발생 및 재생중인 흰쥐 간세포에 Actinomycin-D가 미치는 영향

This experiment observed ultrastructural exchange of developing and regenerating rat hepatocyte injected Actinomycin-D. The result are as fllowed. 1) After inject Actinomycin-D protein synthesis needing hepatocyte differation inhibited, mitochondra damage, RER nomber decrease, nucleous type was irregular and chromatin in nuclear becames dense. 2) After inject Actinomycin-D microorganelles damaged but amount of glycogen did'nt reduce. 3) After inject Actinomycin-D hepatocytes differation inhibited.

高麗末 士大夫의 儒佛一致論과 그 意義

The adoption of Sung-Confucianism brought about a huge ideological change at the end Koryo Dynasty, which is believed to be no less significant change than the replacement of dynastyideological, social transition from the Buddhism-centered society to confucianism-centered one. However, the understandings about this ideological change had been limited to the scope of confrontation between Confucianism and Buddhism. Therefore, it seems within the realm of idea that promoted such an ideological change. So, this dissertation has focused on approaching what the mutual relation between Confucianism and Buddhism was like through the Sa-Dae-Boo(士大夫)'s view on Buddhism. Especially, more focus has gone to the way the idea of harmony was realized. Because Buddhism had totally different values and outlook on the world from the traditional Chinese ideas in the process of its introduction into China, inevitable confronfation and conflict were generated. These confrontation and conflict emerged though the problem regarding such a problem as "Filial Piety" which had been placed ahead of any other value in Chinese society. Buddhism often insisted on its ideological superiority to other religions in dealing with actual challenge from Confucianism and Taoism. As Buddhism started moving forward under the banner of "harmony", and groped its way for "co-existence", however, the idea of harmony of three ideas-Confucianism, Buddhism, Taoism-was spreading widely. This trend was prevalent distinctively when the formation of Sung-Confucianism was underway during Song Dynasty. Especially, the relationship between Buddhism and Confucianism was groping its way for a compromise and harmony based on the idea of Confucian-Buddhist harmony. Such a trend is attributed to the historical background that Buddhism was absolutely subordinate to the nation at the time that Buddhists had no choice but to represent the interests of Sa-Dae-Boo(士大夫) group which took the lion's share of the society both politically and culturally. However, the idea of Confucian-Buddhist harmony had such limitations like failing to get out of the conventional scope, taking passive measure toward the actual interests. In contrast, the idea of Confucian-Buddhist harmony at the end of Koryo came across more active and practical in the matter. In the realm of idea during the end of Koryo, Sung-Confucianism was taking root gradually through the contact with Yuan(元), and criticism on Buddhism was drawn along with social reform policy. In some other sense, Koryo's realm of idea was being largely influenced from liberal and open-minded intellectual atmosphere of Yuan(元) during the early 14th century. So, Sa-Dae-boo group, sharing such intellectual, cultural experiences, fundamentally had positive views on Buddhism and was groping its way for the harmony of Confucianism and Buddhism. Even in the late 14th century, when Sung-Confucianism was being pervasive in the society and the critical voice toward Buddhism was mounting, the idea of Confucian-Buddhist harmony was consistently taking its place. The reason is traced to the fact that the understandings and practice about Buddhism, especially Zen(禪), were shared inside the group of Sa-dae-boo as a ideological basis in understanding a new idea-Sung-Confucianism.

Helicobacter pylori와 대장균의 Shuttle Vector 개발

In this study, a vehicle vector using cryptic plasmids was constructed for gene transfer in Helicobacter pylori. pHP51(3.9 kb) and pHP489(1.2 kb) were selected for constructing vectors from cryptic plasmid of H. pylori isolates in Korea. The HindⅢ-digestedDNA fragment(1.2kb) of pHP489 and 1.6kb DNA fragment of pHP51 were ligated with a kanamycin resistance gene(aph3'-Ⅲ) from C. jejuni to produce the recombinant plasmids pHP489K and pHP51K, respectively. Transformation frequency of pHP51K by electroporation was low. But pHP489K could be effectively transformed into various H. pylori strains. In order to design an intermdiate vehicle vector for gene transfer into H. pylori, pBlueHP489K was prepared by recloning pHP489K DNA into pBluescript and pTZ19R vector. This vector permitted the DNA fragment containing pHP489 sequence, aph3'-Ⅲ, and cloned DNA to be cut and self-ligated in the SacⅠ site after cloning. ureA and ureB gene were inserted into pBlueHP489K, resulting in pBlueHP489K/AB. The DNA fragment containing pHP489, kanamycin resistance gene(aph3'-Ⅲ), and urease structural gene was cut away from pBlueHP489K/AB and self-ligated to generate pBlueHP489K/AB. pBlueHP489K/AB made urease-negative H. pylori strains restore their urease activity. By this experiment, pBlueHP489K was confirmed to be the vehicle system for transferring H. pylori genes.

강제 수영이 흰쥐 골격근에 미치는 영향

This experiment was designed to observe the changes in skeletal muscle(gastrocnemius) of the rat in the swimming training. The subject were divided into three groups : control, 4 weeks swimming and 8 weeks swimming trained group. Myofibrils in the muscle fiber were disaggregated of the forced swimming, although mitochondria between myofibrils were fused with each other and because gigant mitochondria, but the density of cristae decreased. Mitochondria between sarcolemma and myofibrils were divided and thus increased in number. Sarcoplasm was increased, and disaggregated membrane system was observed in swelling formation. Muscle fibers in the skeletal muscle were damaged by stress when the skeletal muscle was affected by forced swimming. The increase in number of mitochondria indicates positive adaptation of the skeletal muscle when training is prolonged.

