cares, including accurate anesthesia, must be applied through all stages of investigation and treatment, i.e. before, during and after the operation. Recently we performed a open heart surgery successfully for a six-years old girl who had suffered from frequent upper respiratory tract infection and mild to moderate exertional dyspnea with congenital heart defects (ventricular septal defect, Kirklin type II, with patent foramen ovale). The operation and postoperative course were very smooth. And now we report this with review of literatures.

      • 역전사 중합효소 연쇄반응을 이용한 사람의 활성 T세포 항원 CTLA-4 유전자 클로닝 및 마우스 L세포에서의 발현

        Occupancy of the T-cell receptor (TCR) by antigen in association with class Ⅱ MHC molecule is required for the initiation of T cell activation. However TCR stimulation is not a sufficient signal to account for all the observed events that occur during T cell activation. And in the absence of a second costimulatory signal provided by abtigen presenting cell antigen specific signal may lead to clonal inactivation or energy. CD28, a T cell surface molecule first defined by monoclonal antibody 9.3 on human cells, appears to function as an alternative, TCR/CD3-independent, activation pathway for T cell. This molecule provides costimulatory signal for T cell proliferation by increasing a number of lymphokines and cytokines production. A T cell activation antigen CTLA-4 is homologous to CD28 with 28% amino acid and 67% of nucleic acid homology in the protein coding region. CTLA-4 was ofiginally identified by the differential screening follosing subtractive hybridization of murine cytotoxic T cell cDNA library and mapped to the same lacation on chromosome 2 in human and chromosome 1 in mouse as CD28. These findings and activation associated expression pattern of CTLA-4 raise question about functional role of this molecule in T cell activation. In this study human CTLA-4 cDNA was amplified from phytohemagglutinin (PHA)-stimulated human peropheral blood lymphocytes mRNA by using reverse transcription-ploymerase chain reaction (RT-PCR) method. And amplified human CTLA-4 cDNA was subsequently cloned into pRC/CMV vector. In double stranded DNA sequencing the 331st base of protein coding region was changed from guanylic acid of human CTLA-4 from alanine to threonine. Murind L cell transfected with this clone expressed 36kD protein. It appears that this dirrerence of molecular weight between native (34kD) and recombinant (36kD) human CTLA-4 molecule may due to glycosylation differences. The cDNA and murine CTLA-4 molecule may due to glycosylation differences. The cDNA and murine L cell lines expressing human CTLA-4 developed by this study would contribute in future work unveiling the biological rile of CTLA-4 in T cell activation.
