Evaluation of Salicornia herbacea as a Potential Antioxidant and Anti-Inflammatory Agent

In this study, the antioxidant and anti-inflammatory activities of Salicornia herbacea were evaluated. The crude $CH_2Cl_2$/methanol extract of S. herbacea showed 52% and 86% scavenging activities of the authentic $ONOO^-$ and $ONOO^-$ from 3-morpholinosydnomimine (SIN-1) at a concentration of $50\;{\mu}g/mL$, respectively, and was subjected to a further fractionation with n-hexane, 85% aqueous methanol, n-butanol, and water. Additional purification of the n-butanol fraction revealed that the most potent scavenging activity led to the isolation of isorhamnetin 3-O-$\beta$-D-glucopyranoside as the active principle. The structure of isorhamnetin 3-O-$\beta$-D-glucopyranoside was elucidated by extensive two-dimensional nuclear magnetic resonance experiments such as $^1H$ correlation spectroscopy nuclear Overhauser effect spectroscopy, heteronuclear single quantum correlation, and heteronuclear multiple-bond correlation as well as by comparison with the published spectral data. Isorhamnetin 3-O-$\beta$-D-glucopyranoside exhibited dose-dependent scavenging activities of the authentic $ONOO^-$ and $ONOO^-$ from SIN-1. The electron spin resonance spin-trap techniques confirmed that reactive oxygen species, including the hydroxyl, superoxide, carbon-centered, and 1,1-diphenyl-2-picrylhydrazyl radicals, were actively quenched by addition of isorhamnetin 3-O-$\beta$-D-glucopyranoside. In addition, isorhamnetin 3-O-$\beta$-D-glucopyranoside suppressed the lipopolysaccharide-induced nitric oxide production and the expression of cytokines such as inducible nitric oxide synthase, tumor necrosis factor-a, and interleukin-$1{\beta}$ in Raw 264.7 cells. Findings from this study should underscore the nutraceutical value of S. herbacea-derived isorhamnetin 3-O-$\beta$-D-glucopyranoside as a potent antioxidative and anti-inflammatory agent via alleviation of radical-induced toxicities and pro-inflammatory responses.
Endovascular Treatment for Lower Extremity Deep Vein Thrombosis: An Overview

Lower extremity deep vein thrombosis (DVT) is a serious medical condition that can result in local pain and gait disturbance. DVT progression can also lead to death or major disability as a result of pulmonary embolism, postthrombotic syndrome, or limb amputation. However, early thrombus removal can rapidly relieve symptoms and prevent disease progression. Various endovascular procedures have been developed in the recent years to treat DVT, and endovascular treatment has been established as one of the major therapeutic methods to treat lower extremity DVT. However, the treatment of lower extremity DVT varies according to the disease duration, location of affected vessels, and the presence of symptoms. This article reviews and discusses effective endovascular treatment methods for lower extremity DVT.
Genetic Polymorphisms of UGT1A and their Association with Clinical Factors in Healthy Koreans

Glucuronidation by the uridine diphosphateglucuronosy-ltransferase 1A enzymes (UGT1As) is a major pathway for elimination of particular drugs and endogenous substances, such as bilirubin. We examined the relation of eight single nucleotide polymorphisms (SNPs) and haplotypes of the UGT1A gene with their clinical factors. For association analysis, we genotyped the variants by direct sequencing analysis and polymerase chain reaction (PCR) in 218 healthy Koreans. The frequency of UGT1A1 polymorphisms, -3279T>G, -3156G>A, -53 $(TA)_{6>7}$, 211G>A, and 686C>A, was 0.26, 0.12, 0.08, 0.15, and 0.01, respectively. The frequency of -118 $(T)_{9>10}$ of UGT1A9 was 0.62, which was significantly higher than that in Caucasians (0.39). Neither the -2152C>T nor the -275T>A polymorphism was observed in Koreans or other Asians in comparison with Caucasians. The -3156G>A and -53 $(TA)_{6>7}$ polymorphisms of UGT1A were significantly associated with platelet count and total bilirubin level (p=0.01, p=0.01, respectively). Additionally, total bilirubin level was positively correlated with occurrence of the UGT1A9-118 $(T)_{9>10}$ rare variant. Common haplotypes encompassing six UGT1A polymorphisms were significantly associated with total bilirubin level (p=0.01). Taken together, we suggest that determination of the UGT1A1 and UGT1A9 genotypes is clinically useful for predicting the efficacy and serious toxicities of particular drugs requiring glucuronidation.

