      • 단백체를 이용한 애기장대 Cytokinin 유도 단백질의 분석

양영실,차준영,네티 엘마와티,정민희,배동원,이창원,손대영 Plant molecular biology and biotechnology research 2005 Plant molecular biology and biotechnology research Vol.2005 No.

Cytokinins are essential plant hormones that play crucial roles in various aspects of plant growth and development. To better understand the molecular mechanisms of cytokinin action, we identified cytokinin related proteins by a proteomic approach. Proteins extracted from control and trans-zeatin treated Arabidopsis seedlings were separated and analyzed by two dimensional gel analysis. Differentially expressed protein spots were identified with peptide mass fingerprinting based on matrix assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry and database searching. We obtained ten up-regulated and one down-regulated proteins upon t-zeatin treatment. The expression of the following proteins was induced; pollen allergen like protein, L-ascorbate peroxidase, tetrapyrrole methylase family protein, SGT1 protein homolog, disease resistance related protein, maternal embryogenesis control protein, paxneb related protein, gluthathione S-transferase and IAA amino acid hydrolase homolog.

Complementary DNA encoding chloroplast outer envelope membrane protein (OEP) from the halophyte Salicornia herbacea has been cloned and sequenced. The full length cDNA is 596 bp and encodes a polypeptide of 91 amino acid residues with a molecular mass of 8.9 kDa. The expression level of ShOEP increased by salt, drought and ABA treatments. ShOEP expression was largely induced in roots and shoots by high salts. The biological function of ShOEP was examined by yeast complementation. ShOEP can suppress Na+ sensitivity of yeast mutant (cnbΔ) in the presence of salt. These results suggest that ShOEP is a salt inducible gene and may have functions in the regulation of plant salt stress.

A BERRY-ESSEEN TYPE BOUND OF REGRESSION ESTIMATOR BASED ON LINEAR PROCESS ERRORS

        Liang, Han-Ying,Li, Yu-Yu Korean Mathematical Society 2008 대한수학회지 Vol.45 No.6

Consider the nonparametric regression model $Y_{ni}\;=\;g(x_{ni})+{\epsilon}_{ni}$ ($1\;{\leq}\;i\;{\leq}\;n$), where g($\cdot$) is an unknown regression function, $x_{ni}$ are known fixed design points, and the correlated errors {${\epsilon}_{ni}$, $1\;{\leq}\;i\;{\leq}\;n$} have the same distribution as {$V_i$, $1\;{\leq}\;i\;{\leq}\;n$}, here $V_t\;=\;{\sum}^{\infty}_{j=-{\infty}}\;{\psi}_je_{t-j}$ with ${\sum}^{\infty}_{j=-{\infty}}\;|{\psi}_j|$ < $\infty$ and {$e_t$} are negatively associated random variables. Under appropriate conditions, we derive a Berry-Esseen type bound for the estimator of g($\cdot$). As corollary, by choice of the weights, the Berry-Esseen type bound can attain O($n^{-1/4}({\log}\;n)^{3/4}$).

Fisetin Inhibits Cell Proliferation and Induces Apoptosis via JAK/STAT3 Signaling Pathways in Human Thyroid TPC 1 Cancer Cells

        Ying Liang,Deyu Kong,Yi Zhang,Siqi Li,Yan Li,Anuradha Ramamoorthy,Junfeng Ma 한국생물공학회 2020 Biotechnology and Bioprocess Engineering Vol.25 No.2

Thyroid cancer is the most important malignant tumor reported in human populations where, its treatment remains undeveloped. Fisetin, a plant flavonoid exhibits several pharmacological properties like antioxidant, antiinflammatory, and anticancer function. In the existing study, we assessed fisetin mediated cytotoxic effects and action potential of fisetin on cell proliferation in TPC-1 human cancer cells. Also, examined the apoptosis in TPC-1 cells by reactive oxygen species (ROS) generation, the mitochondrial membrane potential (MMP) and apoptotic morphological changes through AO/EtBr staining. Additionally, we analyzed the effects of fisetin through ELISA analysis to evaluate the caspases expression and studied JAK 1 and STAT3 signaling molecule in TPC1 cells. Our results demonstrated that fisetin stimulated apoptosis, which confirmed through reduced cell viability, improved ROS generation, altered MMP and cell cycle phases in TPC-1 cells. Further, the fisetin up-regulated the expression of caspase (3, 8, and 9) expressions in TPC-1 cells. Also, we observed the fisetin down-regulated the JAK 1 and STAT3 expression in TPC1 cells. Thus, the fisetin induced apoptosis in TPC-1 cells by the induction of oxidative damage and enhanced caspases expression by down-regulating JAK 1 and STAT3 signaling molecules. Hence, the fisetin would be considered as a beneficial therapeutic agent for the thyroid cancer treatment.

