        The differentiation of cellular structure during encystment in the soil hypotrichous ciliate Australocirrus cf. australis (Protista, Ciliophora)

        Ciliates are able to form resting cysts as a survival strategy in response to stressful environmental factors. Studies on the characteristics of cellular structure during encystment may provide useful information for further understanding of the regulatory mechanism of cellular patterns and supply new clues regarding the phylogeny of ciliates. Scanning and transmission electron microscopies were used to observe the ultrastructure of cells during encystment of the soil ciliate Australocirrus cf. australis. The dedifferentiation of ciliature was revealed for the first time. Ciliary shafts first shortened, and the remaining ciliature, including basal bodies and the fibrillar cirral basket, retracted into the cytoplasm and was surrounded by the autophagic vacuoles and then gradually digested. A large number of autophagic vacuoles were observed in mature resting cysts. Autophagy might not only be necessary for the differentiation of cellular structures during encystment but might also be important to sustain the basic life activities in the resting stage. Australocirrus cf. australis formed a kinetosome-resorbing cyst and contained four layers in the cyst wall: the ectocyst, mesocyst, endocyst and granular layer. The ciliature resorbing state and the number of layers in the cyst wall were consistent with those found in other oxytrichous ciliates. However, the phenomenon wherein the two macronuclear nodules are not fused during encystment is not commonly observed among oxytrichids. Additionally, the octahedral granules in the mesocyst of this species exhibit different morphology from the congeners.

        Probing the Key Binding Sequence and Improvement of the Stability of a β-Bungarotoxin-binding Aptamer in Snake Venom

        Chemical modifications of the nucleotides can improve the stability of aptamers against enzyme degradation in serum, but it is not clear whether these methods are effective in snake venom. In this study, a DNA aptamer, βB-1, which specifically recognize β-bungarotoxin and Bungarus multicinctus venom was chosen, and the key binding sequence of the aptamer was determined. Based on the secondary structure of the truncated aptamer, locked nucleic acids and 2′-O-methyl nucleotides were applied to modify the stem and loop sequences, respectively. In addition, a 3′-3′-thymidine cap was also adopted to block the 3′ end. It was shown that these chemical modifications can all enhance the stability of the aptamer in snake venom. Simultaneously, modified aptamer with the above modifications in one sequence exhibited a significantly elevated biostability, with the half-life improved from several minutes to 210 min while maintaining its binding affinity to the target.

        Selection of high transfer stability and optimal power-efficiency tradeoff with respect to distance region for underground wireless power transfer systems

        Magnetic resonant wireless power transfer (MR-WPT) has become a significant method for powering the measurement units in landslide monitoring boreholes. This paper focuses on the effects of quality factor, coupling distance and load on the power transfer stability, sensitivity and performance of an MR-WPT system. Firstly, through a numerical model of an MR-WPT, the effect of quality factor and coupling distance on the output power, transfer efficiency and frequency splitting phenomenon of MR-WPT systems have been comparatively analyzed. The relationship between the load and the coupling distance region corresponding to optimal transfer performance has been studied. Results show that improving the quality factor of the coils is beneficial for the transfer performance and stability of WPT system. Furthermore, through the selection of the load, a coupling distance region with high transfer stability and an optimal tradeoff between power and efficiency can be obtained. Finally, the above theoretical simulation results have been verified by experimental results.

        Nutrigenomics reveals potential genetic underpinning of diverse taste preference of Chinese men

        Background Taste preference varies geographically in China. However, studies on Chinese people’s taste preference in different regions of China are limited, and are lack of research on the mechanism of diferences in taste preference, especially in genetics. Objective This study aims to investigate the characteristics of taste preference of Chinese men, and estimate whether diverse taste preference in Chinese have genetic underpinning. Methods We conducted a questionnaire survey on taste preferences on 1076 males from 10 regions of China, and collected another 1427 males from the same regions which genotyped by microarray. We compared the correlation between diferent taste preference, and evaluated the correlation between the mutation frequency of inhouse database and diferent taste preference. The putative taste-preference-related genes were further utilized to estimate the candidate relationship on gene and gene network in diferent taste preference. Results There was a correlation between diferent taste preferences in Chinese men. We found 31 SNPs associated with 6 kind of taste preferences. These SNPs located within or nearby 36 genes, and the tastes associated with 4 of these genes (TRPV1, AGT, ASIC2 and GLP1R) are consistent with the previous studies. Moreover, in diferent tastes which were suggested to be associated with each other, some putative related genes were the same or in the same gene network, such as pathways related with blood pressure, response to stimulus and nervous system. Conclusions This study indicates that the diverse taste preference of Chinese men may have genetic underpinning.

