        Molecular characterization of duck enteritis virus CHv strain UL49.5 protein and its colocalization with glycoprotein M

        The UL49.5 gene of most herpesviruses is conserved andencodes glycoprotein N. However, the UL49.5 protein ofduck enteritis virus (DEV) (pUL49.5) has not been reported. In the current study, the DEV pUL49.5 gene was firstsubjected to molecular characterization. To verify thepredicted intracellular localization of gene expression, therecombinant plasmid pEGFP-C1/pUL49.5 was constructedand used to transfect duck embryo fibroblasts. Next, therecombinant plasmid pDsRed1-N1/ glycoprotein M (gM)was produced and used for co-transfection with thepEGFP-C1/pUL49.5 plasmid to determine whether DEVpUL49.5 and gM (a conserved protein in herpesviruses)colocalize. DEV pUL49.5 was thought to be an envelopeglycoprotein with a signal peptide and two transmembranedomains. This protein was also predicted to localize in thecytoplasm and endoplasmic reticulum with a probability of66.7%. Images taken by a fluorescence microscope atdifferent time points revealed that the DEV pUL49.5 andgM proteins were both expressed in the cytoplasm. Overlapof the two different fluorescence signals appeared 12 h aftertransfection and continued to persist until the end of theexperiment. These data indicate a possible interaction between DEV pUL49.5 and gM.

        Hydrogen Peroxide Detection Using a Polypyrrole/Prussian Blue Nanowire Modified Electrode

        A hydrogen peroxide sensor based on a polypyrrole (PPy)/Prussian blue (PB) modified electrode was developed. PPy nanowires were prepared by one step template-free electrochemical polymerization on a gold electrode and a PB layer was deposited using an electrochemical method. The introduction of the PPy nanostructure can provide a high surface area and lead to high sensitivity to hydrogen peroxide. The surface morphology of the PPy/PB electrode was characterized by scanning electron microscopy (SEM). The structural features of the PPy/PB electrode were confirmed by attenuated total reflection infrared (ATR-IR) and X-ray photoelectron spectroscopy (XPS). Chronoamperometry (CA) was used to evaluate the sensitivity of the sensor towards hydrogen peroxide.

        Medical Image Retrieval with Relevance Feedback via Pairwise Constraint Propagation

        Relevance feedback is an effective tool to bridge the gap between superficial image contents and medically-relevant sense in content-based medical image retrieval. In this paper, we propose an interactive medical image search framework based on pairwise constraint propagation. The basic idea is to obtain pairwise constraints from user feedback and propagate them to the entire image set to reconstruct the similarity matrix, and then rank medical images on this new manifold. In contrast to most of the algorithms that only concern manifold structure, the proposed method integrates pairwise constraint information in a feedback procedure and resolves the small sample size and the asymmetrical training typically in relevance feedback. We also introduce a long-term feedback strategy for our retrieval tasks. Experiments on two medical image datasets indicate the proposed approach can significantly improve the performance of medical image retrieval. The experiments also indicate that the proposed approach outperforms previous relevance feedback models.

        Fabrication and Osteoblastic Adhesion Behavior of Regenerated Silk Fibroin/PLLA Nanofibrous Scaffold by Double Syringe Electrospinning

        In this work, regenerated silk fibroin (SF)/poly-L-lactic acid (PLLA) composite electrospun fibers weresuccessfully prepared by electrospinning method with single and double syringe systems. Furtherly, we applied simultaneousand alternative spinning when using double syringe systems. The morphologies of the obtained silk/PLLA hybrid nanofiberswere characterized by scanning electron microscope (SEM). Fourier transform infrared spectroscopy (FTIR) showed thatPLLA has been well incorporated into the SF electrospun fibers using double syringes. Furthermore, the tensile test resultsindicate the tensile strength and Young’s modulus of the silk/PLLA hybrid nanofibers webs were improved by both singleand double syringe system. Moreover, the cell adhesion ratios of simultaneous silk/PLLA nanofibers were much higher thanthat of alternative silk/PLLA nanofibers, and even higher than pure silk nanofibers. Therefore, simultaneous spinning usingdouble syringe system is a potential method for fabrication of nanofiber scaffold.

