        Lipopolysaccharide와 어성초 혼합 추출물의 지질대사 상관성

        본 연구는 어성초 함유 한방원료의 지질대사 기능을 가진 한방차를 개발하는데 도움을 줄 목적으로 LPS로 간독성이 유도된 흰쥐에서 어성초에 오미자. 구기자. 사상자. 음양곽을 혼합해서 추출하고 이 혼합 추출물이 지질대사에 미치는 영향을 관찰하였다. 본 실험에서는 어성초 90 g, 복분자 1 g, 사상자 3 g, 구기자 2 g, 음양곽 4 g을 혼합하고 추출하여 감압 농축하여 실험 샘플로 사용하였으며 생후 7주 암컷 흰쥐를 총 21마리를 7마리씩 3군으로 나누었고 군별로 cage에 분리시키고 고형사료와 물을 자유롭게 섭취하도록 하였다. 정상군은 20일 동안 1.5 ㎖/㎏ of 0.9% saline을 투여하고 대조군은 20일 동안 1.5 ㎖/㎏ of 0.9% saline을 매일 투여한 후 21일째 되는 날 LPS를 5 ㎎/㎏의 농도로 만들어 1.5 ㎖/㎏의 용량으로 복강 내로 투여하였다. 시료군(HM)에는 100 ㎎/㎏ 농도의 각각의 분획을 1.5 ㎖/㎏씩 복강 내에 20일간 매일 투여하고 21일째 되는 날에 LPS를 1.5 ㎖/㎏의 용량을 복강 내로 투여하였다. LPS를 투여하고 절식 시킨 뒤 4시간 후에 ether로 마취하고 해부하여 혈액을 채취하고 간을 적출하여 실험하여 다음과 같은 결과를 얻었다. Total-lipid 수준에서는 LPS를 투여한 대조군이 정상군에 비교하여1.7배 증가하였으며 대조군과 비교해 HM군은 66.30% 감소하였다. Total-cholesterol 수준에서는 LPS를 투여한 대조군이 정상군에 비교하여 약 2.1배 감소하여 LPS로 간에 염증이 유발하여 total cholesterol의 수치가 감소하는 것을 확인하였으며 HM군이 대조군에 비교하여 41.13%의 수치로 회복하였다. HDL-cholesterol 수준에서는 LPS를 투여한 대조군이 정상군에 비교하여 약 2.3배 감소하였으며 HM군이 대조군에 비교하여 72.61%의 수치로 회복하였다. LDH 수준에서는 LPS를 투여한 대조군이 정상군에 비교하여 약 5.8배 증가하였으며 HM군이 대조군에 비교하여 68.91%의 수치로 감소하였다. TG의 수준에서도 LPS를 투여한 대조군이 정상군에 비교하여 약 3.8배 증가하였으며 HM군이 대조군에 비교하여 101.14%의 수치로 감소하였다. MDA는 LPS를 투여한 대조군이 정상군에 비해서 약 3.5배 정도로 높게 나타났고 HM군은 대조군에 비해 58.65%의 감소효과를 보였다. LPS가 간에 염증을 유발하여 혈청 속의 total-cholesterol과 HDL-cholesterol level이 감소하고 total-lipid와 LDH, TG의 수준이 증가한 것으로 사료되며 대조군에 비교하여 HM군이 혈청 속의 total-cholesterol과 HDL-cholesterol level을 증가시키고 total-lipid와 LDH, TG의 수준을 감소시킨 것으로 보아 HM이 동맥경화 등 지질대사 이상에 의한 성인병에 효과가 있을 것으로 사료된다. MDA에서는 대조군에서 LPS가 간 염증을 유발하여 과산화지질함량이 증가한 것으로 사료되며 대조군에 비해 HM군이 과산화지질함량이 감소된 것으로 보아 HM이 활성산소의 생성을 억제하여 높아진 간 조직 과산화지질함량을 감소시킴으로써 간기능 개선효과가 있을 것으로 사료된다. This study was designed to investige the effects of Houttuynia cordata Thunb mixture extract on the lipid metabolism in the lipopolysaccharide (LPS)-induced liver damage of rats. LPS-treatment increased the levels of total-lipid, LDH (lactate, dehydrogenas), triglyceride (TG) and malondialdehyde (MDA). But Houttuynia cordata Thunb mixture extract (HM) pretreatment decreased the levels of total lipid, LDLcholesterol, TG and MDA. Also LPS-treatment decreased total cholesterol and HDL-cholesterol, but HM-pretreatment increased both of them. These results demonstrated that HM-pretreatment had the preventive effects against the dyfunction of lipid metabolism in the LPS-induced liver damage of rats.

