The methanol extracts of flowers obtained from 67 plant species were screened for their inhibitory activity on aldose reductase(AR). Alnus japonica , Aster spathulifolius , Chionanthus retusus , Morus bombycis , Crysanthemum boreale , Persicariatinctoria , Platycarya strobilacea , and Serratula coronata var. insularis exhibited potent aldose reductase inhibitory (ARI)activity. HPLC-UV analysis of quercetin, an AR inhibitory flavonoid, was performed on extracts showing strong ARI activity. Quercetin was detected in C. retusus , C. boreale , P. tinctoria , and S. coronata var. insularis at concentrations of 1.33,1.56, 0.82, and 3.37 mg g −1 extract, respectively, indicating that quercetin contributed to the ARI activity of these extracts. In the samples in which quercetin was absent, other compounds may be responsible for their potent ARI activity. Theseresults serve as a basis for further studies regarding the bioactive components responsible for the inhibitory eff ects of variousfl ower extracts on AR activity.
Closed loop cable robot for large horizontal workspaces

Inspection and maintenance of civil structures are important issues for sustainability of existing and new infrastructures. Classical approach relies on large human activities eventually performed in unsafe conditions. This paper proposed a non-invasive solution for inspecting horizontal surface such as decks of bridges. The proposal presented here is based in cable-driven robots and allows to inspect large surfaces maintaining a very low vertical occupancy in comparison to the conventional architecture of this kind of robot. Using closed cables loop instead of a set of cables a device with low motorization power and very large workspace is designed and prototyped. As example of control an inverse dynamics technique is applied to control the end-effector where inspection tool is located, e.g., a vision system. Experimental results demonstrate that this novel device allows to inspect large horizontal surfaces, with low motorization and low vertical occupancy.

The production of human interferon gamma (hIFN-γ) using a synthetic gene in Escherichia coli BL21-SI was optimized by response surface methodology (RSM) and a Box-Behnken design. The process variables studied were temperature, biomass concentration at induction time and the NaCl concentration as inducer. According to the Box-Behnken design, a second order response function was developed. The optimal expression conditions were a temperature of 32.6℃, induction biomass of 0.31 g/L and 0.3 M NaCl in minimal medium. The model prediction for the maximum hIFN-γ production was 77.3 mg/L, which corresponded satisfactorily with the experimental data. The hIFN-γ concentration attained under optimized conditions was 13-times higher than that obtained using the non-optimized conditions. We conclude that RSM is an effective method for the optimization of recombinant protein expression using synthetic genes in E. coli.

<P>Purpose: We determined the effect of testosterone solution 2% on total testosterone level and the 2 symptoms of hypogonadism, sex drive and energy level. Materials and Methods: This was a randomized, multicenter, double-blind, placebo controlled, 16-week study to compare the effect of testosterone and placebo on the proportion of men with a testosterone level within the normal range (300 to 1,050 ng/dl) upon treatment completion. We also assessed the impact of testosterone on sex drive and energy level measured using SAID (Sexual Arousal, Interest and Drive scale) and HED (Hypogonadism Energy Diary), respectively. A total of 715 males 18 years old or older with total testosterone less than 300 ng/dl and at least 1 symptom of testosterone deficiency (decreased energy and/or decreased sexual drive) were randomized to 60 mg topical testosterone solution 2% or placebo once daily. Results: Of study completers 73% in the testosterone vs 15% in the placebo group had a testosterone level within the normal range at study end point (p < 0.001). Participants assigned to testosterone showed greater baseline to end point improvement in SAID scores (low sex drive subset p < 0.001 vs placebo) and HED scores (low energy subset p = 0.02 vs placebo, not significant at prespecified p < 0.01). No major adverse cardiovascular or venous thrombotic events were reported in the testosterone group. The incidence of increased hematocrit was higher with testosterone vs placebo (p = 0.04). Conclusions: Once daily testosterone solution 2% for 12 weeks was efficacious in restoring normal testosterone levels and improving sexual drive in hypogonadal men. Improvement was also seen in energy levels on HED though not at the prespecified p < 0.01. No new safety signals were identified.</P>

