        Heme oxygenase-1 induced by desoxo-narchinol-A attenuated the severity of acute pancreatitis via blockade of neutrophil infiltration

        <P><B>Abstract</B></P> <P>Heme oxygenase-1 (HO-1) has an anti-inflammatory action in acute pancreatitis (AP). However, its mechanism of action and natural compounds/drugs to induce HO-1 in pancreas are not well understood. In this study, we investigated the regulatory mechanisms of HO-1 during AP using desoxo-narchinol-A (DN), the natural compound inducing HO-1 in the pancreas. Female C57/BL6 Mice were intraperitoneally injected with supramaximal concentrations of cerulein (50 μg/kg) hourly for 6 h to induce AP. DMSO or DN was administered intraperitoneally, then mice were sacrificed 6 h after the final cerulein injection. Administration of DN increased pancreatic HO-1 expression through activation of activating protein-1, mediated by mitogen-activated protein kinases. Furthermore, DN treatment reduced the pancreatic weight-to-body weight ratio as well as production of digestive enzymes and pro-inflammatory cytokines. Inhibition of HO-1 by tin protoporphyrin IX abolished the protective effects of DN on pancreatic damage. Additionally, DN treatment inhibited neutrophil infiltration into the pancreas via regulation of chemokine (C-X-C motif) ligand 2 (CXCL2) by HO-1.         In vitro and in vivo anti-inflammatory effects of pegmatite

        Pegmatite is a coarse-grained intrusive igneous rock rich in rare elements such as uranium, tungsten, and tantalum with Ca, K, Mg, Fe, Se, Ge, and Ho. We tested in vitro and in vivo assays for the anti-inflammatory activity of pegmatites. We firstly evaluated the suppressive effects of pegmatite on macrophage cell line RAW 264.7 cells stimulated with proinflammatory stimuli lipopolysaccharide (LPS) to determine nitric oxide (NO) production and TNF-$\alpha$ and IL-6 release. The $IC_{50}$ values of pegmatite exceeded $5,000\;{\mu}g/mL$. Treatment of RAW 264.7 cells with pegmatite significantly inhibited LPS-stimulated NO production and proinflammatory cytokines such as TNF-$\alpha$ and IL-6 secretion in a dose-dependent manner (P<0.05). In vivo studies were tested with two animal models of arachidonic acid-induced mouse ear edema and an acetic acid-induced increase in capillary permeability. The pegmatite significantly attenuated ear edema induced by arachidonic acid and reduced the acetic acid-induced increase in capillary permeability in mice (P<0.05) when the pegmatite was administered topically (10 mg per ear) for 24 h. Therefore, pegmatite potentially shows an anti-inflammatory activity in the in vitro and in vivo mice and in the development of newer anti-inflammatory agents as mineral materials.

        Antioxidant and hepatoprotective effects of Korean ginseng extract GS-KG9 in a D-galactosamine-induced liver damage animal model

        BACKGROUND/OBJECTIVES: This study was designed to investigate the improvement effect of white ginseng extract (GS-KG9) on D-galactosamine (Ga1N)-induced oxidative stress and liver injury. SUBJECTS/METHODS: Sixty Sprague-Dawley rats were divided into 6 groups. Rats were orally administrated with GS-KG9 (300, 500, or 700 mg/kg) or silymarin (25 mg/kg) for 2 weeks. The rats of the GS-KG9- and silymarin-treated groups and a control group were then intraperitoneally injected Ga1N at a concentration of 650 mg/kg for 4 days. To investigate the protective effect of GS-KG9 against GalN-induced liver injury, blood liver function indicators, anti-oxidative stress indicators, and histopathological features were analyzed. RESULTS: Serum biochemical analysis indicated that GS-KG9 ameliorated the elevation of aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), and lactate dehydrogenase (LDH) in GalN-treated rats. The hepatoprotective effects of GS-KG9 involved enhancing components of the hepatic antioxidant defense system, including glutathione, glutathione peroxidase (GPx), superoxide dismutase (SOD), and catalase (CAT). In addition, GS-KG9 treatment inhibited reactive oxygen species (ROS) production induced by GalN treatment in hepatocytes and significantly increased the expression levels of nuclear factor erythroid-2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1) proteins, which are antioxidant proteins. In particular, by histological analyses bases on hematoxylin and eosin, Masson"s trichrome, α-smooth muscle actin, and transforming growth factor-β1 staining, we determined that the administration of 500 mg/kg GS-KG9 inhibited hepatic inflammation and fibrosis due to the excessive accumulation of collagen. CONCLUSIONS: These findings demonstrate that GS-KG9 improves GalN-induced liver inflammation, necrosis, and fibrosis by attenuating oxidative stress. Therefore, GS-KG9 may be considered a useful candidate in the development of a natural preventive agent against liver injury.

