        Alloying Behavior and Properties of FeSiBAlNiCox High Entropy Alloys Fabricated by Mechanical Alloying and Spark Plasma Sintering

        In this paper, FeSiBAlNiCo x ( x = 0.2, 0.8) high-entropy alloy (HEA) powders were fabricated by mechanical alloying process,and the powders milled for 140 h were sintered by spark plasma sintering (SPS) technique. The microstructures andproperties of as-milled powders and as-sintered samples were investigated. The results reveal that the fi nal milling products(140 h) of both sample powders present the fully amorphous structure. The increased Co contents obviously enhance theglass forming ability and thermal stability of amorphous HEA powders, which are refl ected by the shorter formation time offully amorphous phase and the higher onset crystallization temperature, respectively. According to coercivity, the as-milledFeSiBAlNiCo x ( x = 0.2, 0.8) powders (140 h) are the semi-hard magnetic materials. FeSiBAlNiCo 0.8 HEA powders possessthe highest saturation magnetization and largest remanence ratio. The SPS-ed products of both bulk HEAs are composedof body-centered cubic solid solution, and FeSi and FeB intermetallic phases. They possess the high relative density above97% and excellent microhardness exceeding 1150 HV. The as-sintered bulks undergo the remarkable increase in saturationmagnetization compared with the as-milled state. The SPS-ed FeSiBAlNiCo 0.8 HEA exhibits the soft magnetic properties. The electrochemical corrosion test is carried out in 3.5% NaCl solution. The SPS-ed FeSiBAlNiCo 0.2 HEA reveals the betterpassivity with low passive current density, and the higher pitting resistance with wide passive region.

        Improving Cellulase Production in Trichoderma koningii Through RNA Interference on ace1 Gene Expression

        Ribonucleic acid interference (RNAi) inhibits the expression of target genes in a sequence-specific manner, and shows potential for gene knockdown in filamentous fungi, in which the locus-specific gene knockout occurs in low frequency. In this study, the function of the repressor of cellulase expression I (ACEI) was verified in Trichoderma koningii (T. koningii) YC01 through RNAi, and ace1-silenced strains with improved cellulase productivity were obtained. An expression cassette that transcribed the interfering double-stranded RNA (dsRNA) of ace1 was constructed and transformed into T. koningii, and the transformants, in which the expression of ace1 was successfully silenced, were selected. As a result of the ace1 gene silencing, the expression levels of the main cellulase and xylanase genes were elevated, and the enhanced production of total proteins, cellulase, and xylanase was observed in the cultivation. In addition, the downregulation of ace1 resulted in an increasing expression of xyr1, but no clear variation in the expression of cre1, which suggested that ACEI acted as a repressor of the xyr1 transcription, but was not involved in the regulation of the cre1 expression. The results of this work indicate that ace1 is a valid target gene for enhancing enzyme production in T. koningii, and RNAi is an appropriate tool for improving the properties of industrial fungi.

        Molecular Cloning and Heterologous Expression of an Acid-Stable Endoxylanase Gene from Penicillium oxalicum in Trichoderma reesei

        An endoxylanase gene (PoxynA) that belongs to the glycoside hydrolase (GH) family 11 was cloned from a xylanolytic strain, Penicillium oxalicum B3-11(2). PoxynA was overexpressed in Trichoderma reesei QM9414 by using a constitutive strong promoter of the encoding pyruvate decarboxylase (pdc). The high extracellular xylanase activities in the fermentation liquid of the transformants were maintained 29~35-fold higher compared with the wild strain. The recombinant POXYNA was purified to homogeneity, and its characters were analyzed. Its optimal temperature and pH value were 50oC and 5.0, respectively. The enzyme was stable at a pH range of 2.0 to 7.0. Using beechwood as the substrate, POXYNA had a high specific activity of 1,856 ± 53.5 IU/mg. In the presence of metal ions, such as Cu2+, and Mg2+, the activity of the enzyme increased. However, strong inhibition of the enzyme activity was observed in the presence of Mn2+ and Fe2+. The recombinant POXYNA hydrolyzed birchwood xylan, beechwood xylan, and oat spelt xylan to produce short-chain xylooligosaccharides, xylopentaose, xylotriose, and xylobiose as the main products. This is the first report on the expression properties of a recombinant endoxylanase gene from Penicillium oxalicum. The properties of this endoxylanase make it promising for applications in the food and feed industries.

        KIF11 Functions as an Oncogene and Is Associated with Poor Outcomes from Breast Cancer

        Purpose The study aimed to search and identify genes that were differentially expressed in breast cancer, and their roles in cancer growth and progression. Materials and Methods The Gene Expression Omnibus (Oncomine) and The Cancer Genome Atlas databases (https://cancergenome.nih.gov/) were screened for genes that were expressed differentially in breast cancer and were closely related to a poor prognosis. Gene expressions were verified by quantitative real-time polymerase chain reaction, and genes were knocked down by a lentivirus-based system. Cell growth and motility were evaluated and in vivo nude mice were used to confirm the in vitro roles of genes. Markers of epithelial-to-mesenchymal transition and the associations of KIF11 with the classical cancer signaling pathways were detected by Western blot. Results A series of genes expressed differentially in patients with breast cancer. The prognosis associated with high KIF11 expression was poor, and the expression of KIF11 increased significantly in high stage and malignant tumor cells. Inhibiting KIF11 expression in lentivirussuppressed cells revealed that KIF11 inhibition significantly reduced cell viability and colony formation, inhibited migration and invasion, but promoted apoptosis. The sizes and weights of KIF11-inhibited tumors in nude mice were significantly lower than in the negative controls. Western blot showed that E-cadherin in breast cancer was significantly upregulated in KIFinhibited cells and tumor tissues, whereas N-cadherin and vimentin were significantly downregulated. BT549 and MDA231 cells with KIF11 knockdown exhibited decreased ERK, AMPK, AKT, and CREB phosphorylation. Conclusion KIF11 acts as a potential oncogene that regulates the development and progression of breast cancer.

