The rice field grasshopper, Oxya chinensis sinuosa (OC), has traditionally been utilized in Korea for various purposes; however, its potential benefits in the context of osteoporosis remain unclear. The results revealed that OC ethanol extract (OCE) significantly inhibited the formation and activity of tartrate-resistant acid phosphatase (TRAP)-positive cells in receptor activator of nuclear factor-κB ligand (RANKL)-stimulated RAW264.7 cells. Furthermore, OCE, at concentrations ranging from 100 to 400 μg/mL, demonstrated a dose-dependent reduction in the protein expression of osteoclast-specific markers, including nuclear factor of activated T cell cytoplasmic 1, c-Src, and TRAP, when compared to RANKL stimulation alone. Additionally, OCE significantly inhibited RANKLinduced activation of p38 mitogen-activated protein kinase (MAPK) and c-Jun N-terminal kinase (JNK) but not the activation of extracellular signal-regulated kinase. Collectively, these results indicate that OCE suppresses osteoclastogenesis by attenuating the phosphorylation of p38 MAPK and JNK. Consequently, these findings suggest that OCE holds promise for the prevention of osteoporosis.

Thamnolia vermicularis (TV) is an edible lichen that is prevalent in the alpine zone of East Asia. This study evaluated the feasibility of using TV acetone extracts as a functional food based on experiments using cell line and obese mice. The cellular triglyceride levels and Oil red O staining of 3T3-L1 cells indicated that TV extracts (5 and 10 μg/mL) dose-dependently suppressed adipocyte differentiation and lipid accumulation compared with the control. The TV extract (0.4%, w/w) in a high-fat diet (HFD) was supplemented to C57BL/6N mice for 12 weeks, and TV extract supplement significantly reduced visceral fat mass and body weight compared with HFD feeding alone. The TV extract also induced significant decreases in serum and hepatic lipids, whereas it increased the serum high-density lipoproteins-cholesterol/total cholesterol ratio and fecal lipids levels. Moreover, the TV extract led to significantly lower homeostasis model assessment of insulin resistance in diet-induced obese mice. Taken together, these results suggest that the TV extract may have anti- obesity effects, including lipid-lowering, and it is a natural resource with the potential for use in obesity management.

Previous study showed that mealworm fermentation extract (TMP) ameliorated obesity-induced hepatosteatogenesis and proteomic changes in high-fat diet (HFD)-fed mice compared to soy protein (SP). In this study, we investigated the effect of the SP and TMP mixing ratio on hepatic lipid metabolism in mice fed a HFD for 12 weeks. Male C57BL/6N mice were fed HFD supplemented with 20% SP (control group) or three kinds of mixing ratio of SP and TMP (4:1, 3:2, 2:3; S4T1, S3T2, S2T3) as protein source. In comparison with the control group, body weight and food intake were significantly reduced in the S2T3 group, while the size of adipose tissue cells was decreased in all of the TMP mixed groups. Serum insulin, leptin, and homeostatic model assessment of insulin resistance tended to be lower in the S2T3 group compared to the control group. The S2T3 group decreased hepatic lipid droplets compared to the control group, which was mediated by down-regulation of lipid metabolism-related gene expression (PPARγ, ChREBP, CD36, FAS, SCD1, and DGAT2). In conclusion, SP and TMP mixing ratio of 2:3 had an excellent effect on improving lipid metabolism in the liver tissue of HFD-induced obese mice.

Modulation of osteoclast formation can be a potent therapeutic target for inhibition of pathological bone destruction. This study investigated the effects of 70% ethanol extract of Protaetia brevitarsis seulensis larvae (PBE) on receptor activator of nuclear factor-kappa B ligand (RANKL)-induced osteoclast differentiation in RAW 264.7 cells. PBE exhibited no significant cytotoxic effect on RAW 264.7 cells at concentrations up to 2 mg/mL. PBE decreased the formation of tartrate-resistant acid phosphatase-positive multinucleated cells (TRAP(+) MNCs) and TRAP activity in RANKL-stimulated RAW 264.7 cells. PBE significantly inhibited RANKL-induced gene expression of osteoclastogenic makers, such as nuclear factor of activated T cell cytoplasmic 1 (NFATc1), TRAP, matrix metalloproteinase-9, and cathepsin K. PBE also suppressed protein expression of NFATc1, TRAP, c-Src, and osteopontin compared to RANKL alone. In conclusion, these results suggest that PBE has an inhibitory effects against RANKL-induced bone destruction by suppressing osteoclast differentiation.

Previous studies found that mealworm fermentation extract (TMP) reduced alcoholic hepatic steatogenesis. This study examined how the ratio of TMP and soy protein (SP) mix affected glucose and lipid metabolism in obese mice given a high-fat diet (HFD). Mice were given HFD supplemented with 100% SP or the following three ratios of TMP and SP mix for 12 weeks: 20% (S4T1), 40% (S3T2), and 60% (S2T3) TMP. When compared to the SP group, the S2T3 group had considerably lower body weight gain and food consumption. When compared to the SP group, the S2T3 group had slightly lower blood insulin and leptin levels, as well as a lower homeostasis model assessment of insulin resistance score. The use of TMP instead of SP reduced the size of epididymal adipose tissue cells. An increase in the extent of substitution of SP with TMP inhibited the gene expression of hepatic fructolysis/gluconeogenesis (KHK, ALDOB, DLD, and FBP1), lipogenesis (FAS, SCD1, CD36, and DGAT2), and its transcriptional factors (PPAR and ChREBP). Furthermore, the S2T3 group dramatically reduced the expression of hepatic genes implicated in endoplasmic reticulum stress (PDI) and antioxidant defense (SOD1). The 60% TMP mix, in particular, reduced the expression of hepatic glucose and lipid metabolism-related genes in HFD-fed mice. The manufacturing of functional processed goods may be accomplished by combining SP and TMP in a 2:3 ratio.
