Two new polyketides, chinoketides A and B (1 - 2) with a known compound xylarphthalide A (3), were isolated from the solid medium of the endophytes from the leaves of the relic plant Distylium chinense with the "black-box" co-culture method, and the structures of two new compounds were elucidated by NMR, MS and CD spectra. And the absolute configurations of chinoketides A (1) and B (2) were determined as 2R,3R,8S and 5R,6S by calculating their ECD spectra to compare with the experimental CD spectra. Finally, the antimicrobial activities were evaluated to Erwinia carotovora sub sp. Carotovora (Jones) Bersey et al, and the results showed that compounds 1 - 3 displayed the antimicrobial activities with MIC value at 20.5, 30.4 and 10.2 mg/mL.

A fast gamma-ray dose rate assessment method for complex geometries based on stylized modelreconstruction and point-kernel method is proposed in this paper. The complex three-dimensional (3D)geometries are imported as a 3DS format file from 3dsMax software with material and radiometric attributes. Based on 3D stylized model reconstruction of solid mesh, the 3D-geometrical solids are automaticallyconverted into stylized models. In point-kernel calculation, the stylized source models aredivided into point kernels and the mean free paths (mfp) are calculated by the intersections betweenshield stylized models and tracing ray. Compared with MCNP, the proposed method can implementcomplex 3D geometries visually, and the dose rate calculation is accurate and fast.
Effects of Lactobacillus plantarum SCS2 on blood glucose level in hyperglycemia mice model

In this study, the hyperglycemia mice model was established with 1-week high sugar and fat diet plus with 70 mg/kg body weight of streptozotocin injection for 3 days. Sixty male Kunming mice of 3 weeks old in a specific-pathogen-free grade were divided into six groups randomly, which includes normal group (NG), prevention group (PG), treatment group for low dose (TGL), middle dose (TGM), high dose (TGH), and model group (MG). NG and MG mice were fed with sterile physiological saline (10 mL/kg body weight). PG mice were fed with the concentration of 6.0 × 109 CFU/mL L. plantarum SCS2 suspensions from the second to third week. TGL, TGM, and TGH mice were fed with the concentration of 2.0 × 109, 4.0 × 109, and 6.0 × 109 CFU/mL L. plantarum SCS2 suspensions (10 mL/kg body weight), respectively from fourth to tenth week. The results showed that the fasting and postprandial 2 h blood glucose levels of TGH mice were reduced by L. plantarum SCS2 significantly (p < 0.05) as compared with MG. The body weight of TGH mice came to normal level at tenth week. Content of K+ in plasma of TGH mice was increased and contents of Na+ and Cl− in the plasma of TGH mice were decreased as compared with MG. Meanwhile, content of glycogen in TGH mice was reduced. However, the effect of L. plantarum SCS2 on the prevention of hyperglycemia in PG mice was not significant as compared with NG mice during the experiment. These results suggested that L. plantarum SCS2 showed a hypoglycemic effect on hyperglycemic mice model.

The growth of many breast tumors is stimulated by IGF-1, which activates signal transduction pathways inducing cell proliferation. $ER{\alpha}$ is important in this process. The aim of the study was to investigate relationships in vitro among inhibitory effects of luteolin on the growth of MCF-7 cells, IGF-1 pathway and $ER{\alpha}$. Our results showed that luteolin could effectively block IGF-l-stimulated MCF-7 cell proliferation in a dose- and time-dependent manner and block cell cycle progression and induce apoptosis evidenced by the flow cytometric detection of sub-G1DNA content. Luteolin markedly decreased IGF-l-dependent IGF-IR and Akt phosphorylation without affecting Erk1/2 phosphorylation. Further experiments pointed out that $ER{\alpha}$ was directly involved in IGF-l induced cell growth inhibitory effects of luteolin, which significantly decreased $ER{\alpha}$ expression. Knockdown of $ER{\alpha}$ in MCF-7 cells by an $ER{\alpha}$-specific siRNA decreased the IGF-l induced cell growth inhibitory effects of luteolin. $ER{\alpha}$ is thus a possible target of luteolin. These findings indicate that the inhibitory effect of luteolin on the growth of MCF-7 cells is via inhibiting IGF-l mediated PI3K-Akt pathway dependent of $ER{\alpha}$ expression.

Activation of the innate immune system requires recognitionof pathogen-associated molecular patterns, such as NOD-likereceptors. The NOD-like receptor protein 3 (NLRP3)inflammasome is involved in induction of the proinflammatorycytokine, IL-1β, and subsequent inflammatoryresponses. NLRP3 inflammasome plays important roles in theinflammatory and innate immune responses associated withautoimmune/inflammatory syndrome. However, analysis of thetissue distribution and expression profiles in BALB/c mice is stillincomplete. In this study, we investigated the tissue distributionand expression pattern of NLRP3 in BALB/c mice to furtherelucidate its function in innate immunity in this commonly usedlaboratory animal model. NLRP3 mRNA expression levels andtissue distribution of the protein were investigated by real-timequantitative PCR and immunohistochemical analyses,respectively. NLRP3 mRNA expression was higher in thekidney and inguinal lymph nodes than in other tissues. Cytoplasmic expression of NLRP3 was detected in the epithelialreticular cells of the spleen and thymus, lymphocytes in theinguinal lymph nodes, cardiac muscle cells, cerebral cortexneurons, alveolar macrophages, renal tubule cells and liversinusoidal endothelial cells. The results of this study will assistinvestigators in interpreting site-specific functions and roles ofNLRP3 in inflammatory responses.

