        GLAC-SVM Combined Poselet with Blocked Particle Filter for Human Detection and Tracking

        Human tracking based detection (HAR) in recent years has attracted much attention from the research community due to its challenges as well as wide application. In this paper, we investigate the use of Poselet detector verified by Gradient Local Auto Correlation (GLAC) and Support Vector Machine (SVM) for detecting human boundary. After that, each detected candidate is associated with a suitable tracker following the Probabilistic Data Association Filter(PDAF). For those tracker that has not been assigned, a tracking approach based block Particle Filter is in charge to maintain their existence. We conduct our experiments the open dataset PESTS 2009 and our self-recorded one CNU. We compare our method with Adaptive Boosting (Adaboost) particles filter tracking in the case of CNU dataset. The average accuracy of Blocked Particle Filter is 56.41% which outperforms that of Adaboost tracking with only 25.58%.

      • Silencing of Rac3 Inhibits Proliferation and Induces Apoptosis of Human Lung Cancer Cells

        Background: Rac3, a member of the Rac family of small guanosine triphosphatases (GTPases), regulates a variety of cell functions, including the organization of the cytoskeleton, cell migration, and invasion. Overexpression of Rac3 has been reported in several human cancers. However, the role of Rac3 in lung cancer (LC) has not been determined in detail. The purpose of this study was to investigate the effect of silencing of Rac3 expression in human LC cells and the consequences for cell survival. Materials and Methods: Lentivirus small hairpin RNA (shRNA) interference techniques were utilized to knock down the Rac3 gene. Gene and protein expression was quantified by quantitative real-time polymerase chain reaction (qRT-PCR) and Western blotting. LC cell apoptosis was examined by annexin V-APC /propidium iodide staining. Results: Efficient silencing of Rac3 strongly inhibited A549 cell proliferation and colony formation ability, and significantly decreased tumor growth. Moreover, flow cytometry analysis showed that knockdown of Rac3 led to G2/M phase cell cycle arrest as well as an excess accumulation of cells in the G1 and S phase. Conclusions: Thus, functional analysis using shRNAs revealed a critical role for Rac3 in the tumor growth of LC cells. shRNA silencing of Rac3 could provide an effective strategy to treat LC.

        PREPARATION AND ANTI-CORROSION PERFORMANCE OF ZINC PHOSPHATE NANOCRYSTALS BY ULTRASONIC – HYDROTHERMAL SYNERGISTIC ROUTE

        The zinc phosphate nanocrystals were synthesized by the ultrasonic – hydrothermal synergisticroute. The ultrasonic – hydrothermal synergistic route can not only decrease the size of the zincphosphate material, but also improve the crystallinity of the product. The transmission electronmicroscopy (TEM) image showed that the needle-like zinc phosphate product was 200 – 300 nmin length and 70 – 80 nm in width. The anti-corrosion tests revealed that the salt atmosphere-resistant time about 1056 h was longer than 768 h common zinc phosphate materials in themarket. The mechanisms of ultrasonic – hydrothermal synergistic route and anti-corrosion werediscussed.

        Association of Lipoprotein-Associated Phospholipase A_2 with Characteristics of Vulnerable Coronary Atherosclerotic Plaques

        Purpose: Lipoprotein-associated phospholipase A2 (Lp-PLA_2) is an inflammatory enzyme expressed in atherosclerotic plaques. We investigated the association of circulating Lp-PLA_2 with characteristics of vulnerable coronary atherosclerotic plaques. Materials and Methods: We recruited 113 patients with either unstable angina (UA, n=59) and stable angina (SA, n=54) by coronary angiography. Thirty-six healthy subjects served as controls. Intravascular ultrasound (IVUS) was used to evaluate the characteristics of coronary atherosclerotic plaque, and serum Lp-PLA2 concentration was measured as well. Results: Lp-PLA_2 concentration was significantly higher in both UA and SA patients [(396±36) μg/L and (321±39) μg/L, respectively]compared with the controls [(127±49) μg/L, p<0.01], and higher in UA than SA group. IVUS findings showed that remodeling index (RI) (0.91±0.15 vs. 0.85±0.11, p=0.005) and eccentricity index (EI) (0.73±0.16 vs. 0.65±0.22, p=0.039) were larger in UA than in SA group, and fibrous caps were thicker in SA than UA group [(0.91±0.23) mm vs. (0.63±0.21) mm, p=0.032]. Moreover, Lp-PLA_2 correlated positively with EI (r=0.439, p<0.01) and RI (r=0.592, p<0.05) in UA group. There was an inverse relationship between Lp-PLA_2 and fibrous cap thickness in both UA (r=-0.587, p<0.001) and SA (r=-0.318, p<0.05) groups. The independent risk factors in UA group were Lp-PLA_2 (OR=1.055, 95% CI: 1.03-1.08, p=0.013), LDL-cholesterol (OR=0.032, 95% CI: 0.00-0.05, p=0.041) and fibrous cap thickness (OR=0.008, 95% CI: 0.00-0.45, p=0.019). Lp-PLA_2 was strongly associated with both EI and fibrous cap thickness in both groups. Conclusion: Serum level of Lp-PLA_2 is associated with both eccentricity index and fibrous cap thickness in both UA and SA groups. Elevated levels of circulating Lp-PLA_2 might to be a strong risk factor and more serious for unstable angina than stable angina.

