      • Reclassification of Bacillus beijingensis Qiu et al. 2009 and Bacillus ginsengi Qiu et al. 2009 as Bhargavaea beijingensis comb. nov. and Bhargavaea ginsengi comb. nov. and emended description of the genus Bhargavaea

        <P>We have carried out a polyphasic taxonomic characterization of <I>Bacillus beijingensis</I> DSM 19037<SUP>T</SUP> and <I>Bacillus ginsengi</I> DSM 19038<SUP>T</SUP>, which are closely related phylogenetically to <I>Bhargavaea cecembensis</I> LMG 24411<SUP>T</SUP>. All three strains are Gram-stain-positive, non-motile, moderately halotolerant and non-spore-forming. 16S rRNA gene sequence analyses showed that the strains constituted a coherent cluster, with sequence similarities between 99.7 and 98.7 %. The percentage similarity on the basis of amino acid sequences deduced from partial <I>gyrB</I> gene nucleotide sequences of these three type strains was 96.1-92.7 %. Phylogenetic trees based on the 16S rRNA gene and GyrB amino acid sequences, obtained by using three different algorithms, were consistent and showed that these three species constituted a deeply rooted cluster separated from the clades represented by the genera <I>Bacillus</I>, <I>Planococcus</I>, <I>Planomicrobium</I>, <I>Sporosarcina</I>, <I>Lysinibacillus</I>, <I>Viridibacillus</I>, <I>Kurthia</I> and <I>Geobacillus</I>, supporting their placement in the genus <I>Bhargavaea</I>. All three type strains have menaquinone MK-8 as the major respiratory quinone and showed similar fatty acid profiles. The main polar lipids present in the three type strains were diphosphatidylglycerol and phosphatidylglycerol, and the three strains showed peptidoglycan type A4α with l-lysine as the diagnostic diamino acid. The DNA G<I>+</I>C contents of <I>Bacillus beijingensis</I> DSM 19037<SUP>T</SUP>, <I>Bacillus ginsengi</I> DSM 19038<SUP>T</SUP> and <I>Bhargavaea cecembensis</I> LMG 24411<SUP>T</SUP> were 53.1, 50.2 and 53.7 mol%, respectively. The level of DNA-DNA hybridization among the three strains was 57-39 %, indicating that they are members of different species of the genus <I>Bhargavaea</I>. The phenotypic data are consistent with the placement of these three species in a single genus and support their differentiation at the species level. On the basis of these data, we have emended the description of the genus <I>Bhargavaea</I> and propose the reclassification of <I>Bacillus beijingensis</I> and <I>Bacillus ginsengi</I> to the genus <I>Bhargavaea</I>, as <I>Bhargavaea beijingensis</I> comb. nov. (type strain ge10<SUP>T</SUP> = DSM 19037<SUP>T</SUP> = CGMCC 1.6762<SUP>T</SUP>) and <I>Bhargavaea ginsengi</I> comb. nov. (type strain ge14<SUP>T</SUP> = DSM 19038<SUP>T</SUP> = CGMCC 1.6763<SUP>T</SUP>).</P>

        Bhargavaea indica sp. nov., a Member of the Phylum Firmicutes, Isolated from Arabian Sea Sediment

        A Gram-positive, aerobic, coccoid-rod shaped, non-motile, catalase- and oxidase-positive bacterium, designated strain KJW98T, was isolated from the marine sediment of Karwar jetty, west coast of India. The strain was β-haemolytic, nonendospore-forming and grew with 0–8.5% (w/v) NaCl, at 15–48°C and at pH 6.5–9.0, with optimum growth with 0.5% (w/v) NaCl, at 42°C and at pH 7.0–8.0. Phylogenetic analyses based on 16S rRNA and gyrB gene sequences showed that strain KJW98T forms a lineage within the genus Bhargavaea. The G+C content of the genomic DNA was 55 mol%. The DNA–DNA relatedness values of strain KJW98T with B. beijingensis DSM 19037T, B. cecembensis LMG 24411T and B. ginsengi DSM 19038T were 43.2, 39 and 26.5%, respectively. The major fatty acids were anteiso-C15:0 (37.7%), iso-C15:0 (19.7%), anteiso-C17:0 (17.0%) and iso-C16:0 (11.1%). The predominant menaquinone was MK-8 and the cell-wall peptidoglycan was of A4α type with L-lysine as the diagnostic diamino acid. The major polar lipids were diphosphatidylglycerol and phosphatidylglycerol. The phenotypic, genotypic and DNA–DNA relatedness data indicate that strain KJW98T should be distinguished from the members of the genus Bhargavaea, for which the name Bhargavaea indica sp. nov. is proposed with the type strain KJW98T (=KCTC 13583T=LMG 25219T).

