A number of factors influencing microspore embryogenesis and plant regeneration were examined in five subspecies (rapa, oleifera, niposinica, perviridis,broccoletto) of B. rapa. Addition of 6-benzylaminopurine (BA) in 1/2 NLN-10 medium improved the embryo yield by 2–12 fold. Addition of activated charcoal (AC) in the medium was not effective for microspore embryogenesis. Moreover, AC canceled the positive effect of BA, when the medium containing both BA and AC was used. Of 24genotypes examined for microspore embryogenesis, 22genotypes of all five subspecies produced embryos ranging from 0.02 to 15.0 per 2 9 105 microspores, but two genotypes were not responsive. Low temperature pretreatment of flower buds significantly improved the microspore embryogenesis. When cotyledonary embryos were subcultured on a filter paper placed on top of 0.8 %agar-solidified B5-2 medium and 1.6 % agar B5-2 medium,plant regenerations were increased 4–8 fold compared to 0.8 % agar medium. The ploidy levels of regenerated plants in three genotypes were determined by flow cytometry, revealing that 66–100 % of them were diploid. The results enable the advancement of breeding programs and genetic studies in B. rapa.
Transcriptional Network Controlling Endochondral Ossification

Endochondral ossification is the fundamental process of skeletal development in vertebrates. Chondrocytes undergo sequential steps of differentiation, including mesenchymal condensation, proliferation, hypertrophy, and mineralization. These steps, which are required for the morphological and functional changes in differentiating chondrocytes, are strictly regulated by a complex transcriptional network. Biochemical and mice genetic studies identified chondrogenic transcription factors critical for endochondral ossification. The transcription factor sex-determining region Y (SRY)-box 9 (Sox9) is essential for early chondrogenesis, and impaired Sox9 function causes severe chondrodysplasia in humans and mice. In addition, recent genome-wide chromatin immunoprecipitation-sequencing studies revealed the precise regulatory mechanism of Sox9 during early chondrogenesis. Runt-related transcription factor 2 promotes chondrocyte hypertrophy and terminal differentiation. Interestingly, endoplasmic reticulum (ER) stress-related transcription factors have recently emerged as novel regulators of chondrocyte differentiation. Here we review the transcriptional mechanisms that regulate endochondral ossification, with a focus on Sox9.
Future Design and Success Engineering in Global Society by Creative Self-innovation

In this paper, we proposed “Future Design and Success Engineering in Global Society by Self-Innovation” (an abbreviatedname: FDSE). We started to make a small experiment between Korea Maritime University and Yamagata University at thecurrent year. After inspecting the results, especially, successful and unsuccessful points in our lecture, we have to develop the strong points and amend the week points, through making the analysis of success-failure, which is his favorite management method. We have to level up the contents of our work in the future. The students for this lecture would learn from experiences of “the common knowledge (common field of discussion)”, held common languages, common thought, common system. They filled up the blanks of “The Matrix Frame” which is mentioned in the previous report, and theyexpressed their own way of life (work, study). As a matter of course, their personal private affairs must not be made public.It is quite happy that the cultural, economical exchanges between Korea and Japan are now in full flourish and people doa lot of traveling each other. However, if this project will start on the educational global standard, we have a world-wide significance. Moreover, we can promote goodwill between Korea and Japan, and also we can contribute to a better international understanding and the peace of the world by students in both Universities. We hope that candidates for thislecture will have a rounded personality and be a life worthy of human being for realizing Self-Actualization in “Motivationand Personality” by Abraham Maslow.
Application of Matrix Thinking Method to Introduction Program in Engineering Education

From a lot of survey, it is obvious that most students in universities lose their desire for learning just after enteringtheir universities. In order to solve this problem, we developed a novel educational tool for the students, named “Thethinking method based on matrix diagram”. If they try hard with the help of this tool, they will be able to learn how todesign and manage their splendid university lives in addition to get the basic knowledge and to improvetheir basic abilities.It is also found that they can earn the shared knowledge mutually after learning a common method, which supports to makethem to improve their communication abilities drastically.

high tumors. Therefore, sufficient sampling of histological specimens is necessary in cases of unresectable pancreatic cancer (UR-PC). This multicenter study investigated the efficacy of endoscopic ultrasound-guided fine-needle biopsy (EUS-FNB) using a Franseen needlefor MSI evaluation in patients with UR-PC. Methods: A total of 89 patients with UR-PC who underwent endoscopic ultrasound-guided fine-needle aspiration (EUS-FNA) orEUS-FNB using 22-G needles at three hospitals in Japan (2018–2021) were enrolled. Fifty-six of these patients (FNB 23 and FNA 33)were followed up or evaluated for MSI. Patient characteristics, UR-PC data, and procedural outcomes were compared between patientswho underwent EUS-FNB and those who underwent EUS-FNA. Results: No significant difference in terms of sufficient tissue acquisition for histology was observed between patients who underwentEUS-FNB and those who underwent EUS-FNA. MSI evaluation was possible significantly more with tissue samples obtained usingEUS-FNB than with tissue samples obtained using EUS-FNA (82.6% [19/23] vs. 45.5% [15/33], respectively; p<0.01). In the multivariateanalysis, EUS-FNB was the only significant factor influencing the possibility of MSI evaluation. Conclusions: EUS-FNB using a Franseen needle is desirable for ensuring sufficient tissue acquisition for MSI evaluation.