        Inhibition of lncRNA KCNQ1OT1 Improves Apoptosis and Chemotherapy Drug Response in Small Cell Lung Cancer by TGF-β1 Mediated Epithelial-to-Mesenchymal Transition

        Purpose Drug resistance is one of the main causes of chemotherapy failure in patients with small cell lung cancer (SCLC), and extensive biological studies into chemotherapy drug resistance are required. Materials and Methods In this study, we performed lncRNA microarray, in vitro functional assays, in vivo models and cDNA microarray to evaluate the impact of lncRNA in SCLC chemoresistance. Results The results showed that KCNQ1OT1 expression was upregulated in SCLC tissues and was a poor prognostic factor for patients with SCLC. Knockdown of KCNQ1OT1 inhibited cell proliferation, migration, chemoresistance and promoted apoptosis of SCLC cells. Mechanistic investigation showed that KCNQ1OT1 can activate transforming growth factor-β1 mediated epithelial-to-mesenchymal transition in SCLC cells. Conclusion Taken together, our study revealed the role of KCNQ1OT1 in the progression and chemoresistance of SCLC, and suggested KCNQ1OT1 as a potential diagnostic and prognostic biomarker in SCLC clinical management.

        In Vivo Biocompatibility and Improved Compression Strength of Reinforced Keratin/Hydroxyapatite Scaffold

        A rapid freezing/lyophilizing/reinforcing process is suggested to fabricate reinforced keratin/hydroxyapatite (HA) scaffold with improved mechanical property and biocompatibility for tissue engineering. The keratin, extracted from human hair, and HA mixture were rapidly frozen with liquid nitrogen and then lyophilized to prepare keratin/HA laminar scaffold. The scaffold was then immersed in PBS for reinforcement treatment, and followed by a second lyophilization to prepare the reinforced keratin/HA scaffold. The morphology, mechanical, chemical, crystal and thermal property of the keratin/HA scaffold were investigated by SEM, FTIR, XRD, DSC, respectively. The results showed that the keratin/HA scaffold had a high porosity of 76.17 ± 3%. The maximum compressive strength and compressive modulus of the reinforced scaffold is 0.778 and 3.3 MPa respectively. Subcutaneous implantation studies in mice showed that in vivo the scaffold was biocompatible since the foreign body reaction seen around the implanted scaffold samples was moderate and became minimal upon increasing implantation time. These results demonstrate that the keratin/HA reinforced scaffold prepared here is promising for biomedical utilization.

        NANOSEEDING EFFECT IN ZSM-5CRYSTALLIZATION PROCESS

        ZSM-5 zeolites were hydrothermally synthesized in three different seeding pathways under the direction of tetrapropylammonium bromide (TPABr) template. In order to investigate the seeding effect in ZSM-5 crystallization process, ZSM-5 crystals and pre-fabricated MFI-type nanoseeds were added into the original self-induction system, respectively. The final ZSM-5 zeolites were systematically investigated based on XRD (X-ray diffraction), SEM (scanning electron micrograph), TEM (transmission electron micrograph), nitrogen adsorption characterizations and NH3-TPD (ammonia-temperature programmed desorption). The self-induction system produced ca. 20 μm ZSM-5 zeolite displaying hexagonally uniform prisms. After the addition of ZSM-5 crystal seeds, the crystal sizes were decreased greatly to ca. 5 μm. When MFI-type nanoseeds were adopted, irregular aggregate particles consisting of 20–50 nm primary particles were rapidly synthesized. The varied hydrothermal crystallization kinetics of the three synthesis system was also explored. Adjusting the seed agents alone, ZSM-5 crystals with diverse structural, morphological, textural and hydrothermal behaviors could be fabricated conveniently. The three ZSM-5 zeolites loaded by 0.05 wt.% Pt were assessed for the xylene isomerization reaction to investigate the particle size effect on the catalytic properties.

        Taguchi Approach for Anti-heat Stress Prescription Compatibility in Mice Spleen Lymphocytes In Vitro

        Heat stress (HS) may induce immunosuppression as well as inhibit the proliferation of lymphocytes. This study evaluated the effects on immune function of our prescription on splenic lymphocytes under HS as well as its compatibility. The effects of four herbal extracts from Agastache rugosa, Atractylodes lancea, Cortex Phellodendri, and Gypsum Fibrosum on heat treated splenic lymphocytes were investigated and the compatibility of the prescription was also explored by using the Taguchi method. This study revealed changes in proliferation by traditional Chinese medicines of splenic lymphocytes after HS. Proliferation in the HS group was significantly lower than the control group. Under HS, the effects of higher concentrations of Agastache rugosa (100 and 200 μg/mL), Atractylodes lancea (100 and 200 μg/mL), Cortex Phellodendri (50 and 100 μg/mL) and Gypsum Fibrosum (100 and 200 μg/mL) caused a significant increase on ConA/LPS-induced proliferation of lymphocytes than lower concentrations. We, therefore, conclude that the prescription of traditional Chinese medicines may recover splenic lymphocytes from the immunosuppression induced by HS. The Taguchi design, which allows rapid and high efficiency for the selection of the best conditions for our prescription on HS-treated splenic lymphocytes, demonstrated that Agastache rugosa (200 μg/mL), Atractylodes lancea (200 μg/mL), Cortex Phellodendri (100 μg/mL) and Gypsum Fibrosum (100 μg/mL)were the optimal conditions for the prescription. The validation experiment confirmed that our composition in optimum extraction conditions enhanced effects on ConA or LPS-stimulated lymphocytes under HS. The results showed that the Taguchi optimization approach is a suitable method for optimization of the composition of prescription.