        Formation and Evaluation of the Coalition Government in Korea

        경기도의 연합정치 성과에 대한 분석을 실시함에 있어 정책연합, 정부연합이라는 두 가 지 분류기준을 주요변수로 규정하였다. 아울러, 경기도 연합정치를 평가함에 있어 한국과 같은 지방자치단체의 독임제 기관구성 하에 연합정치의 성과를 분석하여 연합정치의 활성 화를 위한 방안으로서 지방자치제도 개선방안을 제시하였다. 경기도 연합정치의 한계로 행정업무 지연의 문제 등 연합정치로 인한 다양한 사업으로 인해 많은 예산이 반영되고 해당 부서의 중요사업에 대한 검토를 신속하게 이루어져야 함 에도 불구하고, 보고 및 결제단계 증가로 시간소요 또한 증가하는 등의 문제점이 나타났다. 향후 경기도 연합정치를 활성화시키고 나아가 이를 실천하기 위한 지방자치제도의 개선 방안으로 첫째, 기관구성의 다양화이다. 둘째, 지방장관제 도입이다. 셋째, 광역시도 의회의 역량을 강화하는 것이다. 넷째, 지방선거제도의 개편이다. On the other hand, comparing the first and second periods in terms of the governmental coalition, the allocation of power and the improvement of the coalition process can be mentioned. First, from the aspect of power allocation, the first stage handed over to the opposition party the vice governor of social integration, which is responsible for the three countries of health, welfare, environment, and women's family. In the second stage(Cho Sung-Ho et al., 2017), In this way, the opposition party is able to share power with the opposition party. At the same time, it introduced the coalition chairperson system, which further strengthened the power of political parties participating in the coalition. In addition, by establishing a unified political arbitration committee to facilitate the conflict situation that occurred under the coalition government, the coalition government made soft landing. In the future, we will suggest ways to improve the local autonomy system to revitalize the Gyeonggido coalition politics and to practice it. First, diversification of organization. Second, the introduction of local ministers. Third, the metropolitan city should strengthen its capacity. Fourth, it is a reorganization of local electoral system.

        Regulatory Roles of Chrysanthemum zawadskii Roots in Nuclear Factor E2-related Factor 2/Antioxidant Response Element Pathway

        Cellular protection against carcinogens could be achieved by the induction of phase 2 detoxifying and antioxidant enzymes such as glutathione S-transferase (GST), NAD(P)H:quinone oxidoreductase 1 (NQO1) and heme oxygenase 1 (HO-1). Nuclear transcription factor E2-related factor 2 (Nrf2) binds to antioxidant response element (ARE) in the promoter region of these genes and the resulting transactivation occurs. In the present study the effect of gujeolcho (Chrysanthemum zawadskii) roots on the Nrf2-ARE pathway were investigated. C. zawadskii root extract was fractionated with a series of organic solvents and their ability to induce Nrf2-ARE pathway was examined. We separated the most potent dichloromethane (DCM) fraction into 12 sub-fractions and found several sub-fractions with strong effects on the Nrf2-ARE pathway. Fraction 4 strongly induced the ARE-reporter gene activity as well as Nrf2 expression. Sitosterol was isolated as a major compound in fraction 4 although its activity was not as potent as its mother fraction. These results indicate that C. zawadskii roots might be used as a potential natural chemopreventive source.

        <P>Platinum complexes [Pt(bpy)Cl2](bpy: 2,2'-bipyridine) and [Pt(PT)Cl2](PT: 1,10-phenantroline) were reacted with 5,5'-(9,9-dioctyl-9H-fluorene-2,7-diyl)bis(2,2'-bipyrine), bpy-spiro-bpy, to afford mono and diplatinium complexes: [(bpy)Pt(bpy-spiro-bpy)][PF6]2, [(bpy)Pt(bpy-spiro-bpy)Pt(bpy)])[PF6]4, and [(PT)Pt(bpy-spiro-bpy)])][PF6]2, which were characterized by NMR, UV/VIS, mass spectrum, and PL spectroscopy. They showed very high internal quantum yields (0.35-0.67) and emitted light in the blue region (473-520 nm). Herein, we describe the synthesis, characterization of the mono- and di-platinum complexes in detail.</P>

      • Real-Time Gaze Estimator Based on Driver's Head Orientation for Forward Collision Warning System

        <P>This paper presents a vision-based real-time gaze zone estimator based on a driver's head orientation composed of yaw and pitch. Generally, vision-based methods are vulnerable to the wearing of eyeglasses and image variations between day and night. The proposed method is novel in the following four ways: First, the proposed method can work under both day and night conditions and is robust to facial image variation caused by eyeglasses because it only requires simple facial features and not specific features such as eyes, lip corners, and facial contours. Second, an ellipsoidal face model is proposed instead of a cylindrical face model to exactly determine a driver's yaw. Third, we propose new features-the normalized mean and the standard deviation of the horizontal edge projection histogram-to reliably and rapidly estimate a driver's pitch. Fourth, the proposed method obtains an accurate gaze zone by using a support vector machine. Experimental results from 200 000 images showed that the root mean square errors of the estimated yaw and pitch angles are below 7 under both daylight and nighttime conditions. Equivalent results were obtained for drivers with glasses or sunglasses, and 18 gaze zones were accurately estimated using the proposed gaze estimation method.</P>

