Three homologous genes encoding functional D8-sphingolipid desaturase in Populus tomentosa

Δ8-sphingolipid desaturase is characterized by itsability to catalyze desaturation at the C8 position of the longchainbase of sphingolipids in plants. No previous studieshave been conducted on genes encoding Δ8-sphingolipiddesaturases in the woody plant Populus tomentosa. In thisstudy, three genes that encode Δ8-sphingolipid desaturasewere isolated fromP. tomentosa. Among these genes, PtD8Aand PtD8B showed high sequence similarity; whereas PtD8Cexhibited large sequence divergence.RT-PCRresults showedthat PtD8A and PtD8B were expressed in all tissues detected,whereas PtD8C was not expressed in roots. Heterologousexpression in yeast revealed that PtD8A/B/C were functionalΔ8-sphingolipid desaturases, and can catalyze the C18-phytosphingeninedesaturation to produce 8(Z)- and 8(E)-C18-phytosphingenine.However, the conversion rate and ratios ofthe two products differed. Compared with control cells,transgenic yeasts expressing PtD8A/B/C exhibited enhancedaluminum tolerance. Our findings further elucidated thebiochemical functions and evolutionary history ofΔ8-sphingolipid desaturases in plants. Candidate genes forbreeding new poplar germplasm resources with enhancedtolerance ability to aluminium were also provided.

In this study, banana biochar (BB) and cassava biochar (CB) were roasted in a tube furnace at 400℃ for 2 h, and their adsorption effects on malachite green (MG) were studied. The removal of MG with respect to the solution concentration and reaction time was also investigated. The results show that the optimal amount of biochar is 2 g/L. Studies of the adsorption kinetics and adsorption isotherms show that the pseudo-second-order kinetics can most accurately reflect the adsorption process. Freundlich model fits the experimental data well. The theoretical saturated adsorption capacities of BB and CB are 1,092.80 mg/g and 261.42 mg/g, respectively. SEM-EDS, TGA, BET, FTIR, XRD, element analysis and other characterization techniques were used to explore the adsorption mechanism. Based on the characterization results, it is speculated that the adsorption of BB and CB for MG mainly includes chemical adsorption and physical adsorption.
New Triterpenoids from the Fruits of Schisandra wilsoniana and Their Biological Activities

Investigation of an organic extract of the fruits Schisandra wilsoniana led to the isolation of two new highly oxygenated nortriterpenoids, named schilancidilactones V-W (1-2). Their structures were elucidated by spectroscopic evidence. Compounds 1-2 feature a double bond between C-7 and C-8 compared with related known nortriterpenoids isolated from the genus Schisandra. Compounds 1 and 2 were tested for their anti-HIV-1 activities and cytotoxicity. The results revealed that compounds 1 and 2 showed moderate anti-HIV-1 activities with $EC_{50}$ 3.05 and 2.87 ${\mu}g/mL$, respectively, and compound 1 showed high cytotoxicity against KB and MDA-MB-231 cell with $IC_{50}$ values of 3.18 and 5.22 ${\mu}M$, respectively.

Purpose Triple-negative breast cancer (TNBC) is highly malignant and has poor prognosis and a high mortality rate. The lack of effective therapy has spurred our investigation of new targets for treating this malignant cancer. Here, we identified RON (macrophage-stimulating 1 receptor) and MET (MET proto-oncogene, receptor tyrosine kinase) as a prognostic biomarker and therapeutic targets for potential TNBC treatment. Materials and Methods We analyzed RON and MET expression in 187 primary TNBC clinical samples with immunohistochemistry. We validated the targeted therapeutic effects of RON and MET in TNBC using three tyrosine kinase inhibitors (TKIs): BMS-777607, INCB28060, and tivantinib. The preclinical therapeutic efficacy of the TKIs was mainly estimated using a TNBC xenograft model. Results Patients with TNBC had widespread, abnormal expression of RON and MET. There was RON overexpression, MET overexpression, and RON and MET co-overexpression in 63 (33.7%), 63 (33.7%), and 43 cases (23.0%), respectively, which had poor prognosis and short survival. In vivo, the TKI targeting RON ant MET inhibited the activation of the downstream signaling molecules, inhibited TNBC cell migration and proliferation, and increased TNBC cell apoptosis; in the xenograft model, they significantly inhibited tumor growth and shrank tumor volumes. The TKI targeting RON and Met, such as BMS-777607 and tivantinib, yielded stronger anti-tumor effects than INCB28060. Conclusion RON and MET co-overexpression can be significant pathological characteristics in TNBC for poor prognosis. TKIs targeting RON and MET have stronger drug development potential for treating TNBC.
