        미역폐기물로부터 알기네이트의 추출 및 디알기네이트의 이용

        Effective alginate extraction from waste-brown seaweeds studied. The waste-brown seaweeds almost consist of stems of seaweeds. Alginate could be obtained from the stems as well as leaves of seaweed. Ultrasonic vibration(47kHz) facilitated filtering step in the alginate extraction process. Among various alkalies for alginate extraction, NaHCO_3 was most appropriate conceming use of dealginates. The yield of alginate extraction using NaHCO_3 2wt% solution was 19.3% at 60℃. The dealginates from NAHCO_3extraction process have been found most suitable food for red-worms.

        인지과학적 시뮬레이션을 통한 朝鮮 性理學의 禮교육 心性모델 개발(1)

        이 연구는 유교, 특히 조선 성리학의 도덕교육에 의한 인격변화의 과정과 그 결과를 컴퓨터로 모의 실험하기 위하여, 조선 성리학자들이 이해했던 心의 구조와 그 작동의 모형을 작성해보고자 하는 데 그 궁극적인 목적이 있다. 아울러 그것을 통해서 조선 성리학의 큰 줄기를 형성한 퇴계의 심학과 율곡의 심학을 연구대상으로 포용하고, 그 둘 사이에 나타나는 차이점과 공통점을 살피고, 그 결과를 근거로 일종의 성리학 통합심성모델을 작성해보고자 할 때 필요한 이론적 정지작업을 하는 것이 본 연구의 실제 내용이다. 그 작업을 위하여 먼저 통합심성모델 작성의 이론적 관점과 기반으로서 일종의 구성주의의 관점을 채택한 이유를 밝히고, 그것에 의하여 성리학의 심성론의 해석과 그에 따른 심성모델의 구성 방식을 제시하였다. 아울러 서구의 심리철학에서 이해되는 mind, 인지과학에서 연구되고 있는 인간의 마음, 인공지능에서 이해되는 마음을 분석하고 그것과 구성주의에 의하여 재해석된 성리학자들의 마음과의 비교를 행하였다. 중요한 것은 성리학의 역사로 볼 때 똑같이 주자학 계열의 성리학자이면서도 인간의 심성의 관념에 관해서 대립적인 견해를 지녔던 퇴계와 율곡의 관점을 하나로 묶어서 통합심성모델을 작성하는 작업이다. 이에 대해서 본 연구는 퇴계와 율곡이 근본적으로 禮실천에 의하여 인간의 심성의 순수함과 정직함을 기르고 회복하려는 점에서 동일한 입장이라는 점에 착안하여, 사단과 칠정, 인심과 도심 등에 관한 理氣論적 해석에 차이가 있다고 하더라도 그것을 통합해 볼 수 있는 이론적 체제를 확보하고자 하였다. 현대에 논의되는 서구철학적 관점의 윤리학과는 다소 다른 방향과 관점에서 진행되는 이 논의는 인지과학과 비트겐슈타인류의 언어철학 등의 관점과 상호 통하는 면이 있으며, 현대 서구철학의 윤리학이 해결하지 못하는 인간의 도덕적 앎과 행위와의 괴리를 넘어설 수 있는 이론적 가능성을 보여준다고 전망된다. 그 점을 인지과학자인 프란시스코 바레라, 언어철학자인 허버트 핑가레트 등의 이론을 근거로 추론하였다. 결론적으로 퇴계와 율곡의 성리학 심성모델은 현대 윤리교육에 대해서 인간의 심신관계 및 실천지향의 윤리교육의 기초를 제시할 수 있으며, 나아가서 그에 관한 실용성도 풍부하게 지니는 것으로 생각된다. This study has a purpose to make a structure of mind and its working principles, which had been thought and understood by confucians of Chosun Dynasty, for computer simulating processes of peronalyity changes through Chosun Dynasty's moral education. This study includes Simhak(心學) of famous Chosun confucians, Toegye and Yulgok, and compares commons and differences of two scholar's theory. The key of this study prepares a necessary theoretical foundation for making a Neo-confucian integrated mind model from such results. We explained why we adopt radical constructivism as a theoretical framework for the integrated mind model, and proposed an interpretation of Neo-confucian mind theory and a method of constructing mind model. Futhermore, we look around a mind of western psychological philosophy, a mind of cognitive science, and a mind of artificial intelligence, and we have compared the confucian mind which is re-interpreted as radical constructivism with such minds. An important work is to make an integrated mind model of Toegye and Yulgok because two scholars are same Neo-confucian, but assisted own theories about human mind. Although the interpretations of Sadan(四端) and Chiljung(七情), Insim(人心) and Dosim(道心) are different each other, encouraging and recovering purity and honesty human mind with the practice of rituals is very much alike. Therefore we want to prepare theoretical foundation for integrating both. This argument is different from ethics of modern western philosophy, but have something to do with cognitive science and Wittgenstein-like philosophy of language. We expect that this idea show a theoretical possibility beyond the dissociation between ethical thinking and behavior, which modern western ethics can't solve. Such a conclusion is inferred from the concepts of Francisco Valera, the cognitive scientist, and Herbert Figarette, the linguistic philosoper. Now to conclude, the confucian mind model of Toegye and Yulgok can provide the explainable mind-body relationship and the foundation of practical moral education for modern education. We believe that the model has much possibility for practical use.

