        Enhancing the Kasugamycin Production by Streptomyces kasugaensis using Olive Mill Waste

        The purpose of this study was to produce the kasugamycin by using Streptomyces kasugaensis with olive mill waste containing oil as an energy source. Among the various concentrations of oil, only initial concentration of 20 g/l produced the highest concentration (125.7 mg/l) of kasugamycin. This result indicates that the kasugamycin production was strongly dependent with oil concentrations. Among these fatty acids, only oleic acid (0.1 g/l) produced the highest concentration of kasugamycin 69.7 mg/l, which resulted in 3.4 fold increase in the production than the absence of oleic acid. The corn steep liquor concentration (30 g/l) was found to be the most suitable source for kasugamycin production. Also, fed batch cultures using olive mill waste containing oil and glucose as the sole carbon source were carried out in 51 jar fermentor for seven days. In the case of olive mill waste the maximum kasugamycin concentration was 426.8mg/l, which was about 2.7 fold higher than that of the glucose medium.

        Production of α-amylase from Bacillus megaterium in the Air Lift Bioreactor Using Corn Starch as Sole Carbon Source

        This research was to investigate the effective production of α-amylase from a culture of Bacillus megaterium H-829 using corn starch as the carbon source and corn steep liquor (CSL) as the nitrogen source in air-lift bioreactor. α-amylase production was increased from 10.5 to 49.1 DUN/mL when corn starch concentration was increased from 5 to 15 g/L. However, when corn starch concentration was increased from 20 to 30 g/L, the enzyme production was decreased from 44.7 to 28.5 DUN/mL. In the case of CSL concentrations, while CSL concentration was increased from 2.5 to 5.0 g/L the α-amylase production was increased from 48.9 to 58.6 DUN/mL. However, it was decreased from 55.4 to 25.4 DU /mL when CSL concentration was increased from 7.5 to 15.0 g/L. The cell concentration was decreased with the increase of acetic acid concentration produced. Especially, when acetic acid concentration was over 1.5 mg/L, it was sharply decreased. In order to compare the effects of corn starch and glucose on the production of amylase in the air-lift bioreactor, batch cultures were carried out in a 5 L air-lift bioreactor for 48 hr using production media containing corn starch 15 g/L and CSL 5 g/L respectively. When corn starch was used, the maximum α-amylase production after 36 hr of culture was 40 DUN/mL, which was similar to that of the glucose medium. These results indicate that corn starch and CSL can be useful for α-amylase production from Bacillus megaterium H-829 in air-lift bioreactor.

        Proximate Composition, Mineral, Amino Acid, and Antioxidant Component from Fruiting Body of Pleurotus ostreatus Mutant Cultivated on Agricultural and Industrial By-Product

        The purpose of this study was to investigate the proximate composition, mineral amino acid, and antioxidant component from fruiting body of Pleurotus ostreatus mutant cultivated based on agricultural and industrial by-product. The concentrations of dry matters, crude ash, crude protein, crude fat, and organic matter in PR media were 91.2, 5.3 23.4 4.8, and 84.6%, respectively. In the ca e of CAPR media, they were 91.4, 5.0, 25.1, 5.0, and 84.9%, respectively. The mineral concentrations followed the order of potasssium> phosphate calcium> sodium> iron> magnesium. The most major components of essential amino acids were lysine, leucine, and valine in PR and CAPR media. On the other hand, in the case of non­essential amino acids, they were aspartic acid and glutamic acid, respectively. The total phenol concentration in PR and CAPR media were 15.5 and 16.7 mg/g, respectively. The total flavonoid concentration in PR and CAPR media were 1.2 and 1.3 mg/g, respectively. In the case of polysaccharide, they were 2.3 and 2.2 mg/g, respectively. Overall, these results indicate that fruiting bodies of P. ostreatus mutant have been good potential as a source of material or additives for food, cosmetics, and drug compositions, in the agricultural and industrial sector.

