        In order to investigate the pharmacological properties of New Wonbang Woohwangchungsimwon Liquid (NSCL), effects of Wonbang Woohwangchungsimwon Liquid (SCL) and NSCL were compared. In isolated rat aorta, NSCL and SCL showed the relaxation of blood vessels in maximum contractile response to phenylephrine (10^-6M) regardless to intact endothelium or denuded rings of the rat aorta. Furthermore, the presences of the inhibitor of NO synthase and guanylate cyclase did not affect the relaxing effect of NSCL and SCL. NSCL and SCL inhibited the vascular contractions induced by acetylcholine, prostaglandin endoperoxide or peroxide in a dose-dependent manner. In conscious spontaneously hypertensive rats (SHRs), NSCL and SCL significantly decreased heart rate. NSCL and SCL, at high doses, had a negative inotropic effect that was a decrease of left ventricular developed pressure and (-dp/dt)/(+dp/dt) in the isolated perfused rat hearts, and also decreased the contractile force and heart rate in the isolated rat right atria. In excised guinea-pig papillary muscle, NSCL and SCL had no effects on parameters of action potential such as resting membrane potential, action potential amplitude, APD_90 and V_max at low doses, whereas inhibited the cardiac contractility at high doses. These results suggested that NSCL and SCL have weak cardiovascular effects with relaxation of blood vessels and decrease of heart rate, and that this effect is no significant differences between cardiovascular effects of two preparations.

Furthermore, the presence of the inhibitors of NO synthase and guanylate cyclase did not aftect significantly the relaxing effects of NSCH and SCH, NSCH and SCH inhibited the vascular contractions induced by acetylcholine, prostaglandin endoperoxide or peroxide in a dose-dependent manner. In conscious spontaneously hypertensive rats (SHRs). NSCH and SCH decreased significantly heart rate. These, at high doses, had a negative inotropic effect that was a decrease of left ventricular developed pressure and (-dp/dt)/(+dp/dt) in the isolated perfused rat hearts, and also decreased the contractile force and heart rate in the isolated rat right atria. In guinea-pig papillary muscle, these had no effects on parameters of action potential such as action potential amplitude (APA). V_max and resting membrane potential (RMP) at low doses, whereas inhibited the cardiac contractility at high doses. Furthermore, these had a significant inhibitory effects on palpitation of the heart in normotensive rats and SHRs. These results suggest that NSCH and SCH have weak cardiovascular effects, and that there is no significant differences between cardiovascular effects of two preparations.

        Significant Change of Stress-related Proteins and Gene Profile in the Liver of Long-term Ground Transported Animals

        The transportation stress cause rise in the levels of many hormones and proteins to maintain homeostasis in the laboratory animals. In order to study whether the stress induced by long-term ground transport could contribute to change in the stress-related protein and profile of stress-regulated gene expression, these factors were detected in the animals between before and after the ground transportation. Herein, it is concluded that; ⅰ) the transported mice significantly exhibited the increase of corticosterone concentration in serum. ⅱ) the heat shock protein 70 (HSP70) in the liver were significantly decreased in the transported animals. ⅲ) 372 and 272 genes were up- and down-regulated, respectively, by stress in the long-term ground transported mice compare to the controls. ⅳ) genes encoding for immunity, signal transduction response and response to protein metabolism were significantly over-represented in the up-regulated group. ⅴ) among the down-regulated transcripts, those encoding for protein metabolism were dramatically over-represented, followed by those related to lipid metabolism and development process. These results suggested that the stress induced by ground transportation could significantly changed the level of stress-indicator hormone, proteins and gene profile in the mice. Therefor, the animal transported a long distance should be had the adaptation period to recover the normal physical state.

        Survey on Current Status of Laboratory Animals for Establishing New National Policy

        The animal quality control is very important to acquire the trustable results from the animal experiment. To survey the current situation on the quality control in laboratory animals care and use program in Korea, a total of 223 institutions having laboratory animals was chosen to search their general current situations on the microbiological monitoring, the genetic monitoring, and the quality control of diet and water. The only 12% of institutions has been doing the microbiologic monitoring, while 40% of institutions has received the certification for animal quality from the animal company. There is also rare the institutions conducting the quality control of diet, and water and bedding. These results suggested that the levels of animal quality were extremely low spite of increasing of animal users, and thereby recognizing necessity of support at the national level.

