        Oxidative stress-induced circKIF18A downregulation impairs MCM7-mediated anti-senescence in intervertebral disc degeneration

        Low back pain, triggered by intervertebral disc degeneration (IVDD), is one of the most common causes of disability and financial expenditure worldwide. However, except for surgical interventions, effective medical treatment to prevent the progression of IVDD is lacking. This study aimed to investigate the effects of circKIF18A, a novel circRNA, on IVDD progression and to explore its underlying mechanism in IVDD. In this study, we found that oxidative stress was positively correlated with nucleus pulposus cell (NPC) senescence in IVDD and that circKIF18A was downregulated in IVDD and attenuated senescent phenotypes such as cell cycle arrest and extracellular matrix degradation in NPCs. Mechanistically, circKIF18A competitively suppressed ubiquitin-mediated proteasomal degradation of MCM7, and the protective effects of circKIF18A on NPCs were partially mediated by MCM7 under oxidative stress. Intradiscal injection of adenoviral circKIF18A ameliorated IVDD in a rat model. This study revealed that circKIF18A regulates NPC degeneration by stabilizing MCM7 and identified a novel signaling pathway, the circKIF18A-MCM7 axis, for anti-senescence molecular therapy in IVDD.

        Mathematical models for manufacturing a novel gear shaper cutter

        The design principle and models for novel error-free shaper cutters are discussed to improve the accuracy and service life of gear cutting tools. The modified methods for designing the tooth profile of the shaper cutter are developed by including the inverse envelope method, midpoint-midline method and midpoint-displacement method based on the theory of geometrical reverse problem. The universal mathematical models for manufacturing optimal tooth profile of shaper cutters are presented. The feasibility and reliability of the proposed principle, methods and models are proved by combining the numerical examples with practical application. The design method suggested in this paper is superior to the traditional design scheme, and will be helpful in facilitating the design and manufacture of shaper cutters.

        Barrier Optical Waveguide Fabrication in LiNbO3 Crystal by 4.5-MeV LithiumIon Implantation with Low Dose

        A barrier planar waveguide was fabricated in z-cut LiNbdO3 crystals by 4.5-MeV lithium ion implantation at a dose of 3 £ 1014 ions/cm2 at room temperature. Dark modes were observed by the prism-coupling method with wavelengths of both 633 nm and 1539 nm. The refractive index pro¯les were reconstructed by using the re°ectivity calculation method. There were about 1.1 % and 0.7 % decreases at the optical barriers of the ordinary and extraordinary refractive index at the wavelength at 633 nm, and the positions of the optical barriers were close to those of the damage peaks calculated by the TRIM098 (Transport of Ions in Matter) code. It is found that the refractive index change may be partly due to the damage induced by nuclear collision.

        The EGF/hnRNP Q1 axis is involved in tumorigenesis via the regulation of cell cycle-related genes

        Heterogeneous nuclear ribonucleoprotein (hnRNP) Q1, an RNA-binding protein, has been implicated in many posttranscriptional processes, including RNA metabolism and mRNA splicing and translation. However, the role of hnRNP Q1 in tumorigenesis remains unclear. We previously performed RNA immunoprecipitation (RIP)-seq analysis to identify hnRNP Q1-interacting mRNAs and found that hnRNP Q1 targets a group of genes that are involved in mitotic regulation, including Aurora-A. Here, we demonstrate that altering the hnRNP Q1 level influences the expression of the Aurora-A protein, but not its mRNA. Stimulation with epidermal growth factor (EGF) enhances both binding between hnRNP Q1 and Aurora-A mRNA as well as the efficacy of the hnRNP Q1-induced translation of Aurora-A mRNA. The EGF/hnRNP Q1-induced translation of Aurora-A mRNA is mediated by the mTOR and ERK pathways. In addition, we show that hnRNP Q1 up-regulates the translation of a group of spindle assembly checkpoint (SAC) genes. hnRNP Q1 overexpression is positively correlated with the levels of Aurora-A and the SAC genes in human colorectal cancer tissues. In summary, our data suggest that hnRNP Q1 plays an important role in regulating the expression of a group of cell cycle-related genes. Therefore, it may contribute to tumorigenesis by up-regulating the translation of these genes in colorectal cancer.

