        Genetic positioning of Korean viral hemorrhagic septicemia virus (VHSV) from cultured and wild marine fishes

        Viral haemorrhagic septicaemia virus (VHSV) is an epidemic virus in olive flounder Paralichthys olivaceus farms in Korea, since the virus have first isolated in 2001. In the present study, partial glycoprotein (G) gene nucleotide sequences of seven Korean VHSV from cultured olive flounder and wild marine fishes in coastal areas of Korea were analyzed to evaluate their genetic relatedness to worldwide isolates. Phylogenetically, all Korean VHSV formed only one minor cluster including Japanese isolates, in genotype IVa, while the North America isolates formed a different minor cluster in genotype IVa. These results suggest that Korean VHSV could be an indigenous virus in Korean and Japanese coastal areas, but have not been introduced from North America

        Is Koi Herpesvirus (KHV) Related to the Mass Mortality Occurring among Cultured Carp, Cyprinus carpio, in Korea?

        Since 1998, a new viral disease with high mortality has been consistently recorded in Korea in cultured carp, Cyprinus carpio. In this study, we investigated an epizootic of the disease that caused high mortality rates in carp obtained from 11 farms in Korea between 1999 and 2007. Assessment of koi herpesvirus (KHV) levels in diseased carp was carried out to determine if this virus was the etiologic agent of disease in this instance. High mortality rates in carp were recorded mainly in the spring and autumn at water temperatures between 19°C and 24°C. Diseased fish typically showed surface discoloration, with a thick opaque mucus covering the body and gills. Protozoan parasites and bacteria were recovered from 7/29 (24%) and 2/26 (8%) of fish, respectively. Evidence of viral infection was marked; cytopathic effects (CPEs), characterized by cell rounding and an extended cytoplasm in fathead minnow (FHM) cells, were detected in 40/41 fish (98%). A high mortality rate (80%) resulted when supernatants of cell cultures showing CPEs were applied to previously healthy fish. KHV was detected by polymerase chain reaction in 6/41 fish (15%), but was not detected in supernatants obtained from cell cultures showing CPEs. These results suggest that KHV may not be the etiologic agent of the high mortality occurring among cultured carp in Korea; therefore, some other—as yet unidentified—infective agent must be responsible.

        해수 사육 무지개송어(Oncorhynchus mykiss)의 Viral Hemorrhagic Septicemia Virus (VHSV) 모니터링

        Mariculture of rainbow trout Onchorhynchus mykiss has been initiated in or around olive flounder Paralichthys olivaceus farms, where viral hemorrhagic septicemia virus (VHSV) is often detected in some fish. In the present study, we investigated VHSV infection in seawater-reared rainbow trout because VHSV has never been detected in salmonids in Korea. A total of 104 adult fish were tested for the presence of VHSV by reverse transcription-polymerase chain reaction, followed by virus isolation with the fathead minnow caudal trunk cell line. Cytopathic effects were observed in two samples but the virus was identified as infectious hematopoietic necrosis virus. Thus, VHSV was not isolated from seawater-reared rainbow trout.

        양식산 강도다리(Platichthys stellatus)에서의 비정형 Aeromonas salmonicida 분리

        About 6.7% mortality was reported in a starry flounder (Platichthys stellatus) aquaculture farm in 2022. Most of the diseased fish showed a loss of pectoral fin, hemorrhages on muscle and gills, pale gills, enlarged spleen, and nodules on kidney. Parasites, fungi or viruses (viral hemorrhagic septicemia virus and hirame novirhabdovirus) were not detected from diseased fish. However, numerous bacteria were isolated from liver, spleen and kidney. Nucleotide sequences of the A-protein-encoding virulence array protein gene (vapA) of the bacteria showed 99.93% identity with Aeromonas salmonicida subsp. masoucida. This study is the first report of isolation of atypical A. salmonicida in cultured starry flounder in Korea.

        자연산 숭어(Mugil cephalus)의 Myxobolus sp. 감염증

        2002년 2월 광양만에서 자연 서식하는 숭어에서 폐사가 발생하였다. 그 폐사 원인을 조사하는 과정에서 Myxobolus sp.의 감염을 확인하여, 본 연구에서는 병리조직학적 검사를 통한 Myxobolus sp.의 감염증을 보고하게 되었다. Myxobolus sp.의 cyst는 숭어의 장간막, 간, 아가미 및 인두낭에서 관찰되었으며, 병리조직학적 검사에서 Myxobolus sp.의 전신감염을 확인할 수 있었다. 감염 충체는 길이 10-12 ㎛, 폭 9-10 ㎛, 두께 6.4-7.2 ㎛의 크기로서 체내에 길이 4-5.2 ㎛, 폭 2.5-3.3 ㎛의 2개의 극낭을 가지고 있었다. The mortality of wild mullet, Mugil cephalus was detected in Kwang-Yang bay on February, 2002. The mullet were infected with Myxobolus sp., the cysts of Myxobolus sp. were found in the mesentery, liver, gill and pharyngeal pocket. The histological findings suggested a systemic infection by the Myxobolus sp.. The spores were measured 10-12 (10.9)㎛ in length, 9-10 (9.4) ㎛ in width, 6.4-7.2 (6.8) ㎛ in thickness, with polar capsules of 4-5.2 (4.4) ㎛ in length and 2.5-3.3 (2.9) ㎛ in width.

