      • 인체 대장암 및 자궁경부암에서 PCR-SSCP법을 이용한 Ki-ras 암유전자의 점돌연변이에 관한 연구

        인체 대장암 20례와 자궁경부암 10례의 암조직을 대상으로 c-Ki-ras 유전자 codon 12와 13에서의 점돌연변이를 알아보고자 polymerase chain reaction(PCR)과 single-strand conformation polymorphism(SSCP) 검사법을 시행하였다. 대장암의 경우 20례중 9례(45%)에서, 자궁경부암의 경우 10례중 1례(10%)에서 양성으로 나왔다. 대장암에서는 codon 12에서 GGT가 TGT로의 치환이 4례로서 가장 많았으며, AGT, CGT로의 치환이 각 1례였고 codon 13에서는 GGC가 GAC로의 치환이 2례, TGC로의 치환이 1례인 것으로 나타났다. 자궁경부암의 경우 codon 12에서 GGT가 AGT로의 치환이 1례인 것으로 나타났다. In an attempt to clarify the role of genetic alteration in the genesis of human colorectal and cervical cancers, tissue specimens from 20 patients with colorectal cancer and 10 patients with cervical cancer were examined for the presence of point mutation in K-ras2 exon 1 by single strand conformation polymorphism analysis of PCR product. Exon I of c-Ki-ras2 was amplified by polymerase chain reaction(PCR) and comparison was made between the normal and mutated genes by nondenaturing polyacrylamide gel electrophoresis(PAGE) of PCR product and nucleotide sequence analysis using asymmetric PCR with direct sequencing. Genomic DNA from white blood cells were used as normal control and those from A427 cell line were used as mutated control. 1.PCR product from A427 cell line showed a distinct migration shift pattern compared to the normal control in PAGE and the direct sequencing indicated that nucleotide sequence of codon 12 was mutated from GGT to GAT. In addition, there was a loss of normal allele in A427 cell line. 2.PCR product from 9 cases(45%) out of a 20 colon cancer patients showed migration shifts in PAGE, and all of these 9 patients invariably demonstrated mutation of ras gene, either in codon 12(6 cases:30%) or 13(3 cases:15%). The base substitutions in codon 12 were: from GGT to AGT(1), CGT(1), or TGT(4). Base changes in codon 13 were from GGC to TGC(1) or GAC(2). 3.PCR product from 1 cases(10%) out of a 10 cervical cancer patients showed migration shrifts in PAGE, Invariably demonstrated mutation of ras gene in codon 12. The base substitutions was from GGT to AGT(1) TQE incidence of point mutation of c-Ki-ras in colon cancer was high, however, it was low in cervical cancer, which showed the relation of ruts gene mutation with colon cancer. PCR -SSCP analysis is a simple, rapid and efficient method of detection of point mutation, especially when dealing with multiple samples.

      • 폐석재와 석분을 사용한 식생블럭 제품 개발

        본 연구는 폐석재와 석분을 사용하여 친환경적인 식생블럭 제품을 개발하기 위하여 폐석재의 입경과 석분의 첨가량을 각각 3종류씩 총 9종류의 식생블럭에 대하여 단위중량, 공극율, 투수량, 압축강도, 쪼갬인장강도, 휨강도, 내산성, 알카리농도에 대하여 구명하였다. 단위중량, 강도, 내산성은 골재의 입경이 작을수록 석분의 혼입량이 많을수록 크게 나타났으며, 공극율과 투수량은 작게 나타났다. 강도는 중성화 처리 유무에 따라 4∼5% 정도의 미소한 차이가 나타났다. 알카리 농도는 중성화 약품처리, 수중용출, 야외폭로 순으로 효과가 크게 나타났으며, 이는 식생블럭의 중성화 방법 선택에 매우 중요한 내용이라 할 수 있다. 또한, 식생상태는 공극율과 골재의 입경이 큰 식생블럭이 생육에 유리한 것으로 나타났다. This study was performed to develop the environment-friendly plantable block and to evaluate unit weight, void ratio, permeability, strength, acid-resistance and value of pH by the change of the grading of waste stone and the addition amount of stone dust. The unit weight, strength, acid-resistance is increased with small size of waste stone with increase of the addition amount of stone dust. But, the void ratio and permeability is decreased. The strength of neutral plantable block is decreased approximately 4-5% than that of the normal plantable block. The decreased effect of pH increased in order to neutral treatment, underwater leaching and field exposure. It is very important contents in neutral method selection of plantable block. Also, the planting state is grown rapidly with increase of the void ratio and of the waste stone size, approximately.

