        Reconsidering cell-assisted lipotransfer for breast augmentation: effect of stromal vascular fraction enrichment on graft survival assessed with 3-dimensional laser scanning

        Background: Cell-assisted lipotransfer (CAL) has been proposed to be beneficial for improving graft retention. Clinically, CAL involves the isolation of the stromal vascular fraction (SVF) from a portion of the lipoaspirate at the time of surgery. However, most studies related to SVF breast augmentation lacked a rigorous methodology and well-designed control. Objective: We aimed to determine the potential improvement of SVF enrichment in fat grafting for breast augmentation with objective volume assessment. Methods: From April 2015 to January 2016, 169 patients were enrolled after applying the exclusion criteria. Among them, 97 patients who underwent conventional fat grafting for breast augmentation were assigned to group A. The other 72 patients underwent SVFenriched fat grafting for breast augmentation and were assigned to group B. A retrospective comparative study was conducted to evaluate the graft survival using 3-dimensional laser scanning. Results: There was no significant difference between the 2 groups in terms of mean age, original breast volume, grafted fat volume, and postoperative weight change. Breast volume assessments revealed that the percentage of graft survival at 12 months was 69.2% in group A and 71.1% in group B, with no significant difference (p=0.641). The preoperative body mass index was significantly lower in group A than in group B. The volume of suctioned fat was significantly less in group A. The operation time was significantly shorter in group A. The postoperative complication rates were significantly lower in group A than in group B. Conclusion: SVF-enriched fat grafting for breast augmentation was associated with a larger amount of harvested fat, a longer operation time, and a higher incidence of complications. The graft retention rate was not significantly increased. The findings of our study do not support the use of SVF in fat grafting for breast augmentation. Level of Evidence: IV

        Runner system plays a very important role in injection molding process. A quality runner design is helpful in improving product qualities and saving material. However, traditional cold runner systems have certain inherent issues. Moreover, poor product cosmetics are commonly seen with the use of traditional cold runners. As a result, hot runner technology has been widely applied. On the other hand, It has been one of the green molding solutions for material/energy saving and clean environments. But the mechanism behind the hot runner system is too complicated to be fully understood. There exist some critical issues currently. As a result, the simulation technolgy is highly needed to examine hot runner designs before the real manufacturing. Through simulation analyses, designers and manufafctuers are able to catch the potential issues on their hot runner systems and revise their designs. Hot runner simulation technology helps with the investigations into the behavior in hot runner system. In this paper, a true 3D numerical method is proposed and applied to investigate the temperature behavior in a real hot runner system for PC material. The experiment is conducted and the simulating result is compared with that from the experiment for the validation purpose.

        A Neuroprotective Action of Quercetin and Apigenin through Inhibiting Aggregation of Aβ and Activation of TRKB Signaling in a Cellular Experiment

        Alzheimer’s disease (AD) is a neurodegenerative disease with progressive memory loss and the cognitive decline. AD is mainly caused by abnormal accumulation of misfolded amyloid β (Aβ), which leads to neurodegeneration via a number of possible mechanisms such as down-regulation of brain-derived neurotrophic factor-tropomyosin-related kinase B (BDNF-TRKB) signaling pathway. 7,8-Dihydroxyflavone (7,8-DHF), a TRKB agonist, has demonstrated potential to enhance BDNF-TRKB pathway in various neurodegenerative diseases. To expand the capacity of flavones as TRKB agonists, two natural flavones quercetin and apigenin, were evaluated. With tryptophan fluorescence quenching assay, we illustrated the direct interaction between quercetin/ apigenin and TRKB extracellular domain. Employing Aβ folding reporter SH-SY5Y cells, we showed that quercetin and apigenin reduced Aβ-aggregation, oxidative stress, caspase-1 and acetylcholinesterase activities, as well as improved the neurite outgrowth. Treatments with quercetin and apigenin increased TRKB Tyr516 and Tyr817 and downstream cAMP-response-element binding protein (CREB) Ser133 to activate transcription of BDNF and BCL2 apoptosis regulator (BCL2), as well as reduced the expression of pro-apoptotic BCL2 associated X protein (BAX). Knockdown of TRKB counteracted the improvement of neurite outgrowth by quercetin and apigenin. Our results demonstrate that quercetin and apigenin are to work likely as a direct agonist on TRKB for their neuroprotective action, strengthening the therapeutic potential of quercetin and apigenin in treating AD.

