DNA methylation in Streptomyces griseus IFO 13350 was analyzed by high-performance liquid chromatographic analysis and bisulfite-based analysis to reveal two methylation sites, 5`-GC5mCGGC-3` and 5`-GAG5mCTC-3`. The methylation was reconstituted in Escherichia coli by simultaneous expression of S. griseus SGR4675 and S. achromogenes M.SacI. The E. coli cells produced plasmids that mimicked the methylation profile of S. griseus DNA, which was readily introduced into S. griseus. The results of this study raise the possibility of a promising approach to establish efficient transformation in several streptomycetes.
RK-270D and E, Oxindole Derivatives from Streptomyces sp. with Anti-Angiogenic Activity

A chemical investigation of a culture extract from Streptomyces sp. RK85-270 led to the isolation and characterization of two new oxindoles, RK-270D (1) and E (2). The structures of 1 and 2 were determined by analyzing spectroscopic and spectrometric data from 1D and 2D NMR and High-resolution electrospray ionization mass spectrometry (HRESIMS) experiments. Compound 1 exhibited anti-angiogenic activities against human umbilical vein endothelial cells (HUVECs) without cytotoxicity. Results of Western blot analysis revealed that 1 inhibits VEGF-induced angiogenesis in the HUVECs via VEGFR2/ p38 MAPK-mediated pathway.

<P>A chemical investigation of a culture extract from a soil-derived <I>Streptomyces</I> sp. RK88-1441 led to the isolation and characterization of two new glycosylated anthraquinones, aturanosides A (<B>1</B>) and B (<B>2</B>), and a new anthraquinone derivative, aturanocin (<B>3</B>). The structures of these compounds were elucidated by detailed NMR and MS spectroscopic analyses. The absolute configurations of the sugar units, based on the magnitudes of the coupling constants, ROESY correlations, and chemical derivatization, from <B>1</B> and <B>2</B> are 6-<I>O</I>-[<I>N</I>-acetyl-α-<SMALL>D</SMALL>-glucosamino-(1→2)-α-<SMALL>L</SMALL>-rhamnoside] and 6-<I>O</I>-α-<SMALL>L</SMALL>-rhamnoside, respectively. Compounds <B>1</B> and <B>2</B> showed no cytotoxicity against human umbilical vein endothelial cells (HUVECs), but significantly suppressed vascular endothelial growth factor (VEGF)-induced tube formation and invasion of HUVECs. The down-regulation of both the phosphorylation of VEGF receptor 2 and the expression of vascular endothelial cadherin at the protein level were also observed.</P> [FIG OMISSION]</BR>

Two new azaphilone derivatives containing 1,3-dioxolane moiety, penidioxolanes A (1) and B (2), were isolated from marine-derived fungus Penicillium sp. KCB12C078, together with four known compounds (3-6) by chemical investigation. Compounds 1 - 6 were isolated by combination of silica gel, ODS column chromatography and preparative HPLC. Their structures were determined by analysis of spectroscopic data including 1D-, 2D-NMR, and MS techniques. The isolates were evaluated against cancer cell growth inhibition effects and antimicrobial activity.

韓國 南海의 濟州道 城山浦 沿岸 地域에서 非靜力學的인 數値解析的 方法에 依해 熱的內部境界層의 1日 成長과 消滅過程이 硏究되었다. 沿岸實驗을 通해 熱的內部境界層의 成長은 복잡한 沿岸의 불규칙한 地形, 地表거칠기, 地表狀態, 地表上 氣溫, 表層水溫, 太陽輻射 및 長波輻射, 風速, 大氣安定度에 依해 生産됨이 밝혀졌다. 數値實驗의 結果는 熱的內部境界層이 晝間에 成長하며, 內部境界層의 消滅은 日沒 後에 始作되어, 夜間에는 完全히 消滅되어 日出 前까지 持續됨을 나타냈다. 內部境界層의 두께는 部分的으로는 下降流와 後向流에 依한 抑壓에 依해 山의 頂上附近보다 沿岸側 附近에서 훨씬 크다. 특히 亂流運動에 對한 鉛直擴散係數의 큰 變化를 갖는 地域이 沿岸에서 熱的內部境界層을 形成하는 地域과 매우 一致함을 보여준다. The processes of growth and recession of the thermal internal boundary layers at the Songsan-po coastal site of Cheju island in the South Sea of Korea were investigated by a non-hydrostatic numerical method. Through our coastal experiment it was found that a thermal internal boundary layer(TIBL) was produced by the effects of coastal irregularities, surface roughness of complex terrain, ground condition, temperatures of air over the gound and sea surface, solar and long wave radiations, wind speed and atmospheric stability. Results of our numerical simulation indicated that the growth of TIBL took place during daytime, while its recession started after sunset, showing the disapperance of internal boundary layer at night and then it lasted until sunrise. The depth of TIBL is much larger near coastal mountain sites than near the top of mountain, partly due to the surpress by the downslope and back flows of air. In particular, the area with a large change of vertical diffusion coefficients for turbulence momentum well matched the region to form a TIBL in the coastal region.
Octaminomycins A and B, Cyclic Octadepsipeptides Active against <i>Plasmodium falciparum</i>

<P>Two new cyclic octadepsipeptides, octaminomycins A (1) and B (2), were isolated from a microbial metabolite fraction library of Streptomyces sp. RK85-270 based on Natural Products Plot screening. Their structures were elucidated on the basis of HRESIMS, 1D and 2D NMR spectroscopic data, and MS/MS experiments for sequence analysis. The absolute configurations of the constituent amino acid residues were determined by a combination of single crystal X-ray diffraction and Marfey's methodology. Notably, octaminomycins A (1) and B (2) showed good in vitro antiplasmodial activity against chloroquine-sensitive as well as chloroquine-resistant strains with no cytotoxicity up to 30 mu M.</P>

A new homocysteine thiolactone derivative, thiolactomide (1), was isolated along with a known compound, N-acetyl homocysteine thiolactone (2), from a culture extract of soil-derived Streptomyces sp. RK88-1441. The structures of these compounds were elucidated by detailed NMR and MS spectroscopic analyses with literature study. In addition, biological evaluation studies revealed that compounds 1 and 2 both exert neuroprotective activity against 6-hydroxydopamine (6-OHDA)-mediated neurotoxicity by blocking the generation of hydrogen peroxide in neuroblastoma SH-SY5Y cells.
