        Optimal Design for Marker-assisted Gene Pyramiding in Cross Population

        Marker-assisted gene pyramiding aims to produce individuals with superior economic traits according to the optimal breeding scheme which involves selecting a series of favorite target alleles after cross of base populations and pyramiding them into a single genotype. Inspired by the science of evolutionary computation, we used the metaphor of hill-climbing to model the dynamic behavior of gene pyramiding. In consideration of the traditional cross program of animals along with the features of animal segregating populations, four types of cross programs and two types of selection strategies for gene pyramiding are performed from a practical perspective. Two population cross for pyramiding two genes (denoted II), three population cascading cross for pyramiding three genes(denoted III), four population symmetry (denoted IIII-S) and cascading cross for pyramiding four genes (denoted IIII-C), and various schemes (denoted cross program-A-E) are designed for each cross program given different levels of initial favorite allele frequencies, base population sizes and trait heritabilities. The process of gene pyramiding breeding for various schemes are simulated and compared based on the population hamming distance, average superior genotype frequencies and average phenotypic values. By simulation, the results show that the larger base population size and the higher the initial favorite allele frequency the higher the efficiency of gene pyramiding. Parents cross order is shown to be the most important factor in a cascading cross, but has no significant influence on the symmetric cross. The results also show that genotypic selection strategy is superior to phenotypic selection in accelerating gene pyramiding. Moreover, the method and corresponding software was used to compare different cross schemes and selection strategies.

        Surgical Options for Appropriate Length of J-Pouch Construction for Better Outcomes and Long-term Quality of Life in Patients with Ulcerative Colitis after Ileal Pouch-Anal Anastomosis

        Background/Aims: Total proctocolectomy with ileal pouch-anal anastomosis (IPAA) is widely accepted as a radical surgery for refractory ulcerative colitis (UC). Definite results on the appropriate pouch length for an evaluation of the risk-to-benefit ratio regarding technical complications and long-term quality of life (QOL) are still scarce. Methods: Data on UC patients who underwent IPAA from 2008 to 2022 in four well-established pouch centers affiliated to China UC Pouch Center Union were collected. Results: A total of 208 patients with a median follow-up time of 6.0 years (interquartile range, 2.3 to 9.0 years) were enrolled. The median lengths of the patients’ short and long pouches were 14.0 cm (interquartile range, 14.0 to 15.0 cm) and 22.0 cm (interquartile range, 20.0 to 24.0 cm), respectively. Patients with a short J pouch configuration were less likely to achieve significantly improved long-term QOL (p=0.015) and were prone to develop late postoperative complications (p=0.042), such as increased defecation frequency (p=0.003) and pouchitis (p=0.035). A short ileal pouch was an independent risk factor for the development of late postoperative complications (odds ratio, 3.100; 95% confidence interval, 1.519 to 6.329; p=0.002) and impaired long-term QOL improvement (odds ratio, 2.221; 95% confidence interval, 1.218 to 4.050, p=0.009). Conclusions: The length of the J pouch was associated with the improvement in long-term QOL and the development of late post-IPAA complications. A long J pouch configuration could be a considerable surgical option for pouch construction.

      • Redox cycling performance of inert-substrate-supported tubular single cells with nickel anode current collector

        An inert-substrate-supported tubular single cell, with a configuration of porous yttria-stabilized zirconia (YSZ) supporter/Ni anode current collector/Ni-Ce<SUB>0.8</SUB>Sm<SUB>0.2</SUB>O<SUB>1.9</SUB> anode/YSZ/Ce<SUB>0.8</SUB>Sm<SUB>0.2</SUB>O<SUB>1.9</SUB> bi-layer electrolyte/La<SUB>0.6</SUB>Sr<SUB>0.4</SUB>Co<SUB>0.2</SUB>Fe<SUB>0.8</SUB>O<SUB>3-δ</SUB> cathode, has been fabricated by extrusion and dip-coating techniques. Thickness of the Ni layer is modified by controlling the number of dip-coatings from one to four. Electrochemical performance and redox cycling stability of the single cell are investigated with respect to the microstructure of the Ni layer. Increasing the thickness of the Ni layer enhances the maximum power density of the cell, while it is unfavorable for the redox cycling stability. Considering the trade-off between these two aspects, an optimum dip-coating time is determined to be two. The cell shows a reasonable maximum power density of 453 mW cm<SUP>-2</SUP> at 800 <SUP>o</SUP>C, as well as good redox cycling stability within eight redox cycles. Additionally, 10 vol.% Ce<SUB>0.8</SUB>Sm<SUB>0.2</SUB>O<SUB>1.9</SUB> ceramic particle is incorporated into the Ni layer to further improve the redox cycling stability. The cell exhibits enhanced redox cycling performance after the Ce<SUB>0.8</SUB>Sm<SUB>0.2</SUB>O<SUB>1.9</SUB> incorporation. Within seven redox cycles, the cell voltage loss is less than 1% at a current density of 400 mA cm<SUP>-2</SUP>, and it maintains 93% of its initial performance after 11 redox cycles.

