        The Combination of Platelet Rich Plasma Gel, Human Umbilical Mesenchymal Stem Cells and Nanohydroxyapatite/polyamide 66 Promotes Angiogenesis and Bone Regeneration in Large Bone Defect

        BACKGROUND: In the present study, a novel tissue engineering bone graft including platelet rich plasma gel (PRP gel), human umbilical mesenchymal stem cells (HUMSCs) and nanohydroxyapatite/polyamide 66 (nHA-PA66) was constructed. We explored whether the composite scaffolds could enhance the angiogenesis and bone repair capacity in rat femoral large bone defect (LBD). This study aimed to provide evidence for the clinical application of the composite scaffold in LBD treatment. METHODS: PRP was prepared, the platelets and growth factors were measured. HUMSCs were isolated and identified. the osteogenic capacity of PRP in vitro was measured. Then HUMSCs-PRP-gel/nHA-PA66 composite scaffolds were synthesized and observed. The proliferation and osteogenesis differentiation of HUMSCs on the composite scaffold was measured. The angiogenic capacity of PRP in vitro was measured by capillary-like tube formation assay. Finally, the angiogenesis and bone repair capacity of the composite scaffolds was measured in rat LBD. RESULTS: PRP contained high level of platelets and growth factors after activation, and promoted osteogenic and angiogenic differentiation in vitro. The HUMSCs-PRP-gel/nHA-PA66 composite scaffold was porosity and promoted the proliferation and osteogenesis differentiation of HUMSCs. At 12th weeks, more micro-vessels and new bone were formed around the composite scaffolds compared with other groups, the defect was almost repaired. CONCLUSION: Our study for the first time identified that the combination of PRP gel, HUMSCs and nHA-PA66 scaffold could significantly promote angiogenesis and bone regeneration in rat LBD, which may have implications for its further application in clinical LBD treatment.

        Patient preferences and shared decision making for the traditional Chinese medicine treatment of lung cancer: A discrete choice experiment study

        Background: Traditional Chinese Medicine (TCM), integrating patient preferences into decision-making process, has been widely used in the multimodality therapy of lung cancer. This study aimed to estimate patient preferences for treatment and shared decision-making (SDM) modes concerning inpatient TCM treatment of lung cancer in Shanghai in order to provide a basis for clinical decision-making process on TCM therapy for lung cancer. Methods: This study was conducted among patients (n = 347) from nine tertiary hospitals in Shanghai by discrete-choice experiment (DCE) survey and Shared Decision-Making Questionnaire-patient version (SDM-Q-9) survey. The DCE was developed with the inclusion of the most relevant attributes at appropriate levels for the TCM treatment of lung cancer. The empirical data analyses of patients were performed using mixed logit models. Additionally, subgroup analysis was conducted. Results: In total, 347 respondents completed the questionnaire. All attributes’ coefficients were statistically significant for patients’ preferences. Patients showed strong preferences for increasing disease control rate, relieving nausea and vomiting, reducing the risk of side effects, and were concerned about monthly out-of-pocket costs. Subgroup analysis indicated that patients with a lower SDM-Q-9 score and those who were satisfied with medical services emphasized more importance of higher disease control rate. Furthermore, most of the patients (90.20%) self-reported a high willingness to use SDM during the decision-making process. Conclusion: In Shanghai, patients mainly preferred SDM and considered disease control rate as the most essential attribute in the TCM treatment of lung cancer. The study findings could underscore the importance of considering patients’ preferences and promote SDM.

      • A Reliable Information Fusion Algorithm for Reputation Based Wireless Sensor Networks

        In wireless sensor networks (WSNs), cryptographic primitives alone cannot provide a sufficient solution to the secure information fusion problem, therefore reputation systems have been introduced into WSNs. In a cluster, each sensor node has a single reputation value that is evaluated by the other sensor nodes in the same cluster. In this paper, we propose a novel, reliable information fusion algorithm, called reputation-driven information fusion (RDIF). In this work, a clustering algorithm is employed to divide all of the sensor nodes into many clusters. Then, a reputation system is established for each cluster, and an information fusion algorithm driven by reputation values is performed by the cluster head. In addition to the sensor nodes’ reputation values, we also consider the values of the readings collected by the sensor nodes and eliminate the outliers before fusing information. The simulation results show that RDIF can improve the reliability and accuracy of fusion results significantly when some compromised nodes appear in the WSNs.

        Conductivity relaxation in NiTiO3 at high temperatures

        NiTiO3 ceramics were prepared via the traditional solid-state reaction route. The dielectric properties of NiTiO3 ceramics have been systematically investigated in the temperature range from room temperature to 1073 K NiTiO3 ceramics exhibit intrinsic dielectric response in the temperature range below 400 K. Two relaxations were observed in the temperature range higher than 400 K. The relaxation activation energy is 0.95 eV and 1.17 eV for the low- and high-temperature relaxations, respectively. Our results strongly indicate that the two relaxations are related to conductivity relaxation associated with the singly and doubly ionized oxygen vacancies.

      • Load Power Estimation Based Secondary Control for Microgrids

        The well-known active power-frequency and reactive power-voltage amplitude droop control is widely used in the coordinative control of parallel inverters in microgrids. However, this conventional droop method may cause frequency and voltage deviation, which affects the accuracy of power supply. This paper proposes a novel secondary control strategy to compensate this deviation. This strategy mimics the Master-Slave control but requires no communication lines among the parallel inverters. The master inverter adopts conventional droop method (using power to control frequency and voltage amplitude) and is controlled as a voltage source while the slave inverters adopt reversed droop method (using frequency and voltage amplitude to control power) and are controlled as current sources. The droop characteristic bias of slave inverters is designed online based on the estimation of load power demand. Through this method, frequency and voltage deviation can be eliminated and power sharing can be realized among all the slave inverters. Simulation and experimental results are provided to prove the effectiveness of the proposed control strategy.

