        Correlations among Six Hormone-Induced Transcription Factors and the Alcohol Acyltransferase Gene in Apple

        Alcohol acyltransferase (AAT) plays an important role in ester biogenesis in ripening fruit. In apple, MdAAT2 is up-regulated in response to treatment with defense-related hormones. We cloned a novel MdAAT2 promoter via thermal asymmetric interlaced PCR (TAIL-PCR). Sequence analysis indicated the presence of salicylic acid (SA)- and methyl jasmonate (MeJA)-responsive elements in the promoter region. To examine further the operational mechanism for transcription factors (TFs) in regulating expression of the MdAAT2 promoter, we isolated and investigated six potential stress-induced TFs -- MdMYB1, MdMYB2, MdMYB6,MdERF1, MdERF2, and WRKY. These were monitored during various stages of fruit development as well as under several hormonal treatments. Reverse transcription-polymerase chain reaction (RT-PCR) analysis showed that all of these TF genes, as well as MdAAT2, were detectable in the stigmas. Levels of expression by MdMYB1, MdMYB2, and MdERF1were significantly correlated with that by MdAAT2 during fruit ontogeny. Moreover, transcription of three TFs (MdMYB1,MdMYB6, and MdERF1) was significantly correlated with that of MdAAT2 upon exposure to ethephon, SA, or MeJA. Sequence analysis demonstrated that CAAT, CCAAT elements,and several MYB transcription factor binding sites (CNGTTmotif,AACCA, and CTAACCA elements) existed in the MdAAT2 promoter region. Using that analysis, we performed modeling of MdMYB1 and MdMYB6 based on PROTEINDNA complex1 and complex3 (PDB Accession Numbers 1H88 and 1H8A). There, similarities were found with the protein complex of CAAT or CCAAT consensus sequences. Thus, we propose that two TFs -- MdMYB1 and MdMYB6-- are directly or indirectly involved in the activation of MdAAT2 expression in apple fruit.

        Shared Spatio-temporal Attention Convolution Optimization Network for Traffic Prediction

        The traffic flow in an urban area is affected by the date, weather, and regional traffic flow. The existing methodsare weak to model the dynamic road network features, which results in inadequate long-term prediction performance. To solve the problems regarding insufficient capacity for dynamic modeling of road network structuresand insufficient mining of dynamic spatio-temporal features. In this study, we propose a novel traffic flowprediction framework called shared spatio-temporal attention convolution optimization network (SSTACON). The shared spatio-temporal attention convolution layer shares a spatio-temporal attention structure, that isdesigned to extract dynamic spatio-temporal features from historical traffic conditions. Subsequently, the graphoptimization module is used to model the dynamic road network structure. The experimental evaluationconducted on two datasets shows that the proposed method outperforms state-of-the-art methods at all timeintervals.

        Stability analysis of prestressed stayed steel columns with split-up crossarm systems

        A Prestressed stayed steel column is an efficient and lightweight way with regard to enhancing the stability behaviour of a compression column. In the past, researchers primarily concentrated on investigating the behaviour of stayed steel columns with horizontal crossarms. However, this article focuses on prestressed stayed steel columns with split-up crossarm system, in which the crossarms are aslant and rotational symmetrically arranged. A mathematical formula calculating the optimal pretension that corresponds to the maximum critical buckling load was established according to geometric analysis based on the small deformation assumption. It was demonstrated that critical buckling mode of this stayed column is different from the one with horizontal crossarms. The governing imperfection direction that should be adopted in the nonlinear buckling analysis was determined in this work. In addition, the effects of crossarm inclination, stay diameter, and crossarm length on the stability behaviour were investigated. An influencing factor denotes the ratio of the load carrying capacity of the prestressed stayed steel column to the Euler load of the main column was also obtained.