        This study focused on the increase of lime plate strength by adding reinforced material into limestone and chemical reaction with carbon dioxide gas. Also we did experiment to find the optimum content of plaster as a binding component. Carbon fiber as reinforced material has disadvantages. It did not mixed well in the forming process and had low cohesion force with limestone. Content of glass fiber showed optimum at 1.0cm, 4.16 wt% in lime plate. The increase of strength in lime plate was ??, after reaction with CO₂at 70℃ for 6hr. When lime plate consists of 40wt% of plaster, 4.16 wt% of 1.0cm glass fiber, its strength was ??

        미생물을 이용한 미역줄기에서 알긴산염 추출 및 저분자화

        We studied a extraction and degradation of alginate from seaweed-stems using microorganism DS-02. DS-02 has a maximum growth rate at 30 ℃ and the enzyme has a maximum activity of alginate extraction at 35 ℃. The yield of alginate extraction using DS-02 is about 16.0% for 3.0 hour and molecular weight of the alginate decreased to about 1/8 of initial value after 24 hour extraction. Alginate extraction method by DS-02, compared with general alkali-extraction method, has an advantage of decreasing the molecular weight of alginate during extraction.

        미생물을 이용한 미역폐기물의 저장 및 알긴산염 저분자화

        We studied a storage of waste-brown seaweed at room temperature and degradation of alginate in seaweed by microorganism DS-02. The seaweeds, mixed with 5.0 wt% DS-02 and sealed in vinyl package without any other treatment, could be stored longer than 1 year without spoilage at room temperature. During the storage process, the alginate of seaweed was decomposed by enzyme of DS-02 and the molecular weight of alginate decreased to about 1/10 of initial quantity. DS-02 growed as fast as it had maximum weight after 24 hour culture and it's enzyme had a maximum activity of alginate degradation at 40 ℃. The seaweed sample became particles in DS-02 culture solution and the M.W of alginate decreased to about 1/10 of initial value after 24 hour decomposition. The effect of alginate degradation with DS-02 was similar to that of degradation with 3.0 M HCl solution for 24 hour.

      • Thrombin-activatable fluorescent peptide incorporated gold nanoparticles for dual optical/computed tomography thrombus imaging

        <P><B>Abstract</B></P> <P>Thrombosis is an important pathophysiologic phenomenon in various cardiovascular diseases, which can lead to oxygen deprivation and infarction of tissues by generation of a thrombus. Thus, direct thrombus imaging can provide beneficial in diagnosis and therapy of thrombosis. Herein, we developed thrombin-activatable fluorescent peptide (TAP) incorporated silica-coated gold nanoparticles (TAP-SiO<SUB>2</SUB>@AuNPs) for direct imaging of thrombus by dual near-infrared fluorescence (NIRF) and micro-computed tomography (micro-CT) imaging, wherein TAP molecules were used as targeted thrombin-activatable peptide probes for thrombin-specific NIRF imaging. The freshly prepared TAP-SiO<SUB>2</SUB>@AuNPs had an average diameter of 39.8 ± 2.55 nm and they showed the quenched NIRF signal in aqueous condition, due to the excellent quenching effect of TAP molecules on the silica-gold nanoparticle surface. However, 30.31-fold higher NIRF intensity was rapidly recovered in the presence of thrombin <I>in vitro</I>, due to the thrombin-specific cleavage of quenched TAP molecules on the gold particle surface. Furthermore, TAP-SiO<SUB>2</SUB>@AuNPs were successfully accumulated in thrombus by their particle size-dependent capturing property, and they presented a potential X-ray absorption property in a dose-dependent manner. Finally, thrombotic lesion was clearly distinguished from peripheral tissues by dual NIRF/micro-CT imaging after intravenous injection of TAP-SiO<SUB>2</SUB>@AuNPs in the <I>in situ</I> thrombotic mouse model, simultaneously. This study showed that thrombin-activatable fluorescent peptide incorporated silica-coated gold nanoparticles can be potentially used as a dual imaging probe for direct thrombus imaging and therapy in clinical applications.</P>

