        Medium- and long-chain triglyceride propofol reduces the activity of acetyl-coenzyme A carboxylase in hepatic lipid metabolism in HepG2 and Huh7 cells

        Medium- and long-chain triglyceride (MCT/LCT) propofol is widely used as an intravenous anesthetic, especially in the intensive care unit. The present study aimed to assess whether MCT/LCT propofol is safe in the hyperlipidemic population for long-term use. Free fatty acids (FFAs) were used to establish high-fat stimulation of HepG2 and Huh7 cells. Subsequently, these cells were treated with propofol at the concentration of 0, 4, or 8 μg/ml for 24 and 48 h. The results indicated that the cell viability was notably decreased when the cells were stimulated with 2 mmol/L FFAs and treated with 12 μg/ml MCT/LCT propofol. Accordingly, we chose 2 mmol/L FFAs along with 4 and 8 μg/ml MCT/LCT propofol for the subsequent experiments. Four and 8 μg/ml MCT/LCT propofol inhibited FFA-induced lipid accumulation in the cells and significantly reversed acetyl coenzyme A carboxylase (ACC) activity. In addition, MCT/LCT propofol not only significantly promoted the phosphorylation of AMPK and ACC, but also reversed the FFA-induced decreased phosphorylation of AMPK and ACC. In conclusion, MCT/LCT propofol reverses the negative effects caused by FFAs in HepG2 and Huh7 cells, indicating that MCT/LCT propofol might positively regulate lipid metabolism.

        Efficient adsorption capability of banana and cassava biochar for malachite green: Removal process and mechanism exploration

        In this study, banana biochar (BB) and cassava biochar (CB) were roasted in a tube furnace at 400℃ for 2 h, and their adsorption effects on malachite green (MG) were studied. The removal of MG with respect to the solution concentration and reaction time was also investigated. The results show that the optimal amount of biochar is 2 g/L. Studies of the adsorption kinetics and adsorption isotherms show that the pseudo-second-order kinetics can most accurately reflect the adsorption process. Freundlich model fits the experimental data well. The theoretical saturated adsorption capacities of BB and CB are 1,092.80 mg/g and 261.42 mg/g, respectively. SEM-EDS, TGA, BET, FTIR, XRD, element analysis and other characterization techniques were used to explore the adsorption mechanism. Based on the characterization results, it is speculated that the adsorption of BB and CB for MG mainly includes chemical adsorption and physical adsorption.

        Low temperature oxidation characteristics analysis of ultra-heavy oil by thermal methods

        To study the oxidation behavior of heavy oils, low temperature oxidation experiments under differentialtemperature and pressure conditions were carried out for both heavy oils (ordinary heavy and ultraheavyoils) with thermogravimetry (TG–DTG)/differential scanning calorimeter (DSC) analyses. And theresults indicated that the differences of physical properties and oxidation reactions for the heavy oilsascribe to the SARA compositions, H/C ratio and specific oxidation path. Owing to the asphaltenescontent, the ultra-heavy oil after the oxidation has more obvious coke deposition with lower activationenergy demand to make it possible for the in-situ combustion without combustion adjuvants.

      • Association of Genetic Polymorphisms at 1q22 but not 10q23 with Gastric Cancer in a Southern Chinese Population

        Objective: Data from a recent genome-wide association studiesy of gastric cancer (GC) and oesophageal squamous cell carcinoma in Chinese living in the Taihang Mountains of north-central China suggest that 1q22 and 10q23 are susceptibility-associated regions for GC. However, this has not been confirmed in southern Chinese populations. The aim of this study was to investigate whether these polymorphisms at 1q22 and 10q23 are associated with the risk of GC in a southern Chinese population. Methods: We selected seven top significant associated single nucleotide polymorphisms (SNPs) at 1q22 and 10q23 and conducted a population-based case-control study in a southern Chinese population. Genotypes were determined using MassARRAYTM system (Sequenome, San Diego, CA). Results: Two SNPs at 1q22, rs4072037 and rs4460629, were significantly associated with a reduced risk of GC, best fitting the dominant genetic model. Logistic regression models adjusted for age and sex showed that rs4072037 AG and GG (OR=0.64, P=0.017, compared with AA) and rs4460629 CT and TT (OR=0.54, P=0.0016, compared with TT) significantly reduced the risk of GC. However, no significant results for the five SNPs at 10q23 were obtained in this study. Conclusion: These outcomes indicate that 1q22 is associated with GC susceptibility in this southern Chinese population, while an association for the locus at 10q23 was not confirmed.

        Pan-cancer Analysis of Tumor Mutational Burden and Homologous Recombination DNA Damage Repair Using Targeted Next-Generation Sequencing

        Purpose Current variability in methods for tumor mutational burden (TMB) estimation and reporting demonstrates the urgent need for a homogeneous TMB assessment approach. Here, we compared TMB distributions in different cancer types using two customized targeted panels commonly used in clinical practice. Materials and Methods TMB spectra of 295- and 1021-gene panels in multiple cancer types were compared using targeted next-generation sequencing (NGS). The TMB distributions across a diverse cohort of 2,332 cancer cases were then investigated for their associations with clinical features. Treatment response data were collected for 222 patients who received immune-checkpoint inhibitors (ICIs) and their homologous recombination DNA damage repair (HR-DDR) and programmed death-ligand 1 (PD-L1) expression were additionally assessed and compared with the TMB and response rate. Results The median TMB between gene panels was similar despite a wide range in TMB values. The highest TMB was eight and 10 in patients with squamous cell carcinoma and esophageal carcinoma according to the classification of histopathology and cancer types, respectively. Twenty-three out of 103 patients (22.3%) were HR-DDR–positive and could benefit from ICI therapy; out of those 23 patients, seven patients had high TMB (p=0.004). Additionally, PD-L1 expression was not associated with TMB or treatment response among patients receiving ICIs. Conclusion Targeted NGS assays demonstrated the ability to evaluate TMB in pan-cancer samples as a tool to predict response to ICIs. In addition, TMB integrated with HR-DDR–positive status could be a significant biomarker for predicting ICI response in patients.

        Shaking Table Test on Seismic Responses of a Monolithic Precast Concrete Shear Wall Structure

        To examine the seismic performance of Monolithic Precast Concrete Shear Wall (MPCSW) structure, a 1/5-scaled twelve-story model is designed and investigated by shaking table test, and connections between the precast elements and their impact on seismic performance of the overall model are comprehensively inspected and analyzed. Based on the testing results, the structural responses are divided into elastic stage and plastic stage, and the variations of dynamic characteristics, cracking and failure pattern, seismic responses of the model are accordingly presented. The experimental results indicate that most of critical cracks of the MPCSW model locate on the coupling beams and horizontal connections of precast concrete shear walls, and the fundamental frequency exhibits a successively decreasing trend during the entire testing process. Furthermore, The MPCSW model is designed in accordance with Chinese codes meets the requirement for the seismic fortification intensity of eight, and shows an adequate safety margin to resist collapse in the case of 0.90 g.
