        Pharmacokinetics, tissue distribution and excretion of peimisine in rats assessed by liquid chromatography-tandem mass spectrometry

        Peimisine, the common ingredient of ‘‘zhebeimu’’groups and ‘‘chuanbeimu’’ groups, is responsiblefor the expectorant and cough relieving effects. The aim ofthis study was to investigate the pharmacokinetics, tissuedistribution and excretion of peimisine in male and femaleSD (Sprague-Dawley) rats by a rapid and sensitive LC-MS/MS (liquid chromatography-tandem mass spectrometry)method used carbamazepine as the internal standard afteroral administration, carbamazepine was stated as an IS. The results showed that peimisine was slowly distributed,and eliminated from rat plasma and manifested lineardynamics in a dose range of 0.26–6.5 mg/kg. Tested byANOVA, there were gender differences in the pharmacokineticparameters of AUC0-t, AUC0-? among a singledose of 0.26, 1.3, 6.5 mg/kg (P\0.05). Drug blood andtissue levels in male rats were significantly higher than thefemale counterparts after oral administration, while boththe males and the females showed high drug levels inspleen, kidney, lung, liver and heart. On the other hand, thepeimisine levels that can be reached in uterus, ovary, testisand brain is low. The excretion study showed that littleadministered peimisine (\0.7 %) was recovered in themale and female bile. Approximately 13.46 and 15.05 %were recovered in female urine and feces, while 43.07 and7.49 % were recovered in male urine and feces, respectively,which indicated that the major elimination route ofmale rats was urine excretion. In addition, there was significantdifferences in total cumulative excretive ratio ofpeimisine in feces (P\0.05) and no significant differencesin the urine (P[0.05) at a dose of 1.3 mg/kg.

        New Results on Finite-time Stabilization for Stochastic Systems with Time-varying Delay

        The paper deals with the problem of finite-time stabilization for stochastic systems with time-varying delay by defining a new criterion for finite-time stability. Firstly, by use of more appropriate Lyapunov-Krasovskii functional (LKF), the difficulties of finite-time stability confronted in system analysis and synthesis can be overcome. Then, a state feedback controller is constructed to guarantee the closed-loop system finite-time stable. New conditions for finite-time stability analysis as well as controller synthesis are established in terms of linear matrix inequality (LMI). Finally, two practical examples demonstrate the validity of the main results.

        Sodium alginate-based wall materials microencapsulated Lactobacillus plantarum CICC 20022: characteristics and survivability study

        In this study, microcapsules of Lactobacillus plantarum CICC 20022 (L. plantarum CICC 20022) were prepared by extrusion technique using sodium alginate (SA), sodium alginate-sodium caseinate (SA-SC) and sodium alginate-whey protein isolate (SA-WPI) as wall materials, respectively. Results showed that the best encapsulation yield of L. plantarum CICC 20022 was SA-WPI. Morphology and texture analysis showed that the microcapsules prepared by the SA-WPI system presented a more compact internal structure and higher resistance to external pressure. Fourier-transform infrared spectroscopy and low field nuclear magnetic resonance analysis demonstrated that the hydrogen bonding ability and network structure of the SA were improved by the addition of WPI. The survivability of L. plantarum CICC 20022 entrapped with the SA-WPI system was improved during freeze-drying and simulated gastrointestinal digestion. Therefore, the SA-WPI system can potentially be used as the vector of L. plantarum CICC 20022 in food applications.

        Profiling Total Viable Bacteria in a Hemodialysis Water Treatment System

        Culture-dependent methods, such as heterotrophic plate counting (HPC), are usually applied to evaluate the bacteriological quality of hemodialysis water. However, these methods cannot detect the uncultured or viable but non-culturable (VBNC) bacteria, both of which may be quantitatively predominant throughout the hemodialysis water treatment system. Therefore, propidium monoazide (PMA)-qPCR associated with HPC was used together to profile the distribution of the total viable bacteria in such a system. Moreover, high-throughput sequencing of 16S rRNA gene amplicons was utilized to analyze the microbial community structure and diversity. The HPC results indicated that the total bacterial counts conformed to the standards, yet the bacteria amounts were abruptly enhanced after carbon filter treatment. Nevertheless, the bacterial counts detected by PMA-qPCR, with the highest levels of 2.14 × 107 copies/100 ml in softener water, were much higher than the corresponding HPC results, which demonstrated the occurrence of numerous uncultured or VBNC bacteria among the entire system before reverse osmosis (RO). In addition, the microbial community structure was very different and the diversity was enhanced after the carbon filter. Although the diversity was minimized after RO treatment, pathogens such as Escherichia could still be detected in the RO effluent. In general, both the amounts of bacteria and the complexity of microbial community in the hemodialysis water treatment system revealed by molecular approaches were much higher than by traditional method. These results suggested the higher health risk potential for hemodialysis patients from the up-to-standard water. The treatment process could also be optimized, based on the results of this study.

