        인간포배기 배아의 효과적인 유리화 동결법의 개발을 위한 연구

        Objective: The purpose of this study was to evaluate the survival rate of vitrified blastocyst according to the freezing vessels, equilibration time in cryoprotectant and artificial dehydration method. Methods: Human blastocysts were vitrified after loading onto the plastic straw, open-pulled straw (OPS), electron microscopy grid (EM grid) for 1.5 min or 3 min. They also were directly plunged into LN2 within 30sec. For artificial shrinkage of blastocysts, 36 gauge fine needle was pushed at the cellular junction of the trophectoderm into the blstocoele cavity until it shrank without damage of inner cell mass. Results: The survival rate of vitrified blastocysts on plastic straw, OPS, EM grid as freezing vessels were 26.7, 13.0 and 60.5%, respectively. The survival rate of EM grid was significantly higher than that of plastic straw and OPS (p<0.05). For 1.5 min equilibrium, the survival rates of early blastocyst (EB), middle blastocyst (MB) and late blastocyst (LB) were 64.4, 81.0, and 20.0% respectively. For 3 min equilibrium, the survival rates of EB, MB, and LB were 69.9, 50.0 and 57.5% respectively. The survival rates of EB and MB were significantly higher than that of LB in 1.5 min equilibrium group (p<0.05), however, the significance was not observed in 3 min equilibrium groups. In cytoplasmic shrinkage before vitrification, the survival rates of EB, MB and LB were 92.9, 100 and 75.9% respectively. The survival rate of MB was significantly higher than that of LB (p<0.05). The survival rates of vitrified blastocysts by artificial dehydration and slow-frozen blastocysts were not significantly different as 88.9 and 66.7%, respectively. Conclusion: This study showed that the vitrification of human blastocysts using EM grid and artificial dehydration is an effective method. Therefore, these methods would be an useful techniques for blastocyst cryopreservation.

        전문계 고등학교 교사들의 이러닝 활용과 영향요인 간의 구조방정식 모형

        이 연구는 전문계 고등학교 교사들을 대상으로 이러닝을 활용하는데 영향을 미치는 요인들을 파악하고, 이들 요인들간의 인과관계성을 구조 모형적인 측면에서 분석 · 설명하는 데 중점을 두고 있다. 연구모형에 포함된 요인은 교직경력, 컴퓨터 사용경험, 이러닝 지원부문, 이러닝의 사회/조직 관련성 등의 외생변인과 이러닝 활용을 위한 컴퓨터 리터러시, 이러닝 활용 의향도 등의 내생변인 등이며, 요인들간의 상관계수를 산출하여 그 행렬을 이용, LISREL 프로그램에 의해 공변량 구조 이론 모델 분석을 수행하였다. 이 연구에서 제시된 모델 분석을 통한 연구결과는 다음과 같다. 첫째, 교사들의 이러닝 활용을 위한 컴퓨터 리터러시에는 설정된 외생변인 중 컴퓨터 사용경험의 영향을 가장 크게 받고 있으며, 다음으로 이러닝 지원정도, 사회/조직 관련성 등의 순으로 영향을 받고 있다. 둘째, 최종 내생변인인 교사들의 이러닝 활용 의향도에는 외생변인 중 이러닝의 사회/조직 관련성만이 영향을 미치는 것으로 분석되었다. 셋째, 내생변인간의 관계성을 분석한 결과 교사들의 이러닝의 활용 의향도는 이러닝 활용을 위한 컴퓨터 리터러시에 매우 큰 영향을 받고 있음을 보여 주었다. This study was designed and implemented to identify factors affecting e-learning use of vocational high school teachers based on structural equation model. The model career, computer experience, e-learning support, social and organizational norm related to e-learning as exogenous variables and computer literacy and intention to use as endogenous variables. The researcher produces a correlation matrix among the variables and run a structural model using LISREL program. The major findings after are as follows. First, teachers' computer experience was the most significant factors affecting their computer literacy to utilize e-learning, followed by e-learning support and soial and organizational norm. Second, intention to use e-learning, terminal exogenous variable was influenced by e-learning social and orginizational norm among exogenous variables. Third, in terms of the relationships between endogenous variables, intention to use e-learning was largely influenced by computer literacy from the model analysis.

