      • Nationalism Interpretation of the Demarcation Process of Central Asian CIS Countries

        Precise demarcation of territory is one of the symbols of modern nationstate, and it is also the main way for nationalist ideology as an internal mobilization factor to build and consolidate modern nation-state. The demarcation process of Central Asian CIS countries and the settlement of demarcation disputes were influenced by the trend of nationalist ideology in the last century. The demarcation began in the 1920s and 1930s, and the remaining problems of demarcation have not been solved so far. As far as the five Central Asian CIS countries are concerned, nationalism prevails, and the process of demarcation increases the contradictions and frictions among countries. As far as the countries are concerned, the demarcation has established the sovereignty of nation-states and safeguarded national interests, which has become an important form of nationalism ideology in Central Asia in the last century to construct modern nationstates.

        This paper takes Corps Spirit as the main research object, pays attention to the historical origin of Corps Spirit and cultivating and guarding border areas, to roles of historical in exploitation of virgin land in border area by troops in development and construction in Xinjiang’s history and present, promoting national unity, accelerating the development and transformation of Xinjiang’s society, industry and agriculture, and to Xinjiang’s important political, economic and cultural communication as the bridgehead of the Silk Road, and discusses the contemporary value of Corps Spirit in the contemporary frontier society.

        Circ_0075960 targets the miR-202-5p/CTNND1 axis to promote the growth and migration of endometrial carcinoma cells via regulating Wnt/β-catenin signaling activity

        Background: Endometrial carcinoma (EC) is one of the most common malignant tumors of the female reproductive tract, involving multiple molecular alterations. Circular RNA (circRNA) dysregulation is frequently observed in EC tissues, suggesting the involvement of circRNA in EC development. We aimed to investigate the role of circ_0075960 in EC. Methods: Real-time quantitative polymerase chain reaction (RT-qPCR) and western blot assays were applied for expression analysis. CCK-8, EdU, colony formation, flow cytometry and wound healing assays were employed for functional analysis. The predicted binding relationship between miR-202-5p and circ_0075960 or CTNND1 was validated by dual-luciferase reporter experiment. In vivo animal models were constructed in nude mice to verify the role of circ_0075960 in tumor growth. Results: Circ_0075960 and CTNND1 were upregulated, while miR-202-5p was downregulated in EC. Knockdown of circ_0075960 induced EC cell apoptosis, suppressed cell proliferation and migration, and repressed tumor growth in animal models. MiR-202-5p was targeted by circ_0075960 and it directly bound to CTNND1 3’UTR. The inhibition of circ_0075960 knockdown or miR-202-5p enrichment on EC cell proliferation and migration was reversed by miR-202-5p depletion or CTNND1 overexpression, respectively. Circ_0075960 targeted miR-202-5p to positively regulate CTNND1 expression. Moreover, circ_0075960 knockdown weakened the activity of Wnt/β-catenin signaling via targeting the miR-202-5p/CTNND1 axis. Conclusion: Circ_0075960 targets the miR-202-5p/CTNND1 axis to modulate Wnt/β-catenin signaling activity, thus contributing to the malignant development of EC.

        OsBAK1 is involved in rice resistance to Xanthomonas oryzae pv. oryzae PXO99

        OsBAK1 gene belongs to a receptor like kinase gene family in rice and shares a highly conserved gene structure and sequence homology with Arabidopsis thaliana BAK1 gene. To investigate the role of OsBAK1 in rice immunity, the full-length cDNA of OsBAK1 was isolated by RT-PCR and the transgenic rice lines (over expression and RNA-interference lines) were generated using Agrobacterium-mediated transformation. The expression level of OsBAK1 was determined by q-PCR in overexpression and RNAi transgenic rice lines. Based on quantitative polymerase chain reaction (q-PCR) results, two overexpression lines and two RNAi lines were evaluated in bioassays for resistance to Xanthomonas oryzae pv. oryzae PXO99, the causal agent of rice bacterial blight disease. Pathogenicity bioassays showed overexpression OsBAK1 lines exhibited resistance to blight disease whereas OsBAK1 RNAi lines promoted susceptibility. Besides, OsBAK1 can complement the function of AtBAK1 in Arabidopsis bak1 protoplast, activating FRK1 expression, a marker gene in PTI signaling pathway, after treatment by flg22. Furthermore, the transcriptional level of OsBAK1 was induced significantly in rice by defense signaling molecules such as salicylic acid, jasmonic acid, and PXO99 inoculation. Our results illustrated OsBAK1 positively regulates plant defense against rice bacterium pathogen Xanthomonas oryzae pv. oryzae PXO99.

      • The Comparative Study of Xinjiang’s Tuntian System before and after the Founding of the People’s Republic of China

        China’s frontier society is in the process of social transformation. Take Xinjiang as an example, there are two parallel governance modes: local governance and corps governance. The proposal of “the Belt and Road Initiative” strategy reshaped the shape and status of Xinjiang, and made Xinjiang a center of two-way opening to the outside world, which had a far-reaching impact on the stability and development of border areas. In this discourse situation, it is necessary to look at the diachronic changes around “the Belt and Road Initiative” in Xinjiang from the comparison of history and modernity. Taking wasteland as the breakthrough point, this paper discusses the influence of Xinjiang Production and Construction Corps on Xinjiang’s modern social transformation in the ways of ethnic communication, regional development, world trade system, social domination, etc. by combing the comparison of Xinjiang’s wasteland and frontier defense, ethnic exchanges, agricultural development, trade and social development before and after the founding of the People’s Republic of China.

