The present study was done to detect Leptospira canicola antigens in paraffin sections from experimentally infected hamsters with PAP method. The anti-Leptospira canicola serum used as first antibody was prepared by immunizing rabbits with Leptospira canicola. Positive reactions were detected in interstitium, tubular epithelial cells and glomerulus of the kidney, and in the hepatic sinusoid. With PAP method, leptospiral antigens were detected at the 48 hours after bacterial infection, but isolation, silver stain, and dark field microscopic examination of the samples did 48 hours later than PAP method. The results suggested that PAP method is considered as a reliable tool for confirmative diagnosis of this disease.

<P><B>Abstract</B></P> <P>A sensitive and specific swarm primer-based reverse transcription loop-mediated isothermal amplification (sRT-LAMP) assay for the detection of serotype O foot-and-mouth disease virus (FMDV) was developed and evaluated. The assay specifically amplified the <I>VP3</I> gene of serotype O FMDV, but did not amplify the <I>VP3</I> gene of other serotype FMDVs or any other viruses. The limit of detection of the assay was 10<SUP>2</SUP> TCID<SUB>50</SUB>/mL or 10<SUP>3</SUP> RNA copies/μL, which is 100 times lower than that of the RT-LAMP assay without swarm primers. The new assay is 10 times more sensitive than reverse transcription-polymerase chain reaction (RT-PCR) and is comparable to the sensitivity of real time RT-PCR (qRT-PCR). Evaluation of the assay using different serotypes of FMDV strains showed 100% agreement with the RT-PCR results. The previously reported serotype O FMDV-specific RT-LAMP assay did not detect 20 out of 22 strains of serotype O FMDVs, probably due to multiple mismatches between the primer and template sequences, showing that it is not suitable for detecting the serotype O FMDVs circulating in Pool 1 region countries, including Korea. In contrast, the developed sRT-LAMP assay with improved primers can rapidly and accurately diagnose serotype O FMDVs circulating in Pool 1 region countries and will be a useful alternative to RT-PCR and qRT-PCR.</P> <P><B>Highlights</B></P> <P> <UL> <LI> A swarm primer-applied RT-LAMP assay was developed for the detection of serotype O FMDV. </LI> <LI> The assay was 10 times more sensitive than RT-PCR and comparable to the sensitivity of qRT-PCR. </LI> <LI> The swarm primer RT-LAMP assay showed 100% agreement with conventional RT-PCR. </LI> </UL> </P>
백혈구 표면항원 특이 단크론항체를 이용한 한우의 말초혈액 백혈구 아군에 관한 연구

The proportion of leukocyte subpopulation in the host is of a great importance in understanding their functions and disease progress. Many methods have been developed to seperate leukocytes and to measure their activities. Characterization of immune cell subpopulations in Korean native cattle was performed using a set of monoclonal antibodies specific which are specific to bovine leukocyte differentiation antigen. Peripheral blood leukocytes from fifty Korean native and ten Holstein cattle were collected and analyzed for the investigation of leukocyte subpopulation by using monoclonal antibodies and flow cytometry. The result indicated that Korean native cattle have significantly higher proportion of leukocyte subpopulations expressing MHC class II molecules and BoCD4 than Holstein cattle.
돼지에서 감염의 조기진단 지표로서의 Haptoglobin 적용에 관한 연구

Sera of pigs with clinically normal and infectious conditions were analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE) to demonstrate the specific changes in protein profile. In the sera from pigs with infection, haptoglobin with a 40KDa protein was found to be increased as compared to that of sera from normal pigs. As a rapid detection method for monitoring infections at large-scale farms, one of acute phase protein, haptoglobin, was selected to compare the concentrations between infectious and non-infectious conditions. Haptoglobin concentrations were low in pigs with clinically normal conditions but significantly increased in pigs with Aujesky's disease, hog cholera and parvo-virus infection. The studies provide that haptoglobin can be used as an indicator to monitor infections early at farm level.
