        Design and dimension optimization of cutter disassembly mechanism for shield tunneling machine

        This study focuses on configuration and dimension optimization of cutter disassembly mechanism. Firstly, a cutter disassembly mechanism configuration is proposed with four components and simple actions, based on the theory of inventive problem solving (TRIZ) contradiction solutions. Furthermore, a comprehensive dimension optimization strategy was developed, including non-dimensional normalization model, kinematic performance atlases and fuzzy comprehensive evaluation method. Then, the optimal relative dimension of crucial parameters was obtained. Combined with the assembly relationship between components, dimension of all parameters was designed for cutter disassembly mechanism. The locking and disassembling process simulation was provided to illustrate the feasibility of the mechanism. The results show that the angular velocity is very smooth and the locking and disassembling actions are realizable without interference for optimized cutter disassembly mechanism. The effectiveness of the configuration and dimension optimization method is validated.

      • Clinical Efficacy and Prognosis Factors for Advanced Hepatoblastoma in Children: A 6-year Retrospective Study

        Objective: This study aimed to investigate the effect of multimodality treatment of advanced paediatric hepatoblastoma (HB) and the factors affecting prognosis. Methods: A total of 35 children underwent multimodality treatments consisting of chemotherapy, surgery, interventional therapy, and autologous peripheral blood stem cell transplantation. The patients were followed up every month. Results: Serum AFP levels in 33 out of 35 patients in this study were significantly increased (P = 0.0002). According to the statistical scatter plot, the values of serum AFP on the 25th, 50th, and 75th percentages were 1,210, 1,210 and 28,318 ng/dl, respectively. Of the 35 cases, 21 were stage IV. 18 cases were treated with systemic chemotherapy before surgery, and 3 cases with locally interventional chemotherapy before surgery. Statistical analysis showed that the preferred interventional treatment affected prognosis, and that there was a statistically significant difference (P = 0.024). Some 33 patients completed the follow-up, of which 17 were in complete remission (CR), 5 were in partial remission (PR), 1 became disease progressive (DP), and 10 died. The remission and overall survival rates were 66.7% (22/33) and 69.7% (23/33), respectively. Patients with the mixed HB phenotypes had worse prognoses than the epithelial phenotype (P < 0.001), and patients in stage IV had a lower survival rate than those in stage III (P < 0.001). Conclusion: Multimodality treatment can effectively improve remission rate and prolong the survival of children with advanced HB. In addition, alpha-fetoprotein (AFP), a tumor marker of liver malignant tumors, HB pathological classification, and staging are highly useful in predicting prognosis.

        In microarray data mining, one of the key problems is how to handle weak signals. Based on a bent piecewise linear accumulated distribution generally found in the microarray data, a new detectable threshold finding method is proposed to filter genes with unreliable information in this paper. More reliable and reproducible data is produced for the subsequent data mining.

        EBV-miR-BHRF1-1 Targets p53 Gene: Potential Role in Epstein-Barr Virus Associated Chronic Lymphocytic Leukemia

        Purpose The purpose of this study was to investigate the prognostic impact of Epstein-Barr virus (EBV)–microRNA (miRNA, miR)-BHRF1-1 with chronic lymphocytic leukemia (CLL) as well as role of EBV-miR-BHRF1-1 in p53 gene. Materials and Methods Quantitative reverse transcription–polymerase chain reaction and western blotting were used to quantify EBV-miR-BHRF1-1 and p53 expression in cultured CLL. Results p53 aberration was associated with the higher expression level of EBV-miR-BHRF1-1 (p < 0.001) which was also an independent prognostic marker for overall survival (p=0.028; hazard ratio, 5.335; 95% confidence interval, 1.193 to 23.846) in 97 newly-diagnosed CLL patients after adjusted with International Prognostic Index for patients with CLL. We identified EBV-miR-BHRF1-1 as a viral miRNA regulator of p53. EBV-miR-BHRF1-1 repressed luciferase reporter activity by specific interaction with the seed region within the p53 3- untranslated region. Discordance of p53 messenger RNA and protein expression was associated with high EBV-miR-BHRF1-1 levels in CLL patients and cell lines. EBV-miR-BHRF1- 1 inhibition upregulated p53 protein expression, induced cell cycle arrest and apoptosis and decreased cell proliferation in cell lines. EBV-miR-BHRF1-1 mimics downregulated p53 protein expression, decreased cell cycle arrest and apoptosis, and induced cell proliferation in cell lines. Conclusion This study supported the role of EBV-miR-BHRF1-1 in p53 regulation in vitro. Our results support the potential of EBV-miR-BHRF1-1 as a therapeutic target in EBV-associated CLL with p53 gene aberration.

