Proceedings of international Symposium on East-West Medicine, Seoul. 44-64, 1999.-Cordyceps sinensis is a major parasitic fungus on larva of Lepidoptera. It is one of the well know fungi used in traditional Chinese medicine for treatment asthma, lung and kideney diseases. Many fungi belonging to the genus Cordyceps have been demonstrated to produce natural products with various biological activities. Reported biological activities of genus cordyceps include:(a)inhibition of DNA and RNA synthesis;(b)enhancement of cell differentiation;(c)restruction of cytoskeleton;(d)inhibition of protein kinase activity;(e)antitumor activity on bladder, colon, lung carcinoma as well as fibroblastoma;(f)inhibition of the infection and revers transciptase activity of human immunodeficiency virus type 1; and(g) inhibition of methylation of nucleic acid. Thus, it is suggested that the extracts of Cordyceps sp. Contain antitumor, antiviral, and many other biological activities. In our previous studies, we proved that C. sinensis contains the antitumor and immunomodulatory agents. In the present studies, effects of C. sinensis on bronchoalveolar lavage fluids(BAL)cells were demonstrated. The crude methanolic extracts of C. sinensis were fractionated into 15 fractions by silica gel column chromatography. Effects of various fraction on BAL cells proliferation and lipopolysaccharide(LPS) stimulated interleukin-1β(IL-8),platelet activating factor(PAF), interleukin-6(IL-6), tumor necrosis factor-α(TNF-α),and interleukin-8(IL-8) production on BAL cells were determined. The result indicated that the CS-19-22 fraction suppressed BAL cells proliferation activated by LPS. The median inhibitory concentration(IC50) was 6㎍/ml. The CS-19-22 fraction also decreased IL-1β,PAF,IL-6, TNF-αand IL-8 production.The results of revers transcription-polymerase chain reaction(RT-PCR) demonstrated that the CS-19-22 fraction did not affect IL-1β,PAF,IL-6, TNF-α,and IL-8 mRNAs expression in BAL cells activated by LPS. By contrast, it inhibited the IL-10 mRNA expression but enhanced IFN-γand IL-12 mRNAs expression in activated BAL cells. Moreover, the CS-19-22 fraction blocked PAF-induced platelet aggregation. It is unlikely that cytotoxicity was involved. Because no cell deths were observable. We hypothesize that inhibitory mechanisms of CS-19-22 on BAL cells proliferation may be related to the impairments of gene expression and production of cytokines in BAL cells. The explanation of C. sinensis antiasthma function may involved the following(1)C.sinenesis contained immunomodulatory agents that stimulated IFN-γand IL-12 expression in TH1cells.(2)Enhanced secretion of IFN-γand IL-12 will inhibit the Tγcells immune responses Ex. Decreasing of IL-6 and IL-10 production.(3) The impairments of cytokines production in TH2 cells suppressed th BAL cells proliferation then more cytokines production Ex.IL-1β,PAF,IL-6,TNF-αand IL-8 in activated BAL cells were decreased. (4)The suppression of TH2cells immune responses will inhibit the growth and differentiation of B cells then IgE production will be decreased. (5)Reduced production of IgE will decreased asthma attack occurring in individuals. However, the results of the present study suggested that C. sinensis may also have acted to treat asthma in part by inhibiting BAL cells proliferation and cytokine gene expression and production. Future, experiments with treatment of animals with asthma with C. sinensis will be necessary to defined whether C. sinensis can reduce asthma injury. Plans are underway for the isolation of pure principal growth inhibitors and their mechanisms of action are subjected for further study.

        A Comparative Proteomics Survey of Proteins Responsive to Phosphorous Starvation in Roots of Hydroponically-grown Rice Seedlings

        Rice takes up phosphorous (P) as major nutrient source for its growth and development when grown under anaerobic water-logged soil conditions. To better understand the underlying mechanisms and to develop potential protein biomarkers of P-starvation, hydroponically-grown rice seedlings in the complete media and phosphorus absence (P-starvation) of phosphorous nutrient solutions were investigated for physiological and proteome changes. The P-starvation manifested significant reduction in root growth in three-week-old seedlings compared to respective complete media. Furthermore, P-starvation also showed increased activity of acid phosphatase in roots of one- and three-week-old seedlings, suggesting that experimental design is suitable for proteomics survey of P-starvation responsive proteins. Two-dimensional gel electrophoresis coupled with matrix-assisted laser desorption/ionization-time of flight (MALDI-TOF) mass spectrometry analysis of total root protein from three-week-old seedlings identified 10 P-starvation responsive protein spots out of 140 high-quality protein spots. Identified 10 proteins were involved in metabolism and defense/stress response. Out of 10, 2 and 8 protein spots were found to be up and down-regulated, respectively. Semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) analysis of corresponding genes of four randomly selected proteins, including putative glyceraldehydes-3-phophate dehydrogenase (G3PDH, spot R1), S-adenosyl-L-methionine synthetase (SAMS, spot R4), ATP synthase subunit alpha (spot R6), and root-specific pathogenesisrelated protein 10 (PR-10, spot R8), showed that just as protein abundance, these proteins are also regulated at the transcript level. Results suggest identified P-starvation responsive proteins are involved in maintaining nutrient homeostasis and/or associated with changes in root physiology under the absence of P.

