        Dynamic Frequency of Blood CD4+CD25+ Regulatory T Cells in Rats with Collagen-induced Arthritis

        CD4+CD25+ regulatory T cells (CD4+CD25+ Tregs) have been shown to play a regulatory or suppressive role in the immune response and are possibly relevant to the pathogenesis of autoimmune diseases. In the present study, we attempted to investigate the frequency of CD4+CD25+ Tregs in peripheral blood (PB) of collagen-induced arthritis (CIA) rats during the development of arthritis, to determine whether their frequency is involved in the immunoregulation of this disease. The results showed that normal rats had similar frequencies of CD4+CD25+ Tregs in PB during the experiment time, expressed as a percentage of CD4+CD25+Foxp3+ T cells among the CD4+ T lymphocyte population. In contrast, the frequency of CD4+CD25+Foxp3+ T cells in CIA rats was found to change during the development of arthritis. In CIA rats, there is a significant negative correlation between the frequency of CD4+CD25+Foxp3+ T cells and paw swelling (r=-0.786, p< 0.01). The relationship between the frequency of CD4+CD25+Foxp3+ T and immune activation was not found in normal rats. During the time course, the frequency of CD4+CD25+Foxp3+ T was lower in CIA rats than in normal ones. The data suggest that the frequency of PB CD4+CD25+ Tregs may be a promising marker for arthritis activity.

        ONLINE OPTIMAL CONTROL STRATEGY METHODOLOGY FOR POWER-SPLIT HYBRID ELECTRIC BUS BASED ON HISTORICAL DATA

        An online optimal control strategy methodology on the basis of historical data for a power-split hybrid electric bus (HEB) is proposed in this study. This approach aims to fully utilize the fuel-saving capability of power-split HEB under real operating cycles and provide an effective way for solving the optimal calibration problem in the application promotion. Firstly, a procedure for synthesizing real-world driving cycles based on cluster analysis and Markov chain method is constructed. Subsequently, dynamic programming (DP) control algorithm is performed to explore the fuel economy potential. Moreover, a DP-based rule control strategy with an automated implementation foundation is introduced to achieve online approximate optimal effect. Finally, offline simulation and hardware-in-the-loop test are conducted. Simulation results validate that the proposed online optimal control strategy methodology has similar fuel-saving performance to DP optimal results and good real-time application conditions

        Mathematical and computer simulation for Electro-Magneto- Thermo-Elastic Buckling of the Porous Nano system

        Buckling analysis of porous sandwich nanoplate integrated with two piezoelectric face-sheets is presented based on shear and normal deformation theory (SNTD). Effect of small scales of the porous core and actuated face-sheets is accounted based on the nonlocal strain gradient theory (NSGT). Large parametric results are presented to investigate variation of various critical loads in terms of significant parameters such as porosity volume fraction, strain gradient and nonlocal parameter, and dimensionless geometric parameters. It is concluded that increase of porosity volume fraction leads to decrease of critical electric and magnetic potentials and increase of critical temperatures.

        Molecular mechanism of poisoning to honeybees (Apis mellifera L.) after ingestion of Camellia oleifera nectar

        To explore the molecular mechanism of honeybees (Apis mellifera L.) poisoning death by ingestion of Camellia oleifera nectar, feeding experiments were designed to investigate the poisoning effects. Observed the develop ment process of poisoning symptoms, and collected bees at key experimental nodes for transcriptome sequencing analysis. The results showed that the first poisoning symptom was observed within 0.5 h after ingestion of Camellia oleifera nectar, with death occurring as early as 2 h post-ingestion and all honey bees dying within 4 h. There were four stages in the development of poisoning symptoms: normal flying, crawling (abdominal swelling, inability to fly normally), fluttering (inability to crawl normally), and death. Transcriptome sequencing analyzed the causes of poisoning death indicated that honey bee poisoning occurred with the transduction of dietary restriction signals into cells, resulting in the down-regulated expression gene elements that ensure cell health and inhibit apoptosis, including intracellular NAD+-dependent protein deacetylase sirtuin 1, insulin-like growth factor 1, and heat shock proteins. At the same time, the expression of cysteinyl aspartate specific proteinase 7, which protects programmed cell apoptosis, was also downregulated, resulting in the rapid and disorderly deaths of a large number of cells in individuals in a short period of time, and eventually leading to bees fatality.