The Arabidopsis beta-carotene hydroxylase gene promoter for a strong constitutive expression of transgene

        Ying Shi Liang,Hee-Jin Bae,Sang-Ho Kang,Theresa Lee,Min Gab Kim,Young-Mi Kim,하선화 한국식물생명공학회 2009 Plant biotechnology reports Vol.3 No.4

To efficiently express a gene of interest in transgenic plants, the choice of promoter is a crucial factor as it directly affects the expression of the transgene that will yield the desired phenotype. The Arabidopsis b-carotene hydroxylase 1 gene (AtBch1) shows constitutive and ubiquitous expression and was thus selected as one of best candidates for constitutive promoter analysis by both in silico northern blotting and semi-quantitative RT-PCR analysis. To investigate AtBch1 promoter activity, the 1,981-bp 50-upstream region of this gene was fused with b-glucuronidase (GUS) and transformed into Arabidopsis. Through the molecular characterization of transgenic leaf tissues, the AtBch1 promoter generated strong activity that drives 1.8- and 2-fold higher GUS expression than the cauliflower mosaic virus 35S (35S) promoter at the transcriptional and translational levels, respectively. Furthermore, the GUS enzyme activity driven by the AtBch1 promoter was 2.8-fold higher than that produced by the 35S promoter. By histochemical GUS staining, the ubiquitous expression of the AtBch1 promoter was observed in all tissues of Arabidopsis. Semi-quantitative RT-PCR analysis with different tissues further showed that this promoter serves as a strong constitutive driver of transgene expression in dicot plants. The Quantitative Evaluation of Aging State of Field Composite Insulators Based on Trap Characteristics and Volume Resistivity-Temperature Characteristics

        Ying Liang,Li-Juan Gao,Ping-Ping Dong,Ting Gao 대한전기학회 2018 Journal of Electrical Engineering & Technology Vol.13 No.3

In order to obtain a better understanding of the ageing process of the field composite insulators, it is necessary to explore a quantitative-valuation method for the aging state evaluation. And the linear relationship between volume resistivity and temperature is proposed. In this paper, the composite insulators with different lengths of operating lives from two manufacturers were tested. The relationship between trap characteristics and volume resistivity-temperature characteristics were analyzed based on Thermal Stimulated Current (TSC), volume resistivity-temperature test, Scanning Electron Microscope (SEM) and Fourier Transform Infrared Spectroscopy (FTIR). Furthermore, the application of trap characteristics in the quantitative evaluation of aging state of composite insulators was discussed. The results showed that there was a general negative correlation between the relative variation ratio of trap charges and the volume resistivity-temperature characteristics. Meanwhile, the physicochemical properties would change with the aging time, which would result in the increasing of electron traps. Combined with the TSC and volume resistivity test results, the trap characteristic thresholds which indicated the serious age of the composite insulators had been proposed.

Chinese patent herbal medicine (Shufeng Jiedu capsule) for acute upper respiratory tract infections: A systematic review and meta-analysis

        Ying-ying Zhang,Ru-yu Xia,Shi-bing Liang,Xiao-yang Hu,Meng-yuan Dai,Yi-lin Li,Le-yi Zhao,Michael Moore,Yu-tong Fei,Jian-ping Liu 한국한의학연구원 2021 Integrative Medicine Research Vol.10 No.3