        Comparative Study of Recombinant Human Bone Morphogenetic Protein-2 Carriers in Rat Subcutaneous Tissues: Pilot Study

        The purpose of this study was to evaluate and compare the potential of using various biomaterials as a carrier of rhBMP-2 in the subcutaneous layer of rats. 5 subcutaneous defects were made in each of the dorsal surfaces of 10 rats. Then, the rats were divided into the following groups: Control group: sham-surgery control, Experimental group 1: rhBMP-2/Bio-Oss®, Experimental group 2: rhBMP-2/Collatape®, Experimental group 3: rhBMP-2/nCS. Histological and histometrical analysis were performed after the rats were sacrificed at 2 and 4 weeks. Regardless of the type of carrier, the groups treated with rhBMP-2 showed significantly greater bone formation when compared to the control group. The amount of new bone formation was greater at 4 week than 2 week. Total bone formation (%) at 4 week was the greatest in rhBMP/Bio-Oss® (26.76±10.13) group, followed by rhBMP/Collatape® (20.38±5.27) and rhBMP/nCS (4.34±2.58). Within the limitation of this study, the results showed that Bio-Oss®, Collatape®, nanocalcium sulfate all showed new bone formation when used as a carrier of rhBMP-2 in the subcutaneous layer of rats, indicating potential use as carriers for rhBMP-2.         사용자가 방문한 웹 사이트와 연관된 광고를 웹 페이지에 실어주는 문맥광고 관련 연구가 광고 효율성 측면에서 최근 주목을 받고 있다. 이러한 문맥광고 관련 연구의 핵심은 웹 페이지와 웹 광고간의 연관성을 높여주는데 있는데, 연관성 향상 방안으로 최근 사용자 의도 분석을 통한 연관성 향상 기법이 많이 연구되고 있다. 그러나 기존 연구에서는 사용자가 로그인을 해야 하거나, 로그 정보를 일정기간이상 수집해야만 사용자 의도 분석이 가능 하다는 문제점이 존재 한다. 본 논문에서는 로그인이나 많은 양의 로그정보 수집 없이 한 세션 내에서 방문한 웹 페이지 로그만을 이용하여 개인화된 문맥 광고를 제공하는 문맥광고 기법을 제안한다. 실험 결과에서는 제안하는 기법이 기존의 광고 기법에 비해 사용자가 판단하는 웹 페이지와 웹 광고의 연관성 (precison) 이 기존의 기법에 비해 높아짐을 증명한다.

        모바일 네트워크에서는 모바일 노드의 이동성으로 인하여 네트워크가 빈번하게 분리되며, 이로 인해 데이터 접근성이 낮아지는 문제가 발생한다. 기존 연구에서는 이러한 문제 해결하기 위해 다양한 복제본 할당 기법이 연구되었다. 그러나 지금까지 연구된 복제본 할당기법들은 모바일 애드혹 네트워크 환경의 제한된 자원으로 인한 노드들의 이기적인 행동에 대해 고려하지 않았다. 본 논문은 모바일 애드혹 네트워크 상의 이기적인 노드들을 검출하기 위해 이진결정 기법과 레벨화 기법을 제안하고, 이를 통해, 이기적인 노드들의 영향을 최소화하는 기법을 제안하고자 한다. 실험 결과는 제안하는 기법이 기존 기법에 비해 모바일 애드혹 네트워크 내에 이기적인 노드들이 있는 환경에서 데이터 접근성이 향상됨을 보여준다.

        난용성 약물인 Isoflavone계 화합물을 함유한 하이드로콜로이드 패치의 물리화학적 특성

        Genistein과 daidzein은 isoflavone 계열의 약물로써 다양한 암의 치료 및 예방, 항산화, 항염 활성 효과를 나타내는 물질이다. 그러나 이 약물들은 용해성이 좋지 않아 약물을 포함하는 크림 형태의 에멀젼에 적용하여 피부를 통해 전달하기 어렵다. 본 연구에서는 하이드로콜로이드 패치라는 TDDS(transdermal drug delivery system)를 이용하여 genistein과 daidzein과 같은 난용성 약물을 피부에 전달하였다. 패치가 에멀젼보다 피부로 잘 흡수되는 것을 돼지 피부(porcine skin)를 이용한 투과 실험 및 confocal microscopy로 확인하였으며, 이는 패치의 밀폐효과(occlusive effect)에 의해 피부 내로 잘 흡수된 것이다. 하이드로콜로이드 패치는 에멀젼 형태의 크림보다 난용성 약물을 피부내로 전달하는데 더 좋은 방법이며, 사용하기 편리하므로 효과적인 화장품으로써 가치가 있다. Genistein and daidzein, phytoestrogens belonging to the isoflavone class, have a role in the treatment and prevention of various cancers, antioxidant and anti-inflammation activity. However, it is difficult to deliver through the skin from applying the emulsion such as cream containing them because these drugs are nearly insoluble in aqueous solutions. In this study, the hydrocolloid patch including poorly soluble drugs such as genistein and daidzein was used as the transdermal drug delivery system (TDDS) to deliver them through the skin. It was confirmed that the patches are helpful for the drugs to be absorb much and more deeply than the cream due to the occlusive effect of patch, which was verified from the porcine skin and the confocal microscopy. The hydrocolloid patch is a better way to deliver the poorly soluble drugs through the skin than the general cosmetics such as emulsion-based cream, and is valuable as effective cosmetics due to its convenient usage.
