In this study, we investigate the bacterial diversity and quality of madetraditionally made doenjang (TMD). The quality of TMD highly depends on the bacterial composition, which ambient bacteria in the environment and production conditions influence, a complete understanding of the bacteria community from different regions is needed. Twenty-nine TMD samples were collected from different regions and its biogenic amine content, bacteria composition, and metabolic function were analyzed. This result suggests that most TMD contained beneficial bacteria and it’s no similarity among the regional groups. Functional profile results showed that histamine contents were lower in CN and CC, and tyramine contents did not differ significantly. In conclusion, TMD mainly contained various Bacillus spp., and the predominant one was B. velezensis, which had antioxidant and fibrinolytic activity regardless of the regional origin. <▼1>
Hypoxia-inducible factor-2α (HIF-2α) is sufficient to cause experimental rheumatoid arthritis and acts to regulate the functions of fibroblast-like cells from tissue surrounding joints, independent of HIF-1α.
</▼1><▼2>
Rheumatoid arthritis (RA) is a systemic autoimmune disorder that manifests as chronic inflammation and joint tissue destruction. However, the etiology and pathogenesis of RA have not been fully elucidated. Here, we explored the role of the hypoxia-inducible factors (HIFs), HIF-1α (encoded by HIF1A) and HIF-2α (encoded by EPAS1). HIF-2α was markedly up-regulated in the intimal lining of RA synovium, whereas HIF-1α was detected in a few cells in the sublining and deep layer of RA synovium. Overexpression of HIF-2α in joint tissues caused an RA-like phenotype, whereas HIF-1α did not affect joint architecture. Moreover, a HIF-2α deficiency in mice blunted the development of experimental RA. HIF-2α was expressed mainly in fibroblast-like synoviocytes (FLS) of RA synovium and regulated their proliferation, expression of RANKL (receptor activator of nuclear factor–κB ligand) and various catabolic factors, and osteoclastogenic potential. Moreover, HIF-2α–dependent up-regulation of interleukin (IL)-6 in FLS stimulated differentiation of TH17 cells—crucial effectors of RA pathogenesis. Additionally, in the absence of IL-6 (Il6−/− mice), overexpression of HIF-2α in joint tissues did not cause an RA phenotype. Thus, our results collectively suggest that HIF-2α plays a pivotal role in the pathogenesis of RA by regulating FLS functions, independent of HIF-1α.
</▼2><▼3>
Author Summary

Rheumatoid arthritis (RA) is a systemic autoimmune disorder characterized by chronic inflammation in joint tissues leading to destruction of cartilage and bone. Despite some therapeutic advances, the etiology of RA pathogenesis is not yet clear, and effective treatment of RA remains a significant, unmet medical need. Hypoxia is a prominent feature of inflamed tissue within RA-affected joints, and earlier work has implicated limited involvement of hypoxia-inducible factor (HIF)-1 α. We explored the role of a second HIF family member, HIF-2α, in RA pathogenesis. We showed that HIF-2α is markedly increased in the tissue lining the RA-affected joints. Notably and in contrast to HIF-1α, when overexpressed in normal mouse joint tissues, HIF-2α is sufficient to cause RA-like symptoms. Conversely, an HIF-2α deficiency blocks the development of experimental arthritis in mice. We discovered further that HIF-2α regulates RA pathogenesis by modulating various RA-associated functions of joint-specific fibroblast-like cells, including proliferation, expression of cytokines, chemokines, and matrix-degrading enzymes, and bone-remodeling potential. HIF-2α also increases the ability of these cells to promote interleukin-6–dependent differentiation of TH17 cells, a known effector of RA pathogenesis. We thus show that HIF-1α and HIF-2α have distinct roles and act via different mechanisms in RA pathogenesis.
</▼3>

Primary lymphoma of the urinary bladder is rare, comprising 0.2% of extranodal lymphomas. The predominant subtype of mucosa-associated lymphoid tissue (MALT) is extranodal marginal zone lymphoma. We report a case of MALT lymphoma of the urinary bladder in a 53-year-old female patient presenting with a five-year history of persistent hematuria and urinary frequency. A cystoscopy revealed multiple nodular lesions at the posterior wall and trigone of the bladder. The tissue obtained by cold-cup biopsy revealed lymphoid infiltration consistent with low-grade MALT lymphoma. Image studies revealed that the tumor originated from the urinary bladder and there was no evidence of metastases. The patient was positive for a rapid urease test. Because of the relationship between gastric MALT lymphoma and Helicobacter pylori, the patient was treated with antibiotics to eradicate Helicobacter pylori. The lymphoma subsequently disappeared and the patient has remained in persistent complete remission for eight years. (Korean J Med 2014;87:229-233)