        양식산 터봇, Scophthalmus maximus에서의 Irido-like virus 감염

        본 연구는 병리조직학적 및 전자현미경적 방법을 통해 양식산 turbot에서 발생하는 irido-like virus의 감염증을 소개하고자 한 것이다. 2003년 6월 전북 고창 소재 육상수조에서 양식중인 turbot치어에서 50∼70%의 대량폐사가 발생하였다. 병어는 사료섭취가 저하되고 힘없이 유영하다 폐사되었으며, 죽기 직전의 어류는 복부가 팽만되고 안구가 돌출되며 체색이 옅어지는 증상을 보였다. 해부학적 조사 결과, 아가미 빈혈과 간이 퇴색되어 있었으며, 특히 비장조직이 심하게 비대되어 있었다. 병어에 대한 조직학적 검사에서 비대 세포를 특징으로 하는 특이 병변이 주로 신장과 비장의 조혈조직에서 관찰되었으며, 아가미, 심장, 위, 장, 간, 췌장, 지방 조직에서도 관찰되었다. 그리고 전자현미경적 관찰 결과, 비대 세포의 세포질내에서 육각형의 지름이 136∼159 ㎚인 Irido-like virus를 관찰할 수 있었다. The high mortality of cultured juvenile turbot, Scophthalmus maximus occurred in Gochang on June, 2003. The diseased fish was lethargic with reduced feed intake. Grossly, these fish showed pale body, abdominal extension and exophthalmia. The dominant internal gross features of diseased fish were severely enlarged spleen, pale gills and or liver. Diseased fish histologically showed basophilic enlarged cells in the kidney, spleen, gill, heart, stomach, intestine, liver, pancreas and adipose tissue. Transmission electron microscopy (TEM) reveled hexagonal virions in the cytoplasm of necrotic cells. The viral particles lead a central electron-dense core and an electron translucent zone, and were 136-159 nm in diameter. These results suggest that the virus belonging to the iridoviridae was responsible for the mortality of cultured juvenile turbot.

        식용작물을 이용한 전복 박리

        본 연구에서는 29종의 식용작물을 이용하여 전복박리 효과를 조사한 결과, 스피아민트, 레몬밤, 로즈플라워, 양파, 와사비분, 겨자분, 오미자 및 히비스커스 용액에서 박리효과가 관찰되었다. 특히, 오미자와 히비스커스 용액에서는 90% 이상의 박리율이 관찰되었다. 양식장에서 사육중인 전복을 대상으로 오미자 및 히비스커스 용액을 사용하여 전복 박리 및 회복 효과를 조사한 결과, 3%와 4%의 오미자 용액에 노출된 전복은 각각 93.6%(850/908)와 97.1% (810/834)의 박리율을 보였으며 1분 15초 이내에 회복되었다. 3%와 4%의 히비스커스 용액에서는 각각 96.1% (780/812)와 97.4%(700/719)의 박리율을 보였고, 박리된 전복은 1분39초 이내에 회복되었다. 이상의 결과, 오미자와 히비스커스 용액은 양식현장에서 전복을 박리하는데 실용적으로 사용 가능할 것으로 사료된다. Twenty nine edible plants were selected in order to study their exfoliation effect on abalone Haliotis discus hannai from its substrate. Exfoliation was observed in spearmint Mentha spicata, lemon balm Melissa officinalis, rose flower, onion Allium cepa, wasabi powder, mustard powder, omija Schizandra Chinensis, and hibiscus Hibiscus sabdariffa solutions. In particular, both the omija and hibiscus solutions exfoliated over 90% of the abalone. In field tests, 3% and 4% omija solutions exfoliated 93.6% (850/908) and 97.1% (810/834) of abalone, respectively, whereas 3% and 4% hibiscus solutions exfoliated 96.1% (780/812) and 97.4% (700/719), respectively. The detached abalone were recovered within 1 min 39 seconds. These results indicate that the omija and hibiscus solutions (3% and 4%) may be useful in the exfoliation of abalone.

        2010∼2012년 연안에서 서식하는 해산어에서 아니사키스 유충의 감염현황

        A survey was conducted to investigate infection of anisakid nematode larvae in 243 wild marine fishes caught from the southern coastal area of Korea between 2010 and 2012. The samples comprised fishes from 9 orders, 30 families and 50 species. Total infection rate of anisakid nematode larvae was 10.7% (26/243 fish), which comprised from Yeosu, 7.4% (7/95) in 2010 and 22.7% (5/22) in 2011; from Jeju, 8.2% (5/61) in 2011; from Wando, 40.9% (9/22) in 2012. Anisakid nematode larvae were not detected in Tongyoung and Wando samples in 2011. Molecular identification of the 89 worms from 26 fish was conducted by PCR-RFLP and/or sequence analysis of internal transcribed spacer (ITS) region of ribosomal DNA. From the results, 6 kinds of anisakis species were identified: Anisakis pegreffii (infection rate: 53.9%, 48/89 worms), Hysterothylacium aduncum (38.2%, 34/89), H. fabri (3.4%, 3/89), hybird (A. simplex X A. pegreffii) (2.4%, 2/89), A. simplex (1.1%, 1/89) and Raphidascaris lophii (1.1%, 1/89). The rate of single infection was 80.8% (21/26 infected fish), while 19.2% (5/26) showed mixed infection with 2 to 3 different anisakis species.

        조피볼락(Sebastes schlegeli )에 대한 비정형 Aeromonas salmonicida 포르말린 사균 백신의 효과

        Atypical furnculosis caused by atypical Aeromonas salmonicida, is an emerging problem of farming of rockfish (Sebastes schlegeli) in Korea. In this study, protection against atypical furunculosis was compared in rockfish vaccinated with A. salmonicida formalin killed cell (FKC) and adjuvant containing FKC. The formalin inactivated A. salmonicida vaccine provided a low protection of 20% and 10% relative percent survival (RPS) at 44 and 58 days post vaccination. However, addition of adjuvant (squalene and aluminum hydroxide) into inactivated A. salmonicida vaccine clearly enhanced the level of protection showing 70% and 50% RPS at 44 and 58 days post vaccination.
