Production of Bio-Based Isoprene by the Mevalonate Pathway Cassette in Ralstonia eutropha

Isoprene has the potential to replace some petroleum-based chemicals and can be produced through biological systems using renewable carbon sources. Ralstonia eutropha can produce value-added compounds, including intracellular polyhydroxyalkanoate (PHA) through fatty acid and lipid metabolism. In the present study, we engineered strains of R. eutropha H16 and examined the strains for isoprene production. We optimized codons of all the genes involved in isoprene synthesis by the mevalonate pathway and manipulated the promoter regions using pLac and pJ5 elements. Our results showed that isoprene productivity was higher using the J5 promoter (1.9 ± 0.24 μg/l) than when using the lac promoter (1.5 ± 0.2 μg/l). Additionally, the use of three J5 promoters was more efficient (3.8 ± 0.18 μg/l) for isoprene production than a one-promoter system, and could be scaled up to a 5-L batch-cultivation from a T-flask culture. Although the isoprene yield obtained in our study was insufficient to meet industrial demands, our study, for the first time, shows that R. eutropha can be modified for efficient isoprene production and lays the foundation for further optimization of the fermentation process.
Neural Tract Injuries by Penetration of Foreign Body: a Diffusion Tensor Tractography Study

We presented with a patient who showed injury of the cingulum and fornix by penetration of a foreign body into the brain on diffusion tensor tractography (DTT). A 63-year-old man suffered a brain injury by a part of a power saw blade that was suddenly detached from a power saw during work. A part of the power saw blade penetrated his right frontal skull and advanced to the right posterior horn of the lateral ventricle. This penetration caused traumatic intracerebral hemorrhage in the right frontal lobe and intraventricular hemorrhage in the lateral ventricle. He underwent craniotomy and removal of intracranial foreign bodies (bony pieces and saw blade). The patient`s Memory Assessment Scale scores were 74 (4%ile) for global memory, 78 (7%ile) for verbal memory, and 80 (9%ile) for visual memory. DTTs showed disruptions in the anterior portion of the fornical body, right fornical crus, the anterior portion of the right cingulum, and the middle portion of the left cingulum, compared to the control. It seems that the sustained memory impairment of this patient might be related to injury of the cingulum and fornix.

Cyanobacteria are able to produce diverse value-added biochemicals from CO<sub>2</sub> and light. To utilize the potency, it is critical to understand the photosynthetic mechanism under various environmental conditions. Here, we investigated the inhibitory mechanisms of photosynthesis under high light and low temperature stress in Synechocystis sp. PCC 6803. Under each stress condition, the transcript abundance and translation efficiency were measured using RNA-seq and Ribo-seq, and the transcription unit architecture was constructed by transcription start sites and transcript 3'-end positions obtained from TSS-seq and Term-seq. Our results suggested that the mode of photosynthesis inhibition differed between the two stress conditions. The poor translation of photosystem I resulted from ribosome stalling at the untranslated regions, affecting the overall photosynthesis under low temperature. Our multi-omics analysis with transcription unit provides foundational information on photosynthesis for future industrial strain development.

<P>We systematically examined the growth parameters of the hydrothermally grown ZnO nanorods and studied their effects on the external quantum efficiency of 32 × 32 pixelated InGaN/GaN microlight-emitting diodes (<I>μ</I>-LEDs). The growth temperature increased the growth rate of the hydrothermally grown ZnO nanorods in the <I>c</I>-axis direction but not in the <I>a</I>-axis direction. On the other hand, the concentration of reaction solution increased the growth rate in the <I>c</I>-axis direction as well as the lateral growth rate of ZnO nanorods. The pressure of autoclave reactor had little influence on ZnO growth. The external quantum efficiency and light output power of the 32 × 32 pixelated InGaN/GaN <I>μ</I>-LEDs with the hydrothermally grown ZnO nanorods on the sapphire substrate were enhanced by 16.4% and 16.0% at 83 A/cm<SUP>2</SUP>, respectively, compared to those of the <I>μ</I>-LEDs without the ZnO nanorods, which can be attributed to increased light extraction due to the ZnO nanorods.</P>