Background: Shufeng Jiedu capsule has been widely used in China for acute upper respiratory tract infections (AURTIs). The aim of this study was to evaluate its effectiveness and safety for AURTIs. Methods: Randomized controlled trials comparing SFJD with conventional drug for patients with AURTIs were included. Eight databases were searched from their inceptions to February 2021. Data was synthesized using risk ration (RR) or mean difference (MD) with their 95% confidence interval (CI). The primary outcome was resolution time of typical symptoms. Results: Twenty-five RCTs involving 3410 patients were included. SFJD in combination with conventional drug was associated with; in common cold shortening the duration of fever (MD −1.54 days, 95% CI [−2.15,−0.92], I2 = 80%, n = 385, 3 trials) and cough (MD −1.22 days, 95% CI [−1.52, −0.93]); in herpangina, shortening the duration of fever (MD -0.68 days, 95% CI [−1.15, −0.21], I2 = 68%, n = 140, 2 trials) and blistering (MD −0.99 days, 95% CI [−1.23, −0.76], n = 386, 3 trials); in acute tonsillitis and acute pharyngitis shortening the duration of fever (MD −1.13 days, 95% CI [−1.36, −0.90], I2 = 33%, n = 688, 7 trials) and sore throat (MD −1.13 days, 95% CI [−1.40, −0.86], I2 = 84.1%, n = 1194, 10 trials). SFJD also improving their cure rate with a range (1–5 days). No serious adverse events were reported. Conclusion: Low certainty evidence suggests that SFJD appears to shorten the duration of symptoms in AURTIs, improve cure rate and seems safe for application. However, high quality placebo controlled trials are warranted to confirm its benefit.

DNA methylation levels in different tissues in tea plant via an optimized HPLC method

        Ying Gao,Jia-Li Hao,Zhen Wang,Kwan-Jeong Song,Jian-Hui Ye,Xin-Qiang Zheng,Yue-Rong Liang,Jian-Liang Lu 한국원예학회 2019 Horticulture, Environment, and Biotechnology Vol.60 No.6

DNA methylation is one of the most important events in epigenetics and significantly influences plant growth and development. In the present study, we established and optimized a high-performance liquid chromatography method for detecting the base composition in DNA in tea plant (Camellia sinensis L.) tissues by using saline buffers-free mobile phases. The DNA methylation level varied with tea plant tissue, cultivar, and growth stage. A relatively higher DNA methylation level was observed in tender leaf (38.34%) and pistil (38.19%) tissues, while a relatively low level was detected in capillary root (19.45%), stamen (19.61%), and old leaf (20.70%) tissues. The pattern of the methylation level formed a saddle curve during the growth of dormant buds in spring; the lowest point appeared at the stage of one leaf and a bud. The methylation level in the adventitious buds regenerated from the branch after pruning seemed to decrease with an increase in the degree of pruning. These DNA methylation levels might be associated with the development of tea plant.

Identification and Molecular Evolutionary Analysis of TLR5 Gene from the Tibetan macaque (Macaca thibetana)

        Ying Huang,Yong-Fang Yao,Yan Li,Qing-Yong Ni,Liang Zhou,Shuan-Ling Zhang,Wei Liu,Huai-Liang Xu 한국유전학회 2012 Genes & Genomics Vol.34 No.4

In order to identify the Toll-like receptor 5 (TLR5) as a putative candidate disease-resistance gene in Tibetan macaque (Macaca thibetana), two pairs of primers were designed based on the TLR5 gene sequence of rhesus macaque (Macaca mulatta,NM_001130429). The primers were used to amplify the TLR5 gene from Tibetan macaque, by the polymerase chain reaction (PCR). The compiled sequences were analyzed by bioinformatics. The DNA sequencing and additional combined results showed that the Tibetan macaque TLR5 gene is about 2825 bp and contains an open reading frame of 2577 bp encoding for 858 amino acids. Homology analysis of TLR5 in both species showed that the amino acid and nucleotide identity is about 99.7% and 99.8% and their transmembrane and intracellular domains appeared more conservative than the extracellular domains of proteins. However, re-examining the entire Tibetan macaque TLR5 coding sequence we found that a purifying selection was also acting on the TLR5 gene region encoding for its intracellular domain of the protein. Even though the selection tests indicated that the TLR5 gene experienced a strong purifying selection in the process of evolution, most likely because its potential role in the primate adaptive immune defense, the Tibetan macaque still has the highest re-lationship with the rhesus macaque.