      • 후석 승객용 ROOF AIRBAG SYSTEM 개발

        The purpose of this study is the development of a Roof Airbag System for protection of rear seat passenger from frontal impacts. First the development concept has been carried out pre-prototype stage sled tests to confirm the propriety of a roof-mounted airbag system. To predict occupant kinematic and injury potential in various conditions, the occupant analysis by using the Madymo-3D has been performed and the mandatory test matrix has been determined. As the result of sound power and pressure tests, we confirmed the concurrent deployment of several airbags is harmless to human's hearing organs. To evaluate the final design specification of this system, sled tests by using improved samples have been carried out.

        Aspergillus niger SFN-416으로부터 생산한 Xylanase 2의 분리정제 및 특성

        Aspergillus niger SFN-416으로부터 생성된 xylanase Ⅱ를 분리·정제하여 특성을 조사하였다. Aspergillus niger SFN-416의 배양액을 ethanol(70%) 침전, (NH_4)_2SO_4(30~90%) 침전, Sephadex G-100 chromatography 및 DEAE-Sephacel chromatography 등의 정제과정을 거친 결과, specific activity가 38.2 units/ mg protein으로 4.3배 정제되었고, 정제효소의 최적 활성온도는 50℃였다. 최적 pH는 5.5이었고, pH 4.5~6.0 사이에서 비교적 안정하였다. 또한 금속이온에 대한 효소의 활성은 대부분 억제를 보였고, 특히 Hg^2+에서는 3.7%로 강하게 효소활성이 저해됨을 보였고, Fe^2+에서는 100%의 상대활성을 나타내었다. 정제 효소의 분자량은 SDS-PAGE에 의하여 42,000 daltons이었으며, 유기용매에 대한 활성은 10%의 methanol, ethanol, isopropanol 및 1-butanol에 대하여 모두 낮은 활성을 나타었다. Xylanase (EC 3.2.1.8) was purified approximately 4,3 fold from Aspergillus niger SFN-416 by ammonium sulfate fractionation, Sephadex G-100 gel filtration and DEAE-Sephacel ion exchange chromatography. Molecular weight of the enzyme was approximately 42,000 daltons. The optimum pH and temperature of the enzyme activity were 5.5 and 50℃, respectively. The enzyme activity was enhanced by Fe^2+, and inhibited by Hg^2+. The activity was decreased by addition of methanol, ethanol, isopropanol and 1-butanol at a concentration of 10%(v/v).

      • 산포도와 머루중의 Polyphenol Oxidase에 관한 연구¹

        Polyphenol oxidase was partially purified from fresh vitis flexuosa and vitis amurensis and its characteristics were studied. the experimental results were summarized as follows. 1.The enzyme activities were increased during the fruit developement gradually in two sample. 2.The optimum pH and temperature for activity were 5.5 and 30℃, respectively. 3.The enzyme was active with o-phenols and trihydroxyphenol, but inactive toward m-diphenols and monophenols. 4.The ?? value of the enzymes was 15.4 mM in vitis flexuosa and 12.5 mM in vitis amurensis with catechol as substrate. 5.Inhibitors sudies indicated that sodium diethyl-dithiocarbamate, L-cysteine, sodium bisulfite, L-ascorbic acid and 2-mercaptoeyhanol were the most potent. 6.The enzyme activity increament by ?? was much better than those by the other metal ions.

      • 초고층 아파트의 일조 일조환경 분석에 관한 연구

        As the modern city grows larger and larger, overpopulated and over-storied apartments caused violation of its sunlight, view and privacy that became social problems. Especially when after the tower-type block have shown up and the formal type of apartment which is plate-type block had been disappearing ever since. Because of this phenomenon, higher and overcrowded apartment became very common and it even made an apartment which made all the block as tower-type block. As the time goes by, it is very easy to see one person pressing charges against another for his or her right to sunshine nowadays. In this study, we will use Auto-CAD to analyze the sunlight environment of tower-type block apartment for its expected harmful element of sunlight. And we will use this data to present basic data of how sunlight environments effect on buildings when it is being designed as apartment.