        Stable Microbial Community and Specific Beneficial Taxa Associated with Natural Healthy Banana Rhizosphere

        Banana planting altered microbial communities and induced the enrichment of Fusarium oxysporum in rhizosphere compared with that of forest soil. Diseased plant rhizosphere soil (WR) harbored increased pathogen abundance and showed distinct microbial structures from healthy plant rhizosphere soil (HR). The enriched taxon of Bordetella and key taxon of Chaetomium together with some other taxa showed negative associations with pathogen in HR, indicating their importance in pathogen inhibition. Furthermore, a more stable microbiota was observed in HR than in WR. Taken together, the lower pathogen abundance, specific beneficial microbial taxa and stable microbiota contributed to disease suppression.

        Crocin alleviates neurotoxicity induced by bupivacaine in SH-SY5Y cells with inhibition of PI3K/AKT signaling

        Background Bupivacaine, a common local anesthetic, can cause neurotoxicity and permanent neurological disorders. Crocin has been widely reported as a potential neuroprotective agent in neural injury models. Objective The aim of this study was to investigate the role and regulatory mechanism of crocin underlying bupivacaine-induced neurotoxicity. Method Human neuroblastoma SH-SY5Y cells were treated with bupivacaine and/or crocin for 24 h, followed by detecting cell viability using 3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide assay. The effect of crocin or bupivacaine on SH-SY5Y cell proliferation was measured by Ki67 immunofluorescence assay. The levels of apoptosis-related proteins and the markers in the PI3K/Akt signaling pathway were examined using western blot analysis. The activities of caspase 3, catalase (CAT), superoxide dismutase (SOD), malondialdehyde (MDA) and glutathione peroxidase (GSH-Px) were tested using respective commercial assay kits. Flow cytometry analysis was executed for detecting SH-SY5Y cell apoptosis. Result Crocin attenuated bupivacaine-induced neurotoxicity in SH-SY5Y cells. Meanwhile, crocin inhibited SH-SY5Y cell apoptosis induced by bupivacaine via repressing the activity of caspase-3, reducing Bax expression, and elevating Bcl-2 expression. Moreover, crocin mitigated oxidative stress in SH-SY5Y cells by increasing the content of CAT, SOD, GSH-Px and reducing the content of MDA. Additionally, crocin protected against bupivacaine-induced dephosphorylation of Akt and GSK-3β. The protective effects of crocin against bupivacaine-induced neurotoxicity in SH-SY5Y cells were counteracted by the Akt inhibitor. Conclusion These results suggested that crocin may exert a neuroprotective function by promoting cell proliferation and suppressing apoptosis and oxidative stress in SH-SY5Y cells. Thus, crocin might become a promising drug for the treatment of bupivacaine-induced neurotoxicity.       • Cervical Cancer Screening and Analysis of Potential Risk Factors in 43,567 Women in Zhongshan, China

        Objective: The objective of this study was to establish a program model for use in wide-spread cervical cancer screening. :Methods: Cervical cancer screening was conducted in Zhongshan city in Guangdong province, China through a coordinated network of multiple institutes and hospitals. A total of 43,567 women, 35 to 59 years of age, were screened during regular gynecological examinations using the liquid-based ThinPrep cytology test (TCT). Patients who tested positive were recalled for further treatment. Results: The TCT-positive rate was 3.17%, and 63.4% of these patients returned for follow-up. Pathology results were positive for 30.5% of the recalled women. Women who were younger than 50 years of age, urban dwelling, low-income, had a history of cervical disease, began having sex before 20 years of age, or had sex during menstruation, were at elevated risk for a positive TCT test. The recall rate was lower in women older than 50 years of age, urban dwelling, poorly educated, and who began having sex early. Ahigher recall rate was found in women 35 years of age and younger, urban dwelling, women who first had sex after 24 years of age, and women who had sex during menstruation. The positive pathology rate was higher in urban women 50 years of age and younger and women who tested positive for human papillomavirus. Conclusion: An effective model for large-scale cervical cancer screening was successfully established. These results suggest that improvements are needed in basic education regarding cervical cancer screening for young and poorly educated women. Improved outreach for follow-up is also necessary to effectively control cervical cancer.