      • PLCM을 이용한 카오스 블록 암호화 기법

        본 논문에서는 동력학적 특성이 좋은 PLCM을 이용한 256비트 길이의 암호 키와 32비트 데이터 블록의 카오스 블록 암호화 기법을 제안한다. 본 논문에서 제안한 기법은 256비트의 키에서 각 암호화 단계마다 사용될 32비트의 세션 키 두 개를 생성해서 PLCM의 초기 값과 반복회수를 결정하는데 사용한다. 본 제안 기법은 피드백 구조와 대치 기능의 S함수를 통해 암호 강도를 향상시켰다. 또한 실험 결과와 안전성 분석을 통해 본 제안 기법이 통계적(Statistical) 공격에 매우 강함을 보여준다. In this paper, we propose a chaotic block encryption scheme using a PLCM (Piecewise Linear Chaotic Map) having a good dynamical property. The proposed scheme has a block size of 32 bits and a key size of 256 bits. In the proposed scheme we use two 32-bit session keys generated from 256-bit key to decide the initial value and the number of iteration of PLCM. To enhance the strength of security we use a feedback structure and a S (Substitution) function. With results of test and analyses of security we show the proposed scheme is very secure against statistical attacks.

        Facile Synthesis and Radioiodine Labeling of Hypericin

        Hypericin (1,3,4,6,8,13-hexahydroxy-10,11-dimethylphenanthro[1,10,9,8-opqra]perylene-7,14-dione), an antidepressant which is also known to be a potent protein kinase C (PKC) inhibitor was synthesized as a precursor for radioiodine labeling via two step reactions. Malignant glioma cells express higher PKC activity compared to untransformed glial cell. Here we report the synthesis and radioiodine labeling of hypericin as a potential brain tumor imaging radiopharmaceutical. The reference compound, 2-iodohypericin, and its radiolabelled analogues, 2-[$^{123}I$]iodohypericin and 2-[$^{124}I$]iodohypericin have been prepared by the reaction of hypericin with NaI or [$^{123}I$]NaI or [$^{124}I$]NaI. The labeling yield was 60-65% for each analogue and the optimal reaction time was 10 min. The purification and isolation of the labelled products were achieved by a reversed-phase HPLC.