        Experience in Professional Resilience for Nurses Caring for Patients with COVID-19: A Qualitative Descriptive Study

        Purpose During the COVID-19 pandemic, nurses have faced many professional and ethical dilemmas and challenges along with bearing physical, mental, and emotional stress resulting from worrying about themselves or their family being infected and stigmatized. This stress can potentially lead to burnout and resignation. Professional resilience is crucial for nurses to cope with these adverse situations. This study aimed to investigate the process by which nurses adapt, change, and overcome challenges during the COVID-19 pandemic and ultimately demonstrate professional resilience. Methods Descriptive phenomenology was applied. Semi-structured interviews were conducted with 11 nurses working in COVID-19 wards and intensive care units to collect data. Giorgi's phenomenological analysis method was employed. Results Based on the interview responses, four major themes were identified: 1) balancing patient care, self-protection, and passing on experience; 2) providing timely pandemic team resources and social support; 3) nurses' perseverance amid social discourse and constrained lives; and 4) selfless dedication shaping nursing's pinnacle experiences. Conclusions In the face of a sudden pandemic, frontline nurses play a critical role in maintaining medical capacity. Consequently, they must balance their families, lives, and work while adapting to the impact of the pandemic and changing practices and procedures based on the development of the pandemic and policy demands. The study findings provide insights into the challenges and emotional experiences encountered by nurses during a sudden pandemic outbreak and can serve as a reference for developing strategies to help nurses overcome these challenges and enhance their professional resilience.

        Three methods for studying coupled vibration in a multi flexible disk rotor system

        This paper improved and developed Shaft-disk-blade (SDB) rotor system based on the previous studies of the authors in the last decade from Yang and Huang (2005) to Chiu et al. (2017). This paper also explored blade-bending, disk-transverse, and shaft-torsion coupling vibration of a multi flexible disk rotor system. Unlike the previous studies of the authors, this paper adopted three methods: (a) Assumed mode method (AMM), (b) Finite element method (FEM), and (c) experimental method. The first approach is the main method, and the two other methods are complementary. Results generated from the three methods were then compared and analyzed. Based on the previous definition of the authors, a flexible disk rotor system displays three types of coupling vibrations: Inter-blade, SDB, and diskblade modes. The system changes the rules of natural frequencies and mode shapes. This paper presents several interesting results. First, the author determined the change rules of the mode shapes and natural frequencies using the AMM, FEM (including three kinds of software), and the experimental method. Second, numerical calculation results also revealed that two phenomena regarding the distance of disk and flexible disk would be affected by the natural frequencies. Third, the experimental results would be explored in this paper. Last, the flexible disk could affect the system instability in the case study of rotation effects.

        Research on effect of rings on coupling vibration in a rigid-disk rotor

        This paper uses new deductions of the equation of motion and obtains new modes of change that differ from the authors’ previous research. The blade-bending, shaft-torsion and ringbending coupled vibration influence of a single-disk rotor whose blade subsystem was coated with a ring is investigated. In the last two years, the authors adopted two methods, the assumed mode method and finite element method, and compared both parties results. This article continues that work. In this paper, the phenomena of ring joint coupling vibrations are different and interesting. The authors defined two new types, the shaft-blade-ring (SBR) and blade-ring (BR) modes. These two coupling modes can be mutually affected by the shaft, blade, and ring, and no single subsystem predominates. The new change rules of natural frequencies of rotors with two to 15 blades and those of mode shapes are discussed. The influence of the ring thickness and rotational speed are also studied in this paper.