Abrasive flow polishing plays an important role as a type of precision nanomachining technology. In this study, the polishing mechanism of abrasive flow was studied using numerical simulations based on molecular dynamics. In order to study the polishing mechanism of polycrystalline materials, the molecular dynamics simulation of three kinds of polycrystalline materials was carried out. The cutting force, energy, friction coefficient, and surface morphology (during the microcutting process) of the three workpiece materials were analyzed. The analysis shows that the adhesion of atoms in the contact area on the workpiece surface will be affected by liquid phase and the type of abrasive particles, and then the surface quality polished by the abrasive flow will also be influenced. The onset of an abrasive flow polishing process involving plastic deformation (including the nucleation and migration of dislocations, grain boundary shape deformation, and deformation twinning) was observed on the workpiece surface. Defects strongly competed with each other in the form of interactions of dislocations and the grain boundaries, resulting in the migration of twin boundaries. A further analysis of the numerical results for different cutting depths and abrasive properties of the workpiece materials will be useful for understanding the different cutting effects.

A DNA fragment containing 2,079 base pairs from Bacillus circulans CGMCC 1416 was cloned using degenerate PCR and inverse PCR. An open reading frame containing 981 bp was identified that encoding 326 amino acids residues, including a putative signal peptide of 31 residues. The deduced amino acid sequence showed the highest identity (68.1%) with endo-β-1,4-D-mannanase from Bacillus circulans strain K-1 of the glycoside hydrolase family 5 (GH5). The sequence encoding the mature protein was cloned into the pET-22b(+) vector and expressed in Escherichia coli as a recombinant fusion protein containing an N-terminal hexahistidine sequence. The fusion protein was purified by Ni2+ affinity chromatography and its hexahistidine tag cleaved to yield a 31-kDa β-mannanase having a specific activity of 481.55 U/mg. The optimal activity of the purified protein, MANB48, was at 58℃ and pH 7.6. The hydrolysis product on substrate locust bean gum included a monosaccharide and mainly oligosaccharides. The recombinant MANB48 may be of potential use in the feed industry.
Oil film deposition characteristics and judgment of lubrication effect of splash lubricated gears

To study the oil film deposition characteristics of splash lubricated gears and determine whether their meshing area is completely lubricated, this study proposes the concept of judging the splash lubrication-deposition characteristics-inlet oil film thickness-lubrication effect. The critical oil film thickness that characterizes the full lubrication of the gear meshing area was obtained as 14.5428 μm by using elastohydrodynamic lubrication (EHL) theory. Based on computational fluid dynamics (CFD) method, based on the renormalization group (RNG) k-ɛ two-equation turbulence model, dynamic mesh model and volume of fluid (VOF) model, the numerical calculation of the intermediate gearbox in splash lubrication is carried out. The oil film deposition thickness of the gear tooth that was about to enter the meshing area was calculated to be 95.6207 μm. It was found that the meshing area is fully lubricated under the rated operating conditions.

Advanced automotive industries generate large demand for the next generation of high strength and high toughness springsteels. Vanadium-containing 55SiCrV spring steels subjected to rapid-induction heating treatment can fulfil such requirements. However, the effect of vanadium microalloying under online rapid-induction heat treatments is rarely reported. Acomparative study of the microstructure and tensile properties of 55SiCr and 55SiCrV spring steel wires subjected to a novelonline rapid induction heat treatment has been demonstrated herein. It is found that the tensile strength of the 55SiCr springwire decreases with the decrease in the wire speed in online rapid-induction heating, and the plasticity increases. Whereas,the tensile strength of the 55SiCrV steel wire increases with the decrease in the wire speed with the retained high plasticity,which is attributed to the strengthening effect of the dislocations. Through the optimized rapid-induction heating/coolingthermal cycles and intermediate-temperature tempering treatment, the tensile strength of the 55SiCrV steel wire approaches2106 MPa with total elongation of 9.7%. Compared with the 55SiCr spring steel, the addition of V in 55SiCrV spring steelchanges the strengthening and toughening mechanisms via the grain refinement and enhancement in the hardenability andtempering resistance. The finely dispersed V-containing secondary phases are rarely found in the matrix, which indicates thatthe precipitation effect stemming from the addition of V is not the dominant strengthening factor in the online rapid-inductionheat process. The proposed novel online rapid-induction heat treatment provides a promising pathway for the mechanicalproperty improvement of the spring steel.
