Aldose reductase inhibition of Rosa hybrida petals and its active component, kaempferol

The aldose reductase inhibitory activity of extracts from Rosa hybrida petals was evaluated, and a bioactive fl avonoid,kaempferol, was isolated from petals via bioassay-guided isolation. Kaempferol was isolated from the ethyl acetate fractionof R. hybrida and was shown to exhibit strong inhibition against aldose reductase (IC 50 = 0.02 μM). Quantitative analyses ofkaempferol in R. hybrida petals, diff erent fl ower parts, and diff erent Rosa species were also performed using high-performanceliquid chromatography. This study is the fi rst to establish that R. hybrida contains kaempferol and that the light-pinkpetals of this edible rose species exhibit the highest content of the naturally occurring aldose reductase inhibitor. The resultsof this study suggested the potential role of R. hybrida petals in the treatment and management of diabetic complicationsvia aldose reductase inhibition.
Determination of flavonoids in Acer okamotoanum and their aldose reductase inhibitory activities

Plants in the genus Acer include medicinal plants and deciduous tree species in which several bioactive compounds have been isolated. In this study, flavonoids were isolated from the aerial parts of Acer okamotoanum by open column chromatography and identified by spectroscopic analysis as afzelin, quercitrin, and isoquercitrin. Among these flavonoids, quercitrin and isoquercitrin exhibited potent aldose reductase inhibitory activity (IC50 value of 0.40 and 0.63 μM, respectively). Almost all studied tissues of Acer spp. contained high amounts of quercitrin. The leaf of A. mono had the highest amount of quercitrin (61.90 μg g−1). Our results demonstrated that Acer species contain high concentrations of flavonoids with promising aldose reductase inhibitory activity that could be utilized to develop novel therapeutics for diabetic disorders.
Analysis of terpenoids and steroids in the seeds of Panax ginseng

Phytosterols have many benefi ts for human health. Phytosterols are abundant in many plant materials. We determined theamounts of lupeol, stigmasterol, β-sitosterol, and squalene in seeds of Panax ginseng by high-performance liquid chromatographicanalysis. We assayed the contents of these phytosterols in seeds from plants of diff erent ages, seeds from diff erentvarieties, and seeds cultivated from diff erent countries. The contents of lupeol, stigmasterol, β-sitosterol, and squalene rangedfrom 14.15–38.88, 21.33–31.12, 0.00–95.41, to 0.01–0.31 mg g −1 dry weight (DW), respectively. The highest content ofterpenoids and steroids was found in 3-year-old seeds (29.06 mg g −1 DW), seeds of the variety Chunpoong (25.20 mg g −1DW), and seeds from China (34.17 mg g −1 DW). This result will be useful in the application of these terpenoids and steroidsfrom seeds of P. ginseng in the development of nutraceutical, pharmaceutical, and cosmeceutical materials.

Perilla frutescens var. japonica (PF) is an annual aromatic herb has been consumed as a food ingredient and medicinal crop in Asian countries. To evaluate the therapeutic efficacy of aldose reductase (AR) inhibition, we tested the PF seeds. The stepwise polarities of PF were tested for AR inhibition, and we determined the CH2Cl2 and EtOAc fractions to be good inhibitors (5.81 and 3.99 lg/mL, respectively). Compounds 1–3 were isolated from the CH2Cl2 and EtOAc fractions and identified as luteolin (1), apigenin (2), and diosmetin (3) by physicochemical and spectroscopic data. Among them, luteolin (1) and apigenin (2) had high AR inhibitory activity (1.89 and 4.18 lM). Deulsaem, a variety of PF, was determined to have the highest flavonoid content among ten PF seeds tested (2.10 mg/g). This study suggests that PF could be utilized as a natural source to treat diabetic complications.
Validation of an optimized HPLC/UV method for the quantification of flavonoids in lotus

Flavonoids present in the leaves of lotus (Nelumbo nucifera) grown in diferent regions of South Korea (Yeongcheon, Haenam, and Seocheon) and at diferent harvest times (July to September) were determined. Flavonoid contents in lotus extracts were identifed and analyzed using high-performance liquid chromatography (HPLC). The HPLC results revealed that the favonoid contents of the lotus extracts varied at diferent harvesting times, with the highest content in July. Analysis of the favonoid content in the leaves from the diferent regions showed the highest contents of isorhamnetin-3-O-glucoside, quercetin 3-O-glucuronide, and quercetin 3-O-glucoside in Yeongcheon, Korea, and highest contnts of rutin, myricetin, kaempferol 3-O-glucoside, and quercetin in Haenam, Korea. The HPLC method was validated and optimized to quantify quercetin 3-O-glucuronide; it showed good linearity (1000–62.5 µg/mL, r 2=0.9999), accuracy (106%–108%), and precision (RSD≤1.70%). Determination of favonoid content in lotus is valu‑ able for producing medicinal crops and identifying the optimal sources to increase the quantity of clinically available medicines.

The inhibition of aldose reductase (AR) has been shown to prevent the progression of the many complications associated with diabetic hyperglycemia. Several compounds purified from various plant sources have exhibited potent inhibition against AR. In this study, the inhibitory effects of the methanol extracts of the flowers of Synurus excelsus and Weigela subsessilis on AR were determined in vitro. Scopolin and scopoletin are coumarins isolated from the flowers of S. excelsus and W. subsessilis; and quercetin is a known AR inhibitor present in many flowers. To determine and quantify their presence in both plants, HPLC-UV analysis of all three compounds was performed. S. excelsus and W. subsessilis showed potent inhibition against AR having IC<sub>50</sub> values of 0.17 and 0.14 μg/mL, respectively. The concentration of scopolin in S. excelsus and W. subsessilis were 34.71 and 174.14 mg/g extract, respectively. Scopoletin was detected in S. excelsus at 3.41 mg/g extract, whereas quercetin was not detected in both plants. This study shows that S. excelsus and W. subsessilis exhibited promising AR inhibitory effects and are both sources of coumarins.