      • Association Between TP53 Arg72Pro Polymorphism and Hepatocellular Carcinoma Risk: A Meta-analysis

        Background: Previous studies on the association between the TP53 Arg72Pro polymorphism and hepatocellular carcinoma (HCC) risk obtained controversial findings. This study aimed to quantify the strength of the association by meta-analysis. Methods: We searched PubMed and Wangfang databases for published studies on the association between the TP53 Arg72Pro polymorphism and HCC risk, using the pooled odds ratio (OR) with its 95% confidence intervals (95% CI) for assessment. Results: 10 studies with a total of 2,026 cases and 2,733 controls were finally included into this meta-analysis. Overall, the TP53 Arg72Pro polymorphism was not associated with HCC risk (all P values greaterth HCC risk in Caucasians in three genetic models (For Pro versus Arg, OR = 1.20, 95%CI 1.03-1.41; For ProPro versus ArgArg, OR = 1.74, 95%CI 1.23-2.47; For ProPro versus ArgPro/ArgArg, OR = 1.85, 95%CI 1.33-2.57). However, there was no significant association between the TP53 Arg72Pro polymorphism and HCC risk in East Asians (all P values greater than 0.10). No evidence of publication bias was observed. Conclusion: Meta-analyses of available data suggest an obvious association between the TP53 Arg72Pro and HCC risk in Caucasians. However, the TP53 Arg72Pro polymorphism may have a race-specific effect on HCC risk and further studies are needed to elucidate this possible effect.

        Mercury Ions Mediated Phosphorus Containing Carbon Dots as Fluorescent Probe for Biothiols Screening

        In this study, we report the mercury ions (Hg2+) mediated phosphorus-containing carbon dots (PCDs) as a selective "off–on" fluorescence probe for glutathione (GSH), cysteine (Cys) and homocysteine (Hcys). PCDs obtained by hydrothermal reaction are sensitive to Hg2+ ions and its fluorescence can be significantly quenched owing to the electron transfer from the lowest unoccupied molecular orbital (LUMO) of PCDs to Hg2+. Interestingly, the weak fluorescence of Hg2+-mediated PCDs could be gradually recovered with the addition of GSH, Cys and Hcys. This can be attributed to the formation of Hg2+–S complex due to the super affinity of Hg2+–sulfydryl bond. The formation of Hg2+–S complex extremely reduces the oxidation ability of Hg2+ that inhibits the electron transfer from LUMO of PCDs to Hg2+ and re-opens the native electron transition from LUMO to the highest occupied molecular orbital (HOMO) of PCDs. Thus, the green fluorescence of PCDs is switched on. Furthermore, the present Hg2+-mediated PCDs assay exhibits a high selectivity for GSH, Cys and Hcy and has been successfully used to detect the total biothiols content in human urine samples.

        Shear Performance of Short Channel Connectors in a Steel-UHPC Composite Deck

        The steel-ultra high performance concrete (UHPC) composite deck has been increasingly concerned for the fatigue damage ameliorations on the steel bridge deck and the pavement. The UHPC plate in the composite deck featured by the favorable tensile performance and small thickness helps achieving a light self-weight. However, it results to a limited short space for the shear connector to transfer the interlayer shear force. Therefore, this study proposed a kind of short steel channel-section connector for the composite deck with a thin UHPC plate, and investigated its mechanical behavior through push-out tests and parametric FEM analysis. The test results showed the deformation of the channel connector during the loading process generally included the elastic, elastic–plastic, and plastic stages, and the shear fracture of channel connector dominated in the failure mode. The shear stiff ness of the specimen with embedded reinforcement was increased by 39%, but had little infl uence on the shear capacity. On the other side, the parametric analysis told that the shear strength of connector increased signifi cantly as the connector height increased from 50 to 80 mm. Nevertheless, the connector arrangement direction was a sensitive factor to the uplift resistance of the connector.