      • p38 MAPK Signaling Mediates Mitochondrial Apoptosis in Cancer Cells Induced by Oleanolic Acid

        Oleanolic acid (OA) is a nutritional component widely distributed in various vegetables. Although it has been well recognized for decades that OA exerts certain anti-tumor activity by inducing mitochondria-dependent apoptosis, it is still unclear that what molecular signaling is responsible for this effect. In this study, we employed cancer cell lines, A549, BXPC-3, PANC-1 and U2OS to elucidate the molecular mechanisms underlying OA anti-tumor activity. We found that activation of MAPK pathways, including p-38 MAPK, JNK and ERK, was triggered by OA in both a dose and time-dependent fashion in all the tested cancer cells. Activation was accompanied by cleavage of caspases and PARP as well as cytochrome C release. SB203580 (p38 MAPK inhibitor), but not SP600125 (JNK inhibitor) and U0126 (ERK inhibitor), rescued the pro-apoptotic effect of OA on A549 and BXPC-3 cells. OA induced p38 MAPK activation promoted mitochondrial translocation of Bax and Bim, and inhibited Bcl-2 function by enhancing their phosphorylation. OA can induce reactive oxygen species (ROS)-dependent ASK1 activation, and this event was indispensable for p38 MAPK-dependent apoptosis in cancer cells. In vivo, p38 MAPK knockdown A549 tumors proved resistant to the growth-inhibitory effect of OA. Collectively, we elucidated that activation of ROS/ASK1/p38 MAPK pathways is responsible for the apoptosis stimulated by OA in cancer cells. Our finding can contribute to a better understanding of molecular mechanisms underlying the antitumor activity of nutritional components.

      • Robust Inland Waterway Ship Tracking via Orthogonal Particle Filter-based Predator

        Tracking-Learning-Detection (TLD), also known as Predator, has become one of the most popular state-of-the-art algorithms in the domain of visual tracking in recent years. It has demonstrated outstanding performance in the application of long-term tracking of a human face in unconstrained videos. In this paper, we address the problem of tracking a single ship in inland waterway CCTV videos given its location in the first frame and no other prior information. Firstly, we deeply analyze Predator in a perspective of control system and point out the search strategy in detection procedure is the most time-consuming part in Predator system. Secondly, in order to speed up the whole pipeline, we propose a novel motion model based on extended particle filter with orthogonal design. Due to the power of particle optimization and re-combination with orthogonal design, we can relate the motion of object of interest better and obtain the most likely candidates of object regions more effectively. Finally, both qualitative and quantitative evaluations on numerous challenging CCTV videos demonstrate that the proposed algorithm achieves favorable performance in terms of efficiency and accuracy.

        Covalent Immobilization of Arginine onto Polyacrylonitrile-Based Membrane for the Effective Separation of Oil/Water Emulsion

        Membrane technology has been accepted as a very effective approach to purify oily wastewater. However, the serious membrane fouling induced by the adsorption of oily foulants significantly hinders the development of membrane technology in treating oily wastewater. To improve the anti-fouling property and oil/ water separation efficiency of membrane, this work reports on the covalent immobilization of short-chain arginine (Arg) molecules onto polyacrylonitrile/polyacrylonitrile- co-poly(glycidyl methacrylate) (PAN/PAN-co-PGMA) blend membrane surface via a ring-opening reaction between epoxy groups and amine groups. It was found that the covalent immobilization of Arg molecules effectively increased the surface hydrophilicity of the membrane, resulting in a significant decrease of the interaction force between the foulants and the membrane surface. This typical characteristic was revealed by the pure water contact angle and the force-extension curve measurements. The Arg-immobilized membranes exhibited much higher separation efficiency against oil/water emulsions than the pristine PAN and PAN/PAN-co-PGMA membranes. Especially when the grafting amount of Arg was 157.3 μg/cm2, the oil rejection ratio of an Arg-immobilized membrane was as high as 99.2%. In addition, the flux recovery ratio of the membrane still reached 88.8%, even after two cycle filtrations of pure water and oil/water emulsion. These results indicated that the Arg-immobilized membrane may have practical applications for oil/water emulsion separation.

        MAPK4 silencing in gastric cancer drives liver metastasis by positive feedback between cancer cells and macrophages

        Liver metastasis is a major cause of death in gastric cancer patients, but the underlying mechanisms are poorly understood. Through a combination of in vivo screening and transcriptome profiling followed by quantitative RT-PCR and tissue array analyses, we found that mitogen-activated protein kinase 4 (MAPK4) downregulation in gastric cancer tissues from patients is significantly associated with liver metastasis and poor prognosis. The knockdown of MAPK4 in gastric cancer cells promotes liver metastasis in orthotopic mouse models. MAPK4 depletion in gastric cancer cells induces the secretion of macrophage migration inhibitory factor (MIF) to polarize tumor-associated macrophages (TAMs) in orthotopic xenograft tumors. Moreover, TAMs activate epithelial–mesenchymal transition of gastric cancer cells to suppress MAPK4 expression, which further increases MIF secretion to polarize TAMs. Taken together, our results suggest a previously undescribed positive feedback loop between cancer cells and macrophages mediated by MAPK4 silencing that facilitates gastric cancer liver metastasis.