랫드에서 l-muscone의 급성독성 및 아급성독성시험 연구

l-Muscone is synthesized for use as substitutive material of musk which is the active ingredient of woohwangchungsimwon. The objective of this investigation was to evaluate the acute and subacute toxicity of l-muscone in rats. In oral acute toxicity test, SPF Sprague-Dawley male and female rats were gavaged with l-muscone of two doses(0, 5.0g/㎏). No dead animal and abnormal autopsy findings were found in control and treated group. Body weights were slightly decreased in both sexes of rats treated with 5.0g/㎏. Therefore, oral LD_50 of l-muscone was consider to be higher than 5.0g/㎏ in male and female rats. In intraperitoneal acute toxicity test, rats were injected intraperitoneally with dosages of 0, 1,000, 1,316, 1,732, 2,279 and 3,000 ㎎/㎏. Decreased body weights and motor activities were observed at high dose group. Intraperitoneal LD_50 of l-muscone were 1,920 ㎎/㎏ in male and female rats. In the subacute study. l-muscone was administrated orally to both sexes of rats for 4 weeks as several doses(0, 10, 100 and 1,000 ㎎/㎏). There were neither dead animals nor significant changes of body weights during the experimental period. In addition, no differences were found between control and treated groups in clinical signs, urinalysis, hematology, serum biochemical analysis, and other findings. Above data suggest that no observed adverse effect level of l-muscone in rats might be over 1,000 ㎎/㎏/day in this study.

Helicobacter pylori 편모 유전자의 클로닝 및 염기서열 분석

A λgt11 expression libary of H. pylori DNA in E. coli Y1090 was screened with flagellin-specific rabbit antiserum for molecular cloning of the flagellin gene of H. pylori. A positive clone, λHPF4, was obtained and the recombinant antigen expressed from λHPF4 was a fusion protein with the molecular weight of 168kd. Sequence analysis of antigen-encoding DNA showed that an open reading frame composed of 1,536 nucleotides encodes a polypepride with a oredicted molecular size of 54kd. This open reading frame did not show the homology with flaA gene encoding 56kd protein of H. pylori and was confirmed as a unique sequence through homoligy searching. Therefore, the cloned antigen is supposed to be the carboxy-terminal region of the other flagellin protein of H. pylori, flaB, with the molecular weight of 58kd.

Some practical plans are presented to construct a sound life by enjoying sound leisure activitives chiefly consisting of sports good for health, not amusements which are nonproductive, consumptive and pleasure-seeking, which are prevalent in our society today. By the way, in order that these plans should be widely spread as the nation-wide social movement, we should create the atmosphere of sound life by having people realize the basic idea of the sound life campaign and inducing them to participate in the sports activities voluntarily. Therefore, we could not only enhance the quality of individual life, but also fix the sound life culture from the national standing by sublimating the sound life campaign as the centripetal campaign for the community harmony, and enjoying the healthy life thanks to the participation in lots of leisure activities chiefly consisting of sports activities. Based on the above-mentioned basic direction and policy of leisure activities, some practical plans for the sound life are summarized as follows; 1. A high dimensional leisure culture should be fixed in order to fulfill the future-oriented man's hopeful expectations for tomorrow. 2. Sound life campaign should be encouraged independently, and be gradually converted to the voluntary, nongovernmental social movement from the government-oriented policy. 3. More cultural facilities which we can utilize with small expense should be continuously built in a long-term project, and they should be equipped with lights and opened to the common people during the night. 4. We should enjoy the new and fresh taste of life which we can't feel in our routine lives by resorting to the natural environments, such as mountains, beaches, parks, and greenbelt area. 5. Recreational sports, which are sound and enjoyful, should be encouraged instead of the competitive sports. 6. It is quite natural that we pay attention to the juvenile problem, but we should develop and encourage the sports program for the old people against the senescent society together with the programs for the neglected, housewives and the handicapped.

랫드에서 신우황청심원의 급성 및 아급성독성시험

The acute and subacute toxicity of New Woohwangchungsimwon(NWCH) which was used l-muscone as substitutive material of musk were investigated in S.D. rats. In intraperitoneal acute toxicity test, rats(Sprague-Dawley, SPF) were injected intraperitoneally with dosages of 0, 540, 750, 1,070, 1,500 and 3,000 ㎎/㎏. Body weights were significantly decreased at 540 ㎎/㎏ dose group in both sexes and abnormal autopsy findings were founded in both sexes at all treated groups. Intraperitoneal LD_50 of NWCH was 812.3 ㎎/㎏ in male and 872.3 ㎎/㎏ in female rats. In the subacute toxicity study, NWCH was administrated orally to both sexes of rats for 4 weeks as several doses(0, 320, 800 and 2,000 ㎎/㎏). There were neither dead animals nor significant changes of body weights during the experimental period. In addition, no differences were found between control and treated groups in clinical signs, urinalysis, hematology, serum biochemical analysis, and other findings. Above data strongly suggest that no observed adverse effect level of NWCH might be over 2,000 ㎎/㎏/day in this study.

칼라 히스토그램과 엔트로피를 이용한 동영상 장면전환 검출

In content-base video data retrieval, the detection for scene change is needed. Color histogram approaches are generally used. Color histogram approaches are simple and more robust to object and camera movements but they ignore the spatial information and fail when two different frames have similar histograms. We propose a new algorithm to use color histogram and entropy to enhance the false-positive occurred by brightness variation and the miss by similar color histogram. At the experiments, it is more robust to brightness variation and have spatial information.