Cryptogenic organizing pneumonia (COP) is an inflammatory lung disease involving the distal bronchioles, respiratory bronchioles, bronchiolar ducts, and alveolae. The etiology is usually unknown; however, there are several known causes and associated systemic diseases. Corticosteroid therapy is the best treatment option and the prognosis of COP is good, with recovery in up to 80% of patients. We described a patient with in-operable hepatocellular carcinoma (HCC) undergoing chemoembolization with doxorubicin in a drug-eluting bead (DEB). COP developed in the patient after chemoembolization but resolved spontaneously in several months.

<P>This study was performed to evaluate whether endocan expression, which is known to be involved in tumor angiogenesis, was increased in rheumatoid arthritic tissues. In addition, the involvement of adiponectin in the regulation of endocan expression in arthritic joints was examined. Arthritic synovial tissues from patients with rheumatoid arthritis (RA) or osteoarthritis (OA) were immunostained with antibodies to endocan and vascular endothelial growth factor (VEGF). Subsequently, synovial cells and human umbilical vein endothelial cells were cultured and stimulated with interleukin-1 관 (IL-1관) or adiponectin. The mRNA and protein levels of endocan were evaluated by polymerase chain reaction and ELISA, respectively. Endocan expression was markedly increased in the inflammatory sites of RA synovial tissues. In OA tissues, endocan expression was higher in tissues displaying moderate and severe inflammation than in those with mild inflammation. In vitro expression levels of endocan and VEGF in endothelial and synovial cells were differentially increased in response to IL-1관 stimulation. Adiponectin was a more potent stimulant of endocan than IL-1관 at their respective physiological concentrations in synovial cells. Endocan silencing by small interfering RNA transfection of synovial cells decreased in vitro cell migration and invasion. In conclusion, adiponectin is an important factor in the stimulation of endocan expression in synovial cells. Adiponectin-induced endocan expression in synovial cells may stimulate cell migration and invasion as well as angiogenesis in the pannus of arthritic joints.</P>
Antioxidant Efficacy of Extracts from a Variety of Seaweeds in a Cellular System

As a part of an ongoing search for antioxidants from marine sources, antioxidant activities of 24 kinds of seaweeds (4 green algae, 8 brown algae, and 12 red algae) were investigated. The seaweeds were extracted by acetone/dichloromethane and methanol, respectively. The antioxidant properties of both extracts were evaluated using four different activity tests, including degree of occurrence of intracellular reactive oxygen species (ROS), NO, lipid peroxidation, and GSH (glutathione) in mouse macrophage Raw 264.7 cells. The levels of intracellular reactive oxygen species (ROS) and GSH were measured using 2',7'-dichlorofluorescin diacetate (DCFDA) and monobromobimane as fluorescence probe, respectively. Moreover, the generation of NO and lipid peroxidation products were determined by each method based on the Griess reaction and TBARS assay. Solvent extracts from seaweeds such as Scytosiphon lomentaria, Prionitis cornea, Laruencia okamurae, Callophyllis japonica, Sargassum horneri, Dictyopteris divaricata, Lomentaria catenata, Corallina confuse, Ishige okamurae, and Ahnfeltiopsis flabelliformi exhibited high antioxidant activities in cellular oxidizing systems.

Novel bicyclo[3.1.0]hexanyl purine nucleoside analogues were synthesized as potential antiherpetic agents via a bicyclo[3.1.0]hexanol (${\pm}$)-8, which was prepared using a highly efficient carbenoid cycloaddition reaction. A highly diastereoselective reduction of ketone and a Mitsunobu reaction for the condensation of glycosyl donor (${\pm}$)-12 with 6-chloropurine were employed.
