Assessment of the crest cracks of the Pubugou rockfill dam based on parameters back analysis

The crest of the Pubugou central core rockfill dam (CCRD) cracked in the first and second impounding periods. To evaluate the safety of the Pubugou CCRD, an inversion analysis of the constitutive model parameters for rockfill materials is performed based on the in situ deformation monitoring data. The aim of this work is to truly reflect the deformation state of the Pubugou CCRD and determine the causes of the dam crest cracks. A novel real-coded genetic algorithm based upon the differences in gene fragments (DGFX) is proposed. It is used in combination with the radial based function neural network (RBFNN) to perform the parameters back analysis. The simulated settlements show good agreements with the monitoring data, illustrating that the back analysis is reasonable and accurate. Furthermore, the deformation gradient of the dam crest has been analysed. The dam crest has a great possibility of cracking due to the uncoordinated deformation, which agrees well with the field investigation. The deformation gradient decreases to the value lower than the critical one and reaches a stable state after the second full reservoir.
An Investigation into the Prevalence of Voice Strain in Chinese University Teachers

Vocal disorders are very common occupation-related disease in teachers, though it has never been given enough attention in China. As a result, the occupational health care of professional voice users is surprisingly undeveloped compared to the attention given to occupational hearing disorders or many other occupational symptoms. The aim of the present study was to assess the prevalence of voice problems in the general population of Chinese university teachers, and explore whether their voice problems affected their daily life, their social life and their work. A voice strain and voice handicap index questionnaire was administered to university instructors of English (N = 156) in six Chinese universities. Results indicated that voice strain is prevalent among Chinese university instructors. The respondents`` self-perceptions revealed that voice strain was significantly correlated with their job and their daily activities.

A Nano-hydroxyapatite (n-HA)/chitosan (CS)/konjac glucomannan (KGM) composite was prepared by integrating composition and molding. Then, X-ray diffraction (XRD), scanning electron microscopy (SEM) and energy dispersive spectroscopy (EDS) were used to analyze the physical, chemical and degradable properties of the composite before and after immersion in simulated body fluid (SBF). Moreover, an in vitro test for drug delivery revealed that the amount of released pentoxifylline (1-[5-oxohexyl]-3, 7-dimethylxanthine)(PTX) reached a plateau and equaled 80% of the drug loaded in an implant. The newly developed n-HA/CS/KGM composite may serve as a good degradable biomaterial for an implantable drug delivery system (IDDS) in bone tissue engineering. We screened four B. thuringiensis strains whose parasporal inclusions contained the S-layer protein (SLP), and cloned two slp genes from each strain. Phylogenetic analysis indicated these SLPs could be divided into two groups, SLP1s and SLP2s. To confirm whether SLPs were present in the S-layer or as a parasporal inclusion, strains CTC and BMB1152 were chosen for further study. Western blots with isolated S-layer proteins from strains CTC and BMB1152 in the vegetative phase showed that SLP1s and SLP2s were constituents of the S-layer. Immunofluorescence utilizing spore-inclusion mixtures of strains CTC and BMB1152 in the sporulation phase showed that SLP1s and SLP2s were also constituents of parasporal inclusions. When heterogeneously expressed in the crystal negative strain BMB171, four SLPs from strains CTC and BMB1152 could also form parasporal inclusions. This temporal and spatial expression is not an occasional phenomenon but ubiquitous in B. thuringiensis strains. [BMB reports 2011; 44(5): 323-328]

Citrobacter sp. is a cause of significant opportunistic nosocomial infection and is frequently found in human and animal feces, soil, and sewage water, and even in industrial waste or putrefaction. Biofilm formation is an important virulence trait of Citrobacter sp. pathogens but the process and characteristics of this formation are unclear. Therefore, we employed in vitro assays to study the nutritional and environmental parameters that might influence biofilm formation of C. werkmanii BF-6 using 96-well microtiter plates. In addition, we detected the relative transcript levels of biofilm formation genes by RT-PCR. Our results indicated that the capacity of C. werkmanii BF-6 to form biofilms was affected by culture temperature, media, time, pH, and the osmotic agents glucose, sucrose, NaCl, and KCl. Confocal laser scanning microscopy results illustrated that the structure of biofilms and extracellular polysaccharide was influenced by 100 mM NaCl or 100 mM KCl. In addition, nine biofilm formation genes (bsmA, bssR, bssS, csgD, csgE, csgF, mrkA, mrkB, and mrkE) were found to contribute to planktonic and biofilm growth. Our data suggest that biofilm formation by C. werkmanii BF-6 is affected by nutritional and environmental factors, which could pave the way to the prevention and elimination of biofilm formation using proper strategies.

In the process of coal seam water injection, the components of different fracturing fluids have differenteffects on coal. To analyze the effect of water injection fracturing fluid on the physical and chemical propertiesof coal, we studied the relationship between guar-based fracturing fluid and cationic clean fracturingfluid (C-VES). Microscopic experiments show that guar-based fracturing fluid has a certain viscosityeffect on coal, while C-VES can adsorb and capture the primary coal dust in coal. FTIR analysis shows thatC-VES has a great change on the content of coal functional groups, and guar-based fracturing fluid haslittle effect on coal. Industrial analysis shows that C-VES can better reduce the ash content of coal andhas a certain impact on the thermal efficiency of coal than guar based fracturing fluid. Combustion characteristicexperiment shows that guar-based fracturing fluid does not affect the combustion effect of coal,and C-VES can expand the exothermic temperature range of coal but has little effect on the main combustionof coal. Molecular dynamics simulation results show that C-VES has the best wetting effect oncoal. This research provides theoretical guidance for dust prevention and improving the use and conversionefficiency of coal.

Versicolols A and B (1 and 2), two rareprenylated isocoumarin derivatives, along with five knownisocoumarins (3–7) were isolated from the fermentationproducts of an endophytic fungus Aspergillus versicolor. Their structures were elucidated on the basis of extensivespectroscopic analysis, including 1D- and 2D-NMR techniques. Compounds 1 and 2 were evaluated for their cytotoxicityagainst five human tumor cell lines. The resultsshowed that compounds 1 exhibited weak cytotoxicityagainst A549 and MCF7 cells with IC50 values of 9.4 and8.8 lm, and compound 2 exhibited weak cytotoxicityagainst SHSY5Y and MCF7 cells with IC50 values of 8.2and 6.8 lm, respectively.
Curcumin derivative HBC induces autophagy through activating AMPK signal in A549 cancer cells

Curcumin exhibits growth-suppressive activity against a variety of cancer cells, but low bioavailability restricts its application in chemotherapeutic trials. A synthetic curcumin derivative hydrazinobenzoylcurcumin (HBC) had been indicated that it could inhibit cell growth in A549 lung cancer cells via cell autophagy in our previous report. The present research aims to investigate the effect of HBC on the AMP-activated protein kinase (AMPK) signaling and whether AMPK signaling is involved in HBC-induced autophagy in A549 cells. The autophagy inhibitor 3-MA was used to block the inhibitory effect of HBC on the growth of A549 cells by acridine orange assay. The phosphorylations of AMPK and its downstream factor Acetyl CoA Carboxylase (ACC) were characterized in HBCtreated A549 cells with western blot analysis. Moreover, pharmacological inhibition of the AMPK signaling pathway by Compound C impairs the autophagyinducing effect of HBC. Our data indicates that HBC could activate the AMPK signal and induce AMPKmediated autophagy, which plays an important role in the inhibiting effect of HBC in A549 cells.
