        Phase II Trial of Epigallocatechin-3-Gallate in Acute Radiation-Induced Esophagitis for Esophagus Cancer

        Acute radiation-induced esophagitis (ARIE) is among the most serious form of toxicities associated with definitive radiotherapy or chemoradiotherapy used for treatment of patients with esophageal cancer. Our preliminary phase I and II trials of lung cancer patients who received radiotherapy indicated epigallocatechin-3-gallate (EGCG) as a promising therapeutic option against ARIE. Therefore, we conducted a prospective phase II study to validate the efficacy and safety of EGCG in the treatment of ARIE. The patients who received chemoradiotherapy or definitive radiotherapy for treatment of esophageal cancer in the Shandong Cancer Hospital and Institute in China were enrolled for the present study. EGCG (440 μM) was administered with first onset of ARIE and then at weeks after final radiotherapy. The patients were monitored every week for dysphagia, Radiation Therapy Oncology Group (RTOG) score, and esophagitis-related pain. Moreover, tumor response and the effect on survival following the treatment were also evaluated. Comparison of the RTOG score in the first, second, third, fourth, fifth, and even sixth week after EGCG prescription and the first and second week after radiotherapy with baseline indicates a significant reduction. The tumor response rate was 86.3%. The overall survival rate in 1, 2, and 3 years was found to be 74.5%, 58%, and 40.5%. Oral administration of EGCG solution seems to be feasible for treating ARIE in patients with esophageal cancer who receive radiation therapy. EGCG might be an ARIE-reliever without compromising the efficacy of radiation therapy. A randomized study with a control group is needed for further evaluation.

        Exploring the Metabolomic Responses of Bacillus licheniformis to Temperature Stress by Gas Chromatography/Mass Spectrometry

        Owing to its high protein secretion capacity, simple nutritional requirements, and GRAS (generally regarded as safe) status, Bacillus licheniformis is widely used as a host for the industrial production of enzymes, antibiotics, and peptides. However, as compared with its close relative Bacillus subtilis, little is known about the physiology and stress responses of B. licheniformis. To explore its temperature-stress metabolome, B. licheniformis strains ATCC 14580 and B186, with respective optimal growth temperatures of 42℃ and 50℃, were cultured at 42℃, 50℃, and 60℃ and their corresponding metabolic profiles were determined by gas chromatography/mass spectrometry and multivariate statistical analyses. It was found that with increased growth temperatures, the two B. licheniformis strains displayed elevated cellular levels of proline, glutamate, lysine, pentadecanoic acid, hexadecanoic acid, heptadecanoic acid, and octadecanoic acid, and decreased levels of glutamine and octadecenoic acid. Regulation of amino acid and fatty acid metabolism is likely to be associated with the evolution of protective biochemical mechanisms of B. licheniformis. Our results will help to optimize the industrial use of B. licheniformis and other important Bacillus species.

        Fabrication of Ni/Epoxy Resin Functionally Graded Materials via a Reciprocating Magnetic Field

        A reciprocating magnetic field was applied to prepare functionally graded materials (FGM) comprised of magnetic Ni and nonmagnetic epoxy resin. The reciprocating field was achieved by shifting a cylindrical permanent magnet along the axis towards or away from the sample in cycle. Ni decreases gradually with the distance from surface to bottom, which attributes to the continuously changing driving force on magnetic Ni particles by the reciprocating field. The gradient of Ni increases with cycle times, while decreases with the increase of the velocity of the reciprocating magnet. The results reveal that it is a more effective way to prepare FGMs by using reciprocating magnetic field compared with normal static magnetic field.

        CD31 and D2-40 Contribute to Peritoneal Metastasis of Colorectal Cancer by Promoting Epithelial-Mesenchymal Transition

        Background/Aims: Colorectal cancer (CRC) patients often exhibit peritoneal metastasis, which negatively impacts their prognosis. CD31 and D2-40 have recently been suggested to be predictors of breast cancer prognosis, but their role in colorectal peritoneal metastasis (CRPM) remains unknown. Methods: The expression profiles of CD31 and D2-40 were analyzed in CRC patients with or without CRPM and in CRC cell lines with increasing metastatic potential. Overexpression and short hairpin RNA knockdown assays were performed in CRC cells, and the effects of these alterations on epithelial-mesenchymal transition (EMT) in vitro, growth of xenograft tumors in vivo, and peritoneal metastasis potential in a mouse model of CRPM were examined. Results: The expressions of CD31 and D2-40 were upregulated in CRC tumor tissues and was elevated further in tumor tissues from patients with CRPM. CD31 and D2-40 expression levels exhibited increasing trends parallel to the EMT potential of CRC cells. CD31 and D2-40 are essential for CRC cell EMT in vitro as well as for xenograft tumor growth and peritoneal metastasis in vivo. Conclusions: CD31 and D2-40 contribute to CRPM by promoting EMT and may serve as prognostic markers and therapeutic targets for CRC, particularly in patients with peritoneal metastasis.         Metal-Doped In2O3 Nanosphere Arrays with Enhanced Gas-Sensing Property

        In2O3 and metal-doped (Ni or Ce) In2O3 mesoporous three-dimensional (3D) nanospheres arrays were synthesized via nanocasting using mesoporous silica as hard templates. Effects of Ni or Ce doping on the structure, optical and gas-sensing properties of the In2O3 nanospheres were investigated. Both the undoped and the doped In2O3 nanospheres showed single-phase structure without any impurity. The nanospheres were about 20 nm in size and they stacked closely to formrigid 3D mesoporous structure. Achange in the value of optical band gap was observed upon metal doping. The room temperature photo luminescence behavior also showed some differences between pure and doped In2O3. Compared with pure In2O3 nanospheres, the metal-doped In2O3 exhibited superior response, fast recovery and good selectivity to ethanol. The enhanced gas-sensing properties might be related to the doping of metal ion and its effective contribution towards the oxygen vacancies, conductivity and crystallite size of the grains.
