Periodontitis is characterized by the loss of periodontal tissues, especially alveolar bone. Common therapies cannot satisfactorily recover lost alveolar bone. Periodontal ligament stem cells (PDLSCs) possess the capacity of self-renewal and multilineage differentiation and are likely to recover lost alveolar bone. In addition, periodontitis is accompanied by hypoxia, and hypoxia-inducible factor-1α (HIF-1α) is a master transcription factor in the response to hypoxia. Thus, we aimed to ascertain how hypoxia affects runt-related transcription factor 2 (RUNX2), a key osteogenic marker, in the osteogenesis of PDLSCs. In this study, we found that hypoxia enhanced the protein expression of HIF-1α, vascular endothelial growth factor (VEGF), and RUNX2 ex vivo and in situ. VEGF is a target gene of HIF-1α, and the increased expression of VEGF and RUNX2 proteins was enhanced by cobalt chloride (CoCl2, 100 μmol/L), an agonist of HIF-1α, and suppressed by 3-(5′-hydroxymethyl-2′-furyl)-1-benzyl indazole (YC-1, 10 μmol/L), an antagonist of HIF-1α. In addition, VEGF could regulate the expression of RUNX2, as RUNX2 expression was enhanced by human VEGF (hVEGF165) and suppressed by VEGF siRNA. In addition, knocking down VEGF could decrease the expression of osteogenesis-related genes, i.e., RUNX2, alkaline phosphatase (ALP), and type I collagen (COL1), and hypoxia could enhance the expression of ALP, COL1, and osteocalcin (OCN) in the early stage of osteogenesis of PDLSCs. Taken together, our results showed that hypoxia could mediate the expression of RUNX2 in PDLSCs via HIF-1α-induced VEGF and play a positive role in the early stage of osteogenesis of PDLSCs
Mechanical evaluation of polymer microneedles for transdermal drug delivery: In vitro and in vivo

In this study, we reported two types of PMNs based on polylactic acid (PLA) and polyvinyl alcohol (PVA),respectively. Parafilm M film, porcine skin, and rats’ models were operated to evaluate the mechanicalproperties in vitro and in vivo to find optimal parameters for efficient insertion. Insertion depth was measuredusing Digital Force Gauge by changing insertion force and speed, respectively. Results showed thatincreasing the insertion force and speed used for PMNs application led to a significant increase in thedepth of insertion. A force of 18 N under a speed of 330 mm/min was the optimal condition for insertingPMNs array into ParafilmM film and porcine skin. In addition, PLA-MNs exhibited higher robustness andenhanced homogeneity in insertion depth compared with PVA-MNs, but PVA-MNs were able to reachmuch deeper insertion depth. Moreover, Sprague Dawley (SD) rat experiments confirmed the effectivenessof optimal insertion parameters for transdermal drug delivery. This study illustrated not only thedevelopment of novel PMNs but also the mechanical evaluation for the design of PMNs.

The optimization of aldose reductase (AR) expression levels and tracking of the AR expression sites within the cell is an essential step in developing a platform for the effective production of aldose reductase inhibitors (ARIs). In this study, we have demonstrated the use of both immunocytochemistry and quantum dots-based immunofluorescence techniques for observing and detecting the expression level and localization of AR in the cytoplasm and cell membrane of a eukaryotic cell model with high levels of AR protein expression. Our results show that high expression levels of human AR can be achieved using the eukaryotic cell model that we have developed. The overexpressed AR can be used for translational studies of hAR and the screening of ARIs. More importantly, the use of the established quantum dots-based immunofluorescence technique in the intracellular labeling of AR allows the determination of the expression and distribution of the AR gene. Overall, the use of the interdisciplinary approach of both genetic engineering and quantum dot-based immunofluorescence allows not only the effective production of a desired protein, but also the determination of the cellular localization of such an expressed protein.
Case Study of Construction Cost Estimation in China Electric Power Industry Based on BIM Technology