        Patterns and drivers of Vibrio isolates phylogenetic diversity in the Beibu Gulf, China

        Members of the genus Vibrio are ubiquitous in aquatic environments and can be found either in a culturable or a viable but nonculturable (VBNC) state. Despite widespread concerns as to how to define the occurrence and dynamics of Vibrio populations by culture-independent approaches, further physiological research and relevant biotechnological developments will require the isolation and cultivation of the microbes from various environments. The present work provides data and perspectives on our understanding of culturable Vibrio community structure and diversity in the Beibu Gulf. Finally, we isolated 1,037 strains of Vibrio from 45 samples and identified 18 different species. Vibrio alginolyticus, V. cyclitrophicus, V. tasmaniensis, V. brasiliensis, and V. splendidus were the dominant species that had regional distribution characteristics. The correlation between the quantitative distribution and community structure of culturable Vibrio and environmental factors varied with the Vibrio species and geographical locations. Among them, salinity, nitrogen, and phosphorus were the main factors affecting the diversity of culturable Vibrio. These results help to fill a knowledge gap on Vibrio diversity and provide data for predicting and controlling pathogenic Vibrio outbreaks in the Beibu Gulf.

        MiR-214 Regulates the Human Hair Follicle Stem Cell Proliferation and Differentiation by Targeting EZH2 and Wnt/b- Catenin Signaling Way In Vitro

        miR-214 plays a major role in the self-renewal of skin tissue. However, whether miR-214 regulates the proliferation and differentiation of human hair follicle stem cells (HFSCs) is unknown. Primary HFSCs were isolated from human scalp skin tissue, cultured, and identified using flow cytometry. An miR-214 mimic and inhibitor were constructed for transfection into HFSCs. The MTS and colony formation assays examined cell proliferation. Immunofluorescence detected the localization and expression levels of TCF4, b-catenin, and differentiation markers. Luciferase reporter and TOP/FOP Flash assays investigated whether miR-214 targeted EZH2 and regulated the Wnt/b-catenin signaling pathway. Western blot determined the expression levels of enhancer of zeste homolog 2 (EZH2), Wnt/b-catenin signaling-related proteins, and HFSC differentiation markers in cells subjected to miR-214 transfection. miR-214 expression was remarkably decreased during the proliferation and differentiation of HFSCs into transit-amplifying (TA) cells. Downregulation of miR- 214 promotes the proliferation and differentiation of HFSCs. Overexpression of miR-214 led to decreased expression of EZH2, b-catenin, and TCF-4, whereas downregulation of miR-214 resulted in increased expression of EZH2, b-catenin, and TCF-4 as well as TA differentiation markers. Immunofluorescence assay revealed that inhibiting miR-214 triggered the entry of b-catenin and TCF-4 into the nucleus. The luciferase reporter and TOP/FOP Flash assays demonstrated that miR- 214 directly targets EZH2 and affects Wnt/b-catenin signaling. The miR-214/EZH2/b-catenin axis could be considered a candidate target in tissue engineering and regenerative medicine for HFSCs.

      • Prognostic Factors Influencing Clinical Outcomes of Malignant Glioblastoma Multiforme: Clinical, Immunophenotypic, and Fluorescence in Situ Hybridization Findings for 1p19q in 816 Chinese Cases

        Malignant glioblastoma multiforme (GBM) is the most malignant brain tumor and despite recent advances in diagnostics and treatment prognosis remains poor. In this retrospective study, we assessed the clinical and radiological parameters, as well as fluorescence in situ hybridization (FISH) of 1p19q deletion, in a series of cases. A total of 816 patients with GBM who received surgery and radiation between January 2010 and May 2014 were included in this study. Kaplan-Meier survival analysis and Cox regression analysis were used to find the factors independently influencing patient progression free survival (PFS) and overall survival (OS). Age at diagnosis, preoperative Karnofsky Performance Scale (KPS) score, KPS score change at 2 weeks after operation, neurological deficit symptoms, tumor resection extent, maximal tumor diameter, involvement of eloquent cortex or deep structure, involvement of brain lobe, Ki-67 and MMP9 expression level and adjuvant chemotherapy were statistically significant factors (p<0.05) for both PFS and OS in the univariate analysis. Cox proportional hazards modeling revealed that age ${\leq}50$ years, preoperative KPS score ${\geq}80$, KPS score change after operation ${\geq}0$, involvement of single frontal lobe, deep structure involvement, low Ki-67 and MMP9 expression and adjuvant chemotherapy were independent favorable factors (p<0.05) for patient clinical outcomes.