      • Network Information Target Search Model and Strategy Based on Web Vertical Crawler System

        With the explosive growth of the web information, acquiring the target information quickly, precisely and effectively in a large number of network information is restricted by many factors. In response to it, this paper analyzed the key technician part in the network information search engine-web crawler technology and proposed a network information target search model based on the web vertical crawler system with discussion on the implementation of the corresponding search strategy. Firstly, it builds the structure of the web crawler system and analyzes different function models. Next, it discusses some crucial problems including the options of deleting duplicated URL, the strategies to choose duplicated URL deletion method and the control model of error probability estimate model to acquire the closest network information to the target information. Finally, it verifies and discusses the operability and effectiveness of the model and the implementation strategy through case study.

        Experimental research on pressure fluctuation and vibration in a mixed flow pump

        To study the pressure fluctuation and vibration in mixed flow pumps, we chose a mixed flow pump with specific speed of 436.1 tomeasure. The time domains and frequency domain at each monitoring point on diffuser and outlet elbow were analyzed, as well as thevibration frequency domain characteristics at the impeller outlet and near the motor. The results show that the peak value of pressurefluctuation peak decreased gradually with the increase of flow rate. The pressure fluctuation of each monitoring point had periodicity,and the frequency domain dominated by blade passing frequency and multiple shaft frequency. The vibration frequency of each monitoringpoint occurred at shaft frequency and its multiple shaft frequency. The dominant frequency and the second frequency were distributedin shaft frequency and double shaft frequency.

        Blast-Resistance and Damage Evaluation of Concrete Gravity Dam Exposed to Underwater Explosion: Considering the Initial Stress Field

        There is an increasing concern on the vulnerability of concrete gravity dams under blast loads. To a better evaluation of the blast-resistance, the effect of the initial stress field is taken into consideration herein. Firstly, a fluid-solid coupling numerical model is established to investigate the blast-resistance of concrete gravity dams. The numerical results indicate that the initial stress field has non-neglectable effect on the shock wave propagation, blast vibration, and failure mode of concrete gravity dams. The damage of dam base may be overestimated without the consideration of initial stress field. Moreover, a parametric study are carried out for further investigations of the dynamic responses and damage properties under various explosion scenarios (explosive charge, detonation depth, and standoff distance). With the consideration of the initial stress field, it can be concluded that the dam head nearest the explosive source is the weakest part, and the safety of the dam base is another important consideration, which may lead to severe dam-break floods. At last, a new method is proposed and recommended to evaluate the blast resistance of the whole concrete gravity dam, where the blast-induced vibration and penetration depth of cracks are suggested for the dam crest and dam base, respectively.

        Multi-objective optimization of drive gears for power split device using surrogate models

        Power split device (PSD) is a key component in the energy coupling and decoupling of parallel-series hybrid electric vehicle. This paperproposes a multi-objective optimization method to achieve optimal balance solution among the volume, contact stress, and frictionalenergy dissipation of PSD drive gears, some of which are implicit with respect to design variables. To avoid the time-consuming problemof finite element analysis used to solve nonlinear responses, surrogate models are adopted to generate approximate expressions of designvariables. Pareto-optimal solutions of PSD are obtained using multi-island genetic algorithm (GA), non-dominated sorting GA-II(NSGA-II), and multi-objective particle swarm optimization algorithm. The performances of PSD before and after optimization are compared. Results indicate that the proposed method is effective, and NSGA-II achieves higher optimizing efficiency in solving the multiobjectiveoptimization problem of PSD than the other algorithms.

        Development of a ladder-shape melting temperature isothermal amplification (LMTIA) assay for detection of African swine fever virus (ASFV)

        Background: Due to the unavailability of an effective vaccine or antiviral drug against the African swine fever virus (ASFV), rapid diagnosis methods are needed to prevent highly contagious African swine fever. Objectives: The objective of this study was to establish the ladder-shape melting temperature isothermal amplification (LMTIA) assay for the detection of ASFV. Methods: LMTIA primers were designed with the p72 gene of ASFV as the target, and plasmid pUC57 was used to clone the gene. The LMTIA reaction system was optimized with the plasmid as the positive control, and the performance of the LMTIA assay was compared with that of the commercial real-time polymerase chain reaction (PCR) kit in terms of sensitivity and detection rate using 200 serum samples. Results: Our results showed that the LMTIA assay could detect the 104 dilution of DNA extracted from the positive reference serum sample, which was the same as that of the commercial real-time PCR kit. The coincidence rate between the two assays was 100%. Conclusions: The LMTIA assay had high sensitivity, good detection, and simple operation. Thus, it is suitable for facilitating preliminary and cost-effective surveillance for the prevention and control of ASFV.