        폐암과 폐 염증성질환의 동적양전자방출단층검사(Dynamic 18F-FDG PET)의 유용성

          목적: 양전자방출단층검사(Fluorine-18 2-Deoxy-D-Glucose positron emission tomography: ¹?F-FDG PET)는 가시적 영상분석과 반정량적 섭취계수 분석 방법을 사용하여 폐의 염증성 질환과 폐암을 구별 및 진단하는 데 유용하다. 일반적으로 표준화섭취계수(Standardized Uptake Value: SUV)가 폐 질환의 진단에 사용되지만, 이는 폐 조직에 있어서 반정량적, 정량적 분석을 통한 동적인 정보를 포함하지 않는다. 그러므로, 본 연구는 ¹?F-FDG PET 검사에서 폐 질환의 동적 섭취 분석이 보다 정확한 대상질환의 진단을 유용하게 할 수 있을 것이라는 가정하에 시간-방사능 곡선(Time-Activity Curve: TAC), 표준화섭취계수-동적곡선(Standardized Uptake Value-Dynamic Curve: SUV-DC), 패트락 분석법(Patlak analysis: Glucose Metabolic Rate(MRGlu))을 토대로 얻은 분석방법을 이용하여 진단적 유용성을 평가하였다.<BR>  대상 및 방법: 폐 질환을 가진 17명의 환자를 대상을 하여, 각각 60분간 Dynamic ¹?F-FDG PET검사를 시행하였다. 획득한 정보를 통하여 폐질환의 형태를 따라 관심영역(Region of Interest: ROI)를 그린 후, 반정량적 분석인 TAC, SUV-DC와 정량적 분석인 Patlak analysis를 각각의 군에 따라 분석하여 비교 분석하였다.<BR>  결과: Dynamic ¹?F-FDG PET을 통한 분석결과, TAC 형태는 초기 혈류상에서 폐의 염증성 질환과 폐암의 구분이 어려웠으나, 시간이 지날수록 폐암의 곡선이 염증성 질환의 경우보다 확연히 증가하였다. SUV-DC의 경우는 TAC 형태와 거의 유사한 형태를 가졌다. Patlak analysis 분석결과, 대동맥 영역에서는 폐의 염증성 질환이 폐암보다 높은 혈액 방사능을 보였으나 시간이 지남에 따라 염증성 질환의 혈액 방사능이 극히 낮아졌다. 반면, 병변 조직에서는 폐암이 가장 높은 섭취를 보였으며 폐의 염증성 질환은 중간 정도에 머물렀다.<BR>  결론: TAC와 SUV-DC 분석에서 폐암과 폐의 염증성 질환은 각각 고유한 그래프 형태를 띠었으며 Patlak analysis에서 역시 주목할 만한 차이를 보였다. 따라서 이러한 분석법을 토대로 좀더 깊이 있는 연구가 진행된다면 비침습적으로 폐의 질환을 구별하는 데 보다 적절하고 유용한 진단적 정보를 얻을 수 있을 것이다.   Purpose: The diagnostic utility of fluorine-18 2-deoxy-D-glucose positron emission tomograhpy (¹?F- FDG PET) for the non-invasive differentiation of focal lung lesions originated from cancer or inflammation disease by combined visual image interpretation and semi-quantitative uptake value analysis has been documented. In general, Standardized Uptake Value(SUV) is used to diagnose lung disease. But SUV does not contain dynamic information of lung tissue for the glucose. Therefore, this study was undertaken to hypothesis that analysis of dynamic kinetics of focal lung lesions base on ¹?F-FDG PET may more accurately determine the lung disease. So we compared Time Activity Curve(TAC), Standardized Uptake Value-Dynamic Curve(SUV-DC) graph pattern with Glucose Metabolic Rate(MRGlu) from Patlak analysis.<BR>  Methods: With lung disease, 17 patients were examined. They were injected with ¹?F-FDG over 30-s into peripheral vein while acquisition of the serial transaxial tomographic images were started. For acquisition protocol, we used twelve 10-s, four 30-s, sixteen 60-s, five 300-s and one 900-s frame for 60 mins. Its images were analyzed by visual interpretation TAC, SUV-DC and a kinetic analysis(Patlak analysis). The latter was based on region of interest(ROIs) which were drawn with the lung disease shape. Each optimized patterns were compared with itself.<BR>  Results: In TAC patterns, it hard to observe cancer type with inflammation disease in early pool blood area but over the time cancer type slope more remarkably increased than inflammation disease.<BR>  SUV-DC was similar to TAC pattern. In the result of Patlak analysis, In time activity curve of aorta, even though inflammation disease showed higher blood activity than cancer, at first as time went by, blood activity of inflammation disease became the lowest. However, in time activity curve of tissue, cancer had the highest uptake and inflammation disease was in the middle.<BR>  Conclusion: Through the examination, TAC and SUV-DC could approached the results that lung cancer type and inflammation disease type has it’s own difference shape patterns. Also, it has outstanding differentiation between cancer type and inflammation in Patlak and MRGlu analysis. Through these analysis methods, it will helpful to separation lung disease.

      • Spontaneous Additive Nanopatterning from Solution Route Using Selective Wetting

        <P>Nanopatterns of functional materials have successfully led innovations in a wide range of fields, but further exploration of their full potential has often been limited because of complex and cost-inefficient patterning processes. We here propose an additive nanopatterning process of functional materials from solution route using selective wetting phenomenon. The proposed process can produce nanopatterns as narrow as 150 nm with high yield over large area at ultrahigh process speed, that is, the speed of solution dragging, of up to ca. 4.6 m·min<SUP>-1</SUP>. The process is highly versatile that it can utilize a wide range of solution materials, control vertical structures including pattern thickness and multistacks, and produce nanopatterns on various substrates with emerging form factors such as foldability and disposability. The solution patterning in nanoscale by selective wetting is enabled by corresponding surface energy patterns in high contrast that are achieved by one-step imprinting onto hydrophobic/hydrophilic bilayers. The mechanisms and control parameters for the solution patterning are revealed by fluid-dynamic simulation. With the aforementioned advantages, we demonstrate 25 400 pixel-per-inch light-emitting pixel arrays and a plasmonic color filter of 10 cm × 10 cm area on a plastic substrate as potential applications.</P> [FIG OMISSION]</BR>

