A rice ABC transporter, OsABCC1, reduces arsenic accumulation in the grain

<P><B>Significance</B></P><P>Contamination of water and foods with arsenic (As) poses a threat to millions of people worldwide. Because the rice grain is the major source of As intake, reducing the transfer of As from soil to the grain is a pressing public health issue. We found that a member of the <I>Oryza sativa</I> C-type ATP-binding cassette transporter (OsABCC) family, OsABCC1, detoxifies As and reduces the amount of As in the rice grain. OsABCC1 in the upper nodes of rice plants restricts the distribution of As to the grain by sequestering it in the vacuoles of the phloem companion cells of diffuse vascular bundles directly connected to the grain. Our work suggests a strategy for limiting As accumulation in rice grains and thereby reducing human As exposure.</P><P>Arsenic (As) is a chronic poison that causes severe skin lesions and cancer. Rice (<I>Oryza sativa</I> L.) is a major dietary source of As; therefore, reducing As accumulation in the rice grain and thereby diminishing the amount of As that enters the food chain is of critical importance. Here, we report that a member of the <I>Oryza sativa</I> C-type ATP-binding cassette (ABC) transporter (OsABCC) family, OsABCC1, is involved in the detoxification and reduction of As in rice grains. We found that <I>OsABCC1</I> was expressed in many organs, including the roots, leaves, nodes, peduncle, and rachis. Expression was not affected when plants were exposed to low levels of As but was up-regulated in response to high levels of As. In both the basal nodes and upper nodes, which are connected to the panicle, OsABCC1 was localized to the phloem region of vascular bundles. Furthermore, OsABCC1 was localized to the tonoplast and conferred phytochelatin-dependent As resistance in yeast. Knockout of <I>OsABCC1</I> in rice resulted in decreased tolerance to As, but did not affect cadmium toxicity. At the reproductive growth stage, the As content was higher in the nodes and in other tissues of wild-type rice than in those of <I>OsABCC1</I> knockout mutants, but was significantly lower in the grain. Taken together, our results indicate that OsABCC1 limits As transport to the grains by sequestering As in the vacuoles of the phloem companion cells of the nodes in rice.</P>
NVOC D-Limonene의 후각자극이 인체에 미치는 영향

본 연구는 D-Limonene의 흡입 시 인체에 미치는 생리적, 심리적 효과를 밝히고자 실시하였다. D-Limonene은 국내 소나무림의 대표적인 NVOC 중 하나이다. 본 연구에는 여자대학생 13명(21.5 ± 1.0 세)이참가하였으며, 피험자는 의자에 앉아 눈을 감은 상태에서 D-Limonene 60μL와 실내공기(대조군)를 각각 90초간 흡입하였다. D-Limonene과 실내공기(대조군)의 데이터를 분석한 결과, D-Limonene을 흡입하는 동안 자율신경계의 부교감신경활동 지표인 심박변이도의 HF성분(827.2 ± 191.3 msec2)은 대조군(654.4 ± 163.6 msec2)에 비해 유의하게 높았으며(P < 0.05), 심박수(72.8 ± 2.3 bpm)는 대조군(74.1 ± 2.5 bpm)에 비해 유의하게 낮았다(P < 0.05). 또한 주관평가를 실시한 결과, D-Limonene의 흡입은 대조군에 비해 쾌적감이 유의하게 높았다(P < 0.01). 본 연구결과를 통하여 D-Limonene의 후각자극은 인체에 생리적, 심리적으로 안정 효과를 미치는 것을 알 수 있었다. The purpose of this study was to investigate the physiological and psychological effects of olfactory stimulation with D-Limonene. D-Limonene is one of major NVOC (Natural Volatile Organic Compounds) in Pinus densiflora forest of Korea. Thirteen Japanese female university students (21.5 ± 1.0 years old) participated in this study. D-Limonene was used as an olfactory stimulant, and room air was used as a control. And the subjects were exposed to the odor for 90 sec while sitting with their eyes closed. The results of the data after exposure to D-Limonene and control were compared, the HF value increased during the D-Limonene administration (827.2 ± 191.3 msec2) compared with control (654.4 ± 163.6 msec2), indicating that parasympathetic nervous activity was significantly higher during D-Limonene administration (P < 0.05). The heart rate decreased during D-Limonene administration (72.8 ± 2.3 bpm) compared with control (74.1 ± 2.5 bpm), and this difference was significant ( P < 0.05). Subjects reported significantly more comfortable during D-Limonene administration than control (P < 0.01). Our results clearly indicate that olfactory stimulation with D-Limonene induced physiological and psychological relaxation.
중ˑ고령자의 인지기능 검진 및 맞춤형 훈련을 위한 모바일 애플리케이션 알츠톡 A&T의 사용성 평가

치매 유병률은 급격하게 높아지고 있고, 그에 따른 환자 의료비와 돌봄 비용이 가중되고 있다. 이에 따라, 치매를 예방하고 손상된 인지기능 향상을 돕는 인지 프로그램 도입이 시급하다. 본 연구에서는 이미 개발된 중ˑ고령자의 인지기능을 평가하고 맞춤화된 인지 강화 훈련 커리큘럼을 추천하는 알츠톡 A & T (Assessment & Training) 애플리케이션의 사용성을 평가하고자 한다. 사용성 평가를 위해서 지역사회에 거주하는 55 세~74 세 중ˑ고령자 7 명과 IT 및 의료 전문가 5 명은 알츠톡 A & T 애플리케이션을 1 회 사용하였다. 사용 과정 중에 사용자의 이용 행태 관찰, 모바일 애플리케이션 등급 척도(MARS: Mobile Application Rating Scale) 답변, 사후 인터뷰를 진행하였고 정량적, 정성적 데이터를 수집 및 검증하였다. 개발된 알츠 A & T 애플리케이션의 사용성 평가 결과는 전문가는 3.09±0.20 점이었고 중ˑ고령자의 경우 3.81±0.62 점이었다. 전문가 집단과 사용자 집단 모두 심미성(Aesthetics)에서 가장 높은 점수를 주었다. 반면, 전문가는 정보(information)에서 가장 낮은 점수를 주었고, 사용자는 주관적인 앱의 질(subjective quality)에 가장 낮은 점수를 주었다. 개발된 알츠톡 A & T 애플리케이션은 대상자인 전문가와 사용자 그룹 양측의 모든 사용성 평가 영역에서 평균 3.0 에 가까운 점수를 얻었으며 전반적으로 만족스러운 수준을 보였고, 정보 (Information)에서 두 그룹간 유의한 차이가 있었다. 본 연구가 중ˑ고령자를 대상으로 하는 헬스케어 애플리케이션의 접근성, 유용성, 사용성을 향상시키는 근거가 되기를 기대한다.