        A study on liquid culture of Pleurotus florid Mutant L-456 using waste vegetable oil containing high fatty acid

        This research was to investigate the liquid culture of Pleurotus florid Mutant L-456 using waste vgetable oil containing high fatty acid for mycelial growth and exopolysaccharde production. The optimal temperature ranges for mycelial growth werefound to be 25-30 . The optimal initial pH ranges of medium ℃ for mycelial growth and exopolysaccharide production were found to be 6.5-7.0. When glucose (2%) and vegetable oil (0.5%) mixture as the carbon source was used, the maximum mycelium growth and exopolysaccharide production were obtained, 4.9 and 3.0mg/L, respectively. Among various nitrogen sources, when peptone and yeast extract were used, the maximum mycelial growth and exopolysaccharide production were 5.1-5.2 and 3.1-3.6/L, respectively. This results shows that waste vgetable oil can be used as carbon source for effective mycelial growth and exopolysaccharde production from Pleurotus florid Mutant L-456.

        Effect of Astragalus sinicus L. seed extract on antioxidant activity

        Antioxidant activity of various extracts of Astragalus sinicus L. seed was evaluated in vitro and in vivo. Among the various extracts, the acetone extract showed the maximum DPPH radical scavenging activity of 95.1% at 10.0 mg/mL, hydroxyl radical scavenging activity of 89.4% at 15 mg/mL, and superoxide anion scavenging activity of 74.5% at 15 mg/mL. The inhibition of xanthine oxidase and chelating effect at 10.0 mg/mL of acetone extract were found to be 95.1 and 93.2%, respectively. The concentrations of total phenolic and flavonoide were, respectively, 279.9 and 175.2 mg/g in the acetone extract, about 4.1 and 3.4 times higher than those of the water extract. When the administered concentration of acetone extract was 5.0 g/kg/body weight/day in rats, catalase, superoxide dismutase, and glutathione peroxidase activities of the liver were increased approximately 50% compared to those of the control groups. These results suggest that the acetone extract of A. sinicus L. seed might be a valuable antioxidant of natural source applicable in the medicine, cosmetics and the food industry.

        Tissue engineering strategies have the potential to improve upon current techniques for intervertebral disc repair. However, determining a suitable biomaterial scaffold for disc regeneration is difficult due to the complex fibrocartilaginous structure of the tissue. Poly(lactide-co-glycolide)(PLGA) has controllable biodegradability and good biocompatibility. hyaluronic acid(HA) called natural moisturizer can be used as biomaterials. In this study, we evaluated 3-dimensional HA loaded PLGA scaffold on proliferation and phenotype maintenance of annulus fibrosus cells. HA/PLGA scaffolds were prepared by emulsion freeze-drying method. Annulus fibrosus cells were seeded in HA/PLGA scaffold, and then cell viability and proliferation analyzed by 3-(4,5-Dimethylthiazol-2-yl)-2,5- diphenyltetrazolium bromide(MTT) assay. Reverse transcription polymerase chain reaction(RT-PCR) was assessed to measure mRNA expression for type II collagen, aggrecan and glycosaminoglycan(GAG) quantity from annulus fibrosus cells in scaffolds. In MTT assay result, cell viability in HA/PLGA scaffolds were higher than PLGA scaffolds. In addition, annulus fibrosus strongly expressed their specific mRNA and produced well sGAG in HA/PLGA scaffold. This result indicates that HA/PLGA scaffold is useful for intervertebral disc regeneration.

        Effect of Solanum Melongena Extract on the Xanthine Oxidase Inhibitory, Nitrite Scavenging, and Angiotensin I-Converting Enzyme Inhibitory Activities

        This research was to study effect of Solanum melongena extract on the xanthine oxidase (XO) inhibitory, nitrite scavenging (NC), and angiotensin I-converting enzyme (ACE) inhibitory activities. All the extracts inhibited the XO and ACE activities in a dose dependent manner. Especially, among various extracts, when 6.0 mg/mL of acetone extract was used, the maximum activity of XO inhibitory was obtained, 65.2%. On the other hand, in the case of methanol and ethanol extracts, they ranged of 61.5 to 62.4%. Acetone extract had a high phenolics and flavonoid concentration compared to other extraxts. Maximum NC activity using hot water extract at pH 1.2 was occurred, 68.7%. ACE inhibitory activity was significantly different among the various extracts at 6.0 mg/mL, values ranged from 35.26 to 56.7%. Especially, when 6.0 mg/mL of methanol and ethanol extracts were used, the highest ACE inhibitory activities were obtained, 57.3 and 56.7%, respectively. These results suggest that XO inhibitory, NC, and ACE inhibitory activities of S. melongena extract were strongly affected by extractant.

