Melatonin attenuates microbiota dysbiosis of jejunum in short-term sleep deprived mice

Our study demonstrated that sleep deprivation resulted in homeostasis disorder of colon. Our study goes deeper into the positive effects of melatonin on small intestinal microbiota disorder caused by sleep deprivation. We successfully established a multiplatform 72 h sleep deprivation mouse model with or without melatonin supplementation, and analyzed the change of small intestinal microbiota using high-throughput sequencing of the 16S rRNA. We found melatonin supplementation suppressed the decrease of plasma melatonin level in sleep deprivation mice. Meanwhile, melatonin supplementation improved significantly the reduction in OTU numbers and the diversity and richness of jejunal microbiota and the abundance of Bacteroidaeae and Prevotellaceae, as well as an increase in the Firmicutes-to-Bacteroidetes ratio and the content of Moraxellaceae and Aeromonadaceae in the jejunum of sleep deprived-mice. Moreover, melatonin supplementation reversed the change of metabolic pathway in sleep deprived-mice, including metabolism, signal transduction mechanisms and transcription etc, which were related to intestinal health. Furthermore, melatonin supplementation inverted the sleep deprivation-induced a decline of anti-inflammatory cytokines (IL-22) and an increase of the ROS and proinflammatory cytokines (IL-17) in jejunum. These findings suggested that melatonin, similar to a probiotics agent, can reverse sleep deprivation-induced small intestinal microbiota disorder by suppressing oxidative stress and inflammation response.

Background: The study aimed to investigate the analgesic effect of a combination of intravenous flurbiprofen axetil and opioids, and evaluate the relationship between refractory pain relief and plasma ${\beta}$-endorphin levels in cancer patients. Materials and Methods: A total of 120 cancer patients was randomly divided into two groups, 60 patients took orally morphine sulfate sustained-release tablets in group A, and another 60 patients receiving the combination treatment of intravenous flurbiprofen axetil and opioid drugs in group B. After 7 days, pain relief, quality of life improvement and side effects were evaluated. Furthermore, plasma ${\beta}$-endorphin levels were measured by radioimmunoassay. Results: With the combination treatment of intravenous intravenous flurbiprofen axetil and opioids, the total effective rate of pain relief rose to 91.4%, as compared to 82.1% when morphine sulfate sustained-release tablet was used alone. Compared with that of group A, the analgesic effect increased in group B (p=0.031). Moreover, satisfactory pain relief was associated with a significant increase in plasma ${\beta}$-endorphin levels. After the treatment, plasma ${\beta}$-endorphin level in group B was $62.4{\pm}13.5pg/ml$, which was higher than that in group A ($45.8{\pm}11.2pg/ml$) (p<0.05). Conclusions: Our results suggest the combination of intravenous flurbiprofen axetil and opioids can enhance the analgesic effect of opioid drugs by increasing plasma ${\beta}$-endorphin levels, which would offer a selected and reliable strategy for refractory cancer pain treatment.
Study on Metallurgically Prepared Copper-Coated Carbon Fibers Reinforced Aluminum Matrix Composites

Carbon materials, like carbon fiber, carbon nanotubes and graphene, were widely used as promising reinforcements tostrengthen aluminum matrix composites (AMCs). The dispersion of reinforcement in matrix and interface wettability betweenmatrix and reinforcing phase have been key factors affecting AMCs properties. In this study, electroless copper-coatedcarbon fibers reinforced aluminum (Cu-Cf/Al) composites were prepared by spark plasma sintering processing followedby heat treatment. Microstructure and mechanical properties were investigated. Microstructure observation indicated thatfibers distributed uniformly in the composites containing up to 9 wt% copper-coated carbon fibers (Cu-Cf). In addition, aninterfacial layer of 50 nm thickness was formed between the fiber and Al matrix due to mutual diffusion of Cu and Al atoms. Vickers hardness, tensile strength and bending strength of 9 wt% Cu-Cf/Al composite increased from 40 to 93 HV, 59 to190 MPa and 110 to 326 MPa, respectively, compared to Al matrix. The improved mechanical properties are ascribed to thesynergistic effect of dispersion, precipitation and solution strengthening. However, the elongation of composite decreased ascompared to that of Al matrix. Moreover, Cu-Cf/Al composite exhibits superior corrosion resistance over uncoated carbonfiber/Al composite but less than Al matrix.
Active and Passive Smoking, and Alcohol Drinking and Breast Cancer Risk in Chinese Women

To evaluate the relation between smoking, alcohol drinking and risk of breast cancer in Chinese women, we conducted a case-control study with 669 cases and 682 population-based controls in Jiangsu Province of China. A structured questionnaire was used to elicit detailed information. Unconditional logistic regression analysis was performed to calculate odds ratios (ORs) and 95% confidence intervals (CIs). The results revealed that smoking, whether active or passive through the husband, was related to increased risk of breast cancer. The ORs (adjusted for age, menopausal status, educational levels, occupation, body mass index and income) were 3.55 (95%CI: 1.27-9.91) for active smoking and 1.47 (95%CI: 1.18-1.84) for passive smoking from husbands, respectively. A significant positive relationship was observed between breast cancer risk and the degree of husbands' smoking. There were significant increase trend in ORs with the daily smoked number of cigarettes of husbands, the passive smoking years from husbands and the pack-years of husbands' smoking (trend test: p=0.00003, 0.00013 and 0.0001, respectively). Alcohol consumption was also found to be a risk factor. The findings of this study in particular suggest that husbands' smoking increases risk of breast cancer in Chinese women.