        Breeding of Moon Cactus 'Yeonbit' with Pink Color

        농가 선호도가 높은 분홍색 비모란 선인장 ‘Yeonbit’는 분홍색 품종 ‘Yeonsi’을 모본으로, 분홍색 ‘Yeonsil’ 품종을 부본으로 하여 2011년에 교배하여 육성된 교배종이다. 종자를 70% C2H5OH와 2% NaOCl로 멸균처리한 후, 100mL 삼각플라스크 0.1% 활성탄이 첨가된 Kyoto 배지에 파종하였다. 발아된 유묘는 기저부를 잘라 기배계대 배양된 어린 삼각주에 접목한 후 동일한 배지가 첨가된 시험관에 치상하였다. 6개월 재배 후 온실로 옮겨서 순화과정을 거쳤으며, 기외에서 매년 접목을 하였으며 총 4번을 접목하여 2013년부터 2015년까지 총 3회에 걸쳐 생육특성을 조사하였다. ‘Yeonbit’ 품종은 편원형 모양과 분홍색 구색을 가지고 있다. 그리고 7~9개, 평균 8.0개의 능(rip), 갈색의 직립형 짧은 가시, 그리고 혹(tubercle)이 돌출된 형태로, 모구가 구성되어 있다. 정식 10개월 후 ‘Yeonbit’ 품종의 구직경은 34.5mm였으며 자구는 모구와 같은 분홍색으로 평균 15.4개가 생성되었다. 2015년 육성계통 평가회에서 ‘Yeonbit’ 품종은 기호도 점수 3.7을 받았다. 2017년 6월 12일에 이 품종은 국립종자원에 등록되었으며 종자산업법에 의해 품종보호(등록번호 6752)를 받게 되었다. The cultivar of moon cactus, ‘Yeonbit’ with pink color, was bred by National Institute of Horticultural & Herbal Science (NIHHS) in 2015. For breeding a pink cultivar with high preference, the pink color cultivars 'Yeonsi' and 'Yeonsil' were crossed in 2011. After 2 months, the seeds were harvested and separated from the peel. And then they were sterilized by C2H5OH 70% and NaOCl 2%, and then sowed in 100 mL mass flask filled with Kyoto medium + charcoal 0.1%. The bottom of shoot was cut, grafted into a young vine cactus, Hylocereus trigonus in vitro, and then planted into test tube with the same medium. After 6 months, a young moon cactus was secondly grafted into vine cactus in vivo. The growth was surveyed three times from 2013 to 2015, according to the guideline for the conduct of tests for distinctness, uniformity, and stability of Gymnocalycium mihanovichii. ‘Yeonbit’ has flat round shape and pink skin (RP N47B). This cultivar has 8.0 ribs in average. The spine had brown color. The spine averagely grew to 5.2 mm in a straight shape. The shape of tubercle on the rib was projecting. This was 34.5 mm in diameter and averagely produced 15.4 offsets at 10 months from planting. The preference score of this was 3.7 on a five-point scale. On June 12th 2017, ‘Yeonbit’ was registered (registration No. 6752) and is now protected by Korean Seed Industry Law.