      • 사람 정자의 성숙도와 운동성에 따른 세포질내 정자주입 후 전핵형성과 초기 배발생

        In the present study, we investigated the effect of maturity and motility of spermatozoa on the formation of pronucleus and subsequent developmental capacity of the human embryo in vitro. The fertilization was performed by means of intracytoplasmic sperm injection (ICSI) in HEPES-buffered m-TCM-199 medium. In the first part of the experiment, motile or immotile human spermatozoa ejaculated were injected into cumulus-enclosed human oocytes matured in vivo. Significantly(P$lt;0.002)higher proportion of oocytes that was injected with motile spermatozoa formed 2 pronuclei than the oocytes injected with immotile spermatozoa(79.8% vs 51.7%). In the second part of the experiment, cumulus-enclosed human oocytes matured in vivo were injected with motile or immotile spermatozoa collected from testes. There was no difference between motile and immotile spermatozoa. In the third part of the experiment, using modified Tyrode's medium containing 10.0 mM lactate, 0.5 mM pyruvate, 0.2 mM taurine, 1.0 mM glutamine, 2.22 mM MEM amino acids, vitamin and 10% human follicular fluid, we found that development of oocytes that formed 2 pronuclei were able to develop to 9-16 cells regardless of maturity and motility of spermatozoa.

      • 난연성 FREP로 휨보강된 철근콘크리트 보의 구조성능 평가

        The purpose of this study is to evaluate the flexural strengthening performance of the iFREP(incombustible Fiber Reinforced Epoxy Panel) to the reinforced concrete flexural members. The tensile strength of iFREP including mechanical and physical properties of the epoxy resin for injection were tested preliminary to investigate the material properties of iFREP constitutions. Also the bond characteristics of iFREP to the concrete were estimated to acquire the basic design data for strengthening through various bonding tests. Finally full-scale RC beams strengthened by iFREP were tested monotonically upto failure to evaluate the strengthening effects of iFREP to the RC flextural members. The main variable of the four test specimens was the thickness of iFREP. From the test results, it was found that the failure mode of RC beams strengthened by iFREP was governed by the fracture of iFREP indicating the sufficient bond strength of iFREP to the concrete. Also flexural strengthening effects of iFREP to the RC beams is proportional to the thickness of iFREP.

      • 골프스윙시 초보자와 숙련자의 근전도에 관한 비교연구

        Based on the analysis of the EMG(Electromyograpy) level of 3 lesson professional-golf players and 3 beginners under one year career performing drive and iron, pitching swing, this study in EMG analysis between lesson professional golf players and beginners, in club, and EMG contrast left to right muscle at the point of impact, using EMG machine has come to the following conclusion. 1. The active muscle involved at the point of impact turned out to reveal slight difference depending on clubs; yet the EMG of the upper body was noticeable in the left-right triceps brachii - the left biceps brachii - the left trapezius - the right petoralis muscle in sequence while that of the lower body was prominent in the left lectus femoris - the right gastrocnemius muscle. 2. When it comes to the EMG difference in clubs, drive, iron 7 and pitching, the experts' inbetween-club EMG of the left triceps brachii and the left biceps brachii muscle was marked by a statistic significance, novices showing a significance in the left biceps brachii muscle only. 3. To speak of experts only, the right-left EMG difference of the muscle involved in swinging was significant in terms of statistics ; while the involvement of left part was conspicuous in the trapezius muscle, the biceps brachii muscle and the rectus femoris muscle, the gastrocnemius's right part proved to be highly active. 4. Speaking of novices only, the statistical EMG difference of right-left muscle part involved in swinging was significant; the EMG of left part turned out highly significant in the trapezius and the rectus femoris muscle while the gastrocnemius' right part proved highly significant. 5. The EMG difference between experts and novices was remarkable in the triceps brachii and the pectoralis muscle ; the former came up with a high EMG in terms of right side muscle while the latter revealed rather a high EMG in terms of left side muscle.