To evaluate the relationship between alcohol drinking, XRCC1 codon 194 and 399 polymorphisms and risk of colorectal cancer, we conducted a case-control study with 315 colorectal cancer cases (105 colon, 210 rectal) and 439 population-based controls in Jiangsu Province of China. The XRCC1 codon 194 and 399 genotypes were identified using polymerase chain reaction and restrictrion fragment length polymorphism methods (PCR-RFLP). A structured questionnaire was used to elicit detailed information. Odds ratios (ORs) were estimated with an unconditional logistic model. In this study no significant differences were observed among the studied groups with regard to the genotype distribution of the XRCC1 codons 194 and 399 and the risk of colorectal cancer did not appear to be significantly influenced by genotype alone, whereas alcohol consumption showed a positive association (P for trend <0.01). When combined effects of XRCC1 polymorphisms and alcohol consumption were analyzed, we found that the 194Trp or 399Gln alleles further increased the colorectal cancer risk due to high alcohol intake. These findings support the conclusion that colorectal cancer susceptibility may be altered by gene-environment interactions.

This study selected Dendrobium offi cinale protocorms as the experimental material and used diff erent light-quality treatmentsand calcium signalling inhibitors to explore the mechanism of the biological response of calcium to light quality. The results indicate that a 1:3 red:blue (R:B) light ratio promotes the accumulation of biomass and polysaccharides in theprotocorm. Nevertheless, this eff ect can be entirely counteracted by calmodulin (CaM) inhibitors and calcium ion-chelatingagents. The possible mechanisms of this eff ect are as follows: (1) The Ca 2+ -CaM signal-regulated photosynthetic apparatusis improved by the R:B 1:3 light, increasing the CaM content and Ca 2+ -ATPase activity. Thus, R:B 1:3 light increases thedevelopment of the photosynthetic apparatus in the D. offi cinale protocorm, enhances its photosynthetic productivity, andpromotes the activities of sucrose phosphatase; accordingly, biomass and sucrose accumulation increase. (2) R:B 1:3 lightenhances Ca 2+ -CaM signalling and regulates glycometabolic processes to promote the activities of sucrose synthase, thusincreasing fructose and glucose levels and providing suffi cient synthetic precursors to promote polysaccharide production.
Intake of Freshwater Fish and Associated Fatty Acids and Risk of Breast Cancer

To investigate the association between intake of freshwater fish and their fatty acids and the risk of breast cancer in Chinese women, we conducted a case-control study with 669 cases and 682 population-based controls in Jiangsu Province of China. A structured questionnaire was used to elicit detailed information. Unconditional logistic regression analysis was performed to calculate odds ratios (ORs) and 95% confidence intervals (CIs). Total freshwater fish intake was linked to decrease in the adjusted OR for breast cancer, but without dose-dependence. Analyses by freshwater fish species showed that consumption of black carp and silver carp was inversely related to breast cancer risk, with adjusted-ORs for the highest intake category of black carp (${\geq}500g/month$) of 0.54 (95%CI=0.33-0.92; $P_{trend}$ <0.002) and for silver carp (${\geq}1000g/month$) of 0.19 (95%CI=0.11-0.33; $P_{trend}$ <0.001). In contrast, consumption of crucian carp was positively related to breast cancer risk, with an adjusted OR for the highest intake category (${\geq}1000g/month$) of 6.09 (95%CI=3.04-12.2; $P_{trend}$ <0.001). Moderate intakes of SFA, PUFA, n3-PUFA and n6-PUFA from freshwater fish may decrease the risk of breast cancer among premenopausal women. The findings of this study suggest that intake of freshwater fish and their fatty acids may modify risk of breast cancer, and that different species of freshwater fish could have a different actions on breast cancer risk. Future epidemiologic studies are needed to know the effects of freshwater fish intake on breast cancer risk and the cause of these effects.

<P><B>Surface doping of conjugated polymers</B> is realized by depositing a thin layer of graphene oxide (GO) on top of the polymers. The high proton density and the unique 2D structure of GO facilitate the protonic surface doping of conjugated polymers to achieve high conductivities. This finding represents a new strategy for improving charge transport across the metal/conjugated polymer interface to achieve much improved performance in organic solar cells. </P>