        Prevalence of CYP17A1 gene mutations in 17α-hydroxylase deficiency in the Chinese Han population

        Background: 17α-hydroxylase deficiency is a rare autosomal recessive disorder caused by mutations in the cytochrome P450 family 17 subfamily A member 1 gene. The major clinical presentation includes hypertension, hypokalemia, male pseudohermaphroditism and female gonadal dysplasia. Hundreds of pathogenic variants have been reported in this disorder, and some common mutations were found to be race-specific. Case presentation: In this study, we reported 5 Chinese girls with 17α-hydroxylase deficiency from Henan Province. The patients all came to the hospital for hypertension, and they also presented with sexual infantilism. The average age of the patients was 14 years old, ranging from 12 to 17 years old. They all had reduced blood cortisol, estradiol (E2), and testosterone (TESTO) and increased adrenocorticotropic hormone (ACTH), follicle-stimulating hormone (FSH), and luteinizing hormone (LH). They all had the appearance of females; however, three of the chromosome karyotypes were 46XX, and two were 46XY. Conclusions: All of the patients carried a mutation on the 329 amino acid of CYP17A1 exon 6. By summarizing the currently known pathogenic mutations of 17α-hydroxylase deficiency, we demonstrated the prevalence of these gene mutations in Chinese Han and non-Chinese populations.

        the construction of Yunnan-Vietnam railway has an important historical significance in the development history of modern Yunnan, which contains a history of imperialist aggression and the resistance of Yunnan people. This process of mutual struggle and interaction essentially reflects the division of interests. This paper will analyze the choice of different game subjects in this process from the perspective of game theory, and clearly reflect the game process between China and France during the construction of Yunnan-Vietnam railway in the late Qing Dynasty by combing the game process, analyzing the game subjects and the relationship between them. Finally, taking history as a mirror, through the division of international interest a hundred years ago, it will give us some enlightenment to solve the current international problems.

      • 顶山市城市社区养老托育服务供给状况研究——基于QCA 方法的组态分析

        人口老龄化一直都是一个备受关注的话题,近年又随着少子化趋势的出现,中国的人口结构也面临着新的挑战。随着国家提出养老和托育代际融合发展的政策,平顶山市也开始面临着养老和托育如何发展的问题,本文通过fsQCA 和csQCA 的分析方法,分析出适合平顶山养老托育发展的缺陷以及促进发展的路径。其中,问题主要表现为专业人员供给不足、医疗健康服务供给不足、幼儿启蒙教育服务供给不足等。针对这些问题,本文提出了加强人才队伍建设、宣传城市社区养老托育服务优点、连接多方资源以增强可持续发展能力以及加大政府财政和政策支撑力度等多个建议,以促进平顶山市养老托育事业的持续发展。 Population ageing has always been a topic of great concern, and in recent years, with the emergence of the declining birthrate, China's population structure is also facing new challenges. With the national policy of intergenerational integration of old-age care and childcare, Pingdingshan City has also begun to face the problem of how to develop old-age care and childcare. Among them, the problems are mainly manifested in the insufficient supply of professionals, medical and health services, and early childhood enlightenment education services. In view of these problems, this paper puts forward a number of suggestions to strengthen the construction of talent team, publicize the advantages of urban community elderly care services, connect multiple resources to enhance sustainable development capacity, and increase government financial and policy support, so as to promote the sustainable development of elderly care in Pingdingshan City.

      • Automatic Subretinal Fluid Segmentation of Retinal SD-OCT Images With Neurosensory Retinal Detachment Guided by Enface Fundus Imaging

        <P>Objective: Accurate segmentation of neurosensory retinal detachment (NRD) associated subretinal fluid in spectral domain optical coherence tomography (SD-OCT) is vital for the assessment of central serous chorioretinopathy (CSC). A novel two-stage segmentation algorithm was proposed, guided by Enface fundus imaging. Methods: In the first stage, Enface fundus image was segmented using thickness map prior to detecting the fluid-associated abnormalities with diffuse boundaries. In the second stage, the locations of the abnormalities were used to restrict the spatial extent of the fluid region, and a fuzzy level set method with a spatial smoothness constraint was applied to subretinal fluid segmentation in the SD-OCT scans. Results: Experimental results from 31 retinal SD-OCT volumes with CSC demonstrate that our method can achieve a true positive volume fraction (TPVF), false positive volume fraction (FPVF), and positive predicative value (PPV) of 94.3%, 0.97%, and 93.6%, respectively, for NRD regions. Our approach can also discriminate NRD-associated subretinal fluid from subretinal pigment epithelium fluid associated with pigment epithelial detachment with a TPVF, FPVF, and PPV of 93.8%, 0.40%, and 90.5%, respectively. Conclusion: We report a fully automatic method for the segmentation of subretinal fluid. Significance: Our method shows the potential to improve clinical therapy for CSC.</P>
