This study investigated the 70% aqueous acetone extract of the fruits of Schisandra rubriflora which led to the isolation of eight lignans, including a new isolate, rubrisandrin C (1), and seven known lignans (2-8) . The structure of 1 was established by extensive 1D and 2D NMR spectroscopy and its absolute stereochemistry was determined by CD spectrum. Compounds 1-5 and 7-8 were evaluated for their anti-HIV-1 activity that showed inhibitory activity on HIV-1IIIB induced syncytium formation with EC50 values in the range of 2.26~20.4 μg/mL. Compounds 1 and 7 exerted their obvious protection of HIV-1IIIB inducted MT-4 host cells lytic effects with a selectivity index of 15.4 and 24.6, respectively.

Soil liquefaction causes extensive damages to buildings and infrastructures during earthquakes. It is very important to identify the significant parameters controlling seismic liquefaction which will enhance the predictability of evaluation models. In order to identify the significant factors, 203 literatures about soil liquefaction are analyzed by bibliometrics, 12 significant factors, such as: magnitude, epicentral distance, duration of earthquake, fines content, particle size, grain composition, relative density, drainage condition, degree of consolidation, thickness of sand layer, depth of sand layer, groundwater table, are identified among 22 influence factors. And then the interrelationship among the 12 main factors was analyzed, and a multilevel structure hierarchy is developed by interpretive structural modeling, which would help understand more clearly about the relationships among these factors. The results show that: magnitude, epicentral distance, duration of earthquake and drainage condition have a direct impact on soil liquefaction, and fines content plays a key role in all factors. The results can provide guidelines for election of factors and the establishment of model in the risk evaluation of soil liquefaction.

The incorporation of microwave irradiation with the prevalent “click chemistry” is currently of considerable synthetic interest. We describe here the introduction of such laboratorial shortcut into carbohydrate-based drug discovery,resulting in the rapid formation of a series of triazole-linked salicylic β-D-O-glycosides with biological activities. All “clicked” products were achieved in excellent yields (≈ 90%) within only a quarter. In addition, based on the structural characteristics of the afforded glycomimetics, their inhibitory activities were evaluated toward protein tyrosine phosphatases 1B (PTP1B) and a panel of homologous protein tyrosine phosphatases (PTPs). Docking simulation was also conducted to plausibly propose binding modes of this glycosyl salicylate series with the enzymatic target.

Astragali Radix ( A. Radix ) is the dried root of Astragalus membranaceus var. mongholicus (Bge) Hsiao or Astragalusmembranaceus (Fisch.) Bge., belonging to the family Leguminosae, which is mainly distributed in China. A. Radix hasbeen consumed as a tonic in China for more than 2000 years because of its medicinal eff ects of invigorating the spleenand replenishing qi. Currently, more than 400 natural compounds have been isolated and identifi ed from A. Radix , mainlyincluding saponins, fl avonoids, phenylpropanoids, alkaloids, and others. Modern pharmacological studies have shown thatA. Radix has anti-tumor, anti-infl ammatory, immunomodulatory, anti-atherosclerotic, cardioprotective, anti-hypertensive,and anti-aging eff ects. It has been clinically used in the treatment of tumors, cardiovascular diseases, and cerebrovascularcomplications associated with diabetes with few side eff ects and high safety. Post-earthquake fire is a common disaster which causes serious safety issues to infrastructures. This study aims to investigate the residual loading capacities of circular concrete-filled steel tube (CFST) columns under post-earthquake fire experimentally and numerically. The experimental programme contains two loading steps - pre-damage cyclic loading at room temperature and transient state tests with constant compression loads. Three finite element models are developed and validated against the test results. Upon validation, a total of 48 numerical results were generated in the parametric study to investigate the effects of thickness and strengths of steel tube, axial compression ratio and damage degree on the fire resistance of circular CFST columns. Based on the analysis on experimental and numerical results, the loading mechanism of circular CFST columns is discussed. A design method is proposed for the prediction of fire resistance time under different seismic pre-damage and compression loads. The predictions by the new method is compared with the newly generated experimental and numerical results and is found to be accurate and consistent with the mean value close to the unity and a coefficient of variation around 1%.