        Magnetic molecularly imprinted polymer photocatalysts: synthesis, applications and future perspective

        Effective degradation of low-concentration pollutants is a critical challenge during the water purificationprocess. The use of different chemicals can leave residue in samples that can impose potential ecotoxicologicaland adverse impacts on human health. Presently, Magnetic molecularly imprinted polymers(MMIPs) have attracted much attention as the research material comprises a non-magnetic polymerand magnetic material for selective binding for target molecule and recoverability of catalyst via magnetism,respectively. This review explains the synergistic effect of adsorption with photocatalysis tounderstand their recognition mechanism and the possible interaction between the target molecule andMIPs. Then their common imprinting polymerization processes i.e., free radical polymerization andnon-free radical polymerization are briefly discussed with their respective advantages and disadvantages. In addition, this review highlights the photocatalytic degradation mechanism of photocatalyst cumadsorbent is critically discussed by comparing it with non-imprinted polymers. Finally, the applicationsof MMIPs in the removal or degradation of refractory pollutants, sensing, and recognition have alsobeen delineated. This paper summarises progressive future challenges of the technology that need tobe exploited for the preparation of the targeted catalyst.

      • Quitline Activity in Rajasthan, India

        Quitline activity in Rajasthan, India is a voluntary activity of Rajasthan Cancer Foundation (RCF) since April 2013. To kick-off, it took the benefit of the State Government- PIRAMAL SWASTHYA (PS)1 collaborative 104 Health Information Helpline that existed already in public-private partnership. It is a reactive quitline that helps callers through the counselors and nursing staff trained specifically through the weekly sessions held by the first author, the RCF resource on quitline. Besides structuring of the scripts for primary intervention and follow-ups after 1 week, 1 month, 6 months and a year, he also monitors calls, advices and coordinates with the supervisors to manage and analyze the data base, and reports to the PS lead at the Jaipur Center on overall performance and to plan strategic communication with the State Government on its outcomes. The quitline has limitations of its informal existence through a voluntary effort of RCF, no specific resource allocation, suboptimal data management, minimal awareness in the masses due to poor IEC (Information, Education and Communication; except its efforts made by RCF in last 1 year through the government-run State TV and City Radio) and staff shortage and its attrition due to lack of plan for career advancement. Despite these challenges in the year 2013, the quit line has registered a quit rate (for complete abstinence) of 19.93% amongst 1525 callers. The quit rate were 58.01% (304/ 524) among the responders at the 3rd follow-up at 18 months (in September 2014)2. In view of an increase in quit rate by 5- 9 times over the prevailing quit rate in the former ever daily users [both smokers and the users of smokeless tobacco (SLT)], efforts are being made by RCF in concurrence with PS to have this cost-effective model established formally with optimal resource allocation in collaboration with willing agencies (the State and Central Governments and the International Quitline Agencies) and its replication in 4 more states where PS is collaborating with the respective state governments similarly (Assam, Chhattisgarh, Jharkhand and Karnataka).

        Induction of oxidative stress and lipid peroxidation in rats chronically exposed to cypermethrin through dermal application

        Present study was undertaken to study the effect of cypermethrin on oxidative stress after chronic dermal application. The insecticide was applied dermally at 50 mg/kg body weight in different groups of Wistar rats of either sex weighing 150∼200 g. Significant (p < 0.05) increase in catalase activity was observed after 30 days of exposure. However, the superoxide dismutase activity declined significantly after 60 days of exposure. The activity of glutathione peroxidase and blood glutathione levels declined significantly (p < 0.05) after 30 days of cypermethrin dermal application. However, the activity of glutathione S-transferase increased significantly (p < 0.05) in all groups after 60 days of dermal exposure. Significant increase in lipid peroxidation was observed from 30 days onwards and reached a peak after 120 days of application.

