        Highly Selective Production of Compound K from Ginsenoside Rd by Hydrolyzing Glucose at C-3 Glycoside Using β-Glucosidase of Bifidobacterium breve ATCC 15700

        To investigate a novel β-glucosidase from Bifidobacterium breve ATCC 15700 (BbBgl) to produce compound K (CK) via ginsenoside F2 by highly selective and efficient hydrolysis of the C-3 glycoside from ginsenoside Rd, the BbBgl gene was cloned and expressed in E. coli BL21. The recombinant BbBgl was purified by Ni-NTA magnetic beads to obtain an enzyme with specific activity of 37 U/mg protein using pNP-Glc as substrate. The enzyme activity was optimized at pH 5.0, 35°C, 2 or 6 U/ml, and its activity was enhanced by Mn<sup>2+</sup> significantly. Under the optimal conditions, the half-life of the BbBgl is 180 h, much longer than the characterized β-glycosidases, and the Km and V<sub>max</sub> values are 2.7 mM and 39.8 μmol/mg/min for ginsenoside Rd. Moreover, the enzyme exhibits strong tolerance against high substrate concentration (up to 40 g/l ginsenoside Rd) with a molar biotransformation rate of 96% within 12 h. The good enzymatic properties and gram-scale conversion capacity of BbBgl provide an attractive method for large-scale production of rare ginsenoside CK using a single enzyme or a combination of enzymes.

        Relative Inhibitions of 5-Lipoxygenase and Myeloperoxidase and Free-Radical Scavenging Activities of Daidzein, Dihydrodaidzein, and Equol

        The effects of bioavailability and metabolic transformation on the biological activities of daidzein are relatively unknown. The effects of daidzein, dihydrodaidzein, and equol at physiologically relevant concentrations on the production of leukotriene B4 and F2-isoprostanes, and myeloperoxidase enzyme activity in freshly isolated human neutrophils were examined. Equol, at physiological concentrations, inhibited leukotriene B4 production (IC50–200 nmol/L) in human neutrophils significantly more than daidzein and dihydrodaidzein (IC50 values >1000 nmol/L). Daidzein, dihydrodaidzein, and equol did not affect the enzymatic hydrolysis of leukotriene A4 to leukotriene B4, suggesting that they exerted their inhibitory effects on the 5-lipoxygenase activity. Daidzein (IC50 = 600 nmol/L) protected against free radical peroxidation of arachidonic acid significantly more than did equol and dihydrodaidzein (IC50 values >1000 nmol/L). Equol also showed significantly greater inhibition of myeloperoxidase activity (IC50 = 450 nmol/L) when compared to daidzein and dihydrodaidzein. Equol accumulated within the human neutrophils at significantly higher concentrations than daidzein and dihydrodaidzein after incubation with the three compounds at physiologically relevant concentrations. Neutrophils were able to accumulate intracellular daidzein, dihydrodaidzein, and equol up to a concentration of ∼600 nmol/L. Our results provide in vitro evidence that the biological activities of daidzein are profoundly influenced by bioavailability and metabolic transformation.

        Causal Relations between Exposome and Stroke: A Mendelian Randomization Study

        Background and Purpose To explore the causal relationships of elements of the exposome with ischemic stroke and its subtypes at the omics level and to provide evidence for stroke prevention. Methods We conducted a Mendelian randomization study between exposure and any ischemic stroke (AIS) and its subtypes (large-artery atherosclerotic disease [LAD], cardioembolic stroke [CE], and small vessel disease [SVD]). The exposure dataset was the UK Biobank involving 361,194 subjects, and the outcome dataset was the MEGASTROKE consortium including 52,000 participants. Results We found that higher blood pressure (BP) (systolic BP: odds ratio [OR], 1.02; 95% confidence interval [CI], 1.01 to 1.04; diastolic BP: OR, 1.03; 95% CI, 1.01 to 1.05; pulse pressure: OR, 1.03; 95% CI, 1.00 to 1.06), atrial fibrillation (OR, 1.18; 95% CI, 1.13 to 1.25), and diabetes (OR, 1.13; 95% CI, 1.07 to 1.18) were significantly associated with ischemic stroke. Importantly, higher education (OR, 0.69; 95% CI, 0.60 to 0.79) decreased the risk of ischemic stroke. Higher systolic BP (OR, 1.06; 95% CI, 1.02 to 1.10), pulse pressure (OR, 1.08; 95% CI, 1.02 to 1.14), diabetes (OR, 1.28; 95% CI, 1.13 to 1.45), and coronary artery disease (OR, 1.58; 95% CI, 1.25 to 2.00) could cause LAD. Atrial fibrillation could cause CE (OR, 1.90; 95% CI, 1.71 to 2.11). For SVD, higher systolic BP (OR, 1.04; 95% CI, 1.00 to 1.07), diastolic BP (OR, 1.06; 95% CI, 1.01 to 1.12), and diabetes (OR, 1.22; 95% CI, 1.10 to 1.36) were causal factors. Conclusions The study revealed elements of the exposome causally linked to ischemic stroke and its subtypes, including conventional causal risk factors and novel protective factors such as higher education.

        청소년의 기본심리욕구와 희망이 학교적응에 미치는 영향

        본 연구는 청소년의 기본심리욕구와 희망이 학교적응에 미치는 영향을 알아보기 위해 수행되었다. 서울, 경기 소재 5개 중?고등학교 학생 454명을 대상으로 하였다. 기본심리욕구 및 희망과 학교적응 수준은 자기보고식 질문지법을 이용해 평가하였다. 수집된 자료는 평균, 표준편차, t-검증, 상관분석, 회귀분석을 이용해 분석하였다. 주요 연구결과는 다음과 같다. 첫째, 청소년의 기본심리욕구, 희망과 학교적응에서 학교급별 차이가 나타나 중학생들이 고등학생들보다 기본심리욕구, 희망과 학교적응 수준이 높은 것으로 나타났다. 둘째, 청소년의 학교적응은 기본심리욕구 및 희망과 정적 관련성이 있었다. 셋째, 희망은 기본심리욕구와 학교적응의 관계를 부분적으로 매개하였다. 즉, 청소년의 기본심리욕구는 학교적응에 직접적인 영향을 미치기도 하고, 희망을 통해 학교적응에 간접적인 영향도 미친다는 것을 알 수 있었다. 구체적으로 살펴보면, 기본심리욕구와 학교적응의 하위요인인 학교공부에 대한 적응, 학교교사에 대한 적응, 학교생활에 대한 적응과의 관계를 희망이 부분적으로 매개하였다. 하지만 기본심리욕구와 학교친구에 대한 적응과의 관계에서는 희망의 매개효과가 나타나지 않았다. 이러한 연구결과는 청소년의 학교적응을 위한 다양한 지원을 제공하는 데 유용한 자료로 활용될 수 있을 것이다. This study examined the effects of basic psychological needs and hopes on the school adjustment levels of adolescents. The participants were made up of 454 middle and high school students from the Seoul and Gyeongi areas. The data were analyzed using t?tests, Pearson’s correlations and multiple regression analysis. The results were as follows: First, middle school students reported greater basic psychological needs, hopes and school adjustment levels than high school students. Second, the school adjustment levels of these adolescents were positively associated with basic psychological needs and hopes. Third, hope was found to play a partially mediating role in the relationship between basic psychological needs and the levels of school adjustment exhibited by the adolescents in this study. These results suggested that the school adjustment levels of adolescents could be improved by the fulfillment of basic psychological needs and hopes.