Objective: The aim of this study was to investigate possible mechanisms of LOX gene effects on invasion and metastasis of breast cancer cells by RNA interference. Methods: LOX-RNAi-LV was designed, synthesized, and then transfected into a breast cancer cell line (MDA-MB-231). Expression of LOX, MMP-2 and MMP-9 was determined by real-time PCR, and protein expression of LOX by Western blotting. Cell migration and invasiveness were assessed with Transwell chambers. A total of 111 cases of breast cancer tissues, cancer-adjacent normal breast tissues, and 20 cases of benign lesion tissues were assessed by immunohistochemistry. Results: Expression of LOX mRNA and protein was suppressed, and the expression of MMP-2 and MMP-9 was significantly lower in the RNAi group than the control group (P<0.05), after LOX-RNAi-LV was transfection into MDA-MB-231 cells. Migration and invasion abilities were obviously inhibited. The expression of LOX protein in breast cancer, cancer-adjacent normal breast tissues and benign breast tumor were 48.6% (54/111), 26.1% (29/111), 20.0% (4/20), respectively, associations being noted with clinical stage, lymph node metastasis, tumor size and ER, PR, HER2, but not age. LOX protein was positively correlated with MMP-2 and MMP-9. Conclusion: LOX displayed an important role in invasion and metastasis of breast cancer by regulating MMP-2 and MMP-9 expression which probably exerted synergistic effects on the extracellular matrix (ECM).

Background: Low tyrosine-protein phosphatase nonreceptor type 12 (PTPN12) expression may be associated with breast cancer growth, proliferation, and metastasis. However, the prognostic value of PTPN12 in breast cancer has not been clearly identified. Patients and Methods: 51 triple-negative breast cancer (TNBC) patients and 83 non-TNBC patients with a histopathology diagnosis from October 2001 to September 2006 were included in this study. Immunohistochemical staining for PTPN12 on tissue microarrays was conducted. Results: High PTPN12 expression was seen in 39.2% of TNBC and 60.2 % of non-TNBC cases. Low PTPN12 expression was associated with lymph node status (p = 0.002) and distant metastatic relapse (p = 0.002) in TNBC patients. Similarly, low PTPN12 expression in non-TNBC patients was significantly correlated with lymph node status (p = 0.002), stage (p = 0.002) and distant metastatic relapse (p = 0.039). The high PTPN12 expression group was associated with longer DFS and OS compared with low PTPN12 expression group only in TNBC cases (p = 0.005, p = 0.015), according to univariate Cox regression analysis. Conclusion: These findings provide evidence that low expression of PTPN12 is associated with worse prognosis and may be used as a potential prognostic biomarker in TNBC patients.

Purpose: This study was to determine the efficacy and safety of pemetrexed based chemotherapy in treating patients with metastatic gastric cancer who failed to respond to first and (or) second line chemotherapy. Patients and Methods: Metastatic gastric cancer patients who failed first and (or) second line chemotherapy, were enrolled. All patients were recruited from Jiangsu Cancer Hospital & Research Institute, and were treated with pemetrexed $500mg/m2$ (intravenous; on day 1), and a platinum (or irinotecan) every 3 weeks until disease progression, or intolerable toxicity. Evaluation on efficacy was conducted after two cycles of chemotherapy using the Response Evaluation Criteria for Solid Tumors. Toxicity was recorded according to National Cancer Institute Common Terminology Criteria for Adverse Events version 3.0. Results: From Jun 2011 to May 2013, 23 patients were enrolled. All eligible 23 patients completed at least 2 cycles of chemotherapy with pemetrexed based chemotherapy, and were evaluable. Their median age was 55 years (range 40 to 78 years). Seventeen patients were male and 6 female. Three patients (13%) achieved partial response, five patients (22%) stable, 15 patients (65%) with disease progression, and none with complete response. Grade 2 neutrophil suppression occurred in 4.3%, grade 3 in 13% of patients, and no grade 4 was reported. Thrombocytopenia was encountered as follows: 4.3% grade 2, 4.3% grade 3 and 4.3% grade 4. Incidence of anemia was 34.8% in grade 2, 8.7% grade 3 and 0% grade 4. Only 4.3% of patients required packed red blood cell infusion. Elevated transaminase were 4.3% in grade 2 and 0% in grade 3 or 4. Other toxicity included oral mucositis. Conclusions: Pemetrexed based chemotherapy is mildly effective in treating patients with metastatic gastric cancer with tolerable toxicity.

Di-2-ethylhexyl phthalate (DEHP) which belongs to phthalatic-acid esters, is a kind of harmful, global environmental pollutants. It is a known endocrine disrupting chemical and male reproductive toxicant. However, the mechanism by which DEHP exposure result in male reproductive toxicity is still unclear. To elucidate the productive toxicity mechanism of DEHP, we attemptted to investigate oxidative stress, apoptotic effects, mRNA and protein expression of apoptosis-associated genes including FasL, caspase-3, and caspase-8 in GC-2spd (mouse spermatogenic cells). The results showed that, with the increase of DEHP concentration, cell apoptosis rate increased; the activities of relation index of oxidative stress such as malondialdehyde (MDA), superoxide dismutase (SOD) and glutatione peroxidase (GSH-PX) changed significantly; the mRNA and protein expression levels of FasL, caspase-3, -8, altered obviously. These results suggested that DEHP could induce apoptosis of GC-2spd cells through oxidative stress and FasL-dependent pathway.