        Yellow Moon Cactus ‘Ahwang’ Cultivar with Excellent Ability of Producing Offset

        비모란 선인장 ‘Ahwang’ 품종이 2016년에 국립원예특작과 학원에서 육성되었다. 증식력이 우수한 밝은 황색 ‘Ahwang’ 품종 육성을 위해서, 황색 ‘Hwangun’품종을 모본으로, 황색 ‘0930001’ 계통을 부본으로 하여 2012년 6월 25일에 교배하였다. 어린 비모란 선인장을 2014년 이전에 삼각주에 2번 접목하여 계통을 양성하였으며 2014년부터 2016년까지 총 3회에 걸쳐 생육특성을 조사하였다. ‘Ahwang’ 품종의 모구는 편원형 모양에 황색 구색(Y9A)이다. 모구는 평균 8.6개의 능(rip)과 2.7mm의 짧은 직립형 회색 가시를 가지고 있고 혹(tubercle)이 돌출된 형태를 띠고 있다. 10개월 재배 후 ‘Ahwang’ 품종의 구직경은 44.5mm였으며 자구는 평균 26.9개가 생성되었다. ‘Ahwang’ 품종은 모구 능마다 황색의 자구가 3-4개가 착생되었다. 2016년 육성계통 평가회에서 ‘Ahwang’ 품종은 기호도 점수 3.9을 받았다. 이 품종은 2018년 5월 16일 국립종자원에 등록되었으며 식물신품종보호법에 의해 품종보호(등록번호 7193)를 받게 되었다. The ‘Ahwang’ cultivar of the moon cactus was bred by the National Institute of Horticultural & Herbal Science in 2016. To breed the bright yellow, easily propagated ‘Ahwang’,’ the yellow maternal cultivar, ‘Hwangun,’ and the yellow paternal line, ‘0930001,’ were crossed on the June 25, 2012. Before 2014, a young moon cactus was grafted onto the vine cactus, Hylocereus trigonus, in vitro and in vivo, each once. The characteristics of growth were surveyed three times, from 2014 to 2016, according to the Korea Seed and Variety Service guidelines for Gymnocalycium mihanovichii. ‘Ahwang’ has a flat round shape, yellow skin (Y9A), and an average of 8.6 ribs. The spines on the body have a gray color. The spines were straight and 2.7 mm long on average. The tubercles on the rib were projected. This cultivar was 44.5 mm in diameter, and at 10 months after planting, produced an average of 26.9 yellow offsets. This cultivar produced three or four offsets on each rib. The preference score of this was 3.9 on a five-point scale. On the May 16, 2018, ‘Ahwang’ was registered (Registration No. 193) and is now protected by the Act on the Protection of New Varieties of Plants.

        저자들은 좌하복부 동통을 주소로 내원한 환자에서 대장 조영술, 대장 내시경, 하복부 전산화 촬영을 통해 S자 결장 게실염으로 진단하고 약물적 치료를 통해 호전되었으나, 지난 3년간 수회에 걸쳐서 게실염의 재발로 내과적 치료를 받은 병력과 최근 시행한 대장 조영술상 S상 결장의 누공이 관찰되어 재발성 게실염의 근치적 치료 목적으로 수술을 시행하여 합병증 없이 좋은 경과를 보였기에 이에 보고하는 바이다. Diverticular disease of the colon is quite common in developed countries, and its prevalence increases with age. The large majority of patients will remain entirely asymptomatic. Nonetheless, an estimated 20% of those affected may manifest clinical illness, mainly diverticulitis, with its potential complications of abscesses, fistulas, and obstruction as well as lower intestinal hemorrhage. A case was experienced involving of diverticulitis operated due to recurrent left lower abdominal pain in 51-year-old female. We present this case with brief review of the related literature.

        Separation of Human Breast Cancer and Epithelial Cells by Adhesion Difference in a Microfluidic Channel

        A simple, label-free microfluidic cell purification method is presented for separation of cancer cells by exploiting difference in cell adhesion. To maximize the adhesion difference, three types of polymeric nanostructures (50nm pillars, 50nm perpendicular and 50nm parallel lines with respect to the direction of flow) were fabricated using UV-assisted capillary moulding and included inside a polydimethylsiloxane (PDMS) microfluidic channel bonded onto glass substrate. The adhesion force of human breast epithelial cells (MCF10A) and human breast carcinoma (MCF7) was measured independently by injecting each cell line into the microfluidic device followed by culture for a period of time (e.g., one, two, and three hours). Then, the cells bound to the floor of a microfluidic channel were detached by increasing the flow rate of medium in a stepwise fashion. It was found that the adhesion force of MCF10A was always higher than that of MCF cells regardless of culture time and surface nanotopography at all flow rates, resulting in a label-free detection and separation of cancer cells. For the cell types used in our study, the optimum separation was found for 2 hours culture on 50nm parallel line pattern followed by flow-induced detachment at a flow rate of $300{\mu}l/min$.
