Coronavirus enzyme inhibitors-experimentally proven natural compounds from plants

Coronavirus disease (COVID-19) can cause critical conditions that require efficient therapeutics. Several medicines are derived from plants, and researchers are seeking natural compounds to ameliorate the symptoms of COVID-19. Viral enzymes are popular targets of antiviral medicines; the genome of coronaviruses encodes several enzymes, including RNAdependent RNA polymerase and viral proteases. Various screening systems have been developed to identify potential inhibitors. In this review, we describe the natural compounds that have been shown to exert inhibitory effects on coronavirus enzymes. Although computer-aided molecular structural studies have predicted several antiviral compound candidates, the current review focuses on experimentally proven natural compounds.

<B>ABSTRACT</B><P>Replication protein A (RPA) is a single-stranded-DNA-binding protein composed of three subunits with molecular masses of 70, 32, and 14 kDa. The protein is involved in multiple processes of eukaryotic DNA metabolism, including DNA replication, repair, and recombination. In <I>Xenopus</I>, <I>Xenopus</I> RPA-interacting protein α has been identified as a carrier molecule of RPA into the nucleus. In this study, human RPA-interacting protein α (hRIPα) and five novel splice isoforms (named hRIPα, hRIPβ, hRIPγ, hRIPδ1, hRIPδ2, and hRIPδ3 according to the lengths of their encoding peptides) were cloned. Among hRIP isoforms, hRIPα and hRIPβ were found to be the major splice isoforms and to show different subcellular localizations. While hRIPα localized to the cytoplasm, hRIPβ was found in the PML nuclear body. Modification of hRIPβ by sumoylation was found to be required for localization to the PML nuclear body. The results of the present work demonstrate that hRIPβ transports RPA into the PML nuclear body and releases RPA upon UV irradiation. hRIPβ thus plays an important role in RPA deposition in PML nuclear bodies and thereby supplements RPA for DNA metabolism.</P>

The composite of porous silicon (Si) and amorphous carbon (C) is prepared by pyrolysis of a nano-porous Si + pitch mixture. The nano-porous Si is prepared by mechanical milling of magnesium powder with silicon monoxide (SiO) followed by removal of MgO with hydrochloric acid (etching process). The Brunauer-Emmett-Teller (BET) surface area of porous Si ($64.52m^2g^{-1}$) is much higher than that before etching Si/MgO ($4.28m^2g^{-1}$) which indicates pores are formed in Si after the etching process. Cycling stability is examined for the nano-porous Si + C composite and the result is compared with the composite of nonporous Si + C. The capacity retention of the former composite is 59.6% after 50 charge/discharge cycles while the latter shows only 28.0%. The pores of Si formed after the etching process is believed to accommodate large volumetric change of Si during charging and discharging process.

<B>ABSTRACT</B><P>Kaposi's sarcoma-associated herpesvirus (KSHV) is the etiological agent of Kaposi's sarcoma. The open reading frame (K9) of KSHV encodes viral interferon regulatory factor 1 (vIRF1), which functions as a repressor of interferon-mediated signal transduction. The amino-terminal region of vIRF1 displays significant homology to the DNA-binding domain of cellular interferon regulatory factors, supporting the theory that the protein interacts with specific DNA sequences. Here, we identify the consensus sequence of vIRF1-binding sites from a pool of random oligonucleotides. Moreover, our data show that vIRF1 interacts with the K3:viral dihydrofolate reductase:viral interleukin 6 promoter region in the KSHV genome.</P>

<P><B>Purpose</B></P><P>To analyze the preoperative clinical and pathological characteristics of patients with pT0 prostate cancer.</P><P><B>Materials and Methods</B></P><P>We retrospectively reviewed the records of 702 patients who underwent radical prostatectomy (RP) at our institution between January 2004 and July 2008 for clinically localized prostate cancer. If there was no evidence of residual tumor in the pathological specimen of the prostate, a patient was staged as pT0. Patients with pT0 disease were compared with a control group of patients who were operated on during the same period.</P><P><B>Results</B></P><P>Overall, 9 (1.3%) patients were staged as pT0 on the pathologic examination. Significant differences were observed between the pT0 group and the control patients in the biopsy Gleason score (p=0.004), the number of positive cores on biopsy (p=0.018), the tumor length of positive cores (p<0.001), and prostate volume (p=0.015). Cutoff values predictive of pT0 tumor status were defined as a biopsy Gleason score sum ≤6, 2 or fewer positive biopsy cores, tumor length on biopsy ≤2 mm, and prostate volume >30 cm<SUP>3</SUP>. Whereas 8 of the 9 (88.9%) pT0 patients showed all of these characteristics, only 55 of the 693 (7.9%) control patients fulfilled the criteria. The combination suggested above afforded a sensitivity of 88.8% and a specificity of 92.1% for the prediction of pT0 status.</P><P><B>Conclusions</B></P><P>The frequency of pT0 prostate cancer seen on RP was 1.3%. A combination of clinicopathological features, incorporating a biopsy Gleason score, the number of positive biopsy cores, tumor length on biopsy, and prostate volume, was useful to predict pT0 stage on RP.</P>

The purpose of this study is to identify the relationship between the job environment, job commitment, and organizational loyalty of tour guides in order to provide a practical and efficient tour guide management policy for the continuous growth and stable management activities and achievements of the travel industry. The results of this study are summarized as follows. First, the job environment of the tour guide was found to be four dimensions of maintenance, achievement evaluation, decision making, and human relationship. Multiple regression analysis showed that there is a significant influence on dependent variables such as job satisfaction, job satisfaction, job responsibility, and turnover Respectively. The relationship between job environment and job satisfaction was influenced by pay, human relations, and achievement evaluation, and decision making was not significant. In the relationship between job environment and job satisfaction, there were significant influences in terms of human relations, achievement evaluation, and decision making, and remuneration was not significant. In terms of job environment and job responsibilities, human relations and decision making were influenced in order, and there was no meaningful relationship between pay and performance evaluation. Job satisfaction and job desire were influenced by achievement evaluation and human relationship order, and remuneration and decision making were not significant. Second, job involvement is divided into four dimensions : job satisfaction, job responsibility, and turnover need. Multiple regression analysis shows that there is a significant influence on organizational loyalty. The relationship between job commitment and organizational loyalty was influenced by job responsibilities and job satisfaction. Job satisfaction and turnover need were not significant. The Tour Guide is a group of experts devoted to the safe and satisfying tourism activities of the people in other countries, and should continuously improve the travel industry and improve and manage the work environment for stable management activities in a changing business environment.

This study aims to systematically develop a usability evaluation model using the Structural Equation Model (SEM) from experiment of usability on using vehicle. Vehicle developers have been adding many functions for enhance the user satisfaction. But it will be made the trade-off problem of usability and design elements of vehicle interior from attempt to make best usability satisfaction in a restricted space. To solve the trade-off problem, we set a new solution criterion from usability evaluation model. The usability experiment is based on major activity pool from derived user's acts pattern in vehicle for make more accurate usability evaluation model. And this model was built with twenty-nine measurement variables for the evaluation of usability of vehicle user. As a result, the proposed SEM model showed statistical significance as well as a high level of R Square (0.7144). This model shows the relationships of detailed usability and design elements. According to the result, this study introduces the criterion to secure the best satisfaction of usability and design elements.