        Biosynthesis of Three Chalcone β-D-glucosides by Glycosyltransferase from Bacillus subtilis ATCC 6633

        Chalcones exhibit multiple biological activities. Various studies have attempted to modify the structure of chalcones with a special focus on the addition of substituents to the benzene rings. However, these chemical modifications did not improve the water solubility and bioavailability of chalcones. Glycosylation can markedly affect the physical and chemical properties of hydrophobic compounds. Here, we evaluated the ability of a highly promiscuous glycosyltransferase (GT) BsGT1 from Bacillus subtilis ATCC 6633 to biosynthesize chalcone glucosides. Purified BsGT1 catalyzed the conversion of 4'-hydroxychalcone (compound 1), 4'-hydroxy-4-methylchalcone (compound 2), and 4-hydroxy-4'-methoxychalcone (compound 3), into chalcone 4'-O-β-D-glucoside (compound 1a), 4-methylchalcone 4'-O-β-D-glucoside (compound 2a), and 4'- methoxychalcone 4-O-β-D-glucoside (compound 3a), respectively. To avoid the addition of expensive uridine diphosphate glucose (UDP-Glc), a whole-cell biotransformation system was employed to provide a natural intracellular environment for in situ co-factor regeneration. The yields of compounds 1a, 2a, and 3a were as high as 90.38%, 100% and 74.79%, respectively. The successful co-expression of BsGT1 with phosphoglucomutase (PGM) and UDP-Glc pyrophosphorylase (GalU), which are involved in the biosynthetic pathway of UDP-Glc, further improved the conversion rates of chalcones (the yields of compounds 1a and 3a increased by approximately 10%). In conclusion, we demonstrated an effective whole-cell biocatalytic system for the enzymatic biosynthesis of chalcone β-D-glucoside derivatives.

        CD49f Can Act as a Biomarker for Local or Distant Recurrence in Breast Cancer

        Purpose: Metastasis and local recurrence are the primary causes of treatment failure and patient death in breast cancer. The aim of this study was to validate a metastasis- and local recurrenceassociated biomarker for prognostic evaluation and planning treatment strategies. Methods: Formalin-fixed, paraffin-embedded tissues from a cohort of 312 patients (all stage II and III) were used. The prevalence of CD49f+ cells in the patients’ tumors was analyzed and correlated with clinical characteristics to determine its prognostic and clinical implications. Results: CD49f+ tumor cells were found in a minority of tumors, with 62.8% of the samples showing not a single cell of this subtype. In the clinical characteristics analysis, which were performed with t-tests, CD49f+ tumors were not associated with age, tumor size, World Health Organization grade, nodal status, human epidermal growth factor receptor 2 status, progesterone receptor status, or estrogen receptor status, although they were significantly associated with disease recurrence (distant metastasis or/and local recurrence). Univariate survival analysis using the Kaplan-Meier method showed that CD49f+ tumors were associated with markedly decreased disease-free survival (DFS); the same result was found using multivariate Cox analysis, even when only chemotherapy- treated patients were analyzed. Conclusion: Our results indicated that breast tumors with CD49f+ cancer cells are associated with an increased risk for disease recurrence after initial surgery with poor clinical outcomes (decreased DFS). Therefore, as it requires testing for only one additional protein, adding CD49f testing to conventional surgical pathology is a strategy that has great potential for prognostic and treatment-guidance purposes.

        Comparative transcriptome analyses provide insights into the adaptation mechanisms to acute salt stresses in juvenile Sinonovacula constricta

        Background Sinonovacula constricta is an economically important bivalve species in China, Korea and Japan that widely resides in estuarine and coastal areas where salinity fluctuates rapidly. However, little is known about its adaptation mechanisms to acute salt stresses. Objective To reveal the underlying molecular mechanisms involved in acute salt stresses in juvenile S. constricta. Methods Nine cDNA libraries (triplicate each trial) were established from juvenile S. constricta, which were subjected to low salinity (5 psu), optimal salinity (15 psu) and high salinity (25 psu) for 6 h, respectively. Results Illumina sequencing generated 478,587,310 clean reads totally, which were assembled into 427,057 transcripts of 246,672 unigenes. Compared with the control, 1259 and 2163 differentially expressed genes (DEGs) were identified under acute low and high salt stresses, respectively. GO and KEGG enrichment analyses of DEGs revealed that several key metabolic modulations were mainly responsible for the acute salt stresses. According to the significantly highlighted KEGG pathways, some key DEGs were identified and discussed in details. Notably, based on which, some potential osmolytes were further speculated. Conclusion Here, we carried out a unique report of comparative transcriptome analyses in juvenile S. constricta in response to acute salt stresses. The identified DEGs and their significantly enriched GO terms and KEGG pathways were critical for understanding and further investigating the underlying the physical and biochemical performances, and ultimately facilitated S. constricta breeding. Besides, the transcriptome data greatly enriched the genetic information of S. constricta, which were valuable for promoting its molecular biology researches.