      • Pulmonary Nodular Ground-Glass Opacities in Patients With Extrapulmonary Cancers

        <P>BACKGROUND: The clinical significance of pulmonary nodular ground-glass opacities (NGGOs) in patients with extrapulmonary cancers is not known, although there is an urgent need for study on this topic. The purpose of this study, therefore, was to investigate the clinical significance of pulmonary NGGOs in these patients, and to develop a computerized scheme to distinguish malignant from benign NGGOs. METHODS: Fifty-nine pathologically proven pulmonary NGGOs in 34 patients with a history of extrapulmonary cancer were studied. We reviewed the CT scan characteristics of NGGOs and the clinical features of these patients. Artificial neural networks (ANNs) were constructed and tested as a classifier distinguishing malignant from benign NGGOs. The performance of ANNs was evaluated with receiver operating characteristic analysis. RESULTS: Twenty-eight patients (82.4%) were determined to have malignancies. Forty NGGOs (67.8%) were diagnosed as malignancies (adenocarcinomas, 24; bronchioloalveolar carcinomas, 16). Among the rest of the NGGOs, 14 were atypical adenomatous hyperplasias, 4 were focal fibrosis, and 1 was an inflammatory nodule. There were no cases of metastasis appearing as NGGOs. Between malignant and benign NGGOs, there were significant differences in lesion size; the presence of internal solid portion; the size and proportion of the internal solid portion; the lesion margin; and the presence of bubble lucency, air bronchogram, or pleural retraction (p < 0.05). Using these characteristics, ANNs showed excellent accuracy (z value, 0.973) in discriminating malignant from benign NGGOs. CONCLUSIONS: Pulmonary NGGOs in patients with extrapulmonary cancers tend to have high malignancy rates and are very often primary lung cancers. ANNs might be a useful tool in distinguishing malignant from benign NGGOs.</P>

      • FN13762 murine breast cancer: region-by-region correlation of first-pass perfusion CT indexes with histologic vascular parameters.

        <P>PURPOSE: To investigate the correlation between first-pass perfusion computed tomographic (CT) indexes and histologic vascular parameters in FN13762 breast cancer in rats by using region-by-region correlation methods. MATERIALS AND METHODS: The Animal Care and Use Committee approved this study. FN13762 murine breast cancer cells were implanted in 14 female Fischer 344 rats, and first-pass perfusion CT was performed. CT perfusion maps depicting blood flow, blood volume, mean transit time (MTT), and permeability-surface area (PSA) product were generated by using commercial perfusion software. The entire tumor area was divided into six separate regions on perfusion maps, and the regional perfusion indexes were quantified. Histologic vascular parameters, including microvessel density (MVD), luminal vessel number, luminal vessel area, and luminal vessel perimeter, were measured in the histologic region corresponding to the perfusion maps. Correlation analysis was performed between regional tumor perfusion indexes and histologic vascular parameters of the corresponding tumor region. Additionally, mean perfusion values of the entire tumor were correlated with histologic vascular parameters of the hot spot within the tumor. Among 14 rats, four were excluded from the analysis, and results were based on a final total of 10 rats. RESULTS: In tumors, blood flow, blood volume, and PSA product were significantly higher and MTT was significantly shorter (P < .05 for all) than these values in normal neck muscles. At region-by-region correlation, regional blood flow (r = 0.476), blood volume (r = 0.348), and MTT (r = -0.506) were significantly correlated with MVD in the corresponding tumor region (P < .01 for all). After adjustment for biologic variability between rats, regional blood flow (r = 0.614), blood volume (r = 0.515), MTT (r = -0.524), and PSA product (r = 0.228) remained significantly correlated with MVD in the corresponding tumor region. Correlation analysis between CT perfusion indexes of the entire tumor and histologic vascular parameters of the hot spot did not show significant correlations (P > .05). CONCLUSION: Regional blood flow, blood volume, and MTT are significantly correlated with MVD in the corresponding tumor region.</P>