A series of N-(2-amino-6-trifluoromethylpyridin-3-ylmethyl)-2-(3-fluoro-4-methylsulfonylaminophenyl)propanamides were designed combining previously identified pharmacophoric elements and evaluated as hTRPV1 antagonists. The SAR analysis indicated that specific hydrophobic interactions of the 2-amino substituents in the C-region of the ligand were critical for high hTRPV1 binding potency. In particular, compound 49S was an excellent TRPV1 antagonist (Ki(CAP) = 0.2 nM; IC50(pH) = 6.3 nM) and was thus approximately 100- and 20-fold more potent, respectively, than the parent compounds 2 and 3 for capsaicin antagonism. Furthermore, it demonstrated strong analgesic activity in the rat neuropathic model superior to 2 with almost no side effects. Compound 49S antagonized capsaicin induced hypothermia in mice but showed TRPV1-related hyperthermia. The basis for the high potency of 49S compared to 2 is suggested by docking analysis with our hTRPV1 homology model in which the 4-methylpiperidinyl group in the C-region of 49S made additional hydrophobic interactions with the hydrophobic region.

Graphic Abstract

ACS Electronic Supporting Info

Electrochemical Corrosion of STS304 Acupuncture Needles by Electrical Stimulation
황혜숙,Myeong Soo Lee,Yeon Hee Ryu,Eun Jin Yang,Sun-Mi Choi 사단법인약침학회 2010 Journal of Acupuncture & Meridian Studies Vol.3 No.2
We present the first investigation of electrical corrosion in acupuncture needles after electrical stimulation. Using scanning electron microscopy, we observed the occurrence of electrochemical corrosion on the surface of stainless steel 304 acupuncture needles after electrical stimulation in the tibial muscles of rats. Biphasic pulse electrical stimuli with 10-Hz frequency, 1-mA intensity and 1-ms pulse width were applied to the needles. The terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling (TUNEL) method labels fragmented DNA. Positive staining using this test indicates apoptotic cells in electrically stimulated tissues. The risk of electrical corrosion was found to be less in bipolar, short-duration, lowcurrent or voltage and short-period stimulation than in monopolar, long-duration,high-current or voltage and long-period stimulation. Evaluation with a scanning electron microscope revealed that electrical stimulation can increase the electrical corrosion of stainless steel 304 acupuncture needles. In biocompatibility studies of stainless steel 304 acupuncture needles for electrical stimulation, TUNEL-positive cells were detected in the tibial muscle within 5 days after electrical stimulation. The results of this study demonstrate that the corrosion products of stainless steel 304 acupuncture needles might affect the post-electrical stimulation tissue response.

AIMS: S-1, an oral fluoropyrimidine, contains tegafur, which is converted to 5-fluorouracil mainly by CYP2A6. We evaluated the association between CYP2A6 polymorphisms and treatment outcome in metastatic gastric cancer patients treated with S-1 plus docetaxel. MATERIALS & METHODS: Chemonaive patients received S-1 40 mg/m(2) twice daily on days 1-14 and docetaxel 35 mg/m(2) on days 1 and 8 of a 3-week cycle. We analyzed the wild-type (W) allele (CYP2A6*1) and four variant (V) alleles that abolish or reduce enzyme activity (CYP2A6*4, *7, *9 and *10). A total of 50 patients were enrolled. RESULTS: The genotype frequencies were as follows: W/W (n=14, 28%), W/V (n=26, 52%) and V/V (n=0, 20%). Patients having fewer variant alleles had significantly better response rates (W/W vs W/V vs V/V=79 vs 65 vs 30%; p=0.04) and median progression-free survival (W/W vs W/V vs V/V=8.1 vs 6.9 vs 3.1 months; p=0.0009). CONCLUSION: Our findings showed that the CYP2A6 genotype correlated with the treatment efficacy of S-1-based chemotherapy in previously untreated metastatic gastric cancer patients.
