      • Involvement of Cdc25c in Cell Cycle Alteration of a Radioresistant Lung Cancer Cell Line Established with Fractionated Ionizing Radiation

        Cancer patients often suffer from local tumor recurrence after radiation therapy. Cell cycling, an intricate sequence of events which guarantees high genomic fidelity, has been suggested to affect DNA damage responses and eventual radioresistant characteristics of cancer cells. Here, we established a radioresistant lung cancer cell line, A549R, by exposing the parental A549 cells to repeated ${\gamma}$-ray irradiation with a total dose of 60 Gy. The radiosensitivity of A549 and A549R was confirmed using colony formation assays. We then focused on examination of the cell cycle distribution between A549 and A549R and found that the proportion of cells in the radioresistant S phase increased, whereas that in the radiosensitive G1 phase decreased. When A549 and A549R cells were exposed to 4 Gy irradiation the total differences in cell cycle redistribution suggested that G2-M cell cycle arrest plays a predominant role in mediating radioresistance. In order to further explore the possible mechanisms behind the cell cycle related radioresistance, we examined the expression of Cdc25 proteins which orchestrate cell cycle transitions. The results showed that expression of Cdc25c increased accompanied by the decrease of Cdc25a and we proposed that the quantity of Cdc25c, rather than activated Cdc25c or Cdc25a, determines the radioresistance of cells.

      • Electrodeposition mechanism of palladium nanotube and nanowire arrays.

        <P>Palladium nanotube and nanowire arrays were fabricated by electrodeposition into anodic alumina oxide (AAO) templates. By using alumina templates with different pore sizes and hole mouth morphologies, nanotubes with various diameters and lengths were obtained. The mechanism of nano-structure formation was studied by analyzing the current-time curves. It was found that the time needed for the transformation of the nanostructure from a tube to a wire depends on the size and structure of the template. However, the overall behaviour of the current-time transient is similar and can be divided into four main stages, which provides a powerful in-situ method to control nanotube-nanowire transition during growth. The palladium nanotube arrays have a polycrystalline structure, but a preferred orientation along the (110) direction was observed for the arrays of nanowires. Moreover, the degree of texture increases with decreasing alumina pore size.</P>

        A new linear cutting experimental machine for disc-cutter rock breaking with high-pressure water jets assisted

        Aiming at the low rock-breaking efficiency of TBM disc cutters in tough rock environments, A new experimental machine combined with the high-pressure water jet rock-breaking method was designed. It can flexibly change the cutting parameters of the water jet, such as spray position, spray height, pressure, etc. Moreover, the three-direction load of the disc cutter during the rock-breaking process can be obtained accurately by strain testing. The rock-breaking experiment reached the following conclusions. The experimental machine can meet the design function. In this study, the normal and rolling forces when the disc cutter breaks rock are significantly lower than the traditional, 26.90% and 28.39%, respectively. The lateral force increased slightly, about 31.17%. Although the energy consumption of water-jet-assisted is higher than the traditional rock-breaking method, the volume of broken rock per unit of time is approximately 5.85 times that of the traditional. It is feasible to increase the energy cost to increase the excavation speed. On the rock surface after the water jet experiment, a rock ridge appears on the rolling trajectory of the disc cutter, which is not conducive to further breaking the rock. The cutting depth of the water jet needs to be adjusted to reduce the height of the rock ridge.

      • Susceptibility of Lung Cancer with Polymorphisms of CYP1A1, GSTM1, GSTM3, GSTT1 and GSTP1 Genotypes in the Population of Inner Mongolia Region

        Background: To study the relationship of susceptibility to lung cancer with the gene polymorphisms of CYP1A1, GSTM1, GSTM3, GSTT1, GSTP1 and smoking status in Han and Mongolian populations of Inner Mongolia, an autonomous region of China. Materials and Methods: PCR-RFLP, allele-specific and multiplex PCR were employed to identify the genotypes of CYP1A1, GSTM1, GSTM3, GSTT1 and GSTP1 in a case-control study of 322 lung cancer patients diagnosed by bronchoscopy and 456 controls free of malignancy. Results: There is a significant difference in genotypic frequency of GSTT1 of healthy Mongolian and Han subjects. A statistically prominent association was found between CYP1A1 Msp1 (vt/vt) (OR=4.055, 95%CI:2.107-7.578, p=0.000), GSTM1 (-) (OR=2.290, 95%CI:1.467-3.573, p=0.000) and lung cancer in Mongolians. Similarly, in the Han population, CYP1A1 Msp1 (vt/vt) (OR=3.194, 95%CI:1.893-5.390, p=0.000) and GSTM1 (-) (OR=1.884, 95%CI:1.284-2.762, p=0.001) carriers also had an elevated risk of lung cancer. The smokers were more susceptible to lung cancer 2.144 fold and 1.631 fold than non-smokers in Mongolian and Han populations, respectively. The smokers who carried with CYP1A1 Msp1 (wt/vt+vt/vt), exon7 (Val/Val+Ile /Val), GSTM1 (-), GSTM3 (AB+BB), and GSTT1 (-) respectively were found all to have a high risk of lung cancer. Conclusions: CYP1A1 Msp1 (vt/vt) and GSTM1 (-) are risk factors of lung cancer in Han and Mongolian population in the Inner Mongolia region. The smokers with CYP1A1 Msp1 (wt/vt+vt/vt), CYP1A1 exon7 (Val/Val+Ile /Val), GSTM1 (-), GSTM3 (AB+BB), and GSTT1 (-) genotypes, respectively, are at elevated risk of lung cancer.