      • Endoplasmic reticulum stress induces hepatic steatosis via increased expression of the hepatic very low‐density lipoprotein receptor

        <P><B>Abstract</B></P><P>Recent evidence suggests that obese animals exhibit increased endoplasmic reticulum (ER) stress in the liver and adipose tissue. Although ER stress is closely associated with lipid homeostasis, it is largely unknown how ER stress contributes to hepatic steatosis. In this study, we demonstrate that the induction of ER stress stimulates hepatic steatosis through increased expression of the hepatic very low‐density lipoprotein receptor (VLDLR). Among the unfolded protein response sensors, the protein kinase RNA‐like ER kinase–activating transcription factor 4 signaling pathway was required for hepatic VLDLR up‐regulation. In primary hepatocytes, ER stress–dependent VLDLR expression induced intracellular triglyceride accumulation in the presence of very low‐density lipoprotein. Moreover, ER stress–dependent hepatic steatosis was diminished in the livers of VLDLR‐deficient and apolipoprotein E–deficient mice compared with wild‐type mice. In addition, the VLDLR‐deficient mice exhibited decreased hepatic steatosis upon high‐fat diet feeding. <I>Conclusion</I>: These data suggest that ER stress–dependent expression of hepatic VLDLR leads to hepatic steatosis by increasing lipoprotein delivery to the liver, which might be a novel mechanism explaining ER stress–induced hepatic steatosis.         Development of novel two-interconnected fluidized bed system

        Two-interconnected fluidized bed systems are widely used in various processes such as Fisher-Tropsch, hot gas desulfurization, CO₂ capture-regeneration with dry sorbent, chemical-looping combustion, sorption-enhanced steam methane reforming, chemical-looping hydrogen generation system, and so on. However, conventional two-interconnected fluidized bed systems are very complex, large, and difficult to operate because most of these systems require a riser and/or pneumatic transport line for solid conveying and loopseals or seal-pots for gas sealing, recirculation of solids to the riser, and maintaining of pressure balance. To solve these problems, a novel two-interconnected fluidized bed system has been developed. This system has two bubbling beds, solid injection nozzles, solid conveying lines, and downcomers. In this study, the effects of operating variables on solid circulation rate and gas leakage between two beds have been investigated in a cold mode two-interconnected fluidized bed system. The solid circulation rate increased as the hole diameter on the injection nozzle, the diameter of the injection nozzle, the solid height above the holes, and the number of holes on the injection nozzle increased. The gas leakage between the beds was negligible. Moreover, long-term operation of continuous solid circulation up to 60 hours was performed to check the feasibility of stable operation. The pressure drop profiles in the system loop were maintained steadily and solid circulation was smooth and stable.

        Comparison of calcium-channel blockers for long-term clinical outcomes in patients with vasospastic angina

        Background/Aims: Calcium channel blockers (CCBs) are the most widely prescribed medication for patients with vasospastic angina (VA). However, few studies have compared the prognosis of VA patients who are prescribed different CCBs. Methods: We enrolled 2,960 patients who received provocation test prospectively in 11 university hospitals in Korea. We divided 1,586 patients received four major CCBs into two groups: a first generation CCB (diltiazem and nifedipine) group and a second generation CCB (amlodipine and benidipine) group. Primary outcome was time to events of composite of death from any cause, acute coronary syndrome (ACS) and symptomatic arrhythmia during 3-year follow-up. We also compared the effect of each CCB on the control of angina symptoms. Results: There was no difference of the primary outcome among the two groups with a cumulative incidence rate of 5.4%, 2.9%, and a person-month incidence rate of 2.33 and 1.26, respectively (hazard ratio [HR], 0.54; 95% confidence interval [CI], 0.25 to 1.17; p = 0.120, as reference with the 1st generation CCBs). The incidence of ACS was significantly lower in 2nd generation CCBs group with a person-month incidence rate of 1.66 vs. 0.35 (HR, 0.22; 95% CI, 0.05 to 0.89; p = 0.034). Use of benidipine showed a significant better control of angina symptom compared with diltiazem for 3 years (odds ratio, 0.17; 95% CI, 0.09 to 0.32; p < 0.0001 at 3rd year). Conclusions: The first and second generation CCB groups did not differ in terms of composite outcome occurrence. However, the ACS incidence rate was significantly lower in the users of the 2nd generation CCBs.