        Localization of α-, γ-, and δ-tubulin in the hypotrich ciliate Stylonychia pustulata (Hyportrichida, Ciliophora)

        Ciliature microtubular organelles, which organize into ciliary units, are highly complex microtubular systems. Toinvestigate the formation of ciliature microtubular organelles and the role that different tubulins play in this process,we examined the distribution and function of α-, γ-, and δ-tubulins in the hypotrich ciliate Stylonychia pustulata. Immunofluorescence analysis demonstrated that α-tubulin localized to all microtubular organelles, while γ- and δ-tubulinscolocalized at basal bodies. Immunogold labeling revealed that γ- and δ-tubulins colocalize to the proximalpart of basal bodies, suggesting that γ- and δ-tubulins probably interact and may function together. In addition, thepermanent localization of γ-tubulin to the adoral zone of membranelles suggests that γ-tubulin play a role in themaintenance of oral ciliature base-associated microtubules, but does not play a role in the biogenesis of theseciliature. We observe that δ-tubulin is restricted to the basal body and the connective fibers of the basal body,suggesting this protein may be required for the extension and stability of the microtubules in these structures. Dramatically different concentrations of these three tubulins were observed in the cytoskeleton and the insoluble andsoluble fractions of the cell, suggesting that these highly related proteins perform different functions in S. pustulata.

        Water Dispersible Graphene Sheets Produced from Unassembled Graphene–Polyaniline Nanohybrids

        We present an effcient process for producing water dispersible graphene sheets from unassembled graphene–polyaniline nanohybrids. The result of atomic force microscopy reveals that over 80% of thus-prepared graphitic sheets are single layers with typical thickness of approximately 0.8 nm. The proportion of modifying molecules in the product is found to be as low as 3.0 wt.%, as determined by elemental analysis. Along with its fascinating water dispersibility and remarkably high electrical conductivity, such material is anticipated to be very promising for use in graphene-based nanoelectronics and high-performance composites.

        Adsorption characteristics of methylene blue onto the N-succinyl-chitosan-gpolyacrylamide/ attapulgite composite

        N-succinyl-chitosan-g-polyacrylamide/attapulgite (NSC-g-PAM/APT) composite was applied as adsorbent for the removal of methylene blue (MB) from aqueous solution. The initial pH value of the dye solutions, the contact temperature, the contact time and the concentration of the dye solutions on adsorption capacity of the composite for MB dye were investigated. The adsorption kinetics and isotherms were also studied. It was shown that all the sorption processes were better fitted by pseudo-second-order equation and the Langmuir equation. The results indicated that the adsorption capacity of the composite was higher than those of chitosan (CTS) and attapulgite (APT). The desorption studies revealed that the composite provided the potential for regeneration and reuse after MB dye adsorption, which implied that the composite could be used as quite effective adsorbent for the removal of MB from aqueous solution.

        Structural evolution of the thermal conversion products of modified coal tar pitch

        Coal tar pitch is a product with high carbon content and aromatic compounds. Modified coal tar pitch is a high quality raw material for the preparation of intermediate phase pitch, needle coke, carbon microspheres, et al. In this paper, modified coal tar pitch was used as raw material, nitrogen was used as protective gas, and thermal conversion was carried out at constant temperatures (370, 390, 410, 420 °C). Polarized light microscopy, SEM, elemental analysis, FTIR spectroscopy, Raman spectroscopy and XRD diffraction combined with split-peak fitting were used to characterize the microstructures of the thermal transformation products. The results showed that the Iar and CH3/ CH2 contents of the products increased with the gradual increase of the thermal conversion temperature, and the aromatic content increased. And the higher the temperature at the same heating rate, the more the ideal graphite microcrystal content, and the defective graphite microcrystals are converted into ideal graphite microcrystals during the thermal conversion process. When the reaction temperature exceeds 390 °C, the microstructure of the thermal transformation products is anisotropic spheres, and the small spheres fuse with each other and tend to be basin-like and mosaic structure as the temperature increases.

        Highly Efficient Biotransformation of Notoginsenoside R1 into Ginsenoside Rg1 by Dictyoglomus thermophilum β-xylosidase Xln-DT

        Notoginsenoside R1 and ginsenoside Rg1 are the main active ingredients of Panax notoginseng, exhibiting anti-fatigue, anti-tumor, anti-inflammatory, and other activities. In a previous study, a GH39 β-xylosidase Xln-DT was responsible for the bioconversion of saponin, a natural active substance with a xylose group, with high selectivity for cleaving the outer xylose moiety of notoginsenoside R1 at the C-6 position, producing ginsenoside Rg1 with potent anti-fatigue activity. The optimal bioconversion temperature, pH, and enzyme dosage were obtained by optimizing the transformation conditions. Under optimal conditions (pH 6.0, 75℃, enzyme dosage 1.0 U/ml), 1.0 g/l of notoginsenoside R1 was converted into 0.86 g/l of ginsenoside Rg1 within 30 min, with a molar conversion rate of approximately 100%. Furthermore, the in vivo anti-fatigue activity of notoginsenoside R1 and ginsenoside Rg1 were compared using a suitable rat model. Compared with the control group, the forced swimming time to exhaustion was prolonged in mice by 17.3% in the Rg1 high group (20 mg/kg·d). Additionally, the levels of hepatic glycogen (69.9-83.3% increase) and muscle glycogen (36.9-93.6% increase) were increased. In the Rg1 group, hemoglobin levels were also distinctly increased by treatment concentrations. Our findings indicate that treatment with ginsenoside Rg1 enhances the anti-fatigue effects. In this study, we reveal a GH39 β-xylosidase displaying excellent hydrolytic activity to produce ginsenoside Rg1 in the pharmaceutical and food industries.