      • Static and harmonic analysis of moderately thick square sandwich plate using FEM

        In this paper, sandwich plate, constructed with orthotropic and isotropic composite materials, is analyzed to obtain the static and harmonic behavior. The analysis is done by using ANSYS APDL FEM tool. A solid-shell 190 and an 8-node solid 185 elements are employed for face and core material respectively to analyze the plate. Results was attained by using Reissner-Mindlin theory. Effect of increasing thickness ratio of face sheet to depth of the plate is presented on static, vibration and harmonic response on the sheet and the results are discussed briefly. Published work in open domain was used to validate the results and observed excellent agreement. It can be stated that proposed model presents results with remarkable accuracy. Results are obtained to reduce the weight of the plate and minimizing the vibration amplitudes.

        Pharmacokinetics of amikacin in plasma of healthy goats after intravenous injection once daily for three days

        Amikacin is a semisynthetic derivative of kanamycin and primarily active against aerobic Gram-negative-pathogens with limited activity against Gram-positive bacteria. Meager study was reported on pharmacokinetic data on multi-days administration of amikacin. Hence, pharmacokinetics study was done in five clinically healthy goats (n = 5), after intravenous bolus injection of amikacin sulfate at the dose rate of 10 mg/kg body weight daily for three consecutive days. The amikacin concentrations in plasma and pharmacokinetics-parameters were analyzed by using microbiological assay technique and noncompartmental open-model, respectively. The mean peak plasma concentrations (Mean ${\pm}$ SD) of amikacin at time zero ($Cp^{0}$) was $114.19{\pm}20.78$ and $128.67{\pm}14.37{\mu}g/mL$, on day 1st and 3rd, respectively. The mean elimination half-life ($t_{1/2}ke$) was $1.00{\pm}0.28h$ on day 1st and $1.22{\pm}0.29h$ on day 3rd. Mean of area under concentration-time curve ($AUC_{0{\rightarrow}{\infty}}$) was $158.26{\pm}60.10$ and $159.70{\pm}22.74{\mu}g.h/mL$, on day 1st and 3rd respectively. The total body clearance ($Cl_{B}$) and volume of distribution at steady state (Vdss) on day 1st and 3rd were $Cl_{B}=0.07{\pm}0.02$ and $0.06{\pm}0.01L/h.kg$ and $Vdss=0.10{\pm}0.03$ and $0.11{\pm}0.05L/kg$, respectively. No-significant difference was noted in both drug-plasma concentration and pharmacokinetics-parameters, respectively. Amikacin concentration in plasma was found higher up-to 4 h and 6 h onward on down-ward trends favour to reduce toxicity. Which also support the pharmacokinetic-pharmacodynamic way of dosing of aminoglycosides and hence, amikacin may be administered 10 mg/kg intravenously daily to treat principally Gram-negative pathogens and limitedly Gram-positive-pathogens.

        Disposition kinetics and urinary excretion of ciprofloxacin in goats following single intravenous administration

        We evaluated the pharmacokinetics of ciprofloxacin in serum (n = 6) and urine (n = 4) in goats following a single intravenous administration of 4 mg/kg body weight. The serum concentration-time curves of ciprofloxacin were best fitted by a two-compartment open model. The drug was detected in goat serum up to 12 h. The elimination rate constant (β) and elimination half-life (t1/2β) were 0.446 ± 0.04 h-1 and 1.630 ± 0.17 h, respectively. The apparent volume of distribution at steady state (Vdss) was 2.012 ± 0.37 l/kg and the total body clearance (ClB) was 16.27 ± 1.87 ml/min/kg. Urinary recovery of ciprofloxacin was 29.70% ± 10.34% of the administered dose within 36 h post administration. In vitro serum protein binding was 41% ± 13.10%. Thus, a single daily intravenous dose of 4 mg/kg is sufficient to maintain effective levels in serum and for 36 h in urine, allowing treatment of systemic, Gram-negative bacterial infections and urinary tract infections by most pathogens.