        LPS로 유도된 급성 간독성에 대한 백작약 추출물의 보호 효과

        The purpose of this study was to investigate the preventive effects of Paeoniae Radix Extract (PRE) against the acute hepatotoxicity-inducing lipopolysaccharide (LPS) in the liver. PRE of 100 mg/kg concentration was intraperitoneally administered into rats at dose of 1.5 ml/kg for 20 days. On day 21, 5 mg/kg of LPS dissolved in saline was injected 4 hours before anesthetization. We examined the levels of glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT), lactate dehydrogenase (LDH) in serum of rats, superoxide dismutase (SOD) in mitochondrial fractions, and malondialdehyde (MDA), catalase (CAT), glutathione peroxidase (GPx) in liver homogenates. LPS-treatment markedly increased the levels of GOT, GPT, LDH and MDA, and significantly decreased those of SOD, CAT and GPx. But PRE-pretreatment decreased the levels of GOT, GPT, LDH and MDA, by 59.7%, 43.6%, 59.6% and 63.5%, respectively and increased those of SOD, CAT and GPx, by 85.5%, 57.8% and 62.9%, respectively.         LPS로 유도된 급성 간독성에 대한 구기자 추출물의 보호 효과

        The purpose of this study was to investigate the preventive effects of Lycii fructus Extract (LFE) against the acute hepatotoxicity-inducing lipopolysaccharide (LPS) in the liver. LFE of 100 mg/kg concentration was intraperitoneally administered into rats at dose of 1.5 ml/kg for 20 days. On the day 21, 1.5 ml/kg of LPS dissolved in saline was injected 4 hours before anesthetization. We examined the levels of glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT), lactate dehydrogenase (LDH) in serum of rats, superoxide dismutase (SOD) in mitochondrial fraction, and malondialdehyde (MDA), catalase (CAT), glutathione peroxidase (GPx) in liver homogenate. LPS-treatment markedly increased the levels of GOT, GPT, LDH and MDA, and significantly decreased those of SOD, CAT and GPx. But LFE-pretreatment decreased the levels of GOT, GPT, LDH and MDA, by 17.7%, 27.5%, 40.7% and 56.9%, respectively and increased those of SOD, CAT and GPx, by 90.5%, 78.9% and 83.8%, respectively. These results showed that the LFE had the preventive effects against the acute hepatotoxicity-inducing LPS in the liver.

        한방원료의 초임계 추출을 이용한 항노화 및 주름개선 효과

        The reactive oxygen species generated by ultraviolet rays causes various types of cutaneous damage, such as lipid peroxidation and denaturation of the extra-cellular matrix. The accumulation of such damage contributes to skin aging, especially the formation of wrinkles. This study was carried out to develop functional cosmatic by using Oriental herb supercritical extracts (OHSE) for prevention of skin. Effects of OHSE on anti-oxidation, collagenase inhibition and collagen synthesis in normal human fibroblast were investigated. OHSE showed antioxidative activity as high as vitamin C, trolox and DL-penicillamine. Also OHSE showed promotive effect on collagen synthesis and inhibitory effect on collagenase activity. From this results, we conclude that OHSE may have the potential to be conveniently used as an additive in cosmetics for prevention and improvement of skin aging.

        Lipopolysaccharide로 유발된 간독성에 대한 까마중의 효과

        This study aimed to investigate the protective effect of Solanum nigum Linne total extract (SNT), Solanum nigum Linne leaf extract (SNL), Solanum nigum Linne root extract (SNR) on liver injury induced by Lipopolysaccharide(LPS) in Sprague-Dawley rats. SNT, SNL, SNR of 100 mg/kg concentration was intraperitoneally administered into rats at dose of 1.5 ml/kg for 20 days. on the day 1.5 ml/kg of LPS was injected. Four hours later, they were anesthetization with ether and dissected. Glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT) were measured in serum and superoxide dismutase (SOD), catalase and glutathione peroxidase (GPX) were measured in liver homogenate. SNT, SNL, SNR extract inhibited GOT and GPT activities in LPSinduced rats, whereas increased SOD, Catalase and GPX activity in liver tissue of LPS-induced rats. These suggested that SNT, SNL, SNR could be used for functional beverage.

        내독소인 LPS로 처치된 흰쥐에 대한 막걸리의 항산화 활성효과

        Modern people have begun to have the nationwide interest in the rice wine called Makgeolri which is one of the traditional Korean alcoholic liquors. This study was performed to investigate the effects of Sansung Makgeolri extract (SM) on antioxidation together with the determination of pH and dissolved oxygen (DO) in the progress of fermentation in the lipopolysaccharide(LPS)-treated rats. We examined the levels of SOD (superoxide dismutase),CAT (catalase), GPx (glutathione peroxidase) in liver homogenates and the histopathological observations in liver tissue. LPS-treated group markedly decreased the levels of SOD, CAT and GPx. But SM + LPS-treated group significantly increased the levels of them. Furthermore, the antioxidative effects of SM were supported by the histopathological observations in liver tissue which showed severe inflammation and necrosis in LPS-treated group,compared to the attenuated inflammation and necrosis in SM + LPS-treated group. This results suggested that SM could be a candidate of antioxidative material in spite of alcoholic liquors.

        복분자 추출물이 LPS로 유도된 산화적 스트레스와 지질대사에 미치는 영향

The purpose of this study was to investigate the preventive efects of Rubus coreanum Miquel (RCM) inlipid metabolism. RCM of 100 mg/kg concentration was intraperitoneally administered into rats at dose of 1.5 ml/kgfor 20 days. On the day 21, 1.5 ml/kg of LPSrelated functions by measuring the levels of triglyceride (TG), total cholesterol (TC), total lipid (TL), high-densitylipoprotein cholesterol (HDL-C) in serum and malondialdehyde (MDA) in liver tissue. The results showed that LPStreatment increased the values of TG, TC, TL and MDA, decreasing that of HDL-C. But RCM pretreatment decreasedthe high values of TG, TC, TL and MDA to the low values and increased the low value of HDL-C to the high value.These results suggested that RCM could be used as the potential candidate for the lipid metabolism natural supple-ment.