        Effect of Ultrasonic Treatment on Grain Structure, Eutectic Phase and Mechanical Properties of an Al–6.2 wt% Cu Alloy

        The microstructural modification of eutectic phase is a crucial factor for the mechanical properties strengthening of the Al–Cu binary alloy. In this paper, Al–6.2% Cu melts were subjected to ultrasonic treatment from 710 or 660 °C with differentdurations. The microstructural evolutions of primary α-Al grains and α-Al + θ-Al2Cu eutectic phase under ultrasonicationwere investigated based on the nucleation and growth behavior. Results indicated that with the increase of ultrasonic processingtime, the growth of dendritic or petaloid grains were completely suppressed and noticeable refinement to globularmorphology were observed for α-Al grains. Meanwhile, broad lamella-like eutectic structure transformed to debris or evenparticles with continuous ultrasonication. Accordingly, the area fraction, average length and width of coarsening eutecticlamellas were dramatically reduced. In addition, the decrease of coarsening eutectic phase rather than the refinement ofα-Al grains was found to be the main contributor to the enhancement of mechanical properties for Al–6.2% Cu alloy. Themechanisms underlying the transformation of eutectic phase under ultrasonic treatment was also interpreted based on theexperimental results.

        Probiotics Increase Intramuscular Fat and Improve the Composition of Fatty Acids in Sunit Sheep through the Adenosine 5′-Monophosphate-Activated Protein Kinase (AMPK) Signaling Pathway

        This experiment aims to investigate the impact of probiotic feed on growth performance, carcass traits, plasma lipid biochemical parameters, intramuscular fat and triglyceride content, fatty acid composition, mRNA expression levels of genes related to lipid metabolism, and the activity of the enzyme in Sunit sheep. In this experiment, 12 of 96 randomly selected Sunit sheep were assigned to receive the basic diet or the basic diet supplemented with probiotics. The results showed that supplementation with probiotics significantly increased the loin eye area, and decreased plasma triglycerides and free fatty acids, increasing the content of intramuscular fat and triglycerides in the muscle and improving the composition of the fatty acids. The inclusion of probiotics in the diet reduced the expression of adenosine 5’-monophosphate-activated protein kinase alpha 2 (AMPKα2) mRNA and carnitine palmitoyltransferase 1B (CPT1B) mRNA, while increasing the expression of acetyl-CoA carboxylase alpha (ACCα) mRNA, sterol regulatory elementbinding protein-1c (SREBP-1c) mRNA, fatty acid synthase mRNA, and stearoyl-CoA desaturase 1 mRNA. The results of this study indicate that supplementation with probiotics can regulate fat deposition and improves the composition of fatty acids in Sunit sheep through the signaling pathways AMPK-ACC-CPT1B and AMPK-SREBP-1c. This regulatory mechanism leads to an increase in intramuscular fat content, a restructuring of muscle composition of the fatty acids, and an enhancement of the nutritional value of meat. These findings contribute to a better understanding of the food science of animal resources and provide valuable references for the production of meat of higher nutritional value.

        ELECTROCHEMILUMINESCENCE AND ELECTROCHEMISTRY FROM ZnO NANOPARTICLES IN ALKALINE AQUEOUS SOLUTION WITH PEROXYDISULFATE

        A strong electrochemiluminescence (ECL) emission from ZnO nanoparticles (NPs) suspended inaqueous solution at a Pt electrode or in NPs layers modi¯ed on a Pt or graphite electrode, whichoccurred between ? 1 : 0 V(versus Ag/AgCl, Sat. KCl) and ? 2 : 0 V, was observed in 0.25 M NaOHin the presence of peroxydisulfate. The ECL intensity was linear to the concentration of sus-pended ZnO NPs in the range from 1 : 0 ? 10? 5 to 1 : 0 ? 10? 3 g/mL. The ECL spectrum of ZnONPs revealed that both ZnO and peroxydisulfate-dissolved oxygen have been involved in the ECLprocess, and ECL spectrum has a red shift compared with photoluminescence (PL). The elec-trochemical redox behavior of ZnO NPs was also studied at a ZnO NPs modi¯ed carbon pasteelectrode by entrapping ZnO NPs in carbon paste, which reveals that the alkaline mediumchanged the surface state of ZnO NPs. The surface passivation has a signi¯cant e®ect on the ZnONPs'electrochemical behaviors.

        Effects of chronic alcohol exposure on ischemia–reperfusion-induced acute kidney injury in mice: the role of β-arrestin 2 and glycogen synthase kinase 3

        Little is known about the effects of chronic alcohol intake on the outcome of acute kidney injury (AKI). Hence, we examined the effects of chronic alcohol intake on the development of renal fibrosis following AKI in an animal model of bilateral renal ischemia–reperfusion (IR) injury. We first found that chronic alcohol exposure exacerbated bilateral IR-induced renal fibrosis and renal function impairment. This phenomenon was associated with increased bilateral IR-induced extracellular matrix deposition and an increased myofibroblast population as well as increased bilateral IR-induced expression of fibrosis-related genes in the kidneys. To explore the mechanisms underlying this phenomenon, we showed that chronic alcohol exposure enhanced β-arrestin 2 (Arrb2) expression and Akt and glycogen synthase kinase-3 (GSK3)β activation in the kidneys. Importantly, pharmacological GSK3 inhibition alleviated bilateral IR-induced renal fibrosis and renal function impairment. Furthermore, we demonstrated that Arrb2− / − mice exhibited resistance to IR-induced renal fibrosis and renal function impairment following chronic alcohol exposure, and these effects were associated with attenuated GSK3β activation in the kidneys. Taken together, our results suggest that chronic alcohol exposure may potentiate AKI via β-arrestin 2/Akt/GSK3β-mediated signaling in the kidney.