        Comparison Study on the Rock Cutting Characteristics of Disc Cutter under Free-face-assisted and Conventional Cutting Methods

        To improve the excavation efficiency of Tunnel Boring Machines (TBMs), the free-face-assisted rock cutting method induced by TBM disc cutter (FM) was explored and the differences between the FM and the conventional Cutting Method (CM) were also compared based on a series of rock cutting tests. The results show that when the free face distance is less than a critical value, the FM can promote the tensile failure of the rock and the formation of big rock debris effectively. Moreover, the cutting efficiency and the cutting forces including normal force, rolling force and side force of the disc cutter under the FM are also significantly reduced and improved, respectively, compared to that under the CM. The FM can be applied in multi-stage cutterhead TBMs to improve the cutting efficiency and reduce the failure of the disc cutter, the research results indicate that the free face distance should be controlled within 80 mm to use the free face effectively.

        Performance Prediction of TBM Disc Cutting on Marble Rock under Different Load Cases

        A series of disc cutting tests was conducted with a single disc cutter (216 mm diameter) and a single rock type (a marble) byapplying different loads. Normal, rolling and side forces were measured for a series of spacings and penetrations, from which othercutting parameters also were calculated. The rock chips and fines were carefully collected and analyzed. Specific Energy (SE)considerations indicate that the optimum cutting condition is existed with the ratio of S/P between 9 and 12 when applying the staticloading. However, when the combination of static and impact loading is applied, the optimum cutting conditions was estimated withthe ratio of S/P between 16 and 19. It shows that the optimal spaces become larger after adding the combination of static and impactloading. The relationship between rolling force and normal force is close and consistent: whether applying the static loading or thecombination of static and impact loading, a nearly linear rise of the ratio of rolling force to normal force with increased penetration,and, conversely, a nearly unchanged ratio with increases in spacing. The grain size distribution curves show that larger chip size andlesser fines are produced when applying the combination of static and impact loading and the degree of crushing size tends to beuniform distribution.

        Flow field characteristics of an agitator system of a large diameter slurry-water shield machine

        A numerical model was established for calculation in this study on the basis of the standard k-epsilon turbulence model and the discrete phase model of the Euler-Lagrange equation. Results of the particle image velocimetry experiment were used to verify the reliability of the numerical model. The effects of the rotation speeds, slurry densities, and impeller structures on the flow field characteristics, including velocity distribution, ballast fluidity, and power loss, were also investigated. The results indicate that the increase of rotation speed has little influence on the flow field near the wall and bottom of the agitator, and the agitator performs well when the speed is set between 30 rpm and 40 rpm. The increase of the slurry density will increase the pressure loss between the inlet and the outlet, and it will also increase the load of the agitator. Hence, the slurry density must be less than 1400 kg/m 3 . The baffle can improve the velocity distribution of the flow field and the slag discharge capacity of the agitator.

        Smart and versatile biomaterials for cutaneous wound healing

        The global increase of cutaneous wounds imposes huge health and financial burdens on patients and society. Despite improved wound healing outcomes, conventional wound dressings are far from ideal, owing to the complex healing process. Smart wound dressings, which are sensitive to or interact with changes in wound condition or environment, have been proposed as appealing therapeutic platforms to effectively facilitate wound healing. In this review, the wound healing processes and features of existing biomaterials are firstly introduced, followed by summarizing the mechanisms of smart responsive materials. Afterwards, recent advances and designs in smart and versatile materials of extensive applications for cutaneous wound healing were submarined. Finally, clinical progresses, challenges and future perspectives of the smart wound dressing are discussed. Overall, by mapping the composition and intrinsic structure of smart responsive materials to their individual needs of cutaneous wounds, with particular attention to the responsive mechanisms, this review is promising to advance further progress in designing smart responsive materials for wounds and drive clinical translation.

      • Hiwi Knockdown Inhibits the Growth of Lung Cancer in Nude Mice

        Hiwi, a human homologue of the Piwi family, plays an important role in stem cell self-renewal and is overexpressed in various human tumors. This study aimed to determine whether an RNA interference-based strategy to suppress Hiwi expression could inhibit tumor growth in a xenograft mouse model. A rare population of $SSC^{lo}\;Alde^{br}$ cells was isolated and identified as lung cancer stem cells in our previous study. Plasmids containing U6 promoter-driven shRNAs against Hiwi or control plasmids were successfully established. The xenograft tumor model was generated by subcutaneously inoculating with lung cancer stem cell $SSC^{lo}\;Alde^{br}$ cells. After the tumor size reached about 8 mm in diameter, shRNA plasmids were injected into the mice via the tail vein three times a week for two weeks, then xenograft tumor growth was assessed. In nude mice, intravenously delivery of Hiwi shRNA plasmids significantly inhibited tumor growth compared to treatment with control scrambled shRNA plasmids or the vehicle PBS. No mice died during the experiment and no adverse events were observed in mice administered the plasmids. Moreover, delivery of Hiwi shRNA plasmids resulted in a significant suppressed expression of Hiwi and ALDH-1 in xenograft tumor samples, based on immunohistochemical analysis. Thus, shRNA-mediated Hiwi gene silencing in lung cancer stem cells by an effective in vivo gene delivery strategy appeared to be an effective therapeutic approach for lung cancer, and may provide some useful clues for RNAi gene therapy in solid cancers.
