양식산 터봇, Scophthalmus maximus에서의 Irido-like virus 감염

본 연구는 병리조직학적 및 전자현미경적 방법을 통해 양식산 turbot에서 발생하는 irido-like virus의 감염증을 소개하고자 한 것이다. 2003년 6월 전북 고창 소재 육상수조에서 양식중인 turbot치어에서 50∼70%의 대량폐사가 발생하였다. 병어는 사료섭취가 저하되고 힘없이 유영하다 폐사되었으며, 죽기 직전의 어류는 복부가 팽만되고 안구가 돌출되며 체색이 옅어지는 증상을 보였다. 해부학적 조사 결과, 아가미 빈혈과 간이 퇴색되어 있었으며, 특히 비장조직이 심하게 비대되어 있었다. 병어에 대한 조직학적 검사에서 비대 세포를 특징으로 하는 특이 병변이 주로 신장과 비장의 조혈조직에서 관찰되었으며, 아가미, 심장, 위, 장, 간, 췌장, 지방 조직에서도 관찰되었다. 그리고 전자현미경적 관찰 결과, 비대 세포의 세포질내에서 육각형의 지름이 136∼159 ㎚인 Irido-like virus를 관찰할 수 있었다. The high mortality of cultured juvenile turbot, Scophthalmus maximus occurred in Gochang on June, 2003. The diseased fish was lethargic with reduced feed intake. Grossly, these fish showed pale body, abdominal extension and exophthalmia. The dominant internal gross features of diseased fish were severely enlarged spleen, pale gills and or liver. Diseased fish histologically showed basophilic enlarged cells in the kidney, spleen, gill, heart, stomach, intestine, liver, pancreas and adipose tissue. Transmission electron microscopy (TEM) reveled hexagonal virions in the cytoplasm of necrotic cells. The viral particles lead a central electron-dense core and an electron translucent zone, and were 136-159 nm in diameter. These results suggest that the virus belonging to the iridoviridae was responsible for the mortality of cultured juvenile turbot.

An Infectious hematopoietic necrosis virus strain (IHNV-RTK) was isolated from cultured rainbow trout at Kumi in Korea during 2000 and 2001. In the RT-PCR amplification with the specific primer set designed from IHNV G protein region, a 540 bp PCR product was amplified from the RTK strain. The RTK strain showed higher sequence homology with the published IHNV G protein genes (RB-76, LR-73, Col-85, and Carson-89).
Differentiation of Lymphocystis Disease Virus Genotype by Multiplex PCR

Lymphocystis disease virus (LCDV) is the causative agent of lymphocystis disease. The viruses have been divided into three genotypes (genotype I for LCDV-1, II for Japanese flounder isolates, and III for rockfish isolates) on the basis of major capsid protein (MCP) gene sequences. In this study, we developed a multiplex PCR primer set in order to distinguish these genotypes. We also analyzed the MCP gene of a new LCDV isolate from the sea bass (SB98Yosu). Comparison of sequence identities between SB98Yosu and eight Japanese flounder isolates, revealed identity of more than 90.1 % at nucleotide level and 96.5% at deduced amino acid level, respectively. Phylogenetic analyses based on the MCP gene showed that SB98Yosu belongs to genotype II, along with Japanese flounder isolates. Multiplex PCR based on the MCP gene allowed us to identify these genotypes in a simple and rapid manner, even in a sample that contained two genotypes, in this case genotypes II and III.
Antimicrobial Resistance for Chlamydia Trachomatis Genital Infection during Pregnancy in Japan

The current study examined the antimicrobial resistance of Chlamydia trachomatis (CT) genital infection during pregnancy in Japan. We requested 2,146 obstetrical facilities that are members of Japan Association of Obstetricians and Gynecologists to provide information of CT screening tests and antimicrobial resistance in pregnant women between April 2020 and March 2021. The prevalence of CT genital infection in Japan was 2.1%. The antimicrobial resistance was recognized in 2.0 and 2.4% of the cases using azithromycin and clarithromycin, respectively. There were no significant differences in the antimicrobial resistance rate between the 2 analogues (P = 0.28). In Japan, azithromycin and clarithromycin have effectively treated genital CT infections during pregnancy.

In recent years, several southern coastal fish farms in Korea have experienced 2-30% mortality in striped beakperch suffering from bacterial infections during the spring. In this study, we identified a bacterium isolated from diseased striped beakperch as Pseudomonas anguilliseptica via a biochemical test and 16S rDNA sequence analysis. To evaluate the susceptibility of striped beakperch to P. anguilliseptica, $4.39\times10^7$ or $4.39\times10^5$ CFU/fish of bacteria were injected intraperitoneally at $18\pm1^{\circ}C$ into fish weighing 5.5 g. Cumulative mortalities reached 100% and 45% in the $4.39\times10^7$ and $4.39\times10^5$ CFU/fish infected groups, respectively. Experimentally infected fish showed cell-associated inflammation as well as bacteria in the kidneys and spleen. This study is the first report of striped beakperch mortality caused by P. anguilliseptica, which has pathogenicity in striped beakperch.

Macrophages play an important role in aging-related muscle atrophy (i.e., sarcopenia). We examined macrophage density in six striated muscles (cricopharyngeus muscle, posterior cricoarytenoideus muscle, genioglossus muscle, masseter muscle, infraspinatus muscle, and external anal sphincter). We examined 14 donated male cadavers and utilized CD68 immunohistochemistry to clarify macrophage density in muscles. The numbers of macrophages per striated muscle fiber in the larynx and pharynx (0.34 and 0.31) were 5-6 times greater than those in the tongue, shoulder, and anus (0.05-0.07) with high statistical significance. Thick muscle fibers over 80 μm in diameter were seen in the pharynx, larynx, and anal sphincter of two limited specimens. Conversely, in the other sites or specimens, muscle fibers were thinner than 50 μm. We did not find any multinuclear muscle cells suggestive of regeneration. At the beginning of the study, we suspected that mucosal macrophages might have invaded into the muscle layer of the larynx and pharynx, but we found no evidence of inflammation in the mucosa. Likewise, the internal anal sphincter (a smooth muscle layer near the mucosa) usually contained fewer macrophages than the external sphincter. The present result suggest that, in elderly men, thinning and death of striated muscle fibers occur more frequently in the larynx and pharynx than in other parts of the body.
