The effects of Laminaria japonica fucoidan extract (LJFE) on the biochemical parameters of lipid-related function were investigated in liver and serum of CCl_(4)-treated rats. LJFE of 100 mg/kg concentration was intraperitoneally administered into rats at dose of 1.5 ml/kg for 14 days. On the day 15, 3.3 ml/kg of CCl_(4) dissolved in olive oil (1:1) was injected 12 hours before anesthetization. We examined the lipid-related functions and enzyme activitys by measuring the levels of total cholesterol (TC), triglyceride (TG), alkaline phosphatase (ALP) and lactate dehydrogenase (LDH) in serum. The results showed that CCl_(4) treatment increased the high values of TG, LDH and ALP decreased the low values of TC but LJFE pretreatment decreased the high values of TG, LDH and ALP to the low values, increased the TC. It was suggested that LJFE can be used as the potential candidate for the lipid-metabolism natural supplement.

In the present study the hepatoprotective and antioxidative effects of Epimedium koreanum (EK) and Salicornia herbacea (SH) were investigated against carbon tetrachloride (CCl_(4))-induced liver damage in rats. EK and SH (100 mg/kg) were administered into rats intraperitoneally (i.p) for 2 weeks. After CCl_(4) a dose of 3.3ml/kg was administered to induce hepatotoxicity. This antioxidative effects of EK and SH on CCl_(4) -treated rats were measured through the activity of Aspartate aminotransferase (AST), Alanine aminotransferase (ALT) in serum and Superoxide dismutase (SOD), Catalase (CAT) in liver tissue. EK or SH-administered and CCl_(4) -treated (ECT or SCT) groups showed inhibitory effects in AST and ALT activities compared to CCl_(4) -treated control (CTC) group. SOD and CAT in ECT and SCT groups were increased compared to those of CTC group.

The aim of this study was to elucidate the antioxidant properties of fucoidan extracts (FE) against $CCl_4$-induced oxidative stress by monitoring the levels of glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT), alkaline phosphatase (ALP), lactate dehydrogenase (LDH), malondialdehyde (MDA), superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx). Female, Sparague-Dowley rats were administered with FE (100 mg/kg daily) for 14 days and $CCl_4$ on the 15'th day, 12 h before they were sacrificed. The levels of GOT, GPT, ALP and LDH in serum of rats, as well as the levels of MDA, SOD, CAT and GPx in total liver homogenate were analyzed. $CCl_4$-treatment was found to increase the levels of GOT, GPT, ALP, LDH and MDA, as well as decrease levels of SOD, CAT and GPx significantly. The pre-treatment of rats with FE, however, suppressed the increment of levels of GOT, GPT, ALP, LDH and MDA, as well as recovered the levels of SOD, CAT and GPx in CCl4-treated rats. Moreover there was a significant decrease in incidences of necrosis and cirrhosis in the liver tissue of FE-treated rats. These results implied that FE possessed antioxidant properties against $CCl_4$-induced oxidative stress.

<P>Angiotensin II (Ang-II) and vascular endothelial growth factor (VEGF) have an important role in the pathogenesis of diabetic nephropathy, but the signaling cascade of VEGF regulation in response to Ang-II in podocytes is largely unknown. In these experiments, we looked at the effect of Ang-II on the production of VEGF, and investigated whether VEGF production depends on the p38 mitogen activated protein kinase (MAPK) pathway in cultured mouse podocytes. Incubation of podocytes with Ang-II induced a rapid increase in VEGF mRNA expression and protein synthesis as well as its transcriptional activity in an Ang-II dose-dependent manner. To further define the role of angiotensin type 1 (AT1) and type 2 (AT2) receptors involved in Ang-II-mediated VEGF synthesis, the effects of selective AT1 and AT2 receptor antagonists were evaluated. Prior treatment with losartan significantly inhibited VEGF mRNA and protein synthesis induced by Ang-II, which suggests that the AT1 receptor is involved in Ang-II-mediated VEGF synthesis. Furthermore, stimulation of the cells with Ang-II increased both phosphorylation of p38 MAPK and MAP kinase kinase 3/6 (MKK3/6). Additionally, Ang-II enhanced the DNA binding activity to cAMP response element binding protein (CREB) and phosphorylation of CREB. In addition, to investigate the role of p38 MAPK in Ang-II-induced VEGF synthesis, podocytes were pretreated with or without the p38 MAPK inhibitor, SB203580 for 24 h to observe whether Ang-II-mediated VEGF synthesis was inhibited by blocking p38 MAPK. The addition of SB203580 led to a marked inhibition of the increased VEGF mRNA and protein production induced by Ang-II in a dose-dependent manner. Taken together, these results suggest that Ang-II stimulates the synthesis of VEGF in podocytes and the production of VEGF induced by Ang-II is mediated, in part, through the activation of the p38 MAPK pathway.</P>

Fungal perylenequinones have photodynamic activity and are promising photosensitizers for photodynamic therapy (PDT). Here, we investigated the bactericidal and antitumor activities of phleichrome from the fungal perylenequinone family in vitro. Photodynamic bactericidal activity of phleichrome was analyzed by agar-well diffusion method under dark and illuminated conditions. The photodynamic antitumor activity of phleichrome was analyzed in MCF-7, HeLa, SW480, and HepG2 human cancer cell lines using in vitro cytotoxicity assays. Photodynamic bactericidal activities against Gram-negative and Gram-positive bacteria were species-specific. Antitumor activity against all tumor cell lines increased under the illuminated condition. Depending on the results of the analyses, Phleichrome has potential for further drug development related to its antibacterial and antitumor activities.