        Angiotensin I Converting Enzyme (ACE) Inhibitory Activity and Antihypertensive Effects of Grass Carp Peptides

        In vitro and in vivo antihypertensive effects ofgrass carp peptides (GCP) were investigated. The aminoacid composition and the angiotensinconverting enzymeinhibitory activity were evaluated using HPLC. GCP wasadministrated to spontaneously hypertensive rats (SHRs)and Wistar Kyoto rats (WKY) by single and long-termadministration before blood pressure measurements. Plasma levels of angiotensin II (Ang II), rennin (RA), nitricoxide (NO), and plasma angiotensin I converting enzyme(ACE) activity were measured. The GCP molecular weightwas between 725 and 1,228 Da with high levels of Leu,Asp, Phe, Gly, and Pro. The in vitro IC50 value was 0.23mg/mL. GCP lowered SHR blood pressure dose and timedependently. Little change occurred in WKY. The plasmalevel of Ang II and the ACE activity decreased. SHR RAand NO concentrations in plasma increased. Theantihypertensive effect of GCP was associated with NOregulation and the rennin-angiotensin system.

        Effects of pores and temperature on mechanics of gold nanowires using molecular dynamics

        The deformation mechanisms of gold nanowires with different nanopores under tension were simulated by molecular dynamics (MD). The stress–strain curves varied from different porous defects, and the tension caused dislocations to take place and slip along plane (1 1 1). Moreover, the tensile strength of the nanoporous monocrystalline gold was decreased when the simulated temperature increased. The stress concentrations factors of porous nanowires were calculated, and it was found that there was a great influence of size and model effects on the stress concentration factors.

      • The impact of total retrieved lymph nodes on staging and survival of patients with pT3 gastric cancer

        <B>BACKGROUND.</B><P>The incidence of lymph node metastasis is high in patients who have pT3 gastric cancer. However, the impact of total retrieved lymph nodes (tLNs) on staging and survival of these patients is not clear.</P><B>METHODS.</B><P>For this study, the authors examined 1895 patients with pT3 gastric cancer who underwent surgery at Yonsei University Medical College from January 1987 to June 2000.</P><B>RESULTS.</B><P>Four hundred sixty of 1895 patients (24.3%) were diagnosed with pT3N0 gastric cancer. Patients who had < 31 tLNs (25th percentile) had less advanced lymph node (N) stage than the other patients (P < .001). Lymph node metastasis had a positive association with the number of tLNs in a logistic regression analysis (P < .001; hazards ratio, 1.014; 95% confidence interval, 1.006–1.021). With a median follow-up of 61.1 months, the overall 10-year survival rate (10-YSR) was 42.8%. Patients with pT3N0 disease who had < 31 tLNs had a 10-YSR of only 55.4%. Although this 10-YSR did not differ significantly from the rate for patients with N0 disease who had ≥31 tLNs (65.8%; P = .108), it approached the rate for the N1 group (53.3%; P = .207). In multivariable analyses, the number of tLNs emerged as an independent prognostic predictor in patients with pT3N2 and pT3N3 disease, but not in patients with pT3N0 or pT3N1 disease.</P><B>CONCLUSIONS.</B><P>Increasing numbers of tLNs may improve the accuracy of staging in patients who have pT3 gastric cancer. Because preoperative lymph node staging is difficult, a thorough lymph node dissection is mandatory in all serosa-positive patients. Cancer 2007. © 2007 American Cancer Society.</P>
