Malnutrition worsens fluorosis-induced damage in hypothalamic-pituitary-ovarian axis of rats

        Yongmei Liu,Ling Li,Jingfeng Xu,Siwen Yu,Shijun Wang,Maojuan Yu,Wenbing Zou,Mingliang Cheng,Shuhua Xia 대한독성 유전단백체 학회 2019 Molecular & cellular toxicology Vol.15 No.2

Backgrounds: This study aimed to evaluate the effects of malnutrition on the hypothalamic-pituitary-ovarian axis (HPOA) sex hormones in female rats with coal burning-type fluorosis. Methods: Female rats were divided into four groups: control, malnutrition, fluorosis, and fluorosis with malnutrition. Rats in the control and malnutrition groups were fed pollution-free corn with either regular or low protein content. Rats in the fluorosis and fluorosis with malnutrition groups were fed corn roasted with coals from the fluorosis endemic areas of Zhijin, China, with either regular or low protein content. Results: Results revealed that the body weight of rats with protein malnutrition was significantly reduced compared with that of the control and fluorosis rats. Urinary fluoride was significantly decreased and bone fluoride was significantly increased in the fluorosis with malnutrition group compared to the fluorosis group. Moreover, protein malnutrition significantly enhanced the effect of fluoride on gonadotropin-releasing hormone, follicle-stimulating hormone, luteinizing hormone, and testosterone. Histological and ultrastructural analyses revealed that protein malnutrition intensified fluoride-induced ovary damage. Conclusion: Malnourishment could promote the abnormal secretion of HPOA sex hormones in females with fluorosis.

Keratin-based drug-protein conjugate with acid-labile and reduction-cleavable linkages in series for tumor intracellular DOX delivery

Huifang Zhang,Mingliang Pei,Peng Liu 한국공업화학회 2019 Journal of Industrial and Engineering Chemistry Vol.80 No.-

PK-SS-Hy-D drug-protein prodrug was obtained by conjugating doxorubicin (DOX) onto the PEGylatedkeratin (PK) with both bioreducible disulfide linkage and acid-cleavable hydrazone bond in a seriesconnection mode. The controlled release profiles demonstrated the pH and reduction dual-responsivetriggered release of DOX from the proposed drug-protein conjugate nanoparticles, with a low prematuredrug leakage of 5.5% in the simulated physiological medium. The in vitro experiments indicated that theproposed prodrug nanoparticles could delivery DOX into the cell nuclei, with an enhanced anti-tumorefficacy. It is expected as a potential candidate for future tumor chemotherapy with minimized toxic sideeffect.

Non-Contrast Cine Cardiac Magnetic Resonance Derived-Radiomics for the Prediction of Left Ventricular Adverse Remodeling in Patients With ST-Segment Elevation Myocardial Infarction

        A Xin,Liu Mingliang,Chen Tong,Chen Feng,Qian Geng,Zhang Ying,Chen Yundai 대한영상의학회 2023 Korean Journal of Radiology Vol.24 No.9

Objective: To investigate the predictive value of radiomics features based on cardiac magnetic resonance (CMR) cine images for left ventricular adverse remodeling (LVAR) after acute ST-segment elevation myocardial infarction (STEMI). Materials and Methods: We conducted a retrospective, single-center, cohort study involving 244 patients (random-split into 170 and 74 for training and testing, respectively) having an acute STEMI (88.5% males, 57.0 ± 10.3 years of age) who underwent CMR examination at one week and six months after percutaneous coronary intervention. LVAR was defined as a 20% increase in left ventricular end-diastolic volume 6 months after acute STEMI. Radiomics features were extracted from the oneweek CMR cine images using the least absolute shrinkage and selection operator regression (LASSO) analysis. The predictive performance of the selected features was evaluated using receiver operating characteristic curve analysis and the area under the curve (AUC). Results: Nine radiomics features with non-zero coefficients were included in the LASSO regression of the radiomics score (RAD score). Infarct size (odds ratio [OR]: 1.04 (1.00–1.07); P = 0.031) and RAD score (OR: 3.43 (2.34–5.28); P < 0.001) were independent predictors of LVAR. The RAD score predicted LVAR, with an AUC (95% confidence interval [CI]) of 0.82 (0.75–0.89) in the training set and 0.75 (0.62–0.89) in the testing set. Combining the RAD score with infarct size yielded favorable performance in predicting LVAR, with an AUC of 0.84 (0.72–0.95). Moreover, the addition of the RAD score to the left ventricular ejection fraction (LVEF) significantly increased the AUC from 0.68 (0.52–0.84) to 0.82 (0.70–0.93) (P = 0.018), which was also comparable to the prediction provided by the combined microvascular obstruction, infarct size, and LVEF with an AUC of 0.79 (0.65–0.94) (P = 0.727). Conclusion: Radiomics analysis using non-contrast cine CMR can predict LVAR after STEMI independently and incrementally to LVEF and may provide an alternative to traditional CMR parameters.