        Effect of polysaccharide of Salicornia herbacea on antioxidant and nitrite scavenging effects

        This research is to study effect of polysaccharide obtained from S. herbacea on antioxidant and nitrite scavenging effect. Neutral sugar, uronic acid, and protein concentrations of polysaccharide were 52.68, 0.54, and 7.21%, respectively. Among neutral sugars, glucose and mannose were the major ones of the sugar moiety with molar ratio of 14.52 and 11.28, respectively. The maximum DPPH radical scavenging, hydroxyl radical scavenging, chelating, and hydrogen peroxide scavenging activities at 7 mg/mL were obtained, 69.04, 58.07, 60.21, and 62.54%, respectively. However, in the case of reducing power, it was OD 0.62 at 9.0mg/mL. The maximum nitrite scavenging activity was obtained, 76.77 % at 9 mg/mL and pH 1.5. These results indicate that the polysaccharide obtained from S. herbacea has good potential to be used as a source of materials or additives for oxidation suppressant in food, cosmetics and drug compositions.

        갯장어 및 버섯 혼합추출물의 아토피 및 여드름 피부개선 효과

        본 연구에서는 갯장어 및 버섯 혼합추출물을 이용하여 아토피 피부염 및 여드름의 개선효과를 확인하기 위하여 항산화 테스트와 아토피 피부염 임상시험자 15명에게 4주 동안 혼합추출물을 복용과 함께 환부에 도포하도록 하였다. DPPH 소거능은 혼합추출물 농도증가와 함께 증가하였다. 특히 혼합추출물농도가 1.0 mg/L에서 50 mg/L로 증가 할 경우 DPPH 소거능은 10.1에서 81.4%로 증가 하였다. 환원력도 추출물농도가 증가 함에 따라 증가 하였다. 특히 추출물 25 mg/L에서 75 mg/L로 증가 할 경우 0.05 OD (700 nm)에서 0.27 (700 nm)로 급격히 증가 하였고 추출물 농도가 100 mg/L일 경우는 0.31 (700 nm)였다. SOD활성은 추출물 농도가 100 mg/L일 경우는 45.7%였다. 아토피성 피부염의 가려움과 붉은 반점에 대하여 임상 시험 결과 임상시험자 모두 완치에 가깝도록 개선되는 효과를 나타내었다. 여드름 피부의 임상시험자 5명 모두 임상시험 후 염증 면포가 완화되어 여드름의 등급이 낮아지는 결과를 나타내었다. 여드름 피부의 임상시험자 모두 얼굴피부의 유수분 및 거칠기 상태가 정상피부의 수준에 이르도록 긍정적인 변화를 나타내었다. 혼합추출물을 피부에 직접 적용하여 임상시험한 결과, 아토피 피부염 및 여드름 피부의 임상시험자 모두 어떠한 부작용도 나타내지 않았다. 이상의 결과는 갯장어 및 버섯 혼합추출물이 아토피 피부염 및 여드름 피부관리를 위한 의약품 및 화장품의 원료로 개발될 충분한 가치가 있는 것으로 사료된다. This study was conducted to investigate effect of complex extract of various mushrooms and sharp toothed eel on the skin conditions with atopic dermatitis and acne symptoms. The total phenol concentration was increased in order of hot water> ethanol> ethyl acetate> petroleum ether> chloroform extract. Especially, when the hot water extract was used, it was about 2-3 fold higher than that of ethyl acetate, petroleumether, and chloroform extract. When the complex extract concentration was increased from 1.0 to 50 mg/L, the DPPH scavenging rate increased from 10.1 to 81.4%. The reduction power was sharply increased from 0.05 to 0.27 (700 nm) when the complex extract concentration was increased from 25 to 75 mg/L. However, above 100 mg/L, it was not decreased. In the case of SOD-like activity, it was 45.7% at 100 mg/L. Total numbers of patients with atopic dermatitis were 15 and 5 patients with severe acne symptoms. According to photos taken before and after the treatment and questionnaire results, considerable improvements in skin conditions are observed in the patients with atopic dermatitis and acne. For atopic patients, erythema and edema have been improved but the degree of effect was dependent on the individual’s constitution. Concerning acne, the effect of coating of the extracts was prominent for first week and the degree decreased with time till 4 weeks. The complex extracts wereefficient in soothing rash and maturation. Side effects such as a scar were not detected during the application and treatment.