<P><B>Abstract</B></P><P>The synthesis and characterization of new ruthenium(II) carbonyl complexes containing polypyridine and triphenylphosphine ligands is reported. Crystallographic information obtained for the <I>trans-</I>PPh<SUB>3</SUB>-[Ru(biq)(PPh<SUB>3</SUB>)<SUB>2</SUB>(CO)]Cl<SUB>2</SUB> complex (biq=2,2’-biquinoline<B>)</B> reveals five-coordination on the metal. The complexes were studied as catalysts in hydrogen transfer reactions in basic solution. <P>Recently, we identified in normally type 1 diabetes-prone NOD/LtJ mice a spontaneous new leptin receptor (LEPR) mutation (designated Lepr(db-5J)) producing juvenile obesity, hyperglycemia, hyperinsulinemia, and hyperleptinemia. This early type 2 diabetes syndrome suppressed intra-islet insulitis and permitted spontaneous diabetes remission. No significant differences in plasma corticosterone, splenic CD4(+) or CD8(+) T-cell percentages, or functions of CD3(+) T-cells in vitro distinguished NOD wild-type from mutant mice. Yet splenocytes from hyperglycemic mutant donors failed to transfer type 1 diabetes into NOD.Rag1(-/-) recipients over a 13-week period, whereas wild-type donor cells did so. This correlated with significantly reduced (P < 0.01) frequencies of insulin and islet-specific glucose-6-phosphatase catalytic subunit-related protein-reactive CD8(+) T-effector clonotypes in mutant mice. Intra-islet insulitis was also significantly suppressed in lethally irradiated NOD-Lepr(db-5J)/Lt recipients reconstituted with wild-type bone marrow (P < 0.001). In contrast, type 1 diabetes eventually developed when mutant marrow was transplanted into irradiated wild-type recipients. Mitogen-induced T-cell blastogenesis was significantly suppressed when splenic T-cells from both NOD/Lt and NOD-Lepr(db-5J)/Lt donors were incubated with irradiated mutant peritoneal exudate cells (P < 0.005). In conclusion, metabolic disturbances elicited by a type 2 diabetes syndrome (insulin and/or leptin resistance, but not hypercorticism) appear to suppress type 1 diabetes development in NOD-Lepr(db-5J)/Lt by inhibiting activation of T-effector cells.</P>
Determination of Silybin B in the Different Parts of Silybum marianum using HPLC-UV

Silymarin is the standardized extract from Silybum marianum which consists mainly of flavonoids and polyphenols. It is highly regarded for its hepatoprotective ability. Silybin B is a flavonolignan and one of the active components of silymarin. The content of silybin B in various parts of S. marianum was analyzed by HPLC-UV. Results show that the extract of seeds contain the highest amount of silybin B (7.434 mg/g DW). The petioles of S. marianum showed a low content of silybin B. This study revealed that seeds of S. marianum contain high amount of silybin B and could be a good source of the compound.

Simultaneous determination of three methoxyflavones, namely, cirsimarin (1), hispidulin (2), and cirsimaritin (3) in selected Korean thistles was performed via reversed-phase high performance liquid chromatography system. Compound 1 was present in all the thistle species examined, whereas 2 and 3 were only detected in Cirsium japonicum and C. japonicum var. maackii (CJM). The concentration of compounds 1-3 in CJM varied according to the time of harvest. Plants collected in the spring (CJMS) and fall (CJMF) had the highest contents of 3 and 1, respectively. A lower content of 2 was observed in CJMF than in CJMS. This indicates that seasonal variation affects the flavonoid content of CJM. The results of this study show that CJM is an excellent source of compounds 1-3 and it can potentially be cultivated for industrial and pharmaceutical applications involving these compounds.

Perilla frutescens var. japonica (PF) is an annual aromatic herb has been consumed as a food ingredient and medicinal crop in Asian countries. To evaluate the therapeutic efficacy of aldose reductase (AR) inhibition, we tested the PF seeds. The stepwise polarities of PF were tested for AR inhibition, and we determined the CH2Cl2 and EtOAc fractions to be good inhibitors (5.81 and 3.99 lg/mL, respectively). Compounds 1–3 were isolated from the CH2Cl2 and EtOAc fractions and identified as luteolin (1), apigenin (2), and diosmetin (3) by physicochemical and spectroscopic data. Among them, luteolin (1) and apigenin (2) had high AR inhibitory activity (1.89 and 4.18 lM). Deulsaem, a variety of PF, was determined to have the highest flavonoid content among ten PF seeds tested (2.10 mg/g). This study suggests that PF could be utilized as a natural source to treat diabetic complications.