        Lactoferrin Increases Antioxidant Activities and Ameliorates Hepatic Fibrosis in Lupus-Prone Mice Fed with a High-Cholesterol Diet

        Lactoferrin (LF) has beneficial effects against various diseases. However, the effects of LF on liver fibrosis in systematic lupus erythematosus (SLE) are unknown. In this study, NZB/W F1 mice were utilized to investigate the effects of LF on SLE. Experiments reveal that LF significantly increases glutathione and 1,1-diphenyl-2-picryl-hydrazyl levels and significantly decreased malondialdehyde levels in both serum and liver in NZB/W F1 mice. LF also lowered matrix metalloproteinase-9 activity and liver inflammatory indices, such as aminotransferase and alanine aminotransferase. Notably, significantly decreased expression of fibrotic related molecules, including transforming growth factor (TGF)-β1, tumor necrosis factor-α, interleukin-1β, and TGF-β1 receptor, were observed in the livers of NZB/W F1 mice that had been treated with LF. Significantly, suppressed Smad2/3 signaling, α-smooth muscle actin, and collagen deposition were also detected. These findings reveal that LF has beneficial effects on SLE by increasing antioxidant activities and ameliorating liver inflammation and fibrosis, suggesting the therapeutic effectiveness of LF against SLE.

      • A meta-analysis of genome-wide association studies for adiponectin levels in East Asians identifies a novel locus near <i>WDR11-FGFR2</i>

        <P>Blood levels of adiponectin, an adipocyte-secreted protein correlated with metabolic and cardiovascular risks, are highly heritable. Genome-wide association (GWA) studies for adiponectin levels have identified 14 loci harboring variants associated with blood levels of adiponectin. To identify novel adiponectin-associated loci, particularly those of importance in East Asians, we conducted a meta-analysis of GWA studies for adiponectin in 7827 individuals, followed by two stages of replications in 4298 and 5954 additional individuals. We identified a novel adiponectin-associated locus on chromosome 10 near <I>WDR11-FGFR2</I> (<I>P</I> = 3.0 × 10<SUP>−14</SUP>) and provided suggestive evidence for a locus on chromosome 12 near <I>OR8S1-LALBA</I> (<I>P</I> = 1.2 × 10<SUP>−7</SUP>). Of the adiponectin-associated loci previously described, we confirmed the association at <I>CDH13</I> (<I>P</I> = 6.8 × 10<SUP>−165</SUP>), <I>ADIPOQ</I> (<I>P</I> = 1.8 × 10<SUP>−22</SUP>), <I>PEPD</I> (<I>P</I> = 3.6 × 10<SUP>−12</SUP>), <I>CMIP</I> (<I>P</I> = 2.1 × 10<SUP>−10</SUP>), <I>ZNF664</I> (<I>P</I> = 2.3 × 10<SUP>−7</SUP>) and <I>GPR109A</I> (<I>P</I> = 7.4 × 10<SUP>−6</SUP>). Conditional analysis at <I>ADIPOQ</I> revealed a second signal with suggestive evidence of association only after conditioning on the lead SNP (<I>P</I><SUB>initial</SUB> = 0.020; <I>P</I><SUB>conditional</SUB> = 7.0 × 10<SUP>−7</SUP>). We further confirmed the independence of two pairs of closely located loci (<2 Mb) on chromosome 16 at <I>CMIP</I> and <I>CDH13</I>, and on chromosome 12 at <I>GPR109A</I> and <I>ZNF664</I>. In addition, the newly identified signal near <I>WDR11-FGFR2</I> exhibited evidence of association with triglycerides (<I>P</I> = 3.3 × 10<SUP>−4</SUP>), high density lipoprotein cholesterol (HDL-C, <I>P</I> = 4.9 × 10<SUP>−4</SUP>) and body mass index (BMI)-adjusted waist–hip ratio (<I>P</I> = 9.8 × 10<SUP>−3</SUP>). These findings improve our knowledge of the genetic basis of adiponectin variation, demonstrate the shared allelic architecture for adiponectin with lipids and central obesity and motivate further studies of underlying mechanisms.</P>