        Study on the Pressure-Volume-Temperature Properties of Polypropylene at Various Cooling and Shear Rates

        To understand the pressure-volume-temperature (PVT) properties of polymer in injection process, a dilatometer with high cooling rate (up to 25 oC/s) and shear rate (up to 320 s-1) was developed. The working pressure and temperature of the dilatometer range from 0 to 100 MPa, and from 30 to 300 oC, respectively. With this instrument, a crystalline polymer polypropylene (PP) was employed to study the effects of cooling rate, shear rate, pressure and their coupling effect on its PVT properties. It was demonstrated that the cooling rate showed a significant effect on the PVT properties. With the increase of cooling rate, the transition temperature of PP from melt state to crystallization decreases gradually and the temperature range of crystallization was also extended. Shear increased the transition temperature of PP from melt state to crystallization. Meanwhile, the initial temperature of shear and crystalline segment was shifted to a higher temperature region with increasing shear duration or shear rate. Above the transition temperature, the effect of shear on the PVT properties weakened gradually with increasing the initial temperature. The coupling analysis of cooling and shear processes showed that increase in cooling rate was enhanced the effect of shear on the transition temperature of PP from melt state to crystallization.

      • Self-assembly and sensing-group graft of pre-modified CNTs on resonant micro-cantilevers for specific detection of volatile organic compound vapors

        <P>This paper reports MWCNT (multi-wall carbon nano-tube)-modified resonant micro-cantilever chemical sensors for detection of trinitrotoluene (TNT) vapor. The MWCNTs are pre-modified and then area-selectively self-assembled at the free-end gold pad of a micro-cantilever, in which a resonance-exciting heater and a signal-readout piezoresistive Wheatstone bridge are integrated. Featuring a high specific surface area, the MWCNTs are further functionalized with TNT-sensitive groups by grafting onto the sidewalls of the MWCNTs. To lower the non-specific absorption of water and other small organic molecules, the SiO<SUB>2</SUB> surface of the micro-cantilever was also pre-treated for hydrophobicity and oleophobicity by self-assembling a monolayer of heptadecafluorodecyltrimethoxysilane. The results of our sensing experiments have shown a capability to rapidly detect ppb-level TNT vapor, and a high specificity of the functionalized groups to TNT molecules. The experiment has also confirmed a good long-term stability in detecting sensitivity.</P>

        Chemerin reverses the malignant phenotype and induces differentiation of human hepatoma SMMC7721 cells

        Chemerin exhibits an inhibitory eff ect on hepatocellularcarcinoma; however, the underlying mechanismis unclear. Here, low chemerin expression was confi rmedin samples of liver cancer patients and hepatoma cells. Chemerin altered hepatoma cell morphology but had noeff ect on normal hepatocytes. Chemerin inhibited proliferationof several human hepatoma cell lines. Real-time PCRdetection of hepatocellular carcinoma markers showed thatmRNA levels of albumin and A-type gamma-glutamyltransferase increased whereas those of alpha-fetoprotein,alkaline phosphatase, B-type gamma-glutamyl transferase,insulin-like growth factor II, and human telomerase reversetranscriptase decreased in chemerin-treated SMMC7721cells. Western blotting revealed that chemerin up-regulatedalbumin and vimentin expressions, and downregulated alpha-fetoprotein expression. Phosphorylated STAT3 wassignifi cantly up-regulated, whereas phosphorylated ERKand AKT were signifi cantly downregulated by chemerin. Chemerin decreased phosphorylated ERK and AKT expressionand the cell proliferation induced by PI3K activator 740Y-P but could not signifi cantly alter phosphorylated STAT3expression and the cell growth induced by STAT3 inhibitorNSC74859. In conclusion, chemerin reversed the malignantphenotype and induced SMMC7721 cell diff erentiation byinhibiting MAPK/ERK and PI3K/AKT signaling; growthinhibition by chemerin is not directly related to the JAK/STAT signaling pathway. Our study provides novel evidencethat chemerin could be utilized for liver cancer treatment.

        Histone demethylase KDM4A plays an oncogenic role in nasopharyngeal carcinoma by promoting cell migration and invasion