        Effectiveness and safety of acupuncture for angina pectoris: An overview of systematic reviews

        Background: The number of systematic reviews or meta-analyses (SRs/MAs) on the effectiveness of acupuncture for angina pectoris (AP) is increasing. Due to the inconsistent conclusions and unknown quality of these SRs/MAs, this overview aimed to systematically evaluate and synthesize the existing SRs/MAs, attempting to provide more reliable evidence for the effectiveness and safety of acupuncture in the treatment of AP. Methods: SRs/MAs were searched via eight databases from inception to March 14, 2022. The risk of bias was evaluated using the Risk of Bias in Systematic reviews (ROBIS) tool. The quality of the methodology, reporting, and evidence were assessed by the Assess Systematic Reviews 2 (AMSTAR-2), the Preferred Reporting Item for Systematic Review and Meta-analysis for Acupuncture (PRISMA-A), and the Grading of Recommendations Assessment, Development, and Evaluation (GRADE) system, respectively. Results: Sixteen SRs/MAs were included and fifteen SRs/MAs were considered being of critically low quality according to AMSTAR-2. Only three SRs/MAs were rated at low risk of bias. No study reported all the items listed in the PRISMA-A checklist. No high-quality evidence with GRADE assessment was found. With the low-quality evidence, acupuncture combined with other interventions was superior to monotherapy (medications or Chinese medicine) in the angina symptom and electrocardiogram recovery. No adverse effects owing to acupuncture were reported. Conclusions: Owing to the lack of high-quality evidence provided by the current SRs/MAs, the effective-ness of acupuncture for AP still warrants further proof. Further researches with more critical design and methodology are needed for providing more convincing evidence.         Effect of Shadow Banking Activities on Firm Risk and Performance: Entrusted Loan Evidence from Chinese Listed Firms

        This study investigates the effect of non-financial firms’ activities in shadow banking on firm risk and performance. Using manually collected data of entrusted loans from Chinese listed firms, we find that lending firms’ bankruptcy risk and performance increases from their engagement of entrusted loan businesses in the year the loans are issued and in the following year. Further, firms’ risk increases and performance improves significantly when firms are financially healthy, financially constrained, and non-state-owned. Overall, our findings provide policy implications that the risk of shadow banking activities must be cautious.

        A Ringed Dwarf LINER 1 Galaxy Hosting an Intermediate-mass Black Hole with Large-scale Rotation-like ${\rm{H}}\alpha $ Emission

        <P>We report the discovery of a 20 kpc sized H alpha emission in SDSS J083803.68+540642.0, a ringed dwarf galaxy (M-V = -17.89 mag) hosting an accreting intermediate-mass black hole at z = 0.02957. Analysis of the Hubble Space Telescope images indicates that it is an early-type galaxy with a featureless low-surface brightness disk (mu(0) = 20.39 mag arcsec(-2) in the V band) and a prominent, relatively red bulge (V - I = 2.03, R-e = 0.28 kpc or 0 48) that accounts for approximate to 81% of the total light in the I band. A circumgalactic ring of a diameter 16 kpc is also detected, with a disperse shape on its south side. The optical emission lines reveal the nucleus to be a broad-line LINER. Our MMT longslit observation indicates that the kinematics of the extended H alpha emission is consistent with a rotational gaseous disk, with a mean blueshifted velocity of 162 km s(-1) and mean redshifted velocity of 86 km s(-1). According to our photoionization calculations, the large-scale H alpha emission is unlikely to be powered by the central nucleus or by hot evolved (post-AGB) stars interspersed in the old stellar populations, but by in situ star formation; this is vindicated by the line-ratio diagnostic of the extended emission. We propose that both the ring and large-scale H alpha-emitting gas are created by the tidal accretion in a collision-and then merger-with a gas-rich galaxy of a comparable mass.</P>

        Rapid Detection and Monitoring Therapeutic Efficacy of Mycobacterium tuberculosis Complex Using a Novel Real-Time Assay

        We combined real-time RT-PCR and real-time PCR (R/P) assays using a hydrolysis probe to detect Mycobacterium tuberculosis complex (MTBC)-specific 16S rRNA and its rRNA gene (rDNA). The assay was applied to 28 nonrespiratory and 207 respiratory specimens from 218 patients. Total nucleic acids (including RNA and DNA) were extracted from samples, and results were considered positive if the repeat RT-PCR threshold cycle was ≤35 and the ratio of real-time RT-PCR and real-time PCR load was ≥1.51. The results were compared with those from existing methods, including smear, culture, and real-time PCR. Following resolution of the discrepant results between R/P assay and culture, the overall sensitivity, specificity, positive predictive values (PPV), and negative predictive values (NPV) of all samples (including nonrespiratory and respiratory specimens) were 98.2%, 97.2%, 91.7%, and 99.4%, respectively, for R/P assay, and 83.9%, 89.9%, 72.3%, and 94.7%, respectively, for real-time PCR. Furthermore, the R/P assay of four patient samples showed a higher ratio before treatment than after several days of treatment. We conclude that the R/P assay is a rapid and accurate method for direct detection of MTBC, which can distinguish viable and nonviable MTBC, and thus may guide patient therapy and public health decisions.