Building Information Modeling (BIM) is mainly being promoted and applied in civil and public building cost management, but it is seldom used in power construction industry. Electric power construction project involves many professionals and different types of buildings and structures. Coupled with Chinese specialized quantity calculation rules, these factors led to some obstacles encountered when it comes to the application of BIM. In this paper, a substation is used as the object of the study, Revit software is used to explore 5D modeling method and other problems related to cost. Especially, quantity statistics according to 2013 edition’s power quota rules is the most difficult, three solutions were provided and one particularly detailed. It can be concluded that as long as it achieves data transfer, BIM 5D technology can be used in the power construction industry and it will bring huge impact on power engineering cost.

Bioassay-guided fractionation of $CHCl_{3}$ extract from the fermentation broth of a sponge Mycale plumose-derived actinomycete Saccharopolyspora sp. nov., led to the isolation of two known prodigiosin analogs - metacycloprodigiosin (1) and undecylprodigiosin (2). These compounds exhibited significant cytotoxic activities against five cancer cell lines: P388, HL60, A-549, BEL7402, and SPCA4. This is the first report on the significant cytotoxicity of metacycloprodigiosin (1) against human cancer cell lines.
No Association Between the GSTM1 Null Genotype and Risk of Renal Cell Carcinoma: A Meta-analysis

Background: Many studies have focused on possible associations between the glutathione S-transferase M 1 (GSTM1) null genotype and risk of renal cell carcinoma (RCC), but the impact remains unclear owing to obvious inconsistencies among the findings. The present study aimed to quantify the strength of any association in a meta-analysis. Methods: We searched the PubMed, Embase and CBM databases for studies concerning the association between the GSTM1 null genotype and risk of RCC. We estimated the summary odds ratio (OR) with its 95% confidence intervals (95% CI) to assess the association. Results: The meta-analysis showed the GSTM1 null genotype was not associated with risk of RCC overall (OR = 1.04, 95% CI 0.92-1.18, P = 0.501). For Caucasians, the GSTM1 null genotype was also not associated with risk of RCC (OR=1.02, 95% CI 0.90-1.16, P = 0.761). The cumulative meta-analyses showed a trend of no obvious association between GSTM1 null genotype and risk of RCC as information accumulated. Sensitivity analyses by omitting those studies also did not materially alter the overall combined ORs. No evidence of publication bias was observed. Conclusion: Meta-analyses of available data show that the GSTM1 null genotype is not significantly associated with risk of renal cell carcinoma.

To investigate the effects of electrolyzed watertreatment on the qualities of rice noodles prepared withsemidry- milled rice flour, pasting properties and thermalproperties of rice flour, and the cooking and texturalproperties of rice noodles were determined. Higher peakviscosity and lower melting enthalpy were observed inelectrolyzed water (EW) treated rice flour. The hardness,gumminess and chewiness of rice noodle in slightly acidicelectrolyzed water treated rice noodles with availablechlorine concentration (ACC) 20.32 mg/L were increasedsignificantly (p\0.05). The cooking loss decreased significantlyin strong acidic electrolyzed water treated noodleswith ACC 10.09 mg/L treatment (p\0.05). Theresults indicated that EW could promote the gelatinizationof rice flour, and improve the textural qualities of ricenoodles. Therefore EW was appropriate to be used in ricenoodle production.

A novel core–shell polymer (HBAPAM) with functionalized nano-SiO2 as core, multibranched associativepolymeric chains as shell was synthesized through in-situ free radical polymerization strategy. TGA, IR,1H NMR, and SEM proved the notion and method of preparing of HBAPAM was applicable. The core–shellstructure and multibranched morphology enhanced the supramolecular interaction of HBAPAM, whichdisplayed the desirable viscosified efficiency and viscoelasticity, especially, long-term stability in harshtemperature and salinity condition. HBAPAM featured the rheology in multiscale consistently indeionized water and formation water. Coreflooding experiments confirmed that HBAPAM had high costperformance in the variable permeability of oilfields.