        Tazarotene-Induced Gene 1 Interacts with DNAJC8 and Regulates Glycolysis in Cervical Cancer Cells

        The tazarotene-induced gene 1 (TIG1) protein is a retinoidinducible growth regulator and is considered a tumor suppressor. Here, we show that DnaJ heat shock protein family member C8 (DNAJC8) is a TIG1 target that regulates glycolysis. Ectopic DNAJC8 expression induced the translocation of pyruvate kinase M2 (PKM2) into the nucleus, subsequently inducing glucose transporter 1 (GLUT1) expression to promote glucose uptake. Silencing either DNAJC8 or PKM2 alleviated the upregulation of GLUT1 expression and glucose uptake induced by ectopic DNAJC8 expression. TIG1 interacted with DNAJC8 in the cytosol, and this interaction completely blocked DNAJC8-mediated PKM2 translocation and inhibited glucose uptake. Furthermore, increased glycose uptake was observed in cells in which TIG1 was silenced. In conclusion, TIG1 acts as a pivotal repressor of DNAJC8 to enhance glucose uptake by partially regulating PKM2 translocation.

        Impact of Refined Land Surface Properties on the Simulation of a Heavy Convective Rainfall Process in the Pearl River Delta Region, China

        The location and occurrence time of convective rainfallshave attracted great public concern as they can lead to terribledisasters. However, the simulation results of convective rainfalls inthe Pearl River Delta region often show significant discrepanciesfrom the observations. One of the major causes lies in the inaccurategeographic distribution of land surface properties used in the modelsimulation of the heavy precipitation. In this study, we replaced thedefault soil and vegetation datasets of Weather Research andForecasting (WRF) model with two refined datasets, i.e. theGlobCover 2009 (GLC2009) land cover map and the HarmonizedWorld Soil Database (HWSD) soil texture, to investigate the impactof vegetation and soil on the rainfall patterns. The result showed thatthe simulation patterns of convective rainfalls obtained from thecoupled refined datasets are more consistent with the observationsthan those obtained from the default ones. By using the coupledrefined land surface datasets, the overlap ratio of high precipitationdistricts reached 36.3% with a variance of 28.5 km from theobserved maximum rainfall position, while those of the defaultUnited States Geological Survey (USGS) dataset and ModerateResolution Imaging Spectroradiometer (MODIS) dataset are 17.0%/32.8 km and 24.9%/49.0 km, respectively. The simulated totalrainfall amount and occurrence time using the coupled refineddatasets are the closest to the observed peak values. In addition, theHWSD soil data has improved the accuracy of the simulatedprecipitation amount, and the GLC2009 land cover data also didbetter in catching the early peak time.

      • The Measurement of People’s Satisfaction on Public Safety and Police Service

          In American police history, supervisor’s judgment, crime reports and public opinion investigation have been used as indicators to measure police performance. Contemporarily, public opinion investigation is the most important indicator because it is the instrument to measure citizen satisfaction, a major dimension of police performance in community problem-solving era.<BR>  Page and Shapiro (1992) noted that public opinion is rational and stable; public opinion does change, but the change is incremental and predictable; a sudden change of public opinion can be traced to a certain events. Longitudinal data of polls conducted by independent institutions demonstrated that Taiwanese attitude and perception toward public safety is stable, incremental and predictable too. This paper shows the national trend of polls on Taiwanese sense of public safety. In addition, differences among counties and cities will be compared.

        Drilling Temperature and Hole Quality in Drilling of CFRP/Aluminum Stacks Using Diamond Coated Drill

        Composite/metal stack materials, most often consisting of carbon fiber reinforced polymer (CFRP) composites and aluminum or titanium alloy (Al or Ti), have been widely used in the aviation industry. It is still a challenge to make holes on the composite/metal stack materials with high quality. This paper aims to investigate the influence of drilling parameters on the drilling of stack materials consisting of T800/X850 CFRP and 7075-T651 Al with regard to drilling force, drilling temperature, hole diameter and hole surface quality. Diamond coated drill bits with double point angles were employed in the drilling tests. The drilling temperature was measured utilizing a rotational motion temperature measuring system. It is indicated that maximum drilling temperature increases with the increase of spindle speed, while, it decreases with increasing feed rate. The thrust force and drilling temperature increase abnormally during drilling of the Al layer, which can be attributed to the evacuation difficulty of the long and flexible Al chips. It is found that sudden drops in the surface profile of CFRP are caused by the cavities owning to the resin degradation effects. The influences of different stack sequences on the drilling temperature and hole surface quality have also been discussed.