      • Metabolism of an Insecticide Fenitrothion by <i>Cunninghamella elegans</i> ATCC36112

        <P>In this study, the detailed metabolic pathways of fenitrothion (FNT), an organophosphorus insecticide by <I>Cunninghamella elegans</I>, were investigated. Approximately 81% of FNT was degraded within 5 days after treatment with concomitant accumulation of four metabolites (M1–M4). The four metabolites were separated by high-performance liquid chromatography, and their structures were identified by mass spectroscopy and/or nuclear magnetic resonance. M3 is confirmed to be an initial precursor of others and identified as fenitrothion-oxon. On the basis of their metabolic profiling, the possible metabolic pathways involved in phase I and II metabolism of FNT by <I>C. elegans</I> was proposed. We also found that <I>C. elegans</I> was able to efficiently and rapidly degrade other organophosphorus pesticides (OPs). Thus, these results will provide insight into understanding of the fungal degradation of FNT and the potential application for bioremediation of OPs.         The effects of paeoniflorin injection on soluble triggering receptor expressed on myeloid-1 (sTREM-1) levels in severe septic rats

        Paeoniflorin (PAE) is the most abundant compound in Xuebijing injection widely used to treat sepsis. We aimed to investigate effect of PAE on expression of soluble triggering receptor expressed on myeloid cells-1 (sTREM-1) in a rat model of sepsis. Wistar rats were divided into Normal, Model, and PAE groups (n=20 each). Endotoxin was administrated at 5 mg/ml/kg in Model and PAE rats to establish rat sepsis model. 1 h after endotoxin administration, PAE was administrated at 4 ml/kg in PAE group once per day for 3 days. Routine blood tests and biochemical indexes were assessed, including aspartate aminotransferase (AST) and creatine kinase-MB (CK-MB). The plasma sTREM-1 level was measured using quantitative ELISA. At the end of experiment, the small intestine, liver, kidney and lung were subjected to pathological examinations. A rat model of sepsis-induced multiple organ dysfunction syndrome (MODS) was established successfully with endotoxin administration (5 mg/ml/kg), evidenced by histo-pathological examinations, routine blood tests and biochemical indexes: platelet count decreased and white blood cell count increased (p<0.05), CK-MB and AST increased (p<0.05). PAE treatment significantly reduced the plasma levels of AST, CK-MB, and sTREM-1, compared to Model group (p<0.05). Meanwhile, sepsis-induced damages in the liver, lung, stomach and intestinal mucosa were also markedly ameliorated by PAE treatment. PAE demonstrated a significantly protective effect in a rat model of sepsis by decreasing plasma sTREM-1 level, reducing inflammation, preventing MODS and protecting organ functions.

      • Study on the Lethal Reason of Overwintering Weedy Rice Seed in Cold Rice Production Area in China

        The outdoor trails were carried out to investigate overwintering survival rate of different weedy rice seeds in two latitude areas including Longjing city located in Jilin province and Donggang city located in Liaoning province in China, respectively. The survival rates of all weedy rice seeds which were frozen under the condition of saturated soil for 3 months are about 0%. The germination rates of weedy rice seeds overwintering in air-dried soil are more than 50%. The death of overwintering weedy rice seed in the two production areas was positively related to its dormancy characteristic, seed water content and temperature during seed frozen period.

      • Quantitative Presentation of the Systematic Relationships Between S. sudanense and S. bicolor

        Fifty-four biological characters were used for quantitative classification of sorghum and sudangrass and their relationship. Discriminant analysis of all 240 individuals of 48 accessions suggested that 100% of originally grouped cases were correctly classified. Analysis of variance results showed that the differences among 48 accessions in characters were significant at the 0.01 level. Cluster analysis showed that the 48 accessions were obviously divided into two clusters by a Euclidean distance of 17.46 as the classification standards, i.e. the genus maize and genus sorghum. The 48 accessions were divided into four clusters by a Euclidean distance of 9.86, i.e. Sorghum-Sudangrass, Sorghum related species (S. propinquum and S. halepense), Zea mexicana and Z. mays. All accessions were divided into five clusters by a Euclidean distance of 8.91, namely group Ⅰ (Sudangrass-Chinese sorghum), group Ⅱ (Non-Chinese sorghum), group Ⅲ (Sorghum related species), group Ⅳ (Zea mexicana), and group Ⅴ (Z mays).