        Compelling evidence has indicated the vital role of lysine-specific demethylase 4 A (KDM4A), hypoxia-inducible factor-1α (HIF1α) and the mechanistic target of rapamycin (mTOR) signaling pathway in nasopharyngeal carcinoma (NPC). Therefore, we aimed to investigate whether KDM4A affects NPC progression by regulating the HIF1α/DDIT4/mTOR signaling pathway. First, NPC and adjacent tissue samples were collected, and KDM4A protein expression was examined by immunohistochemistry. Then, the interactions among KDM4A, HIF1α and DDIT4 were assessed. Gain- and loss-of-function approaches were used to alter KDM4A, HIF1α and DDIT4 expression in NPC cells. The mechanism of KDM4A in NPC was evaluated both in vivo and in vitro via RT-qPCR, Western blot analysis, MTT assay, Transwell assay, flow cytometry and tumor formation experiments. KDM4A, HIF1α, and DDIT4 were highly expressed in NPC tissues and cells. Mechanistically, KDM4A inhibited the enrichment of histone H3 lysine 9 trimethylation (H3K9me3) in the HIF1α promoter region and thus inhibited the methylation of HIF1α to promote HIF1α expression, thus upregulating DDIT4 and activating the mTOR signaling pathway. Overexpression of KDM4A, HIF1α, or DDIT4 or activation of the mTOR signaling pathway promoted SUNE1 cell proliferation, migration, and invasion but inhibited apoptosis. KDM4A silencing blocked the mTOR signaling pathway by inhibiting the HIF1α/DDIT4 axis to inhibit the growth of SUNE1 cells in vivo. Collectively, KDM4A silencing could inhibit NPC progression by blocking the activation of the HIF1α/DDIT4/mTOR signaling pathway by increasing H3K9me3, highlighting a promising therapeutic target for NPC.

        Bidirectional‑friendly rectifier control strategy for advanced traction power supply system under unbalanced supply

        A bidirectional-friendly control strategy of three-phase NPC rectifier under unbalanced voltage for advanced traction power supply system (ATPSS) is proposed in this study. Negative-sequence current and DC voltage ripple are restrained effectively with very low total harmonic distortions (THDs) on the basis of predictive current control theory. First, power balance equation containing only positive-sequence voltage of the three-phase grid voltage is derived, and the secondary fluctuation of instantaneous active and reactive powers on the grid side is analyzed and calculated. Second, expression of the DC-side voltage fluctuation is deduced to fix the calculation of Pref for the optimization of the control system. Third, command current that can achieve bidirectional-friendly control is calculated according to the power expression and the reference voltage value is obtained on the basis of the predictive current control principle. Finally, simulations and experimental results verify the correctness of the theoretical analysis and the effectiveness of the proposed strategy.

        Development of mechanistic cladding rupture model for severe accident analysis and application in PHEBUS FPT3 experiment

        Cladding ballooning and rupture are the important phenomena at the early stage of a severe accident. Most severe accident analysis codes determine the cladding rupture based on simple parameter models. In this paper, a FRTMB module was developed using the thermal-mechanical model to analyze the fuel mechanical behavior. The purpose is to judge the cladding rupture with the severe accident analysis code. The FRTMB module was integrated into the self-developed severe accident analysis code ISAA to simulate the PHEBUS FPT3 experiment. The predicted rupture time and temperature of the cladding were basically consistent with the measured values, which verified the correctness and effectiveness of the FRTMB module. The results showed that the rising of gas pressure in the fuel rod and high temperature led to cladding ballooning. Consequently, the cladding hoop strain exceeded the strain limit, and the cladding burst. The developed FRTMB module can be applied not only to rod-type fuel, but also to plate-type fuel and other types of reactor fuel rods. Moreover, the FRTMB module can improve the channel blockage model of ISAA code and make contributions to analyzing the effect of clad ballooning on transient and subsequent parts of core degradation.

        Erdheim-Chester Disease with Emperipolesis: A Unique Case Involving the Heart

        Histiocytosis is an uncommon disease characterized by excessive accumulation of histiocytes. Here, we report a rare case of non-Langerhans-cell histiocytosis in a 51- year-old woman who presented with severe symptoms of pericardial effusion. Radiologic investigation also detected multiple bone (lower limbs, vertebrae, ribs, and ilium) lesions. Resected pericardium showed abundant mono- or multi-nucleated non-foamy histiocytes (CD68+/CD163+/S-100+/CD1/langerin) in a fibroinflammatory background. The histiocytes demonstrated emperipolesis of lymphocytes, a hallmark feature of Rosai-Dorfman disease (RDD). However, molecular analysis revealed a BRAF V600E mutation of the proliferating histiocytes, highlighting the neoplastic features frequently observed in another non-Langerhans-cell histiocytosis known as Erdheim- Chester Disease (ECD). We consider this case to be a unique presentation of ECD harboring some RDD-like cells with emperipolesis, but not a case of RDD with a BRAF mutation concerning its clinical manifestation (involvement of the heart and bones) and neoplastic features.