Study on the Influence of Stiffeners on the Wind Resistance Performance of Aluminum Panel Curtain Wall

        Cun Hui,Siyan Wang,Mingliang Liu,Weichi Han,Ran Hai 한국강구조학회 2022 International Journal of Steel Structures Vol.22 No.4

In order to study the infl uence of stiff eners on the wind resistance performance of the aluminum panel curtain wall unit (APCU), two APCU specimens with diff erent structures were designed and fabricated. The deformations under positive and negative wind loads were studied. The defl ection, stress, and mode were analyzed by fi nite element software. The results indicate that the stiff eners and the aluminum panels work well with each other under positive and negative wind pressure. Adequately designed stiff eners can eff ectively improve the wind resistance of the APCU, reduce the deformation of the aluminum curtain wall, and make the stress distribution uniform. The shedding of aluminum welding nails has a signifi cant impact on the overall wind resistance performance of the component. The aluminum panels and the stiff eners reduce the wind resistance performance of the APCU when the aluminum panels and stiff eners are not fi rmly connected. The modal analysis points out the weak point of the specimens. The fi nite element calculations and analysis results are consistent with the actual measurement results. The research results can guide engineering applications. The research in this paper can provide guidance for wind resistance of aluminum plate, defl ection deformation of stiff ening rib, failure of aluminum plate under wind pressure and engineering research of aluminum curtain wall system under wind load.

Amphiregulin secreted by cartilage endplate stem cells inhibits intervertebral disk degeneration and TNF-α production via PI3K/AKT and ERK1/2 signaling pathways

        Chen Qin,Wu Yaohong,Zhong Mingliang,Xu Chanhua,Chen Rongchun,Liu Ning 대한독성 유전단백체 학회 2023 Molecular & cellular toxicology Vol.19 No.2

Background Intervertebral disk degeneration (IDD) is a common orthopedic disorder, and nucleus pulposus cells (NPCs) serve to stabilize the intervertebral disk. Objective This study was implemented to explore the protective eff ect of endplate stem cells (EPSCs) on IDD. NPCs and EPSCs were isolated from rats and tert -Butyl peroxide (TBHP) was used to treat NPCs and simulate IDD. EPSC-derived conditioned medium (CM) was collected and mixed with a culture medium at diff erent proportions, which was then used to treat NPCs. SOD assay was employed to examine the oxidative stress level in the NPCs. To assess apoptosis and caspase-3 activity, fl ow cytometry and western blots for cleaved PARP1 and BAX were carried out. The mRNA expression levels of diff erent genes were detected by quantitative real-time polymerase chain reaction. Furthermore, the concentrations of amphiregulin (Areg) and infl ammatory factors were measured with ELISA kits and cell signaling pathways were evaluated using western blotting. Results Following TBHP treatment, caspase-3 activity in the NPCs increased. However, exposing TBHP-induced NPCs to the EPSC-derived CM reduced oxidative stress, caspase-3 activity, cell apoptosis, fi brotic response, and cytokine production. Additionally, the EPSC-derived CM contained a higher concentration of Areg compared to that in the NPCs. Furthermore, Areg decreased oxidative stress, cell apoptosis, fi brosis, and infl ammation via both PI3K/AKT and ERK1/2 signaling pathways. Conclusion ECSCs reduce NPC apoptosis, oxidative stress, production of infl ammatory factors, and fi brotic response via the release of Areg. Moreover, Areg can exert these protective eff ects on NPCs via PI3K/AKT and ERK1/2 signaling pathways.
